The objective of this work was to investigate the effect of nitric oxide (NO) like protective agent in sesame seeds submitted to different stresses. Therefore, two experiments were conducted. In the first one, sesame seeds were placed in soaked substrate with water or with different concentrations of cadmium chloride and cadmium chloride plus sodium nitroprusside (SNP) in the following treatments: 1) water (control), 2) +200 μM of SNP, 3) 800 μM of CdCl2, 4) 800 μM of CdCl2 +200 μM of SNP, 5) 600 μM of CdCl2, 6) 600 μM of CdCl2 +200 μM of SNP, 7) 400 μM of CdCl2 and 8) 400 μM of CdCl2 +200 μM of SNP. Were done the following determinations: germination (G), first count of germination (PC), speed germination index (IVG), hipocotyl length (CH), radicle length (CR), dry mass of hipocotyl (MSH) and dry mass of radicle (DMR) and imbibition curve. It were quantified the activity of antioxidative enzymes, superoxide dismutase (SOD), catalase (CAT), ascorbate peroxidase (APX) and total peroxidase (POX). It was adopted the completely randomized design with five replicates. The data were submitted to variance analysis and the means obtained for treatments were compared by Tukey test at 5% of significance. The means obtained in treatments with and without SNP were compared by F test at 5% and the times of imbibition by Tukey test at 5%. For the realization of imbibition curve was realized a polynomial regression fit 3. In the second experiment, sesame seeds were placed in soaked substrate with water or with different concentrations of PEG 6000 and PEG 6000 plus SNP in the following treatments: 1) water (control), 2) +200 μM of SNP, 3) 0,1MPa, 4) -0,1MPa +200 μM of SNP, 5) - 0,2MPa, 6) -0,2MPa +200 μM of SNP, 7) -0,3MPa and 8) -0,3MPa, +200 μM of SNP. The same determinations done in experiment one, was made in the experiment two. Both stress caused the reduction of germination and vigor, being the highest concentrations, the most harmful to sesame seeds. NO was able to minimize the damage caused by the stresses, reversing partially all the biochemical parameters evaluated, what provides higher and faster germination, beyond the higher initial development of xiii seedlings. Cadmium and PEG 6000 provoked less absorption and less gain of mass in higher concentrations in relation to water, and an extension of Phase II. In relation to the antioxydative system, there was an increaseof enzymatic activity in period of 0 to 24 houras, showing the organization of the antioxidant system with the sesame seeds over time. Was observed that in higher potentials and in solutions more concentrated, the activity enzymatic was higher, what suggest detoxifying activity of these molecules in seeds submitted to water deficit and solution of cadmium more strict. The use of SNP, increased the activity of antioxidative enzymes, evidencing an efficient of elimination of EROs formed. / Sementes estão frequentemente expostas a diversos estresses bióticos e abióticos que prejudicam o seu crescimento, desenvolvimento e produtividade. O objetivo desse trabalho foi investigar o efeito do óxido nítrico (ON) como agente protetor em sementes de gergelim (Sesamum indicum L.) submetidas à diferentes estresses. Para tanto, foram conduzidos dois experimentos. No primeiro, as sementes foram semeadas em substrato umedecido com água ou com diferentes concentrações de cloreto de cádmio (CdCl2) e cloreto de cádmio acrescido de nitroprussiato de sódio (SNP) nos seguintes tratamentos: 1) água (controle), 2) +200 μM de SNP, 3) 800 μM de CdCl2, 4) 800 μM de CdCl2 +200 μM de SNP, 5) 600 μM de CdCl2, 6) 600 μM de CdCl2 +200 μM de SNP, 7) 400 μM de CdCl2 e 8) 400 μM de CdCl2 +200 μM de SNP. As seguintes determinações foram feitas: germinação (G), primeira contagem de germinação (PC), índice de velocidade de germinação (IVG), comprimento de hipocótilo (CH) e radícula (CR), massa seca de hipocótilo (MSH) e radícula (MSR), curva de embebiçao, além da quantificação da atividade das enzimas antioxidativas, superóxido dismutase (SOD), catalase (CAT), ascorbato peroxidase (APX) e peroxidases totais (POX). O delineamento estatístico utilizado foi o inteiramente casualizado com cinco repetições. Os dados foram submetidos à análise de variância (ANOVA) e as médias obtidas para os tratamentos foram comparadas pelo teste Tukey a 5% de significância. As médias obtidas nos tratamentos com e sem SNP foram comparadas pelo teste F a 5% de significância e os tempos de embebição na análise das atividades das enzimas, também pelo teste Tukey a 5%. Para a realização da curva de embebição realizou-se regressão com ajuste polinomial de grau 3. No segundo experimento, as sementes de gergelim foram semeadas em substrato umedecido com água ou em diferentes concentrações de polietileno glicol (PEG 6000) e PEG 6000 acrescido de SNP nos seguintes tratamentos: 1) água (controle), 2) +200 μM de SNP, 3) 0,1Mpa, 4) -0,1MPa +200 μM de SNP, 5) - 0,2MPa, 6) -0,2MPa +200 μM de SNP, -7) 0,3MPa e 8) -0,3MPa, +200 μM de SNP. As mesmas determinações e análises estatísticas do primeiro experimento foram realizadas. Conclui-se que ambos os estresses causaram a redução da germinação e do vigor, sendo Xi as maiores concentrações as mais prejudiciais às sementes de gergelim. O ON foi capaz de amenizar os danos causados pelos estresses invertendo parcialmente todos os parâmetros fisiológicos avaliados, o que proporcionou maior e mais rápida germinação além de maior desenvolvimento inicial das plântulas. O cádmio e o PEG 6000 provocaram menor absorção e menor ganho de massa nas maiores concentrações em relação à água assim como um prolongamento da Fase II da embebição. Em relação ao sistema antioxidante, houve aumento da atividade enzimática no período de 0 a 24 horas, demonstrando organização do sistema antioxidante nas sementes de gergelim com o decorrer do tempo. Observou-se que nos maiores potenciais e nas soluções mais concentradas, a atividade enzimática foi maior, o que sugere atividade desintoxicante dessas moléculas em sementes submetidas ao déficit hídrico e solução de cádmio mais rigorosa. O uso do SNP, de maneira geral, aumentou a atividade das enzimas antioxidantes, evidenciando um eficiente sistema de eliminação das espécies reativas de oxigênio formadas.

Nanoemulsões são emulsões especiais com glóbulos de tamanho na faixa de 20 a 500 nm. São sistemas que apresentam transparência ou fluidez, favorecendo o aspecto sensorial de espalhabilidade e umectação muito interessantes para produtos cosméticos destinados aos cuidados da pele e produtos para a saúde, atuando como sistemas de liberação de fármacos, além da característica nanométrica, que previne processos de instabilidade física. A queimadura da pele é o quadro resultante da ação direta ou indireta do calor sobre o organismo humano causando inflamação proporcionando intensa ação de radicais livres. A presente pesquisa teve como objetivo desenvolver nanoemulsões à base de óleo de gergelim aditivadas de óleo de framboesa para aplicação em queimaduras de pele. Os óleos de gergelim e framboesa apresentaram atividade antioxidante nos testes de DPPH e Xantina Oxidase. No desenvolvimento das dispersões o método de preparo por baixa energia, fundamentado na mudança de curvatura da molécula do tensoativo, foi induzido pela temperatura de inversão de fase. O resultado desta pesquisa foram duas nanoemulsões utilizando dois tensoativos derivados do óleo de rícino com diferentes etoxilações. A avaliação da estabilidade foi realizada pelo cálculo do principal mecanismo de instabilidade das nanoemulsões, chamado Ostwald ripening. O projeto também propõe uma nova metodologia para a avaliação in vitro- HET-CAM e, in vivo do eritema através da medida pelo Chroma Meter e transformação em escala dos resultados. A pesquisa demonstrou que as nanoemulsões não preveniram o processo de queimadura da pele dos camundongos hairless, mas sugere uma tendência mais rápida à redução do eritema. / Nanoemulsions are special systems with uniform and extremely small droplet size, in the range of 20500 nm. Due to their characteristic size, some nanoemulsions are optically transparent or translucent and have kinetic stability, low viscosity, that result in excellent espalhability and humectation making them of increasing use in many applications, for example, in cosmetics as personal care formulations, and in pharmaceutical field as drug delivery systems. The skin burn is the result of direct or indirect hot effect over human organism. This situation provokes an inflammatory process with intense action of reactive oxygen species. The aim of this research was the development of sesame indicum oil-based nanoemulsions with raspberry oil for application in skin burn. The sesame and raspberry oils presented antioxidant activity in DPPH test and Oxidase Xantine system. The dispersions were obtained by low energy method, that was based on curvature change of surfactant molecule induced for temperature. It was obtained two nanoemulsions using different surfactant castor oil derivative etoxilated. The stability assessement was realized with the calculation of main instability mechanism, the Oswald ripening rate. This Project has been carried out with an in vitro method to evaluate the toxicity (HET-CAM) and antioxidant activity (DPPH and Xantine Oxidase), and in vivo erytem evaluation with Chroma Meter. The results of in vivo test was transformed in a score, that reflected the level of erythema. This work demonstred nanoemulsion hasn´t prevented the skin burn in hairless rats, but there was a tendency to decrease the erythema.

Constituting Sesame : a minor field study of a cross-cultural cooperation

Internationally, universities are undergoing renewal because of technological and social changes that both increases the importance of open and flexible learning, as well as makes it practicable. Net-based education makes it possible to create courses where students can collaborate and share knowledge globally. Collaborative Learning in Virtual Communities, with the working title Sesame, is a cooperative project between Blekinge Institute of Technology, BTH, and University of Pretoria, UP that has intention to provide a course like this. The aim of this collaboration is to initiate research in, as well as to test, and evaluate methods for net-based collaborative learning to see how this can provide new perspectives for students and lecturers in both countries. One key concept in the project is ?internationalisation at home? and this means interaction and knowledge sharing between people from different countries, and cultures, without them having to physically leave their country. In this thesis we describe the phase of constituting Sesame focusing on the cooperation between the involved parties from the two countries. / Sesame är ett projekt mellan Blekinge Tekniska Högskola och Universitet i Pretoria som syftar till att skapa kurser där studenter från de båda universiteten ska samarbeta och därigenom utbyta kunskaper och erfarenheter. Målet med projektet är att initiera forskning, såväl som att testa och utvärdera metoder för kollaborativt lärande. Ett nyckelbegrepp inom Sesame är ? internationalisation at home?, vilket betyder möjlighet till interaktion och kunskapsutbyte mellan människor i olika länder och kulturer, utan att de fysiskt måste lämna sina länder. I den här rapporten beskriver vi planeringsfasen av Sesame där vi fokuserar på samarbetet mellan deltagarna i Sverige och Sydafrika.

Comparative study of open source and dot NET environments for ontology development.

However, none of these studies have included the dot NET-based semantic web platforms in the empirical investigations. This study conducted a comparative analysis of open-source and dot NET-based semantic web platforms for ontologies development. Two popular dot NET-based semantic web platforms, namely, SemWeb.NET and dotNetRDF were analyzed and compared against open-source environments including Jena Application Programming Interface (API), Protégé and RDF4J also known as Sesame Software Development Kit (SDK). Various metrics such as storage mode, query support, consistency checking, interoperability with other tools, and many more were used to compare two categories of platforms. Five ontologies of different sizes are used in the experiments. The experimental results showed that the open-source platforms provide more facilities for creating, storing and processing ontologies compared to the dot NET-based tools. Furthermore, the experiments revealed that Protégé and RDF4J open-source and dotNetRDF platforms provide both graphical user interface (GUI) and command line interface for ontologies processing, whereas, Jena open-source and SemWeb.NET are command line platforms. Moreover, the results showed that the open-source platforms are capable of processing multiple ontologies’ files formats including Resource Description Framework (RDF) and Ontology Web Language (OWL) formats, whereas, the dot NET-based tools only process RDF ontologies. Finally, the experiment results indicate that the dot NET-based platforms have limited memory size as they failed to load and query large ontologies compared to open-source environments.

Neste contexto objetivou-se, com este trabalho, a elaboração de misturas especiais de óleos vegetais para fins alimentícios, com base nos óleos de amendoim, gergelim, maracujá e soja. Os óleos de amendoim e gergelim como alternativa por serem culturas produzidas no Brasil, constituem uma fonte de ácidos graxos e antioxidantes tão pouco explorados na alimentação dos brasileiros, tal como o óleo de maracujá como co-produto, descartado pela indústria de suco e ainda o óleo de soja cuja matéria-prima abundante, tornam o produto acessível aos consumidores de todas as classes sociais. Os óleos de amendoim e gergelim foram extraídos de sementes das cultivares BRS-Havana e BRS-Seda, respectivamente, cedidas pela Embrapa Algodão enquanto o óleo de maracujá foi adquirido no comércio de São Paulo e o de soja no de Campina Grande. As misturas foram elaboradas com base em uma matriz de planejamento experimental e, em seguida, submetidas às análises de composição de ácidos graxos e ensaios físico-químicas (umidade, índice de acidez, índice de iodo, índice de refração, densidade e viscosidade) segundo a metodologia do IAL (2008). As medidas obtidas foram utilizadas para estudar e otimizar a elaboração das misturas de óleos vegetais e como variáveis para predição por espectrometria no infravermelho próximo (NIR) foram utilizados os mínimos quadrados parciais (PLS) como metodologia de calibração multivariada. Os resultados foram submetidos à análise de variância (ANOVA), à análise de variância multivariada (MANOVA), à análise de correlação canónica e à análise de correlação de Pearson, pelo software SAS 9.1.3; na análise de componentes principais (PCA) e regressão por mínimos quadrados parciais (PLS) utilizou-se o software Uncrambler 9.8®. Concluiu-se que a mistura equivalente a 40% de óleo de amendoim, 20% de óleo de gergelim, 20% de óleo de maracujá e 20% de óleo de soja apresentou-se como a melhor mistura para fins alimentares, com base na composição de ácidos graxos. Os métodos de predição por NIR foram eficazes para a estimativa não destrutiva, rápida, de baixo custo e direta, dos ácidos graxos palmítico, esteárico, oleico, linoleico, araquídico e linolênico e para as medidas físico-químicas de densidade, índice de refração, viscosidade, índice de acidez e índice de iodo. / Brazil has great potential to produce vegetable oils as a source of raw materiais for food and energy. But in the last five years has been the largest importer of olive oil in the worid due to its health benefits and wide divulgation. In this context, the aim of this work was the development of special blends of vegetable oils as an alternative to olive oils based on peanut, sesame, soy and passion fruit. The peanut and sesame oils because they are a source of fatty acids and antioxidants in the diet of little explored in Brazil, passion fruit oil as co-product of the juice industry, and soybean oil abundant raw material which makes the product available to consumers of ali social classes. The peanut and sesame oils were extracted from seeds of BRS-Seda and BRS-Havana, while passion fruit oil was purchased commercially of city São Paulo-SP and trade soybean in the city of Campina Grande-PB. The mixtures were prepared based on an array of experimental design and then submitted to analysis of fatty acid composition and physico-chemical (moisture, acid value, iodine value, refractive index, density and viscosity). Moreover, were used as reference variables for prediction via infrared spectroscopy (NIR) using the partial least squares (PLS) and multivariate calibration methodology. The results were subjected to analysis of variance (ANOVA), multivariate analysis of variance (MANOVA), canonical correlation analysis and Pearson correlation analysis by SAS 9.1.3 software, the principal component analysis (PCA) and Partial least squares regression (PLS) was used Uncrambler 9.8 ® softwarlt was concluded that the mixture equivalent to 40% peanut oil, 20% sesame oil, 20% passion fruit oil, and 20% soybean oil was presented as the best mix for food composition based on fatty acids. The prediction by NIR methods were effective for estimating non-destructive, rapid, inexpensive and direct fatty acids palmitic, stearic, oleie, linoleic, linolenic and arachidic and physico-chemical measurements of density, refractive index, viscosity, acid value and iodine value.
