O objetivo deste trabalho foi analisar o perfil de resistência antimicrobiana e genotitipificar os genes da coagulase e espécie-específicos dos S. aureus nas propriedades do Distrito federal e Entorno. Foram analisadas 47 amostras de leite de 116 animais de diversas raças que apresentaram um escore de 3+ no teste do Californian Mastit Test. Foi incorporado mais 28 amostras do banco de germoplasma do Laboratório de Microbiologia (MicroMedVet) da Universidade de Brasília aos testes, totalizando 75 amostras. O teste do perfil de resistência antimicrobiana demostrou que a penicilina (71,2%), ampicilina (52,9%) e enrofloxacina (39,9%) foram os antibióticos que mais apresentaram resistência. Os maiores índices de sensibilidades antimicrobianas foram visto na gentamicina (79,8%), neomicina (76,9%), norfloxacina (76,9%), cefalexina (74,1%), cefazolina (74,1%), lincomicina (74,1%), oxacilina (74,1%), tetraciclina (74,1%), tobramicina (74,1%), ceftiofur (71,2%). A sensibilidade da enrofloxacina apresentou uma discrepância muito elevada quando comparada a estudos anteriores, pois no presente trabalho seu índice foi de apenas 51,3%. Os genes da coagulase (CoA) obtiveram quatro diferentes tamanhos de bandas do produto da PCR nas amostras testadas. Foram elas 490bp (2,66%), 570bp (33,35%) 680bp (41,33%), e 780bp (22,66%). O gene de espécie-específico amplificou todas as 75 amostras com fragmentos de 1153bp e mostrou ser uma ferramenta útil na identificação rápida de S. aureus. Os resultados apresentados nesse estudo demostra que o perfil de resistência antimicrobiana vem se tornando algo mundial e que técnicas simples podem facilitar a sua identificação e também mapear o polimorfismo ocorrido nos genes da coagulase. ______________________________________________________________________________ ABSTRACT / Staphylococcus aureus is one of the most commonly microorganisms isolated from bovine subclinical mastitis, it is very damaging to the producing milk tissues due to the release of toxins and difficulty of treatment. The objective of this study was to analyze the antimicrobial resistance profile and genotyping genes of coagulase and species-specific of S. aureus in the properties of the Distrito Federal and Entorno. We analyzed 47 milk samples from 116 animals of several breeds that showed score of 3+ in the Californian Mastit Test. Another 28 samples from the germplasm bank of the University of Brasília Microbiology Laboratory (MicroMedVet) were incorporated to the tests, totaling 75 samples. Testing of the antimicrobial resistance profile showed that penicillin (71.2%), ampicillin (52.9%) and enrofloxacin (39.9%) were the antibiotics that showed more resistance. The sensitivity of enrofloxacin showed a very high discrepancy when compared to previous works, at the present study its index was only 51.3%. Coagulase genes (CoA) had four different PCR product bands sizes in the tested samples. These were 490bp (2.66%), 570bp (33.35%) 680bp (41.33%), and 780bp (22.66%). The species-specific gene amplified all the 75 samples with fragments of 1153bp and proved to be a useful tool for S. aureus rapid identification. The results presented in this study shows that the antimicrobial resistance profile is becoming worldwide and that simple techniques can facilitate their identification and also map the polymorphism occurred in the coagulase genes.

Embora seja extremamente eficaz na proteção contra intempéries do ambiente externo, infecções na pele são comuns, podendo ser causadas por bactérias, fungos ou vírus. A principal bactéria associada à infecção cutânea consiste na Staphylococcus aureus. Embora a maioria das infecções possa ser facilmente tratada, outras não apresentam a mesma facilidade. Isto tem sido observado especialmente em infecções adquiridas em feridas cirúrgicas, que são o tipo de infecção hospitalar mais comum e uma importante causa de morbidade e mortalidade. Essas infecções cada vez mais vêm sendo ocasionadas por patógenos resistentes aos tratamentos convencionais enfatizando a importância de se identificar novas terapias antimicrobianas mais eficazes. Peptídeos antimicrobianos (PAMs) têm emergido como um novo grupo promissor para serem avaliados na intervenção terapêutica de doenças infecciosas. Dentre os PAMs se destacam os ciclotídeos que são peptídeos cíclicos isolados inicialmente de plantas que possuem em sua estrutura seis resíduos de cisteína altamente conservados, que formam três pontes de dissulfeto interligadas. Essa estrutura faz com que estes peptídeos sejam altamente estáveis e tem atraído um interesse como modelos baseados em peptídeos para aplicações de concepção de drogas. Aqui os ciclotídeos CyO2, KB1 e KB2 foram avaliados com o objetivo de caracterizar suas atividades citotóxica, imunomodulatória e antiestafilocócica in vitro e in vivo em um modelo de infecção subcutânea. Nos ensaios in vitro foi observado que os ciclotídeos não apresentaram atividade citotóxica para os monócitos RAW 264.7 e apresentaram atividade antiestafilocócica variando de forte (CyO2 e KB2) a moderada (KB1). Nos ensaios de avaliação da atividade imunomodulatória, a citocina TNF-α apresentou variação significativa somente nos grupos tratados com CyO2, onde os níveis desta citocina foram reduzidos. Nos ensaios in vivo, os ciclotídeos reduziram significativamente a carga bacteriana total corroborando com a atividade antiestafilocócica in vitro. Além disso, o ciclotídeo KB2 induziu a um aumento na secreção de TNF local na maior concentração testada (3,0 mg.kg-1). Em conclusão, os ciclotídeos CyO2, KB1 e KB2 podem constituir uma nova alternativa terapêutica para o tratamento local de infecções, sendo necessários mais estudos para avaliar a aplicabilidade destes PAMs. _______________________________________________________________________________________ ABSTRACT / The skin is the largest and the most exposed organ of the human body. While it is extremely effective in protecting against external environment setbacks, skin infections are common and may be caused by bacteria, fungi or virus. The main bacterium associated with skin infection is Staphylococcus aureus. Although the majority of infections can be easily treated, others did not demonstrate the same facility. This could occurs, especially in infections acquired in surgical wounds, which are the most common hospital-acquired infections and are major cause of morbidity and mortality. These infections are increasingly caused by pathogens resistant to conventional treatments, which emphasizes the importance of identifying new and more effective antimicrobial therapies. Antimicrobial peptides (AMPs) have emerged as a promising new therapy to be evaluated in the treatment of infectious diseases. Among the AMPs, the cyclotides have been focused, being cyclic peptides originally isolated from plants sources, showing six highly conservated cysteine residues in their structure, which form three interconnected disulfide bonds. This structure produced highly stable peptides and has attracted interest as a peptide-based model for drugs designing. Here the cyclotides CyO2, KB1 and KB2 were evaluated in order to characterize their cytotoxic, immunomodulatory and antistaphylococcal activities in vitro and in vivo by using in a model of subcutaneous infection. In vitro essays showed no cytotoxicity activity to RAW 264.7 monocytes with all cyclotides. Otherwise antistaphylococcal activities ranging from strong (CyO2 and KB2) to moderate (KB1) were also observed. In trials evaluating the immunomodulatory activity, the cytokine TNF-α showed a significant variation only in groups treated with CyO2, where cytokine levels were reduced. At in vivo assays, the cyclotides significantly reduced the total bacterial load corroborating with the in vitro antistaphylococcal activity. Furthermore, the KB2 cyclotide induced an increase in local TNF secretion at the highest concentration tested (3.0 mg.kg-1). In conclusion, the cyclotides CyO2, KB1 and KB2 could represent a novel therapeutic approach for the local treatment of infections, more studies being needed to evaluate the applicability of these AMPs.

O objetivo desse trabalho foi detectar a presença dos genes de enterotoxinas e da toxina da Síndrome do Choque Tóxico dos Staphylococcus aureus isolados do leite de animais em mastite clínica. Foram analisadas 75 amostras de S. aureus do banco de germoplasma do Laboratório de Microbiologia Médica Veterinário da Universidade de Brasília. As amplificações obtidas apresentaram valores maiores que o esperado para os genes de enterotoxinas. Foram identificados entre os isolados de S. aureus um percentual de 10,68% de amplificações com utilização de primers isoladamente (uniplex) e não houve amplificações em combinação com diferentes primers (multiplex). Analisando a PCR-uniplex, houve maior número de amplificações da enterotoxina SEA, presente em 8% dos isolados. Os genes SEB e SEC, foram detectados em 1,34% das amostras e os genes SED e TSST-1 não apresentaram nenhuma amplificação nesse estudo. O tamanho do fragmento para SEA foi de 1.200 pb em 4 amostras amplificadas, 1 amostra com 800 pb e outra com 620 pb. Em relação a SEB e a SEC, houve apenas amplificação de uma de suas amostras, com tamanho da banda de 1.200 pb e 1100 pb respectivamente. O conhecimento do perfil molecular dos S. aureus auxilia em estudos epidemiológicos de infecções intramamárias de rebanhos leiteiros e conhecendo o potencial toxigênico das estirpes de S. aureus, corrobora para prevenir e controlar as infecções nos rebanhos para melhorar a qualidade dos produtos oferecidos à população. _______________________________________________________________________________________________ ABSTRACT / Staphylococcus aureus is the most frequently isolated organism in cases of bovine mastitis and stands by having resistance to conventional antibiotic treatments and due to its potential to produce toxins that cause diseases. The aim of this study was to detect the presence of enterotoxin genes and the Toxic Shock Syndrome toxin gene of Staphylococcus aureus isolated from the milk of animals with clinical mastitis. Seventy-five samples of S. aureus, from the gene bank of the Medical Veterinary Microbiology Laboratory of the University of Brasilia, were analysed. The obtained amplifications showed higher values than expected for the enterotoxin genes. We identified 10.68% amplifications when using singly primers (uniplex), but there were no amplification when combining different primers (multiplex). Analyzing the uniplex PCR, there were more amplifications of the SEA enterotoxin gene (8% of the isolates). The SEB and SEC genes were detected in 1.34% of the samples and the SED and the TSST-1 genes showed no amplification in this study. The size of the SEA fragment was 1.200 pb in 4 amplified samples, 800 pb in 1 sample and 620 pb in another one. Regarding the SEB and the SEC genes, there was the amplification of only one of the samples, that had a band size of 1.200 pb and 1100 bp, respectively. Knowledge about the molecular profile of S. aureus assists in epidemiological studies about mammary infections of dairy herds. Moreover, knowledge about the toxigenic potential of S. aureus strains helps to prevent and control infections in herds in order to improve the quality of the products offered to the population.

Assim, dividiram-se as amostras em quatro ensaios: 1 e 2, com leite desnatado e concentrações de 34.7 mg L-1 e 44.8 mg L-1 de ozônio, respectivamente; e ensaio 3 e 4, com leite integral (34.7 mg L-1 e 44.8 mg L-1 de ozônio, respectivamente). Os tempos de ozonização em todos os ensaios foram de 5 (T1), 10 (T2), 15 (T3), 20 (T4) e 25 (T5) minutos, com três repetições. Cada tratamento foi repetido três vezes. As contagens bacterianas foram feitas imediatamente antes e após o processo de ozonização das amostras, em agar Baird Parker. Nos tratamentos 1 e 2, as maiores reduções foram de 0.42 log10 UFC/mL, enquanto nos tratamentos 3 e 4, foram de 0.19 e 0.21 log10 UFC/mL, respectivamente. Os resultados de todos os ensaios mostram que, em T1, não houve redução da carga microbiana, enquanto que a partir de T2, há uma crescente queda da microbiota, que é acentuada nos tempos T4 e T5. Tanto para leite desnatado quanto para o integral, nas concentrações de 34,7 e 44,8 mg L-1, a partir dos 20 minutos de ozonização, as reduções foram significativamente maiores. Os resultados do trabalho confirmam a eficácia do ozônio sobre o patógeno bacteriano Staphylococcus aureus inseridos em leite fluido e que os lipídios do leite exercem influência sobre tal eficiência bactericida. ______________________________________________________________________________ ABSTRACT / In order to evaluate the efficiency of the action of ozonation of milk on the reduction in bacterial counts of Staphylococcus aureus, an experiment was undertaken in which milk samples inoculated with the microorganism were subjected to ozonation with different binomials time / concentration of ozone. Thus, the samples were divided in four assays: 1 and 2, with skim milk, and concentrations of 34.7 mg L -1 and 44.8 mg L-1 of ozone, respectively, and assay 3 and 4, with whole milk (34.7 mg L -1 and 44.8 mg L- 1 of ozone, respectively). The time of ozonation in all assays were 5 (T1), 10 (T2), 15 (T3), 20 (T4) and 25 (T5) minutes. Each treatment was replicated three times. The bacterial counts were made immediately before and after the ozonation process, plating the samples on Baird Parker agar. In treatments 1 and 2, the greatest reductions were 0.42 log10 CFU / mL, while in treatments 3 and 4 were 0.19 and 0.21 log10 CFU / mL, respectively. The results of all the tests show that, on T1, there was no reduction in microbial load, whereas, on T2, there is a sharp decrease of the microbiota, which is remarkable in the times T4 and T5. In both skim and whole milk, at ozone concentrations of 34.7 and 44.8 mg L-1, after 20 minutes ozonation, the reductions were significantly greater. The findings confirm the effectiveness of ozone on the bacterial pathogen Staphylococcus aureus inoculated into fluid milk. In addition, the milk lipids have an influence on the ozonation’s effects.
