Characterization of Escherichia coli Single-gene Deletion Mutants Impaired in Bacteriophage Reproduction

An assay was designed to measure the sensitivity of Escherichia coli mutants to bacteriophage infection via growth curves, using a Tecan temperature controlled plate reader. I screened 3985 single-gene deletion strains in Escherichia coli K12 from the Keio collection and identified 43 strains displaying varying degrees of resistance to four different phages, three non-contractile tailed phages (λcI857, HK97, and HK243) and the myoviridae T6, including 20 genes not previously implicated in phage infection. Additional assays, such as adsorption and tests of DNA-injection, were designed to further characterize resistant strains. The use of these assays helped identify varying sensitivities to LPS structure and LamB receptor concentration in the three non-contractile tailed phages, showing HK97 is the most sensitive to changes and HK243 the least. I also found that the periplasmic chaperone, FkpA, is required for HK97 DNA-injection.

Experimental Investigation Of Energy Dissipation Through Screens

Screens may be utilized efficiently for dissipating the energy of water. In this study, water flowing beneath a gate is used to simulate the flow downstream of a hydraulic structure and screens are used as an alternative mean for energy dissipation. Investigations are done conducting a series of experiments. The porosity, thickness, and the location of the screens are the major parameters together with the Froude number of the upstream flow. The scope of this thesis covers the situation where there is a pseudo-jump formation. The experiments covered a range of Froude numbers between 5 and 18, porosities between 20% and 60%, and location of the screen up to 100 times of the undisturbed upstream flow depth. The thicknesses of the screens used are in the order of the undisturbed upstream flow depth. The results show the importance of each parameter on the energy dissipating performance of the screens and the system. It is observed that screens dissipate more energy than a jump within the range covered in these studies.

Nosso estudo é pioneiro ao tentar relacionar a qualidade de vida com os resultados do FMS.Objetivo Avaliar o equilíbrio postural estático e dinâmico, nível de atividade física e a qualidade de vida de universitários jovens e sadios. Métodos Trata-se de um estudo transversal, caracterizado pela aplicação do método FMS em indivíduos jovens e sadios. Os voluntários foram submetidos a testes de equilíbrio e questionários de nível de atividade física e qualidade de vida. Participaram do presente estudo 58 voluntários, de ambos os sexos, universitários, de 18 a 35 anos e sem histórico de trauma ou lesões que pudessem gerar algum impedimento na realização das atividades do trabalho. Resultados O presente estudo demonstrou que o nível de atividade física, domínios de qualidade de vida e índices de equilíbrio unipodal de universitários sadios não discriminaram os grupos FMS1 e FMS2. Considerações finais Tais achados reforçam a importância de se rever o uso do FMS enquanto única estratégia para prevenção de lesões ou análise de desempenho durante atividades físicas e esportivas. / IntroductionIt is understood that the evaluation of functional movements is a way to identify deficiencies of mobility and stability, sometimes neglected in classical and traditional assessments even in the asymptomatic population. Our study is the first to try to relate the quality of life with the results of FMS. Objective To evaluate static and dynamic postural balance, physical activity and quality of life of young students and healthy.Methods This was a cross-sectional study, characterized by the method FMS in young and healthy individuals. The volunteers underwent balance tests and questionnaires level of physical activity and quality of life. The study included 58 volunteers of both sexes, students, 18-35 years and with no history of trauma or injury that could generate some impediment in carrying out work activities. Results The study showed that the level of physical activity, quality of life domains and single leg balance indexes of healthy university did not discriminate the FMS1 and FMS2 groups. Conclusion These findings reinforce the importance of reviewing the use of FMS as the sole strategy for prevention of injury or performance analysis for physical and sporting activities.

TITLE: THE EFFECTS OF A COMMUNITY-BASED LIFESTYLE INTERVENTION ON SNACKING PATTERNS, SCREEN TIME PATTERNS, AND PHYSICAL ACTIVITY LEVELS AMONG ADOLESCENTS WHO ARE AT-RISK FOR TYPE 2 DIABETES MAJOR PROFESSOR: Dr. Sharon Peterson Previous research has shown that overweight and obesity in adolescents is one of the leading risk factors for developing Type 2 Diabetes Mellitus (T2DM). There are several risk factors for obesity that are modifiable in adolescents. Of these, snacking patterns, screen time, and lack of physical activity have an impact on the body weight and overall health of adolescents. The design of this "R.U.A. Healthy Kid?" study was a prospective cohort of 16 free-living adolescents at-risk for T2DM. Each participant was previously screened and found to have two or more risk factors for T2DM. The intervention targeted several components, however snacking patterns, physical activity levels and screen time levels mold the foundation for the current study. An overall "snacking score" was developed to measure the frequency and quality of snacking patterns of participants. After three months of intervention, participants' overall snacking score increased. Many significant relationships were found between the overall snack score, physical activity, and screen time questions, such as negative relationships between: "time on TV" and total snacking score and "time spent walking, biking, or jogging" and "unhealthy" snacking score. These results show how community-based lifestyle interventions that address adolescents' snacking patterns, screen time levels, and physical activity levels are crucial in the fight to overcome the T2DM epidemic now prevalent in adolescents.

Philosophiae Doctor - PhD / Tuberculosis (TB) is an infectious disease with adverse effect on a global scale. The disease is one of the major causes of death in sub-Saharan Africa. Nearly 70% of TB-infected persons are co-infected by the human immunodeficiency virus (HIV). About 50% of TB/HIV patients are smear negative and up to 28% are sputum scarce, which is a significant problem in South Africa since sputum smear microscopy is the most widely used diagnostic test for TB. The detection of Mycobacterium tuberculosis (MTB) and resistance to the TB drug rifampicin (RIF) are the basis of the GeneXpert MTB/RIF protocol. The GeneXpert MTB/RIF is an automated nucleic acid amplification technique for detecting the DNA that originates from MTB. However, low sensitivity and low concentrations of MTB for DNA amplification are a serious issue associated with the protocol. Therefore, other TB diagnostic methods, such as the ones involving biochemical markers of TB, are becoming very important. / 2020-08-31

The role of alpha-synuclein oligomers in Parkinson's disease pathophysiology and biology

Accumulating evidence links oligomeric species of the protein alpha-synuclein to the neuronal death associated with Parkinson's disease. However, the direct detection of alpha-synuclein oligomers in post-mortem brain has been challenging and this has limited our understanding of their structure, distribution and effects in Parkinson's disease. The work presented in this thesis addresses two aspects of the role of alpha-synuclein oligomers in Parkinson's disease. Firstly, I describe the development of a novel technique, the alpha-synuclein proximity ligation assay (AS-PLA), which specifically detected alpha-synuclein oligomers in vitro and in post-mortem brain tissue. In a blinded study with post-mortem brain tissue from eight Parkinson's disease patients and eight controls, AS-PLA revealed widespread, previously unrecognised pathology in the form of extensive diffuse deposition of alpha-synuclein oligomers. Furthermore, AS-PLA preferentially detected early-stage, loosely compacted Parkinson's disease lesions such as pale bodies, whereas Lewy bodies, considered heavily compacted late lesions were only very exceptionally stained. The oligomeric species detected by AS-PLA displayed a unique, intermediate proteinase K resistance profile, suggesting the detection of a conformer that is different from both physiological pre-synaptic alpha-synuclein (proteinase K sensitive) and highly aggregated alpha-synuclein within Lewy bodies (proteinase K resistant). In addition, AS-PLA revealed the age-dependent accumulation of alpha-synuclein oligomers in the substantia nigra of a BAC transgenic mouse model of Parkinson's disease that overexpresses human wild-type alpha-synuclein, SNCA-OVX. Secondly, the detection of early pathology in Parkinson's disease brain tissue using AS-PLA suggests that oligomeric species of alpha-synuclein could represent a potential target for therapeutic intervention. Therefore, I undertook a screen to identify compounds that can prevent the formation of alpha-synuclein oligomers in vitro. Using bimolecular fluorescence complementation constructs, I identified nine compounds capable of reducing the fluorescence indicative of the formation of alpha-synuclein oligomers. Two of these compounds showed dose-dependent activity. Future work will confirm the hits in vitro before studying whether Parkinson's-like phenotypes in the SNCA-OVX mice can be ameliorated or reversed by treatment with the compounds.
