Estudo dos efeitos de extratos de glândulas salivares de fêmeas de Rhipicephalus sanguineus (Latreille, 1806) (Acari : Ixodidae) na resposta imunológica de hospedeiros pós inoculadores /

Resumo: Este estudo abordou os efeitos de extratos de glândulas salivares de fêmeas de Rhipicephalus sanguineus com 2, 4 e 6 dias de alimentação (EGS2-EGS6) em coelhos New Zealand White virgens de infestação inoculados três vezes a intervalos de 21 dias. Os animais inoculados foram desafiados com carrapatos R. sanguineus, para verificar a influência dos extratos sobre a resposta imune-inflamatória dos hospedeiros. Foram estudadas as alterações histopatológicas na pele dos hospedeiros na região da lesão de fixação dos carrapatos, os parâmetros alimentar e reprodutivo das fêmeas obtidas da infestação desafio, bem como suas glândulas salivares aos 2, 4 e 6 dias de alimentação. Para obtenção dos extratos glandulares utilizou-se 55 fêmeas (110 glândulas salivares) com 2 dias de alimentação, 36 (72 glândulas) com 4 dias e 25 (50 glândulas) com 6 dias, de onde obteve-se os extratos: EGS2, a partir de glândulas salivares de fêmeas com 2 dias de alimentação; EGS4 com 4 dias, e, EGS6 com 6 dias. Os resultados revelaram que os diferentes extratos glandulares estimularam o desenvolvimento de resposta imuneinflamatória na pele dos hospedeiros comprometendo os processos alimentar e reprodutivo das fêmeas dos carrapatos. Os hospedeiros do grupo GT (EGS4) mostraram a formação de um intenso e precoce infiltrado de células inflamatórias sinalizando que este foi o extrato mais eficaz no desenvolvimento da resposta imune-inflamatória local. Por outro lado, a resposta dos hospedeiros do grupo GT (EGS6) à infestação foi mais tardia e com menor ocorrência desse infiltrado. Já o processo inflamatório local nos hospedeiros do GT (EGS2) foi o menos intenso. O teste de desafio mostrou médias para os parâmetros alimentar e reprodutivo das fêmeas oriundas da infestação desafio que não foram significativamente diferentes, com exceção do peso médio de ingurgitamento das fêmeas... (Resumo completo, clicar acesso eletrônico abaixo) / Abstract: This study addressed the effects of salivary glands extracts of Rhipicephalus sanguineus females with 2, 4 and 6 days of feeding (EGS2-EGS6) in New Zealand White rabbits naive inoculated three times at intervals of 21 days. The inoculated animals were challenged with R. sanguineus ticks to verify the influence of extracts on the host's immune-inflammatory response. Changes in the hosts' skin at the feeding lesion, feeding and reproductive parameters as well salivary glands with 2, 4 and 6 days of feeding of females obtained from the challenge infestation were studied. To obtain the glandular extracts, we used 55 females (110 salivary glands) with 2 days of feeding, 36 (72 glands) with 4 days and 25 (50 glands) with 6 days, which were obtained the extracts: EGS2, from salivary glands of females with two days of feeding; EGS4 with 4 days and EGS6 with 6 days. The results revealed that the different glandular extracts stimulated the development of immune-inflammatory response in the hosts' skin compromising the feeding and reproductive processes of the female ticks. The hosts of the TG group (EGS4) showed the formation of an early and intense infiltration of inflammatory cells, indicating that this extract was more effective in developing the local inflammatory-immune response. On the other hand, the response of the TG group hosts (EGS6) to the infestation was delayed and with lower incidence of this infiltrate. The local inflammatory process in the GT hosts (EGS2) was the less intense. The challenge test showed averages for feeding and reproductive parameters of females from infestation challenge that were not significantly different, with the exception of the engorgement average weight of females from TG group (EGS6). Bioinformatic analysis of pea aphid salivary gland transcripts

Master of Science / Biochemistry and Molecular Biophysics Interdepartmental Program / Gerald Reeck / Pea aphids (Acyrthosiphon pisum) are sap-sucking insects that feed on the phloem sap of some plants of the family Fabaceae (legumes). Aphids feed on host plants by inserting their stylets between plant cells to feed from phloem sap in sieve elements. Their feeding is of major agronomical importance, as aphids cause hundreds of millions of dollars in crop damage worldwide, annually. Salivary gland transcripts from plant-fed and diet-fed pea aphids were studied by RNASeq to analyze their expression. Most transcripts had higher expression in plant-fed pea aphids, likely due to the need for saliva protein in the aphid/plant interaction. Numerous salivary gland transcripts and saliva proteins have been identified in aphids, including a glutathione peroxidase. Glutathione peroxidases are a group of enzymes with the purpose of protecting organisms from oxidative damage. Here, I present a bioinformatic analysis of pea aphid expressed sequence tag libraries that identified four unique glutathione peroxidases in pea aphids. One glutathione peroxidase, ApGPx1 has high expression in the pea aphid salivary gland. Two glutathione peroxidase genes are present in the current annotation of the pea aphid genome. My work indicates that the two genes need to be revised.

The recombinant protein c90 was prepared and polyclonal antibodies against this protein were raised. The dsRNA was made for the experiments with RNA interference. The samples from dissected tissues of dsRNA silenced ticks were tested by RT-PCR and Western blot. Results suggest that protein c90 plays a role in the tick body during the reaction to injury. Finally, another experiment with injection of water, G+ and G- bacteria into the ticks was realized. It was found that the members of the 18,7 kDa protein family can create multimers. The overexpression of silenced genes was observed during RNAi experiments despite of expected inhibition of c90 production. These results together with the bioinformatics analysis could mean that these proteins are important the physiology of tick probably as a reaction to injury. However c90 protein is produced only in the first phase of feeding which could mean that it has some role in the tick-host interaction as well.

A gengivite e a periodontite constituem doenças periodontais infecciosas comuns do homem, nas quais as bactérias periodontais e seus produtos participam ativamente para indução da inflamação local e efeitos sistêmicos (ex.: coração). Sabendo que a saliva representa a primeira e grande barreira às infecções orais, este trabalho se propôs a: i) avaliar a perda óssea induzida pela doença periodontal em ratos submetidos à indução de doença, via implante de ligadura, após os intervalos de 3, 7 e 14 dias; ii) Investigar possíveis alterações de fluxo (estimulado ou não com pilocarpina) e composição salivar nesses animais; iii) avaliar a concentração / expressão de marcadores de estresse oxidativo e inflamatórios em glândulas salivares e em amostras de saliva; iv) avaliar o papel funcional da função das glândulas salivares ex-vivo na produção da amilase. Para isto, ratos Wistar machos (180 -200g) foram submetidos à indução da periodontite através do implante da ligadura, e os parâmetros inflamatórios e bioquímicos foram avaliados nesses animais. Ratos com periodontite no dia 3, quando comparado ao grupo sham, exibiram aumento significativo do fluxo salivar (estimulada com pilocarpina), produção de Ca2+, secreção de proteínas e produção de amilase na saliva, bem como aumento do conteúdo de TBARs em glândulas parótida e da amilase liberada das glândulas submandibulares (GSM). Observou-se, ainda, aumento da expressão de mRNA para iNOS e nNOS em GSM. Em contrapartida, ratos com periodontite após 7 dias exibiram redução da taxa de salivação estimulada (e não estimulada), da produção de proteínas totais e da concentração e secreção de amilase na saliva, muito embora o conteúdo sérico e salivar de TBARs e da atividade de MPO na saliva desses animais mostrou-se elevado em relação ao grupo sham. Não foram observadas diferenças significativas quanto ao conteúdo de TBARs em glândulas salivares, secreção e concentração de Ca2+ na saliva e tampouco sobre o conteúdo de proteínas nitradas em amostras de GSM desses animais. Já no 14<font face=\"Symbol\">&#176; dia visualizou-se um aumento da atividade de NOS Ca2+ dependente e da expressão mRNA das iNOS e nNOS em GSM. Ratos com periodontite, após 14 dias de indução, não exibiram aumento significativo na taxa de salivação, concentração e secreção de Ca2+ salivar, produção e concentração de proteínas totais, amilase na saliva e conteúdo de TBARs em amostras de saliva e glândulas salivares. Ainda, observou-se aumento das atividades da peroxidase/MPO, concentração de nitrato em saliva e proteínas nitradas em GSM e maior concentração de citocinas Th1 / Th2 (IL-4, IL-13 e IL-10) em amostras de GSM. Conclui-se que a indução experimental da doença periodontal em ratos , influencia o funcionamento de glândulas salivares de acordo com os dias de indução, inicialmente estimulando, em um segundo momento inibindo e posteriormente retornando aos níveis basais. Após 7 dias, caracteriza-se como o tempo ideal para a manifestação dos efeitos inibitórios na glândula. / Gingivitis and periodontitis are common infectious periodontal diseases in man, in which periodontal bacteria and their products participate actively to induce local inflammation and systemic effects (eg heart). Knowing that saliva represents the first major barrier to oral infections, this study aimed to: i) to assess bone loss due to induced periodontitis in rats, after intervals of 3, 7 and 14 days, ii) to investigate possible changes in flow (or not stimulated with pilocarpine) and salivary composition in these animals, and iii) evaluate the concentration and expression of markers of oxidative stress and inflammation in the salivary glands and saliva samples, and iv) assess the functional role of salivary gland function in ex vivo production of amylase. For this purpose, male Wistar rats (180-200g) underwent induction of periodontitis by implanting the ligature, and biochemical and inflammatory parameters were assessed. Rats with periodontitis on day 3 when compared to sham group, exhibited a significant increase in salivary flow (stimulated with pilocarpine), production of Ca2+, protein secretion and production of amylase in saliva, as well as increased contents of TBARS in the parotid and amylase released from submandibular glands (GSM). There was also increased expression of mRNA for iNOS and nNOS in GSM. In contrast, rats with periodontitis after 7 days exhibited a reduction in stimulated saliva (not stimulated), production and total protein concentration and secretion of salivary amylase, although the content of serum and salivary TBARS and the activity of MPO in saliva of these animals was high compared to the sham group. There were no significant differences in TBARS content in salivary gland secretion and Ca2+ concentration in saliva, nor on the content of nitrated proteins in samples from these animals GSM. By the 14th day envisioned an increase of NOS activity and Ca2+ dependent mRNA expression of iNOS and nNOS in GSM. Rats with periodontitis, 14 days after induction, exhibited a significant increase in the rate of salivation, concentration and salivary secretion of Ca2+, production and concentration of total protein, salivary amylase and content of TBARS in samples of saliva and salivary glands. Still, there was increased activity of peroxidase / MPO, nitrate concentration in saliva and proteins nitrated in GSM and higher concentration of Th1 / Th2 (IL-4, IL-13 and IL-10) in samples of GSM. We conclude that the experimental induction of periodontal disease in rats, influence the functioning of the salivary glands according to the days of induction, initially stimulating, in a second time and subsequently inhibiting return to baseline levels. After 7 days, is characterized as the ideal time for the expression of an inhibitory effect on the gland function.

Vinte e um animais foram divididos em 2 grupos experimentais, controle e irradiado, sendo o primeiro grupo composto por animais não expostos à radiação e o segundo formado for animais que tiveram a região de cabeça e pescoço irradiada com uma dose única de 15 Gy. As glândulas parótidas dos animais irradiados foram removidas nos tempos 4, 8, 12, 24, 48 e 72 horas após sua exposição e, posteriormente, submetidas ao processamento tecidual por meio de métodos imunohistoquímicos, para a avaliação da proteína laminina, e pela coloração por hematoxilina-eosina, para avaliação morfológica. Não foram observadas diferenças estatisticamente significantes entre a intensidade demarcação da laminina do grupo controle e do grupo irradiado. Por outro lado, verificou-se o espessamento e a presença de conglomerados de laminina nas membranas basais acinares dos subgrupos irradiados, tendo sido detectadas diferenças significativas entre o grupo controle e o subgrupo 72 horas (p = 0,0315). A análise morfológica revelou a presença de figuras de necrose nuclear, vacuolizações e severa atrofia dos ácinos, especialmente nos grupos 8 e 12 horas. O início do processo de reparo se deu no tempo 24 horas, mas não foi observada recuperação completa nos últimos tempos avaliados. Os resultados sugeriram que os efeitos deletérios da radiação ionizante podem ser um dos fatores responsáveis pelo remodelamento da matriz extracelular / Abstract: The aim of this study was to evaluate the morphological and immunohistochemical characteristics of gamma radiation acute effects on rat parotid glands. Twenty-one animals were divided into two groups: control and irradiated ones. The former was composed by non-exposed animals, while the latter was formed by rats which had their necks and heads irradiated with 15 Gy. The parotid glands were excised at 4, 8, 12, 24, 48 and 72 hours after irradiation and, then, were submitted to immunohistochemical methods, for the evaluation of the laminin, and to hematoxylin-eosin staining, for the analysis of the morphology. Laminin staining intensities between irradiated and non-irradiated salivary glands showed no statistically significant differences. Nevertheless, it was observed that laminin thickening and laminin-positive conglomerations demonstrated significant differences between the control and the 72-hour-subgroup (p= 0,0315). The morphological analysis revealed the presence of vacuolation, acinar atrophy and necrosis, especially by 8 and 12 hours after irradiation. Regeneration of the tissue started by 24 hours, but it was not complete at 48 and 72 hours after exposure. No presente estudo utilizamos microscopia eletrônica de transmissão e métodos bioquímicos para estudar alterações que ocorrem na matriz extracelular durante atrofia experimental de glândulas submandibulares de ratos. Estudos imunohistoquímicos da laminina e do colágeno IV mostraram distribuição irregular dessas proteínas ao redor das estruturas ductais e acinares atróficas. Análise em "Western blot" mostrou que glândulas submandibulares de ratos expressam laminina contendo apenas cadeias S, enquanto a cadeia A é presente em pequena quantidade. Isso foi observado tanto em glândulas normais como atróficas. Investigação da expressão da colagenase fibroblástica (MMP-1) por técnica zimográfica mostrou altos níveis desta enzima após 5 dias de atrofia. Com a evolução da atrofia, a atividade gelatinolítica regrediu gradativamente até 30 dias de atrofia. Microscopia eletrônica mostrou extensivas alterações morfológicas nas células glandulares. Células mioepiteliais de glândulas atróficas mostraram áreas com espessamento da membrana basal. A atrofia experimental de glândulas salivares pode ser usada como modelo in vivo para estudar eventos moleculares que ocorrem durante remodelação da matriz extracelular / Abstract: Obstructive atrophy of salivary glands is a common disease. It may occur by ductal obstruction caused by calculus, infection or neoplastic processes. In the present work we have used electron microscopy and biochemical methods to study the changes that occur in the expression laminin, type IV collagen and metaloproteinases during the experimental atrophy of the submandibular gland in rats. Immunohistochemical studies of laminin and type IV collagen showed an irregular distribution of there proteins around atrophies ducts and acini. The composition of laminin chains was not altered during glandular atrophy. Western blot analysis showed that rat submandibular gland laminin contain B chains, while A chain seems to be present only in small amounts. Investigation of the expression of fibroblast collagenase (MMP1), by zimographic analysis showed. high levels of this enzyme after 5, days of ligation. Gelatinolytic activity decrease progressively until 30 days of atrophy. Electron microscopy showed extensive morphologic changes in glandular cells. Myoepithelial cells showed "wrinkling" of outer plasma membrane leading to a thickening of the associated basement membrane.
