Diversidade e aspectos bioecológicos de simulídeos (Diptera: Simuliidae) que ocorrem nos municípios adjacentes a construção da usina Hidrelétrica de Peixe-Angical, TO, BrasilAna Margarida Ribeiro do Amaral 08 October 2012 (has links)
O objetivo desta Tese é realizar o levantamento taxonômico e avaliar a dinâmica
populacional de Simuliidae em localidades sob influência do Aproveitamento Hidrelétrico de
Peixe Angical, TO. Os simulídeos possuem abrangente distribuição geográfica, e os estágios
imaturos utilizam ambientes lóticos como sítios de criação. Algumas espécies podem atuar
como vetores de vírus, protozoários e helmintos, o que confere ao grupo importância médica e
veterinária. O hábito hematofágico das fêmeas de simulídeos pode acarretar sérios prejuízos
ao turismo; ocasiona baixa no rendimento escolar; e na agropecuária dificulta a execução do
trabalho o que reduz a produtividade. Durante a construção de grandes empreendimentos
ocorre em pouco tempo à introdução de contingente populacional com drástica transformação
do meio. A intervenção do homem sobre os ecossistemas e o crescimento desordenado pode
provocar desequilíbrio ecológico que propicia a proliferação de espécimes vetores com
consequentes problemas médico sanitários. A maior parte dos trabalhos realizados com
insetos vetores em áreas sob influência da construção de hidrelétricas se refere aos culicídeos.
O estudo dos aspectos taxonômicos permitirá o levantamento da biodiversidade e o
diferencial deste projeto está no estabelecimento da sazonalidade e dinâmica das populações
de imaturos e adultos de simulídeos. As amostras foram obtidas em áreas de influência direta
e indireta da UHE Peixe no rio Tocantins, em 12 pontos diferentes de coleta, nos municípios
deJaú do Tocantins, Peixe, Palmeirópolis, Paranã e São Salvador do Tocantins. Foram
realizadas bimestralmente de 2004 a 2007, um total de 24 campanhas para coleta em
criadouros pré-selecionados, que acompanharam todas as fases de construção início das
obras, formação do lago, funcionamento da Usina. OS dados abióticos foram aferidos, e os
imaturos removidos do substrato manualmente por 10 minutos e posteriormente preparados
para eclosão dos adultos. Parte do material foi identificado no Laboratório de Simulídeos e
Oncocercose do Instituto Oswaldo Cruz, onde foram verificados novos registros específicos
para a ocorrência de Simuliidae em Tocantins, além do assinalamento de espécies
antropofílicas e/ou vetores de Onchocerca volvulus. Nas áreas usadas para a formação do
lago houve desaparecimento de criadouros. O desmatamento ocorrido aliado ao vigor dos
simulídeos que conseguem realizar voos de longas distâncias na procura de alimento ou locais
adequados a oviposição devem ter contribuído para a dispersão de espécimes. Há relatos
sobre a da ocorrência de oncocercose na área estudada, um foco foi demarcado na divisa de
Goiás com Tocantins, municípios Paranã e Minaçu investigado a partir de um caso autóctone
de oncocercose. Este estudo é relevante uma vez que o Brasil possui potencial hidroenergético
e prevê a construção de inúmeras hidrelétricas nos próximos anos. É importante estudas as
áreas impactadas, conhecer a sua biodiversidade e os aspectos bioecológicos de Simuliidae no
país. / This project aims to carry out the taxonomic survey and population dynamics of
Simuliidae in locations under the use influence of hydroelectric power station at Peixe
Angical (Tocantins). Immature black-flies use lotic environments as rearing sites. Some
species are vectors of viruses, protozoa or helminths, which assigns them medical as well as
veterinary importance. The blood feeding habits of females affect rural life causing financial
damages to tourism, declining of school performance and reducing of agricultural
productivity. During the construction of large enterprises the need of hiring extra personnel
drastically affects the environment. Human intervention on ecosystems as well as disorderly
population growth can cause ecological imbalance added to significant vector proliferation
causing health as well as sanitary problems. Deforestation and exploitation in the river beds
also affect population of black-flies dynamics influencing their geographical distribution. In
specialized literature, most of the work carried out in hydroelectric construction areas is
referred to culicidae. Studies that mention black-flies in impacted areas by hydroelectric
power stations in Brazil are restricted to taxonomic survey and brief comments on the
formation of lakes with consequent disappearance of breeding sites in lotic environments.
This research work is part of a larger project that studies the black-flies and their local
associated entomological fauna, which will allow us to carry out its biodiversity surveillance.
The differential of this project is to establish the seasonality and population dynamics of
immature as well as adult black-flies. Samples were obtained in areas of direct and indirect
influence of Peixe Hydroelectric power station in 12 different collecting sites in the
municipalities of Peixe, Paranã, Palmeirópolis, Jaú and São Salvador do Tocantins. Were
conducted bimonthly from 2004 to 2007, a total of 24 campaigns, which accompanied
building phases: start of groundwork, lake formation and operation of the plant. All abiotic
data were registered and immature black-flies removed from the substrate for 10 minutes and
prepared for the outbreaking of adults. Part of the material has been identified at the
Simuliidae and Onchocerciasis laboratory at Oswaldo Cruz Institute, where new records of
specific occurrence of Simuliidae in Tocantins have been registered, as well as anthropophilic
species and / or vectors of Onchocerca volvulus. Mansonella ozzardi of vector species were
not collected in the study area. It is expected that the prevalence is higher in the rainy months
of the year. In areas where there was a lake formation there was loss of habitats. Black-flies
can fly long distances in search of food or oviposition sites and that fact, coupled with
deforestation may lead to the dispersal of species. There is a report on the occurrence of an
autochthonous case of onchocerciasis in Paranã and Minaçu, municipality of Goiás and
boarding area of the studied sites. This study is relevant to knowledge of biodiversity and
bioecological aspects of Simuliidae in Brazil.

volvulus Leuckart, which is the ethiologic agent of oncocerciasis in the Amazonian focus of
the disease, located in the northern portion of South América. The objectives of this study
were to analyze different populations of this species using polythene chromosomes and
mitochondrial gene sequences of the COI region to evaluate the relationships among these
populations. The specimens for the cytotaxonomic study were collected in the Brazilian states
of Amazonas, São Paulo and Goiás (Rio Ponte de Pedra) and for the molecular study the
specimens were collected in the states of Amazonas, Amapá, Goiás (Rios Ponte de Pedra and
Verdão), Maranhão and Roraima (one population from the Homoxi oncocerciasis focus and
two from sites outside of the focus: Caroebe and Surumu). The polythene chromosomes from
the analyzed populations were compared with a standard map that had been previously
established for this species. The cytological analysis indicates the presence of polymorphic
inversion in the Amazonas and São Paulo populations; the population from Goiás (Rio Ponte
de Pedras) had two fixed inversions. Comparing 702 bp from the COI mithochondrial gene in
the nine studied populations, a considerable number of polymorphic sites were observed,
thereby allowing population and phylogeographic analyses to be undertaken. A representative
cladogram was constructed to analyse the relationships among the observed haplotypes. The
cladogram suggests that the populations from the northern region of Brazil (Amapá,
Amazonas and the Caroebe site in Roraima) share one haplotype, while the populations from
the Amazonian focus (Homoxi site in Roraima) is most closely related to the population from
the Surumu site in Roraima, as these populations share a common haplotype. The populations
from São Paulo and Goiás, Rio Verdão (from the southeast and centro-west regions of Brazil,
respectively) have high haplotypic variability, each specimen being represented by one
haplotype. The population from the Rio Ponte de Pedra site in Goiás did not share haplotypes
with any of the other populations studied, including the population from the Rio Verdão site
in Goiás, located approximately 50 Km away in the same hydrological basin. The populations
from Amazonas and the Caroebe site in Roraima had negative values of F ST, suggesting that
they exchange alleles freely and do not show any genetic structure. The population from
Maranhão is well sructured in relation to the others with a low number of migrants; this
population appears to be derived from the population in São Paulo. The NJ phylogeographic
tree constructed using the F ST values indicates the division of the analyzed populations into
two groups separated by the Amazon River. The northern group is represented by the
populations in Amapá, Roraima (Surumu, Homoxi and Caroebe) and Amazonas, while the
southern group is represented by Maranhão, the Rio Verdão site in Goiás, and São Paulo. The
results obtained by cytological and molecular techniques suggested that the S. guianense s.l.
populations analyzed, in general, do not constitute a species complex, as has been suggested
by some authors, even though the populations had high genetic variability. The only exception
is the population from Goiás (Rio Ponte de Pedra), which probably represents a species that is
new to science. / A espécie Simulium guianense s.l. Wise é incriminada como principal vetor de Onchocerca
volvulus Leuckart, causadora da oncocercose no foco Amazônico, localizado no norte da
América do Sul. Os objetivos do presente estudo foram analisar diferentes populações dessa
espécie utilizando cromossomos politênicos e seqüências da região do gene mitocondrial COI,
para avaliar as relações entre essas diferentes populações. Para a análise cromossômica, os
espécimes foram coletados nos estados do Amazonas, São Paulo e Goiás (Rio Ponte de Pedra)
e para a análise molecular foram coletados nos estados do Amazonas, Amapá, Goiás (Rios
Ponte de Pedra e Verdão), Maranhão e Roraima (uma população na área do foco de
oncocercose-Homoxi e, duas fora da área do foco, Caroebe e Surumu). Os cromossomos
politênicos das populações analisadas foram comparados com mapas estabelecidos
anteriormente para essa espécie. As análises citológicas do padrão de bandas dos
cromossomos politênicos mostraram inversões polimórficas, na condição heterozigota nas
populações do Amazonas e São Paulo, e a de Goiás (Rio Ponte de Pedras) apresentaram duas
inversões fixas. Comparando 702 pb da região do gene mitocondrial COI das nove
populações estudadas, um valor considerável de sítios polimórficos foram observados,
permitindo estudos populacionais e filogeográficos. Um cladograma representativo foi
construído a fim de relacionar os haplótipos detectados, permitindo observar que as
populações do norte, Amapá, Amazonas e RR-Caroebe compartilham um mesmo haplótipo,
enquanto a área do foco Amazônico (RR-Homoxi) está mais relacionada com as populações
de RR-Surumu, compartilhando um haplótipo em comum. As populações de São Paulo e
Goiás, Rio Verdão (das regiões sudeste e centro-oeste, respectivamente) apresentam elevada
variabilidade haplotípica, onde cada indivíduo é representado por um haplótipo. A população
do Rio Ponte de Pedra, de Goiás não compartilhou haplótipos com as outras populações
estudadas, nem mesmo com a do Rio Verdão, também em Goiás, localizada a
aproximadamente 50 Km de distância. As populações do Amazonas e RR-Caroebe
apresentaram valores negativos de F ST, sugerindo que elas, trocam alelos livremente,
representando uma população sem estrutura genética. A população do Maranhão, que se
mostrou bem estruturada em relação às outras, tem baixo número de migrantes, mas parece
ser derivada da população de São Paulo. Usando os valores de F ST uma árvore filogenética de
NJ foi gerada e pode-se observar uma separação entre os grupos ao norte da calha do Rio
Amazonas, representados por indivíduos do Amapá, RR (Surumu, Homoxi e Caroebe) e
Amazonas, e outro ao sul da calha do Rio Amazonas (Maranhão, GO-Rio Verdão e São
Paulo). Os resultados obtidos, utilizando técnicas citogenéticas e moleculares, sugerem que as
populações de S. guianense s.l. analisadas, no geral, não fazem parte de um complexo de
espécie, como sugerido por alguns autores, apesar de apresentar acentuada variabilidade
genética. A exceção é a população de Goiás (Rio Ponte de Pedra) que, provavelmente,
representa uma nova espécie para a Ciência.

Molecular Evidence for Vector Implication of Onchocerca lupi in Los Angeles County, CABolcen, Shanna June 27 March 2014 (has links)
Onchocerca is a genus of roundworm most commonly associated with the human infection onchocerciasis, or river blindness. While typically a zoonotic infection of ungulate populations, canine cases (Onchocerca lupi) have been identified in the United States and Greece. In 2012, Los Angeles County, Veterinary Public Health Program identified 3 cases of Onchocerca spp. infections in domestic canines. Samples from the ensuing blackfly collections were sent to the Global Health Infectious Disease Research Unnasch Laboratory for parasite isolation and vector species identification. Species-specific primers were designed and optimized for O. lupi using a non- specific cytochrome oxidase (COI) gene target (689bp) previously utilized in Onchocerca identification as a base. A secondary, nested PCR primer set (115bp) was additionally designed to aid in the screening process. Extracted DNA samples from the collected blackflies were screened for the presence of the parasite and positive samples were further tested against the larger fragment for confirmation. The primers designed for the study were determined to be specific to O. lupi and not cross-reactive with other Onchocerca species or Dirofilaria immitis. Of the 213 blackflies screened, 6 samples tested positive for parasite presence. The blackfly species was identified as Simulium spp. The presence of the O. lupi parasite in the black fly population implicates that this is the likely vector in the LA County cases. While the blackfly identification was unable to be determined to the species level, the identification of Simulium spp. confirms the typically suspected vector in Onchocerca infections. Concerns of cross-species transmission make O. lupi an important parasite for public health study.
