Comparison of Root Resorption in Patients Treated With .018 Slot Brackets Versus Those Treated With .022 Slot Brackets

Indiana University-Purdue University Indianapolis (IUPUI) / Understanding the factors that increase patients susceptibility to orthodontically induced root resorption is of the utmost importance to the practicing clinician. Numerous studies have been conducted that investigated contributing and etiological factors that tend to increase the amount of external apical root resorption a patient may incur during orthodontic treatment. However, there has been little research that has attempted to determine if patients treated with different slot size orthodontic brackets exhibit the same amount of external apical root resorption. The purpose of this research was to determine if patients treated with the 0.018 x 0.025 slot size bracket and patients treated with the 0.022 x 0.028 exhibited similar amounts of external apical root resorption during orthodontic treatment. Pre and Post treatment panoramic films from 91 consecutively treated orthodontic patients from a private orthodontic practice were used for this study. Each case that was included in the study had been treated with standard edgewise brackets using the Tweed-Merrifield philosophy of treatment. Forty-three cases from the 0.018 group and 48 from the 0.022 group were obtained. Each film that was analyzed was blinded prior to measuring to minimize observer bias. Mitutoyo Digimatic® calipers accurate to the nearest tenth of a millimeter were used for obtaining tooth measurements. Measurements were made from the Cemento-enamel junction and from incisal/occlusal to most apical portion of each incisor and all first molars. Statistical analysis was performed and the results showed no significant correlation between the size of the bracket and the amount of root resorption. No significant correlations existed between the groups for patient age, time in treatment, gender, and angle classification. Statistically significant differences were noted for cases in which extraction of four bicuspids was performed. Subjects belonging to the extraction group demonstrated significantly more external apical root resorption than those in which extractions were not done. This study demonstrated that the incidence of EARR that a patient may incur during treatment is independent of the size of the slot of the orthodontic bracket.

Root resorption during the retention phase after orthodontic treatment with fixed appliance – a randomized controlled trial

SAMMANFATTNING Syfte: Syftet med studien var att granska periapikala intraorala röntgenbilder avseende extern apikal rotresorption på överkäkens incisiver, före och efter retentionsbehandling med avtagbar retentionsapparatur. Syftet var också att registrera intra och inter-observatör överrensstämmelse vid bedömning av rotresorption. Metod: Studien utfördes som en randomiserad klinisk prövning. Patientmaterialet omfattade75 patienter och insamlades på specialistkliniken för ortodonti i Ystad, Sverige. Patienterna behandlades för trångställning mellan år 2001-2007 och behandlingen involverade extraktion av fyra pre-molarer följt av fast ortodontisk apparatur i båda käkarna. Patienterna randomiserades till tre olika retentionsgrupper för retentionsbehandlingen (25 patienter i varje grupp). Intraorala periapikala röntgenbilder togs i överkäksfronten före ortodontiska behandlingens start (T0), vid retentionsbehandlingens start (T2) samt efter retention (T2.1). Genom observation av intraorala periapikala röntgenbilder angavs ett index (0-4) för rotresorptionen för respektive överkäksincisiv. Fyra observatörer granskade var för sig samtliga röntgenbilder, och vid ett senare tillfälle granskades 20 stycken slumpvis utvalda bilder. Resultat: Studien visar att rotresorption förekommer under den ortodontiska behandlingen med fast apparatur och signifikanta skillnader i rotresorption registrerades av två av fyra observatörer under retentionsbehandlingen (p<0.05). Inter-observatörs överrensstämmelsen var generellt låg, medan intra-observatör överrensstämmelsen var något högre. Konklusion: Enligt denna studie förekommer rotresorption under den ortodontiska behandlingen med fast apparatur, och till viss del även under retentionsperioden med avtagbar apparatur. / ABSTRACTAim: The overall aim of the study was to register external apical root resorption in maxillary incisors before and after retention phase with removable appliance. A secondary aim was to register intra- and inter-observer agreement during the assessment of root resorption. Method: The study was carried out as a randomized clinical trial. The patient sample consisted of 75 patients with dental crowding that were treated at specialist orthodontic clinic in Ystad, Sweden. The patients were treated between year 2001-2007 and the treatment involved extraction of four premolars followed by fixed appliance in both jaws. The patients were randomized into three retention groups with 25 patients in each group. Intraoral periapical radiographs were taken on maxillary incisors before orthodontic treatment (T0), before start of retention phase (T2) and after retention (T2.1). An index ranging from 0-4 was used to assess root resorption for each maxillary incisor. At the first assessment, four observers individually analyzed all radiographs. 20 patients were randomly selected for a second assessment. Results: The study shows that root resorption occur during orthodontic treatment with fixed appliance (p<0.05). Statistical significant root resorption occurred during the retention period according to two of four observers. The inter-observer agreement was generally low, while the intra-observer agreement was higher. Conclusion: According to this study, root resorption occurs during fixed orthodontic treatment and to some extent during the retention period with removable appliances.

Genetic Factors in External Apical Root Resorption Associated with Orthodontic Treatment

Indiana University-Purdue University Indianapolis (IUPUI) / External apical root resorption (EARR) is a common sequela of orthodontic treatment, although it may also occur without orthodontic treatment. Despite rigorous investigation, no single factor or group of factors that directly causes root resorption has been identified. Experiment 1. A sample of 83 pairs of full siblings who had undergone orthodontic treatment was studied. Measurements were made of the longest maxillary central incisor, mandibular central incisor and mesial and distal roots of the mandibular first molars. Heritability estimates were generated by generalized liner models. Our results showed that the heritability estimate of the EARR was 64% on average. It was concluded that there was sufficient heritability for EARR to pursue genetic analysis. Experiment 2. Five polymorphic markers flanking or lying within the IL-IA , IL-JB, TNSALP, TNFA, and TNFRSFJ JA genes were used in a candidate gene approach to assess linkage and association with EARR in 38 pedigrees. Suggestive evidence for linkage between EARR and the polymorphic marker D18S64 was obtained with the analysis program MAPMAKER/SIBS (LOD score 2.51). The Q-TDT program showed highly significant (p = 0.0003) evidence of linkage disequilibrium of IL-1 B polymorphisms with EARR. Our analysis indicates that the JL -1 B polymorphism accounts for 15% of the total EARR variation. Experiment 3. Nine-week-old male mice were randomly selected as controls or for placement under anesthesia of an open coil spring ligated to the left maxillary first molar producing a force of approximately 25 g. The control (C) or treated (T) per strain were A/J (C=3,T=9), C57BL/6J (C=7,T=8), C3H/HeJ (C = 4,T=6), BALB/cJ (C=4,T=6), 129P3 /J (C=6,T=8), DBA/2J (C=8,T=9), SJL/J (C=8,T= 10), and AKR/J (C=9,T =8). Animals were sacrificed after nine days of treatment or control; maxillae were immediately removed, prepared, sectioned, mounted, stained with H&E, and observed microscopically at 1 OOX to determine root resorption. Mice were grouped into root resorption resistant (A/J, C57BL/6J and SJL/J); intermediate (C3H/HeJ and AKR/J); and susceptible (BALB/cJ, DBA/2J, and 129P3/J) strains. It was concluded that there were differential susceptibility or resistance to root resorption among inbred mouse strains, indicating that genotype is an influencing factor.

Efeito do ácido zoledrônico usado no tratamento de dentes reimplantados tardiamente : estudo em ratos /

Frente a isso, este trabalho teve como objetivo avaliar o ácido zoledrônico, um inibidor da reabsorção, como medicação para o tratamento da superfície radicular para dentes reimplantados tardiamente. Vinte e quatro incisivos centrais superiores direitos de ratos foram avulsionados e mantidos a seco por 30 minutos. Todos os dentes tiveram o ligamento periodontal removido mecânicamente. Na seqüência, os dentes foram divididos em dois grupos. No grupo I, a superfície radicular foi tratada com solução de ácido zoledrônico a 10-6M por 20 minutos; no grupo II, foi usado o fluoreto de sódio a 2% fosfato acidulado, por 20 minutos. Todos os canais foram preenchidos com pasta de hidróxido de cálcio. Após esse procedimento, os dentes foram reimplantados em seus respectivos alvéolos. Passados 15 e 60 dias do reimplante, os animais foram eutanasiados e as peças obtidas, processadas em laboratório para análise microscópica e morfométrica. Os resultados demonstraram que o ácido zoledrônico foi eficiente para o tratamento da superfície radicular. Apesar da ausência de formação de um tecido à semelhança do ligamento periodontal, o ácido zoledrônico foi eficaz em retardar a reabsorção radicular. / Abstract: The use of substances that inhibit root resorption may be an alternative for cases of unsuccessful tooth reimplanted. Hence, the purpose of this study was to test the zoledronic acid, a resorption inhibitor, as an intracanal therapeutic agent for teeth submitted to late reimplantation. Twenty-four rat maxillary right central incisors were avulsed and kept dry for thirty minutes. The periodontal ligament were removed mechanically with aid of blade. Thereafter, the teeth were divided in two groups. In group I, the root surfaces was treated with 10-6M solution of zoledronic acid for 20 minutes; in group II, 2% sodium fluoride was used for 20 minutes. The canals were filling with calcium hydroxide paste. Teeth were then reimplanted in their respective socket. The animals were killed at 15 and 60 days after reimplantation and the samples processed for morphometric and microscopic analysis. The results showed that the zoledronic acid limited the root resorption. Although the absence of formation of periodontal tissue-like, zoledronic acid was efficient for restricted to root resorption. / Mestre

Este estudo objetivou analisar a influência do ácido acetilsalicílico e do acetaminofeno na movimentação dentária induzida e nas reabsorções radiculares associadas do ponto de vista celular e tecidual ao microscópio óptico. Os grupos experimentais constituíram-se de 18 ratos com administração de soro fisiológico; 36 ratos, com ácido acetilsalicílico e os outros 36 com acetaminfeno. Os animais foram subdividos de acordo com o tempo de tratamento movimentação dentária induzida: 5, 7 e 9 dias. O aparelho para a movimentação ortodôntica foi o preconizado por Heller e Nanda e modificado por Martins-Ortiz. As drogas foram submetidas em períodos curtos e longos. Aplicou-se análise microscópica histomorfométrica quantitativa na raiz distovestibular do primeiro molar superior esquerdo do rato. Esta análise foi dividida em duas etapas: quantificação do percentual de áreas hialinas no ligamento periodontal e quantificação do percentual de reabsorção radicular no lado de pressão. Os resultados do teste t de Student nos permitiram as seguintes constatações: 1) a administração de ácido acetilsalicílico (AAS) e do acetaminofeno (ACT) comparado ao controle, tanto na forma longa quanto curta, não alterou os percentuais de áreas hialinas e reabsorções radiculares nos três períodos de tempo avaliados; 2) a administração de AAS longa comparado ao AAS - curta e ACT - longa comparado ao ACT - curta, não alterou os percentuais de áreas hialinas e reabsorções radiculares nos três períodos de tempo avaliados. Não houve diferenças nos percentuais de áreas hialinas e reabsorções radiculares nos três períodos de tempo avaliados quando da administração de AAS e ACT, tanto na forma longa quanto curta. Por extrapolação, pode se afirmar que o uso do ácido acetilsalicílico e acetaminofeno após a ativação do aparelho ortodôntico para controlar a dor e o desconforto do paciente não afeta a movimentação dentária e nem influencia na freqüência e severidade de reabsorções dentárias associadas. / This study aimed at analysing the influence of acetylsalicylic acid and acetaminophen on induced dental movement and associated root resorption at cell and tissue levels by means of optic microscopy. The experimental groups consisted of 18 rats receiving physiological saline solution; 36 rats receiving acetylsalicylic acid; and 36 receiving acetaminophen. The animals were subdivided according to the duration of induced dental movement during treatment: 5, 7, and 9 days. The orthodontic appliance was conceived by Heller and Nanda and then modified by Martins-Ortiz. The drugs were administered during short and long periods of time. Quantitative histomorphometric microscopic analysis of the distal buccal root of the upper left first molar was performed in two steps: percentage quantification of hyaline areas of periodontal ligaments and percentage quantification of root resorption on pressure side. The results of the Student t test allow us to find the following: administration of acetylsalicylic acid and paracetamol compared to control group did not change the percentage amount of hyaline areas and root resorption for the three periods of time, regardless of the administration frequency. Long-administration versus short administration of acetylsalicylic acid as well as long-administration versus short administration of paracetamol did not change the percentage amount of hyaline areas and root resorptions for the three periods of study; one can conclude that there was no difference in the percentage amount of hyaline areas and root resorptions during the experimental periods in which acetylsalicylic acid and paracetamol were administered in all forms. By extrapolation, one can state that the use of acetylsalicylic acid acetaminophen following activation of orthodontic appliance for controlling pain and discomfort in patients does not affect dental movement nor influence frequency and severity of associated dental resorption.

Anti-inflamatórios não esteroidais e sua interferência na movimentação dentária induzida em ratos / Antiinflammatory drugs and their impact on tooth movement induced in rats

O objetivo deste estudo foi avaliar quais os anti-inflamatórios não esteroidais (AINEs) mais prescritos entre 100 ortodontistas pesquisados, para alívio da dor durante o tratamento ortodôntico corretivo. E analisar a Influência do mecanismo de ação dos fármacos selecionados e do fármaco nimesulida (seletivo COX-2) durante a movimentação dentária induzida em ratos e nas reabsorções radiculares associadas do ponto de vista celular e tecidual ao microscópio óptico. Foi utilizada uma amostra de 80 ratos albinos da linhagem Wistar com cerca de 300g, os quais obtiveram a instalação do aparelho ortodôntico preconizado por Heller e Nanda que era constituído por uma mola de secção fechada ancorada aos incisivos centrais superiores, movimentando mesialmente o primeiro molar superior esquerdo. Os animais foram divididos em quatro grupos: paracetamol, ibuprofeno, nimesulida e controle (livre de medicamentos) em cada um dos grupos havia 20 animais, os quais foram divididos em subgrupos de acordo com o tempo de tratamento da movimentação dentária induzida: três, cinco e sete dias, submetidos à medicação diariamente. Posteriormente, os animais receberam doses letais da mistura de relaxante muscular e anestésico por via intramuscular, para coleta do material, o qual foi devidamente processado e submetido à análise microscópica por meio de cortes transversais dos molares. Os resultados mostraram que os fármacos de primeira opção na prescrição medicamentosa para dor durante o tratamento ortodôntico foi o paracetamol e a segunda opção o ibuprofeno. As análises microscópicas revelaram durante a movimentação dentária induzida que não houve diferença estatisticamente significante para as reabsorções radiculares e áreas hialinas com relação ao uso do paracetamol, ibuprofeno, nimesulida e grupo controle. / This aim of this study was to investigate which non-steroidal anti-inflammatory drugs (NSAIDs) the most prescribed for pain relief during orthodontic treatment among 100 surveyed orthodontists; also to analyze the influence of the mechanism of action of drugs selected and the drug nimesulide (selective COX-2) during induced tooth movement in rats and associated root resorption in terms of cellular and tissue under an optical microscope. We used a sample of 80 Wistar rats (Rattus norvergicus, albinus) weighting about 300g, which obtained the installation of braces recommended by Heller and Nanda who was constituted by a spring anchored to the closed section of the upper central incisors, moving mesially the first left molar. The animals were divided into four groups: paracetamol, ibuprofen, nimesulide and control (drug free). Each group had 20 animals, and they were divided into subgroups according to duration of treatment of induced tooth movement: three, five and seven days, subjected to daily medication. Posteriorly, the animals received lethal doses intramuscular of mixture of muscle relaxant and anesthetic, to collect the material, which was duly processed and subjected to microscopic analysis using transverse sections of the molars. The results showed that the first choice drug prescribed for pain during orthodontic treatment was paracetamol and second option was ibuprofen. Microscopic analysis revealed during induced tooth movement that there was no statistically significant difference in the root resorption and hyaline areas in relation to the usage of paracetamol, ibuprofen, nimesulide and control groups.

A movimentação dentária induzida em ratos representa um bom modelo para estudar a aposição e reabsorção óssea induzidas na normalidade e nas doenças ósseas metabólicas, bem como nas avaliações quanto aos efeitos de medicamentos. Desta maneira, no primeiro capítulo estudaremos a influência do alendronato no percentual de reabsorções radiculares durante a movimentação dentária induzida, utilizando o modelo experimental em ratos em cortes transversais. O alendronato é um tipo de bisfosfonato, uma classe de drogas amplamente utilizada na medicina, no controle da osteopenia, prevenindo a osteoporose humana e na prevenção das fraturas em osteoporose pós-menopausal. O modelo experimental empregado nos trabalhos é o movimento de inclinação mesial do primeiro molar superior. Variações metodológicas são observadas neste modelo experimental em ratos, quanto aos períodos de movimentação e aos cortes empregados para a análise microscópica. Desta maneira, no segundo capítulo compararemos o percentual e a freqüência das reabsorções radiculares em cortes transversais e longitudinais, e sugeriremos o plano de corte ideal para o estudo da movimentação dentária induzida em ratos. A biologia da movimentação dentária induzida modifica-se por uma série de condicionantes, por exemplo, por diferentes períodos de observação e por planos dos cortes microscópicos Desta maneira, no terceiro capítulo avaliaremos os fenômenos teciduais e celulares da movimentação dentária induzida nos períodos de três, cinco, sete e nove dias de observação e nos planos dos cortes microscópicos transversais e longitudinais. / Induced tooth movement in rats is a good model to investigate the induction of bone resorption and apposition in normality and in metabolic bone diseases, as well as to evaluate the effects of drugs. Thus, the first chapter addresses the influence of alendronate on the percentage of root resorptions during induced tooth movement, using the experimental model of rats on transverse sections. Alendronate is a type of bisphosphonate, a class of drugs widely used in Medicine for osteopenia control, human osteoporosis prevention, as well as for fracture prevention in postmenopausal osteoporosis. Investigations employ the experimental model of mesial inclination of the maxillary first molar. Methodological variations are observed in this experimental model in rats concerning the periods of tooth movement and sections employed for microscopic analysis. Thus, on the second chapter, the percentage and frequency of root resorptions on transverse and longitudinal sections are compared, and the ideal direction of section for investigation of induced tooth movement in rats is suggested. The biology of induced tooth movement is changed by several factors, e.g. the different study periods and directions of microscopic sections. For this reason, the third chapter comprises evaluation of tissue and cellular phenomena of induced tooth movement at study periods of three, five, seven and nine days, on transverse and longitudinal sections.

Estudo histomorfométrico da ação do laser de baixa potência na reabsorção radicular ortodonticamente induzida em ratos / Histomorphometric study of low-level laser action on orthodontic induced root resorption

A reabsorção radicular inflamatória induzida pela movimentação dentária é uma consequência da lesão dos cementoblastos devido à aplicação da força sobre o dente deixando a raiz exposta à ação dos osteoclastos. Estudos vêm sendo feitos a fim de se desenvolver um meio de evitar, controlar ou reduzir este tipo de reabsorção. A fototerapia com laser tem demonstrado efetividade como agente antiinflamatório e, considerando a origem inflamatória desta patologia este estudo se propôs a avaliar a ação do laser sobre as reabsorções radiculares. Para isto realizou-se uma análise histomorfométrica da raiz disto-vestibular dos primeiros molares superiores de ratos após a indução da sua movimentação mecânica, sendo irradiados ou não com laser de baixa potência. Para a fototerapia foi utilizado um laser diodo, calibrado com comprimento de onda de 808 nm, potência de saída de 100mW, energia total de 2,1J ou 96J e área de 0,0028cm2. A aplicação foi contínua, pontual e com contato. Foram utilizados 54 ratos Wistar, machos, com aproximadamente 80 dias, pesando entre 280±40g. Para a movimentação utilizou- se uma mola fechada super-elástica, com força constante, pré-calibrada em 25g. Os espécimes foram irradiados a cada 48 horas conforme o grupo a que pertenciam. O sacrifício se procedeu no sétimo ou décimo dia após o início da movimentação. O exame histológico foi realizado com cortes de 6 &#x3BC;m corados em HE. Os resultados revelaram que, com as dosimetrias estudadas, o laser de baixa potência, não teve efeito estatisticamente significante sobre as reabsorções radiculares. Concluindo-se que não há diferença na área reabsorvida, espessura ou extensão da reabsorçãoradicular entre os grupos controle e experimental. / Inflammatory root resorption induced by tooth movement is a consequence of the cementoblasts injury due to application of force on the tooth leaving the root exposed to the action of osteoclasts. Studies have been made to develop a means of preventing, controlling or reducing this kind of resorption. Phototherapy by laser has demonstrated effectiveness as an antiinflammatory and considering the inflammatory origin of this pathology this study evaluated the effects of laser on root resorption. For this we carried out a histomorphometric analysis of maxillary first molars distobuccal root of rats after being moved, irradiated or not with low-level laser. Order to phototherapy it was used a diode laser, calibrated with a wavelength of 808 nm, output power of 100mW, 2.1J or 96J of energy and area of 0.0028 cm2. The application was continuous, punctual and with contact. It was used 54 male Wistar rats, approximately 80 days, weighing 280 ± 40g. For tooth movement, it was used a super elastic closed spring, with pre-calibrated and constant force of 25g. The specimens were irradiated every 48 hours depending on the group they belonged to. The sacrifice was made in the seventh or tenth day after the onset of movement. Histological examination was performed with sections of 6 &#x3BC;m stained with HE. The results revealed that, considering the dosimetry studied, low-level laser, has no effect statistically significant on the root resorption. Concluding it does not have difference in the root reabsorption area, thickness or extension, between the control and the experimental groups.

Introdução: As reabsorções radiculares internas (RRI) são lesões de difícil prognóstico e o diagnóstico é baseado em exames clínicos e de imagem. Estudos que avaliam a acurácia dos exames por imagem para a detecção de RRI, utilizam simulações realizadas por brocas, criando cavidades com forma e limites definidos diferentes da realidade clínica. Além disso, sabe-se que tão importante quanto a detecção da lesão de RRI é a capacidade de visualização do envolvimento da estrutura dentinária adjacente. Assim, este trabalho objetivou desenvolver uma nova metodologia para simulação de RRI com base na desmineralização ácida e, após, testar a influência de diferentes protocolos de aquisição de imagens tomográficas na medição do volume dessas lesões. Materiais e métodos: Foi testada uma metodologia de simulação de RRI, com ação progressiva de ácido nítrico 5% e hipoclorito de sódio 8%, em onze dentes monorradiculares. As cavidades geradas foram avaliadas em Microscopia Eletrônica de Varredura (MEV) e as medidas de diâmetro e profundidade foram correlacionadas com os tempos de ação do ácido. Imagens de Tomografia Computadorizada de Feixe Cônico (TCFC) foram adquiridas de todos os dentes da amostra em dois aparelhos, de FOV amplo e restrito, com três protocolos de tamanho de voxel cada: I-Cat Next Generation (voxel 0,200; 0,250; 0,300 mm) e Kodak 9000 3D (voxel 0,076; 0,100; 0,200 mm). As simulações de RRI foram mensuradas com uma ferramenta de cálculo de volume do software Dolphin 3D. Os valores médios de volume das cavidades foram comparados com Análise de Variância (ANOVA) e Teste Tukey ( =0,05). A validação das mensurações de volume obtidas pelo software foi realizada por meio de moldagem das cavidades com silicona de adição. Resultados: As simulações de RRI reproduziram lesões de diferentes tamanhos e com profundidades de 0.22 mm a 1.59 mm. Em relação às medidas volumétricas, não houve diferença estatística significante (P<0,05) entre os volumes de RRI mensurados a partir dos dois menores tamanhos de voxel em cada aparelho de TCFC, mas ambos foram diferentes estatisticamente (P>0,05) do volume calculado com os maiores tamanhos de voxel. Ainda, as médias de volume a partir das impressões em silicona foram menores que as mensuradas pelo voxel 0,200 mm. Conclusão: A metodologia de simulação de RRI proposta gerou cavidades com limites irregulares e com razão de diâmetro:profundidade mais semelhante ao processo de RRI in vivo. Diferentes protocolos de TCFC influenciam nas medidas de volume de RRI simuladas, ressaltando a importância da padronização na aquisição das imagens para acompanhamento dessas lesões. / Introduction: The internal root resorption (IRR) injuries are difficult to predict and the diagnosis is based on clinical and radiographic examination. Studies that evaluated the accuracy of imaging tests for the detection of IRR using simulations produced by burs, created cavities with different forms and limits of clinical reality. Furthermore, it is known that as important as the IRR detection is the ability to preview the involvement of adjacent dentinal structure. Thus, this study aimed to develop a new methodology for IRR simulation-based with acid demineralization and after investigate the influence of different tomographic protocols in the measurement of the IRR lesions volume. Materials and methods: A methodology was desenvolved for simulation of IRR with progressive action of 5% nitric acid and 8% sodium hypochlorite in eleven teeth single-rooted. IRR cavities produced were evaluated by scanning electron microscopy (SEM), measured in diameter and depth and after correlated with the acid times. Cone Beam Computed Tomography (CBCT) images were acquired of all teeth in two tomography devices, with large and restrict field of view (FOV) using three voxel sizes protocols: I-Cat Next Generation (voxel 0.200, 0.250, 0.300 mm) and Kodak 9000 3D (voxel 0.076, 0.100, and 0.200 mm). IRR simulations were measured with a tool for calculating volume of the Dolphin 3D software. Volume means of the IRR cavities were compared using Analysis of Variance (ANOVA) and Tukey test ( = 0.05). A validation of the tomographic measurements was performed with addition silicone impression. Results: The simulations reproduced IRR lesions with different sizes and depths (0.22mm to 1.59mm). Regarding volumetric measurements, there was no statistically significant (P<0,05) difference between the IRR volumes measured from the two smaller voxel sizes in each CBCT device, but both were statistically different (P>0,05) from the calculated volume with larger voxel sizes. Still, the volume means from silicone impressions were lower than those measured at 0.200 mm voxel. Conclusion: The IRR simulation methodology generated cavities with irregular borders and diameter:depth ratio more similar to in vivo process of IRR. Different protocols of CBCT influences the volume measures of IRR simulated, emphasizing the importance of standardization in the acquisition of CBCT images for monitoring these lesions.

Este estudo objetivou analisar a influência do ácido acetilsalicílico e do acetaminofeno na movimentação dentária induzida e nas reabsorções radiculares associadas do ponto de vista celular e tecidual ao microscópio óptico. Os grupos experimentais constituíram-se de 18 ratos com administração de soro fisiológico; 36 ratos, com ácido acetilsalicílico e os outros 36 com acetaminfeno. Os animais foram subdividos de acordo com o tempo de tratamento movimentação dentária induzida: 5, 7 e 9 dias. O aparelho para a movimentação ortodôntica foi o preconizado por Heller e Nanda e modificado por Martins-Ortiz. As drogas foram submetidas em períodos curtos e longos. Aplicou-se análise microscópica histomorfométrica quantitativa na raiz distovestibular do primeiro molar superior esquerdo do rato. Esta análise foi dividida em duas etapas: quantificação do percentual de áreas hialinas no ligamento periodontal e quantificação do percentual de reabsorção radicular no lado de pressão. Os resultados do teste t de Student nos permitiram as seguintes constatações: 1) a administração de ácido acetilsalicílico (AAS) e do acetaminofeno (ACT) comparado ao controle, tanto na forma longa quanto curta, não alterou os percentuais de áreas hialinas e reabsorções radiculares nos três períodos de tempo avaliados; 2) a administração de AAS longa comparado ao AAS - curta e ACT - longa comparado ao ACT - curta, não alterou os percentuais de áreas hialinas e reabsorções radiculares nos três períodos de tempo avaliados. Não houve diferenças nos percentuais de áreas hialinas e reabsorções radiculares nos três períodos de tempo avaliados quando da administração de AAS e ACT, tanto na forma longa quanto curta. Por extrapolação, pode se afirmar que o uso do ácido acetilsalicílico e acetaminofeno após a ativação do aparelho ortodôntico para controlar a dor e o desconforto do paciente não afeta a movimentação dentária e nem influencia na freqüência e severidade de reabsorções dentárias associadas. / This study aimed at analysing the influence of acetylsalicylic acid and acetaminophen on induced dental movement and associated root resorption at cell and tissue levels by means of optic microscopy. The experimental groups consisted of 18 rats receiving physiological saline solution; 36 rats receiving acetylsalicylic acid; and 36 receiving acetaminophen. The animals were subdivided according to the duration of induced dental movement during treatment: 5, 7, and 9 days. The orthodontic appliance was conceived by Heller and Nanda and then modified by Martins-Ortiz. The drugs were administered during short and long periods of time. Quantitative histomorphometric microscopic analysis of the distal buccal root of the upper left first molar was performed in two steps: percentage quantification of hyaline areas of periodontal ligaments and percentage quantification of root resorption on pressure side. The results of the Student t test allow us to find the following: administration of acetylsalicylic acid and paracetamol compared to control group did not change the percentage amount of hyaline areas and root resorption for the three periods of time, regardless of the administration frequency. Long-administration versus short administration of acetylsalicylic acid as well as long-administration versus short administration of paracetamol did not change the percentage amount of hyaline areas and root resorptions for the three periods of study; one can conclude that there was no difference in the percentage amount of hyaline areas and root resorptions during the experimental periods in which acetylsalicylic acid and paracetamol were administered in all forms. By extrapolation, one can state that the use of acetylsalicylic acid acetaminophen following activation of orthodontic appliance for controlling pain and discomfort in patients does not affect dental movement nor influence frequency and severity of associated dental resorption.
