The diploma thesis aims the attention on reduction diets at pupils at choosing school. The teoretical part describe the development of the pubety age, the accurate regime according to the pupils' proces, the types of reduction diets, the alternative food and the following motion aktivity. The thesis deals with the illneses, which can be caused by reduction diets, which are kept quite often. At the end of the teoretical part it is manchened the under topic the reduction diet, which is involved in the part Health Education and PE of the topic according to the the Framework Education Programme and the School Education Programme ZŠ Dobiášova. The issue that conception of educational lesson aim on reduction diets, alternative way of eating and the illneses, that can be caused by reduction diets The practical part of thesis contains the amount of pupils in second level of primary school (6. - 9. grade) who keep or kept reduction diet at choosing liberecká school, afterwads there were the conversations with the choosing pupils which lead solving, why pupils want to be on diet and what is the reason to be on diet. The thesis "Service Design of Zátiší Catering Group, a.s." is focused on the management of innovations and service design. The aim is to analyse and identify possible potential and opportunities for improvement of catering services in Zatiší Catering Group, a.s. Content of the thesis includes research of service design and innovation process, study based on application of the design service approaches, introduction of the study outputs, actual project of a new service design and recommendations. Following methods and techniques were used in research: persona, contextual interview, service safari, benchmarking, service blueprints, empathy map, product box. The thesis should contribute to the theoretical determination of service design and its following aplication in the design of a specific service. Final outcome of the study is a service proposal for a canteen in Lycée Français de Prague.

The thesis consists of two parts. The first part introduces the topic of human chemical communication and reviews current evidence on individual variation in human body odour and its perception. This part is framed by sexual selection theory. In the first chapter, the concept of the theory of communication is introduced followed by a discussion on the specifics of chemical communication. Next, the formation of individually specific body odour signatures with reference to skin glands, their volatile products and the subsequent metabolization by skin microflora is described. The next chapters are dedicated to selected interindividual body odour cues such as sex and kin recognition, genetic compatibility in genes of Major Histocompatibility Complex, and health and reproductive status in a mate choice context. Furthermore, interactions between perfumes and body odours are discussed. Finally, methods of body odour sampling are introduced and a rationale behind presenting individual samples or body odour blends is discussed. The second part is comprised of six scientific papers, specifically three reviews and three empirical studies. Title: Active lifestyle of young school-age children Objectives: The aim of this study is to explore, whether the younger school age children are kept to an active and healthy lifestyle. To find out, whether the kids have enough physical activity, how they eat and how much time they spend at the computer or TV. Methods: The quantitative and qualitative methods were used in my research. The qualitative data were collected through surveys and quantitative data were obtained via step counting by using a pedometer. A measurement was used as an empirical method. Measurements were held in the months of May-June and October-November 2015. Results: It was examined a total of 160 children. 14 of them are obese and 9 are overweight. The rest of the students has an optimal weight, eat adequately and has enough physical activity. Obese children spend their free time at the computer or television and have no hobbies or sports activity. Ecological connectivity in the Alpine anthropic matrix. Natural reserves and corridors for the conservation of brown bear in the Alps (ABC - AlpBearConnect)

Large carnivores are among the most challenging species to conserve in our modern and crowded world. Having large spatial requirements and living in low density, they generally require wide and relatively undisturbed areas. In Europe, one of the most anthropized areas of the planet, these needs must be fulfilled in a complex human-dominated landscape. The reintroduced brown bear population living in the Central Alps represents one of the most emblematic examples of a constrained carnivore: despite a steady population increase in the first few years after reintroduction, the population did not substantially expand its range, nor has the Alpine-Dinaric metapopulation been reestablished as envisioned. Although humans have lived in the Alps for centuries, little is known about their impact on the bear population. In other environments humans are known to function as a “super-predator” by changing habitats, competing for space, consuming resources, and harvesting, which alters the ecological niche of animals, especially large carnivores. This dissertation aims to evaluate this phenomenon by assessing the effects of human disturbance on brown bears in the Alps. Anthropogenic disturbance is generally assessed by structural proxies, such as infrastructure and land use, which overlook the impact of human presence. In the first Chapter, we developed the Cumulative Outdoor activity Index (COI) to derive anthropogenic disturbance using crowdsourced data by Strava and validated it with ground truth observations derived from a local camera trapping survey. The intensity of COI provided an effective measure of functional anthropogenic disturbance, and it outperformed all commonly-used proxies of structural disturbance in predicting bear habitat use. When displacement is not an option because of habitat limitations and social mechanisms, bear mobility may clash with human activity. During the moments of lowest mobility, such as resting periods, animals have decreased ability to cope with risky situations, and therefore the selection of suitable resting areas is crucial for the long-term survival of individuals. In the second Chapter, we measured multi-scale response to risk perception (i.e., COI) and resource proximity using bedding sites by GPS radio-collared adult brown bears in the Alps. To map resources across the study area, we developed a GIS-database combining spatial and non-spatial ecological information to map fruit availability. We observed that bears apply a security-food trade-off strategy, avoiding functional anthropogenic disturbance while in proximity to resources. In the third Chapter, we explicitly tested the effect of an abrupt interruption of human mobility during COVID-19 lockdown on bears’ use of ecological corridors. Using bear occurrences reported to local authorities during the recent COVID-19 outbreak, we observed that bears used human-dominated areas more frequently, approached more intensively hot spots for road crossing network, and used areas further from the population core areas more often than previous years, suggesting that connectivity increased with reduced human mobility. In a comparatively human-free system, for the fourth Chapter we used longitudinal morphometric data to analyze drivers of changes in body mass as part of an international collaboration with biologists studying the grizzly bear in the Greater Yellowstone Ecosystem. Specifically, we analyzed changes in lean body mass and fat percentage during years of major ecosystem perturbations. We observed that individual lean body mass during the last two decades was primarily associated with population density, but not body fat percentage, showing density-dependent factors. Our combined findings (Chapters 1-3) showed that brown bears have to adapt their space use, movement, and resource proximity as a result of functional anthropogenic disturbance. In Chapter 4 we explored one effect of unconstrained bear space use on individuals, as manifested through density-dependent effects on body size. In the Alps, however, we found multiple instances of the human-super predator outcompeting bears so as to make density-dependent effects likely less significant as compared to human-caused mortality. These effects could occur in a variety of socio-ecological contexts across Europe, jeopardizing the long-term establishment of both newly reintroduced bear populations, as well as spatially limiting those naturally present in the environment. In response to disturbance, bears have had to reduce their ecological niche in human-dominated landscapes. Allowing humans and bears to coexist in the same landscape is a challenging task, but it is essential for the long-term survival of this newly reintroduced population that are otherwise at risk of extinction.

The next part is devoted to compiled training plan and to the testing of dynamic and endurance skills. Acquired knowledge is discussed and its summary is stated in conclusion.

Tvorba biofilmu u probiotických kultur a možnosti jeho využití ve farmacii / Biofilm formation in probiotic cultures and its application in pharmacy

The work was comprehensively focused on the development of adhesive forms of probiotics in the form of a biofilm on combined carriers with a prebiotic component. The second part dealed with the influence of food on the multiplication and survival of selected types of probiotic bacteria. Subsequently, the effect of individualized probiotic supplements on changes in the human intestinal microbiome was monitored. Suitable adherent probiotic strains for biofilm formation were selected and tested. Methods have been introduced and different variants of carriers for culturing and binding bacteria have been tested. In vitro experiments verified the stability of biofilm stucture and its resistance to low pH, bile and antibiotics in comparison with the planktonic cell form. The antimicrobial effect of probiotic strains in the form of a biofilm was studied. The cultivation of the multispecies biofilm on the combined carrier was optimized and the stability of the biofilm and the final viability of probiotic bacteria were confirmed. Furthermore, the influence of various foods and beverages on the viability of probiotic bacteria was evaluated with emphasis on the simulation of passage through the gastrointestinal tract. Both models, solutions with standardised concentrations of alcohol, sugar, salts, proteins or different pH and different types of real foods and beverages were tested. The effect of food and beverages was tested on monocultures of Lactobacillus acidophilus, Bifidobacterium breve and on probiotic capsules containing a mixed culture of probiotic microorganisms. The survival of probiotics in various food matrices in the simulated gastrointestinal tract was quantitatively different. We managed to define foods suitable for supporting the multiplication of probiotic bacteria. A separate part of the work was focused on the targeted modulation of the intestinal microbiome by individualized probiotics that were prepared on the basis of molecular biological analyzes of the intestinal microbiome aimed at detecting the percentage of lactobacilli, bifidobacteria and phylum Firmicutes and Bacteroidetes. Personalized probiotic supplementation confirmed the positive effect of this approach on microbiome changes, especially on the increase of the content of lactobacilli, bifidobacteria and the overall diversity of the microbiome.
