Kustovnice čínská a maka v nových potravinových výrobcích / Lycium chinense and Lepidium meyenii in new food products

This diploma thesis deals with application of wolfberry (Lycium chinense) and maca (Lepidium meyenii) in food products. Both plants have high nutrition content and are studied for their health benefits to humans. In case of wolfberry it’s mainly immune-stimulating and antioxidant properties. Maca is known for increasing fertility and there’s also reported lowering of cholesterol and blood sugar. In theoretic part are summarized findings about chemical composition, science studies about effects of these plants, situation on the Czech trade and project of use. Aim of this thesis was to create practical demonstration of use and consumption both plants and to evaluate it by methods of sensory analysis. For this purpose three samples of wolfberry cereal bars and three samples of chocolate with maca powder were produced. These samples were sensory evaluated using ranking test, scale test and profile test. Higher amount of maca in chocolate had worse rating, while use of wolfberry was rated quite well. Therefore three wolfberry cereal bars were produced as the final product in cooperation with bakery and chocolate factory Fikar – bar with fruit filling glazed with chocolate, classical bar glazed and classical bar not glazed with chocolate, as a demonstration of possible industrial use.

Stanovení aromaticky aktivních látek v odrůdách jablek / Assessment of aroma active compounds in cultivars of apples

The aim of this master‘s thesis was to determine the aroma active compounds and monitor the sensory quality of apples. The theoretical part deals with the description, composition and properties of apples, information about the aroma compounds, their determination and possibilities for sensory evaluation of apples is also given. The gas chromatography method combined with the solid-phase microextraction, utilized for the specification of aroma compounds, is described. In the experimental part, the SPME-GC-FID method was applied on selected cultivars of apples specially cultivated against apple scab (Venturia inaequalis), i.e. 24 genotypes and Golden Delicious as the control variety. In total 60 various aroma compounds were identified, 22 alcohols, 12 aldehydes, 11 ketones, 10 esters and 5 organic acids. The number and concentration of compounds identified differed according to the cultivars. Sensory analysis was performed using category ordinal scales and the profile test.

La valorisation des déchets par le compostage et la méthanisation est encouragée par les politiques de l'UE. Cependant, ces technologies de recyclage des déchets sont confrontées à un défi crucial en matière de libération et de contrôle des émissions de gaz odorants. En effet, la pollution par les odeurs est la principale cause d'opposition du public au traitement des déchets, ce qui a entraîné la fermeture et des poursuites en justice pour les unités de traitement des déchets. À l'heure actuelle, les inventaires des émissions d'odeurs et de produits chimiques pour appuyer les plans de gestion des odeurs et de la pollution atmosphérique font défaut, en raison de la complexité des évaluations des odeurs, impliquant généralement des analyses sensorielles et chimiques. L'objectif de cette thèse était donc de caractériser de manière détaillée les émissions de gaz et les odeurs lors du processus de compostage. À cette fin, les émissions d’odeurs et de polluants ont tout d’abord été étudiées au cours des différentes étapes opérationnelles du processus de compostage, à savoir le stockage, la phase active, le retournement et la maturation. Les résultats ont montré que la phase active et le retournement jouent un rôle essentiel dans les générations de nuisances olfactives causées principalement par les composés soufrés volatils (CSV). Des efforts supplémentaires ont ensuite été consacrés à la caractérisation des émissions de gaz au cours de la phase active de compostage de quinze déchets solides et du digestat, permettant ainsi de corroborer l’importance des CSV sur la production d’odeurs lors du compostage et de différencier l’impact des émissions de gaz sur les odeurs en fonction des matières premières des déchets. La dernière partie de ce travail de recherche a été consacrée à la corrélation des mesures sensorielles et chimiques, par le biais de méthodes de régression simple et multiple visant à faciliter et à renforcer le contrôle des odeurs dans les unités de traitement des déchets. Notamment, un modèle des moindres carrés partiels améliore la prévision de la concentration d'odeurs grâce à la composition chimique des gaz émis. / Waste valorization through composting and methanization are actively promoted by the EU policies. However, these waste recycling technologies face a crucial challenge regarding the release and control of odorous and hazardous gas emissions. Indeed, odor pollution is the principal cause for public opposition to waste processing, leading to the closure and lawsuits for waste treatment units. Currently, there is a lack of odor and chemical emissions inventories to support odor and air pollution management plans due to the complexity of odor assessments, generally involving sensory and chemical analysis. Hence, the aim of this PhD thesis was to characterize extensively the gas emissions and odors released during the composting process. To this end, the patterns of odor and pollutant emissions were first traced throughout different operational stage of composting process, namely, storage, active phase, turning and curing. The results showed that the active phase and turning played a pivotal role in odor nuisance generations which were mainly caused by volatile sulfur compounds (VSC). Then, further efforts were focused on characterizing the gas emissions released along the composting active phase of fifteen solid wastes and digestate. The results corroborated the relevance of VSC on odors production during composting and enabled to differentiate the odor impact of gas emissions as a function of the wastes’ feedstock. The last part of this research work was devoted to correlate both, sensory and chemical measurements, through univariate and multivariate regression analysis to ease and strength odor monitoring in waste treatment units. Notably, a partial least squares model improved odor concentration prediction based on the chemical composition of emitted gases.

Les polysaccharides et les ellagitanins du bois de chêne : influence sur la qualité sensorielle des vins / Oak wood polysaccharides and ellagitannins : the impact on sensory quality of wines

Il est généralement admis que la qualité des vins et des eaux de vie dépend de leur vieillissement au contact du bois de chêne. La sélection du bois de chêne et sa maturation constituent des étapes essentielles dans la fabrication de barriques. Compte tenu des nombreux facteurs impliqués dans le choix de la matière première et dans la phase de séchage naturel du bois de chêne, il est très complexe de déterminer l’impact de la durée de séchage et l’incidence des facteurs de sélection combinés. Nous avons pu mettre en œuvre un plan d’essai qui nous a permis d’étudier l’évolution de la composition chimique du bois de chêne pour quatre stades de séchage : bois frais, 12 mois, 18 mois et 24 mois. Pour ce plan d’essai, l’échantillonnage a été réalisé en prenant en compte trois facteurs de sélection d’intérêt en tonnellerie : l’âge du bois, le type de grain et la classe d’Indice Polyphénolique mesurée par Oakscan®. En plus de la partie extractible (~10%), trois polymères constituent la majeure partie (~90%) du bois de chêne : la cellulose, les hémicelluloses et les lignines. Ces composés sont susceptibles de subir des hydrolyses ou des réactions chimiques au cours des différents processus de tonnellerie et notamment lors de la chauffe, libérant ainsi des composés aromatiques ou certains précurseurs aromatiques ayant un véritable intérêt sensoriel pour le vin vieilli en fût ou en contact avec du bois de chêne. Aucune étude n’a, à ce jour, montré un lien entre les proportions de ces composés dans le bois de chêne et l’impact au niveau chimique et organoleptique dans des vins élevés au contact de bois de chêne. Nous avons pu montrer que leurs proportions évoluent de façon significative au cours du séchage du bois de chêne et nos résultats soulignent l’incidence des différents facteurs de sélection. Respectivement, les proportions en composés extractibles, lignines, hémicelluloses et cellulose apparaissent majoritaires aux stades bois frais, 12 mois, 18 mois et 24 mois de séchage. La mise en place d’un plan d’essai sur un vin de Merlot à partir d’un deuxième échantillonnage de bois de chêne, selon les mêmes modalités que le plan d’essai sur bois, nous a permis d’évaluer l’impact du temps de séchage du bois sur la composition chimique d’un vin élevé au contact de morceaux de bois de chêne. Les résultats ont montré entre autres une baisse de près de 8% de teneur en ellagitanins moléculaires du vin entre les modalités 12 et 24 mois de séchage mais aussi des impacts sur les concentrations en composés volatiles du vin : les aldéhydes furaniques sont corrélés positivement à des temps de séchage longs (18 et 24 mois), tandis que les aldéhydes phénoliques le sont avec un temps de maturation plus court (12 mois). Nos résultats soulignent aussi l’influence des facteurs de sélection sur la composition chimique du vin. Sur le même vin de Merlot, la réalisation d’analyses sensorielles a mis en évidence des différences significatives perçues par notre panel entre les modalités de séchage 12 et 24 mois, tandis que les modalités 18 et 24 mois n’ont pas été distinguées lors de tests triangulaires. Des profils sensoriels ont également été établis pour tenter d’associer les différences perçues à un ou plusieurs descripteurs. Pour cette épreuve, les résultats soulignent la difficulté pour notre panel de dégustateurs de s’accorder sur l’existence d’une différence globale et significative entre les modalités 12 et 24 mois. Cependant, ils contribuent à expliquer les différences mises en évidence lors des tests triangulaires pour certaines modalités de séchage pour les descripteurs grillé/fumé, amertume, astringence, rondeur en bouche et perception sucrée. / It is widely accepted that alcoholic beverages quality depends on their ageing in premium quality oak wood. Oak wood selection and maturation are essential steps in the course of barrel fabrication. Given the existence of many factors involved in the choice of raw material and in natural seasoning of oak wood, it is very difficult to determine the real impact of seasoning and selection factors on oak wood composition. A sampling was done to study the evolution of oak wood chemical composition during four seasoning steps: non matured, 12 months, 18 months and 24 months. For this sampling, three selection factors were taken into account: age, grain type and the Polyphenolic Index measured by Oakscan®. Besides extractables (~10%), three polymers constitute the main part of oak wood: cellulose, hemicelluloses and lignins. These compounds may undergo hydrolysis or chemical reactions during cooperage processes, especially during heat treatment, which release some aromatic compounds or aromatic precursors having a genuine sensorial interest on wine aged in barrel or in contact with oak products. To date, no studies revealed a link between the proportions of these compounds in oak wood and the chemical and sensorial impact in wines ageing with oak wood. Our study showed that the proportions of these compounds evolved significantly during oak wood seasoning and the results highlighted the impact of selection factors. Respectively, extractables, lignins, hemicelluloses and cellulose proportions were mostly for non-matured, 12 months, 18 months and 24 seasoning months. The development of a test plan with a Merlot wine from a second oak wood sampling, using similar modalities as the previous test plan allowed the evaluation of oak wood seasoning impact on the chemical composition of a wine ageing with oak wood pieces. Results showed a lowering of 8% in ellagitannins content of wine between 12 and 24 months modalities. An impact on volatile composition in wine has also been established: furanic aldehydes were positively correlated with a long maturation time (18 and 24 months), whereas phenolic aldehydes were positively correlated with a shorter seasoning time (12 months). Our results highlighted also the impact of selection factors on wine chemical composition. Sensorial analysis on this Merlot wine led to significant differences detected by the panel between 12 and 24 seasoning modalities, but not between 18 and 24 months modalities with triangular tests. Sensorial profiles were also established to attempt to associate the differences detected to one or more descriptors. For this test, results highlighted the difficulty for the panel to make a significant difference between 12 and 24 months modalities. However they contribute to explain the differences demonstrated with triangular tests for some seasoning modalities: toasted, bitterness, astringency, roundness and sweet perception.

PHYSICOCHEMICAL AND SENSORY EVALUATION OF INVASIVE SILVER CARP (Hypophthalmichthys molitrix) FISH NUGGETS

<p>Silver carp (<i>Hypophthalmichthys molitrix</i>)<b> </b>are an underutilized, invasive fish threatening native species throughout major water systems in the United States. The goal of this research was to use silver carp meat to create a value-added product, to analyze the changes in physicochemical structure and consumer liking over time, and to evaluate the benefits of adding soy, pea, and a combination of soy and pea protein isolates to the formulations. Fish nuggets were prepared from minced meat in four treatments consisting of 3% soy protein isolate (SPI), 3% pea protein isolate (PPI), a combination of 1.5% SPI and 1.5% PPI, and a control without plant protein isolate. Nuggets from each treatment were stored frozen for 1.5, 8.5, and 13.5 weeks. Proximate composition, pH, cook loss, textural hardness, expressible moisture, color, microbial counts and lipid oxidation were evaluated. Sensory acceptability was also evaluated for each frozen storage time period and treatment. A descriptive (QDA<sup>™</sup>) trained sensory panel was also conducted on all treatments independent of the storage testing. Results showed that lipid oxidation and textural hardness significantly (P < 0.05) increased with frozen storage time. PPI had significantly lower expressible moisture compared to the control at week 1.5, but there were no statistically significant differences between treatments at weeks 8.5 and 13.5. Similarly, formulations with PPI improved (p<0.05) cook loss for week 1.5, but not week 8.5 or 13.5. Overall, sensory acceptability did not change (P > 0.05), with the exception of decreased degree of liking scores for SPI aroma (p=0.03) and flavor (p=0.03)) during the frozen storage period; all degree of liking scores remained above 6.5 throughout analysis, indicating that consumers’ acceptability of the sample treatments over time despite the changes in physicochemical structure. The descriptive panel created an attribute lexicon for the aroma, flavor, and mouthfeel of the fish nuggets and did not find significant differences in intensities for those attributes between the treatments. Although there were measurable changes in oxidation, texture, expressible moisture, and cook loss over 13.5 weeks, these changes did not impact sensory acceptance. The addition of protein isolates improved water holding capacity initially but did not maintain those benefits over extended shelf life and had little impact on consumer liking during any time period. Overall, this study demonstrated that value-added products such as silver carp nuggets can be created using an otherwise under-utilized fish. The fish nuggets had high sensory acceptability, and the addition of protein isolates did not significantly improve their sensory characteristics; therefore, silver carp nuggets can be formulated without the need of additional protein additives .</p>

Hur påverkar olika lokala jäststammar aromutveckling och smak vid fermentering av äppeljuice? / How does different indigenous yeast strains influence the aroma development and taste during fermentation of apple juice?

Intresset för lokalproducerad dryck ökar. Konsumenten efterfrågar produkter med mer komplexa smaker. Främst gäller detta produktkategorin öl, vin och cider. Det har i flera studier visats att lokala stammar av mikroorganismer kan användas för att tillverka produkter med en större smakkomplexitet än de produkter som baseras på konventionella stammar. Detta har fått bryggare och vinmakare att experimentera med såväl val av råvara som mikroorganismer i syfte att ta fram unika produkter. I denna studie har tre lokala stammar av Saccharomyces cerevisiae undersökts avseende eventuella skillnader i fermenteringsförlopp och aromutveckling samt smak vid fermentering av äppeljuice. Resultatet från studien visar att alla stammarna överlag beter sig lika inklusive enskilda sensoriska attribut och att en fermentering på tre dagar ger upphov till högre totalt gillande av produkt än vid sju dagars fermentering. / There is an increasing interest in locally produced drinks. Consumers demand products with complex taste profiles. Especially when it comes to wine, beer and cider. Several studies have shown that indigenous strains of microorganisms can be used to produce products that’s deemed more complex than the products based on conventional strains. This has led brewers and winemakers to experiment with both the raw material as well as the local microorganisms in order to develop unique products. In this study, three local strains of Saccharomyces cerevisiae have been studied for potential differences regarding aroma development and taste during fermentation of apple juice. The results from the study showed that the different strains generally behaved the same, and that fermentation during three days resulted in greater taste liking compared to seven days.

L’âge de la vigne et sa relation avec la qualité du vin sont des sujets d’intérêt récurrents, tant scientifiques qu’économiques. Les consommateurs et acteurs de la filière vitivinicole semblent s’accorder à propos de la capacité des vieilles vignes à produire des vins de caractère supérieur. Malgré les recherches en cours, la validité de ce point de vue reste débattue et les questions concernant les mécanismes à travers lesquels de vieilles vignes aboutiraient à des vins qualité supérieure restent nombreuses. Pour tenter d’y répondre, l’impact de l’âge des vignes sur la physiologie, la tolérance au stress hydrique, ainsi que la qualité des baies et du vin ont été étudiés dans un vignoble expérimental constitué de plants de Vitis vinifera L. cv. de matériel génétique identique (Riesling de clone Gm 239 greffé sur 5C Teleki) mais aux dates de plantation différentes.En 2014 et 2015, les vignes plantées en 2012 n’avaient pas encore atteint leur plein potentiel et avaient une productivité végétative et un rendement significativement inférieurs à ceux des vignes plantées en 1995 et 1971. Par ailleurs, les vignes plantées en 2012 n’ont pas été soumises au même traitement d’enherbement que les vignes plus âgées pendant cette période afin de prévenir une compétition excessive pendant leur établissement. La capacité inférieure de ces vignes et l’absence d’enherbement ont mené à une plus grande exposition des grappes à la lumière et une plus grande accumulation d’azote, ce qui s’est traduit par une plus grande concentration en acides aminés, monoterpènes, norisoprénoides, et flavonols en 2014 et 2015. Les années suivantes (2016 et 2017), le rendement et le poids des bois de taille de ces vignes, ainsi que la composition des baies, étaient comparables à ceux des vignes plus âgées. Les paramètres de maturité technologique (°Brix, l’acidité totale et le pH de moûts) n’ont pas été significativement affectés par l’âge des vignes. […]Des analyses sensorielles et chimiques ont été réalisées en 2017 sur des vins de millésimes précédents. Les vins des plus jeunes vignes ont été associés à des arômes de fruits mûrs et de l’arôme de pétrole typique du Riesling. Ces vins ont aussi été identifiés par de plus hautes concentrations de monoterpènes et norisoprénoides potentiels et de composés soufrés volatils, en 2014 et 2015 uniquement. Les profils sensoriels et chimiques de vins issus des vignes plantées en 1995 et 1971 étaient dépendants du millésime mais pas de l’âge des vignes. Les profils des vins produits en 2016 étaient en superposables pour les trois groupes d’âge.Les travaux décrits dans ce manuscrit de thèse sont uniques, du fait notamment que le vignoble dans lequel ils ont été conduits a été conçu spécifiquement pour étudier l’effet de l’âge de la vigne dans des conditions environnementales comparables. Une fois que les vignes les plus jeunes ont atteint leur potentiel fructifère et ont été conduites de la même manière que les vignes plus âgées, leur productivité, la composition de leurs baies et la qualité des vins qu’elles produisent ont convergé avec celles des deux autres groupes. Plus intéressant encore, des vignes âgées de 19 et 43 ans se sont comportées de la même façon tout au long de l’étude et ont abouti à des vins comparables en termes d’analyses sensorielles, ce qui va à l’encontre de l’idée reçue qui veut que les vignes les plus âgées produisent des vins de qualité différente.Des travaux précédents ont démontré que la productivité des vignes, quel que soit leur âge, pouvait être expliquée par les réserves de bois et par la taille du tronc. Pour avoir une meilleure idée des différences liées aux réserves, la technique dite « structure-from-motion with multi-view stereo-photogrammetry » (SfM-MVS) a été testée pour mesurer l’épaisseur des troncs et leur volume. Cette technique qui permet la création de modèles tridimensionnels géo-référencés et à l’échelle a pu générer des modèles précis de tronc de vignes plantées en champ. / Vine age and its relation to the quality of the wine are topics of recurring interest, both scientific and economic. Consumers and actors in the wine sector seem to agree on the ability of old vines to produce wines of superior character. Despite ongoing research, the validity of this point of view remains debated and questions about the mechanisms through which old vines would end up with superior quality wines remain numerous. To try to answer them, the impact vine age on physiology, tolerance to water stress, and berry and wine quality were studied in an experimental vineyard planted with Vitis vinifera L. cv. of identical genetic material (Riesling Gm 239 grafted on 5C Teleki) but planted in different years.In 2014 and 2015, the vines planted in 2012 had not yet reached their full potential and had a significantly lower vegetative productivity and yield than the vines planted in 1995 and 1971. Moreover, the vines planted in 2012 were not subjected to the same grass treatment as older vines during this period to prevent excessive competition during establishment. The lower capacity of these vines and the absence of cover crop led to greater exposure of clusters to light and greater nitrogen accumulation, which resulted in a higher concentration of amino acids, monoterpenes, norisoprenoids, and flavonols in 2014 and 2015. In the following years (2016 and 2017), the yield and pruning weight of these vines, as well as their berry composition, were comparable to those of the older vines. The parameters of technological maturity (° Brix, total acidity and must pH) were not significantly affected by vine age.Vines planted in 1995 and 1971 showed similar physiological characteristics throughout the study with the exception of a higher incidence of esca syndrome in the older group. This disease was responsible for the decline in the total yield of vines planted in 1971, but individual yield per vine was equivalent for both groups.Sensory and chemical analyzes were conducted in 2017 on wines from previous vintages. The wines of the youngest vines were associated with aromas of ripe fruit and the kerosene aroma that is typical of Riesling. These wines were also identified by higher concentrations of potential monoterpenes and norisoprenoids and volatile sulfur compounds in 2014 and 2015 only. The sensory and chemical profiles of wines from vineyards planted in 1995 and 1971 were dependent on the vintage but not on the age of the vines. The wine profiles produced in 2016 were overlapping for the three age groups.The works described in this thesis manuscript are unique, particularly because the vineyard in which they were conducted was designed specifically to study the effect of the age of the vine under comparable environmental conditions. Once the youngest vines reached their fruiting potential and were conducted in the same way as the older vines, their productivity, the composition of their berries and the quality of the wines they produce converged with those of the two other groups. More interestingly, vines aged 19 and 43 years behaved similarly throughout the study and resulted in wines comparable in terms of sensory analysis, which goes against the an idea that the older vines produce wines of a different profile.Previous studies have shown that the productivity of the vines, whatever their age, could be explained by the wood reserves and the size of the trunk. To have a better idea of differences linked to reserves, the structure-from-motion with multi-view stereo-photogrammetry (SfM-MVS) method was tested to measure trunk thickness and volume. The technique, which allows the creation of scaled, georeferenced 3D models based on photographs, was able to produce accurate models of field-grown grapevine trunks.

Classifying South African Chenin blanc wine styles

Thesis (MSc)--Stellenbosch University, 2011 / ENGLISH ABSTRACT: South African Chenin blanc wine is characterised by its style diversity and has been receiving more industry limelight over the past few years. Three dry Chenin blanc wine styles are currently described by the wine industry as Fresh and Fruity (FF), Rich and Ripe unwooded (RR-unwooded) and, Rich and Ripe wooded (RR-wooded). According to the South African wine industry, however, consumers seem to be somewhat confused by these various Chenin blanc wine styles, which have not yet been clearly defined and classified. During this study we investigated the following perspectives of this wine style classification confusion: Currently, no specified sensory method is being used to differentiate between wine styles of a single wine cultivar, such as Chenin blanc. The sorting method can be used as a more cost and time effective classification and profiling method than general quantitative descriptive analysis (QDA). It was therefore investigated whether both uninstructed and instructed sorting, with consumers and wine experts, can be used to classify and describe the Chenin blanc wine styles similarly to QDA. The increase in young wine consumers, Generation Y, introduces new wine marketing opportunities for the wine industry which need to be investigated. Young consumers are not aware of the various Chenin blanc wine styles, which may influence their wine preferences. The influence of wine style knowledge on these consumers’ wine style preferences were investigated during blind and wine style informed preference test. Any segmentation within this Generation Y group was also investigated with regards to their Chenin blanc wine style preferences and/or exploratory data. The following scientific and industrial findings were made: QDA was successfully substituted by instructed and uninstructed sorting tasks. Although the sorting tasks easily differentiated between wooded and unwooded wines, QDA differentiated more successfully between the unwooded wines. Wine tasting experience did not significantly influence the sorting results of the consumers versus the wine experts, but more experienced panellists consistently used fewer descriptors than untrained consumers. All panellists sorted products slightly better when they were provided with wine style sorting instructions. However, there were no significant differences between the instructed and uninstructed sorting task results, i.e. a wine style descriptor list did not improve panellists’ abilities to describe their sorted wine groups. South African Chenin blanc wines can definitely be divided into wooded and unwooded wine styles, i.e. two wine style groups, but the unwooded Chenin blanc wine spectrum formed a wine-style-continuum. This finding is apparent from both the sorting and QDA methods. The unwooded continuum wines can further be differentiated from one another with the use of a fresh-to-mature scale and/or a light-to-full body scale during QDA. Generation Y wine consumers preferred all the Chenin blanc wine styles equally and moderately during the blind tasting, but their knowledge of the wine style concepts significantly increased their preferences for the wines during the informed wine style tasting. The cognitive influence of wine style knowledge is therefore powerful in terms of influencing a consumer’s wine preferences. Other interesting correlations were also made between Generation Y consumers’ wine style preferences and their exploratory data. / AFRIKAANSE OPSOMMING: Suid-Afrikaanse Chenin blanc wyn word gekarakteriseer deur styl diversiteit en is die afgelope paar jaar meer in die kollig. Drie droë Chenin blanc wynstyle word tans deur die wynbedryf beskryf as: Vars en Vrugtig, Ryk en Ryp ongehout en, Ryk en Ryp gehout. Volgens die Suid- Afrikaanse wynbedryf is daar heelwat verwarring is tussen hierdie Chenin blanc wynstyle weens die feit dat hierdie wynstyle nog nie duidelik gedefinieer en geklassifiseer is nie. Tydens die huidige studie het ons die volgende perspektiewe van hierdie wynstyl-verwarring ondersoek: Huidiglik word daar geen gespesifiseerde sensoriese metode gebruik om te onderskei tussen verskillende wynstyle van ’n enkele wyn kultivar, soos Chenin blanc, nie. Die sorteringsmetode kan gebruik word, instede van kwantitatiewe beskrywende analise, as ’n meer koste- en tyd-effektiewe produk klassifikasie en profileringsmetode. DIt was dus ondersoek of beide geïnstrukteerde en ongeïnstrukteerde sorteringstake, met verbruikers en wynkundiges, gebruik kan word om die verskeie Chenin blanc wynstyle kan profileer en klassifiseer soortgelyk soos kwantitatiewe beskrywende analise. Die toename in jong verbruikers, Generasie Y, skep nuwe wynbemarkingsgeleenthede wat ondersoek moet word. Jong verbruikers se onbewustheid van die verskeidenheid Chenin blanc wynstyle mag hulle voorkeure vir hierdie wyne beïnvloed. Die invloed van wynstyl kennis op hierdie jong verbruikers se wynstyl voorkeure was ook ondersoek gedurende ’n blinde en ’n wynstyl-ingeligte voorkeur toets. Enige segmentasie binne hierdie Generasie Y verbruikersgroep was ook ondersoek in verband met hul Chenin blanc wynstyl voorkeure en/of verkennende data. Die volgende wetenskaplike en industriële bevindings is gemaak: Kwantitatiewe beskrywende analise kan suksesvol vervang word deur beide geïnstrukteerde en ongeïnstrukteerde sorteringsmetodes. Alhoewel die sorteringsmetodes maklik gebruik kon word om te onderskei tussen gehoute en ongehoute wyne, kan kwantitatiewe beskrywende analise fyner onderskeid tref tussen die individuele ongehoute wyne. Die wynproe-ervaringsvlak het geen invloed op die sorteringsresultate gehad nie, maar meer ervare paneellede het konsekwent gebruik gemaak van minder beskrywende terme as onopgeleide verbruikers wat baie meer en verskillende soorte terme gebruik het. Beide panele het effens beter gesorteer wanneer hulle spesifieke wynstyl sorteringsinstruksies ontvang het. Daar was egter geen beduidende verskille tussen die geïnstrukteerde en ongeïnstrukteerde sorteringsresultate nie, wat daarop dui dat sorteringsinstruksies en ’n lys van wynstyl beskrywings nie die paneellede se wyse van sortering verander het nie. Suid-Afrikaanse Chenin blanc wyne kan definitief verdeel word in gehoute en ongehoute wynstyle, d.w.s twee wynstyl groepe, maar die ongehoute Chenin blanc wyn spektrum vorm ’n wynstyl-kontinuum. Laasgenoemde is bevind tydens beide die sorteringstake asook kwantitatiewe beskrywende analise. Die ongehoute kontinuum wyne kan verder van mekaar onderskei word met behulp van ‘n vars-tot-volryp en/of ‘n ligte-tot-volmond skaal tydens kwantitatiewe beskrywende analise. Alle jong wyn verbruikers het ewe veel van al die wynstyle gehou tydens die blinde wynstyl voorkeur toets, maar tydens die wynstyl-ingeligte voorkeur toets het hulle voorkeure vir al die wynstyle drasties verbeter. Die kognitiewe invloed van Chenin blanc wynstyl kennis kan dus lei tot hoër voorkeure vir die wynstyle. Ander interessante korrelasies is ook bevind tussen hierdie Generasie Y wyn verbruikersgroep se wynstyl voorkeure en hul verkennende data.

The influence of different winemaking techniques on the extraction of grape tannins

These building blocks are called monomers which consist of catechins, epicatechins, epigallocatechins and epicatechin-gallate. Tannin is important in wine as it contributes to bitterness, mouth feel (astringency) and maturation potential of the wine. Futhermore it has a health benefit as an antioxidant. Anthocyanins are responsible for the colour of red wine. The anthocyanins combine with tannins to form stable polymeric pigments. Due to the importance of tannins and anthocyanins in wine, it is imperitative that different winemaking techniques are used to extract as much of these components as possible and that the analysis is done quickly and accurately. The aim of this study was to evaluate different winemaking techniques and their extraction of tannins and anthocyanins into the wine. Too much tannin extraction can have a negative effect on the sensory quality of the wine. Therefore a second aim was to evaluate the mouth feel properties of a Shiraz wine. A third aim was to compare the two tannin precipitation methods in terms of time efficiency, repeatability and the ease of practice. To investigate the amount of tannin concentration extracted by different winemaking techniques, two cultivars (Cabernet Sauvignon and Shiraz) were used. These treatments included the addition of an enzyme during fermentation [E], cold maceration [CM], post maceration [PM] and the combination of cold and post maceration [CM+PM]. The grapes were harvested in two different climatic areas during the 2008 and 2009 vintages. The two climatic areas were classified according to the Winkler scale as a III (Morgenster) and a IV (Plaisir de Merle). The grapes were harvested at two different ripeness levels in order to evaluate the effect of the different winemaking processes on the extraction of tannins and anthocyanins. One harvest was before (LB) and the other after (HB) the commercial harvest. The results of this study showed significant differences in the phenolic composition of the wines. It was found that the warmer area showed higher tannin concentrations than the cooler area for both cultivars. In the 2008 Cabernet Sauvignon the CM extracted higher concentrations of tannin from the cooler area at both ripeness levels. In the warmer area, CM extracted the highest tannin concentration HB, but the CM+PM extracted the highest tannin concentration from Cabernet Sauvignon at the LB and CM at the HB of the warmer area. In 2009 the PM extracted the highest concentration of tannin at the lower ripeness level, while the E treatment extracted the highest concentration from the warmer area. In the cooler area the CM+PM extracted the highest concentration of tannin at a lower ripeness level, while there were no siginicant differences between the different treatments at the higher ripeness level. The highest anthocyanin concentration was found in the cooler area. The CM treatment was found to have no effect on anthocyanin extraction. Different methods are available to quantify the tannin concentration in wine. Two of the most popular tannin analytical methods are the bovine serum albumin (BSA) and the methyl cellulose precipitable tannin (MCP) methods. The BSA method is a very complex method which uses at least 3 times more reagents than the MCP method. The MCP method only analyzes tannins, while the BSA method analyzes tannins, monomeric pigments (MP), small polymeric pigments (SPP) and large polymeric pigments (LPP). In this study a good correlation was found between the two tannin precipitation methods (R2 – 0.88). There is controversy regarding the variability of these methods. Some scientists found that the two methods show a good correlation with HPLC, while others found that there was no such correlation between the precipitation methods and the HPLC. The MCP method had a practical advantage as it could be performed in half the time required for the BSA method. This has a significant impact in scenarios where a high sample throughput is required although it only measures total tannin. The phenolic composition and mouth feel of the wine was strongly influenced by the climatic area. In the warmer area the effect of tannin concentration on mouth feel was much less than in the cooler area. The wine made of riper grapes, was more grippy, bitter and numbing than the wines made from greener grapes. The E treatment was especially associated with a dry, grippy sensation. / AFRIKAANSE OPSOMMING: Druif en wyn fenole bestaan uit flavanole wat weer die boublokke is van tanniene. Hierdie boublokke, wat bekend staan as monomere, betsaan uit katesjiene, epikatesjiene, epigallokatesjiene an epikatesjien-gallaat. Tanniene is belangrik in wyn aangesien dit bydra tot bitterheid, mondgevoel (vrankheid) asook die verouderingspotensiaal van wyn. As antioksidante hou dit ook gesondheidsvoordele in. Antosianiene dra by tot die kleur van rooiwyn. Antosianiene kombineer met tanniene om meer stabiele polimeriese pigmente te vorm. As gevolg van die belangrikheid van tanniene en antosianiene is dit van uiterse belang dat verskillende wynmaak tegnieke gebruik word om ekstraksie in die wyn te bevoordeel en dat die analitiese metode so vinnig en akkuraat as moontlik gedoen word. Die eerste doel van hierdie studie was om die ekstraksie van tanniene en antosianiene deur middel van verskillende wynmaak tegnieke te evalueer. Te veel tanniene in die wyn kan negatiewe sensoriese kwaliteit tot gevolg het. Daarom is die tweede doel om die sensoriese kwaliteit van Shiraz wyn te evalueer. Die derde doel van hierdie studie was die twee tannien presipitasie metodes met mekaar te vergelyk in terme van die moeilikheidsgraad van die metode, tyd doeltreffendheid en herhaalbaarheid. Verskillende wynmaak tegnieke (ensiem byvoegings [E], koue maserasie [CM], verlengde dopkontak [PM] en ‘n kombinasie van koue maserasie en verlengde dopkontak [CM+PM]) is vergelyk ten opsigte van tannien en antiosianien ekstraksie. In 2008 en 2009 is twee kultivars (Cabernet Sauvignon en Shiraz) in twee verskillende klimatologiese areas gepars. Hierdie areas is geklassifiseer in die Winklerskaal as ‘n IV (Plaisir de Merle) en ‘n III (Morgenster). In 2009 het PM die meeste tanniene geëkstraheer by LB terwyl E die meeste tanniene geëkstraheer in die warmer area. In die koeler area het CM+PM die meeste tanniene geëkstraheer, terwyl geen van die behandelings ‘n effek gehad het by HB. Die meeste antosianien konsentrasie was in die koeler area gevind as in die warmer area. In beide 2008 (LB en HB) en 2009 (LB) het CM die meeste antosianiene geëkstraheer, terwyl geen behandeling ‘n effek gehad het by HB. Twee van die mees populêre tannien analitiese metodes is die BSA (bovine serum albumien) en die MCP (metielsellulose presipitasie) metodes. Die BSA metode is ‘n baie meer ingewikkelde metode waarvoor drie keer meer reagense gebruik word as vir die MCP metode. Maar waar die MCP net tanniene ontleed, ontleed die BSA metode tanniene, monomere (MP), klein polimeriese pigmente (SPP) en groot polimeriese pigmente (LPP). Dit help indien daar gekyk wil word na die evolusie van polimeriese pigmente. In hierdie studie is bevind dat daar ‘n redelike korrelasie (R2 – 0.88) tussen die BSA en MCP metode bestaan. Die herhaalbaarheid van die metodes het redelike kontroversie veroorsaak, waar sommige navorsers bevind het dat die BSA metode nie so herhaalbaar is soos eers bevind is nie. Die MCP metode het ’n praktiese voordeel aangesien dit in die helfde van die tyd van die BSA metode uitgevoer kan word. Dit het ‘n groot impak indien ‘n groot hoeveelheid monsters ontleed moet word. Die fenoliese samestelling en mondgevoel word sterk beïnvloed deur die klimatologiese area. In die warmer area was die effek van tannien konsentrasie op mondgevoel kleiner as in die koeler area. Die wyn van ryper druiwe het meer harder, verdowingseffek en bitter nasmaak gehad as by die wyn van groener druiwe. Die ensiem behandeling was meer geassossieerd met droë mond gevoel.
