Com o crescimento populacional e aumento das atividades humanas, grandes quantidades de poluentes são despejadas diariamente no meio ambiente. Nos ecosssistemas aquáticos, organismos são expostos por longos períodos à baixas concentrações de contaminantes. Assim estudos que focam em novas metodologias que sejam rápidas e não invasivas, são necessários para a avaliação de efeitos subletais. Um exemplo é a espectroscopia no infravermelho com transformada de Fourier (FTIR). Esta técnica é capaz de detectar alterações macromoleculares em amostras biológicas de forma rápida e não destrutiva, sendo uma ferramenta atrativa para análises ecotoxicológicas. Assim, o objetivo deste estudo foi verificar a aplicabilidade do FTIR como metodologia alternativa, utilizando perfis de composição macromoleculares como indicadores de efeitos subletais, em organismos expostos a elementos-traço. Para tal, ostrácodes (Chlamydoteca sp.) foram expostos aos elementos Cu, Cd, Hg e Mn. As alterações em perfis biomoleculares foram avaliadas em ostrácodes expostos por 48h ao Mn. Os resultados revelaram uma correlação negativa entre concentração de exposição e proporção de ácidos graxos saturados e insaturados com relação à proteína em organismos expostos ao Mn e a mesma correlação foi observada para o conteúdo de ácidos nucléicos. Isto sugere que os organismos tiveram seu metabolismo lipídico e sua capacidade de produção de proteína celular alterados devido à exposição ao Mn. Ademais, foram realizados testes crônicos (9 dias) com ostrácodes expostos ao Cu, Cd, Hg e Mn. Foi observada a diminuição de lipídios saturados e insaturados em relação à proteína em organismos expostos ao Cd, sugerindo o uso de reservas lipídicas devido ao estresse fisiológico. Ostrácodes expostos ao Cu, tiveram o aumento dos ácidos nucléicos, no qual o DNA pode ter sido o componente dominante. Alterações significativas relacionadas ao conteúdo de carbonato de cálcio foram reveladas nos organismos expostos ao Cd e ao Cu, o que sugere mudanças na composição da carapaça e possível alteração na capacidade de muda e crescimento dos organismos. Therefore, studies that focus on fast and non-invasive new methodologies are necessary for the assessment of sublethal effects in organisms. This technique can detect macromolecular changes of biological samples in a fast, inexpensive and non-destructive way, all of which lend to the attractiveness of this methodology for ecotoxicological analyzes. Thus, the aim of this study was to verify the applicability of FTIR as an alternative methodology, using the composition of macromolecular profiles as an indicator of sublethal effects in organisms exposed to trace elements. For this, ostracods (Chlamydoteca sp.) were exposed to the elements Cu, Cd, Hg and Mn. Changes in biomolecular profiles were evaluated in ostracods exposed for 48h to Mn. The results showed a negative correlation between the concentration of exposure and the proportion of saturated and unsaturated fatty acids for protein in organisms exposed to Mn and the same correlation was observed for the content of nucleic acids. This suggests that the organisms had their lipid metabolism and their cellular protein production capacity altered due to Mn exposure. Sublethal Effects of Crude Oil and Chemical Dispersant on the Eastern Oyster (<em>Crassostrea virginica</em>) at Multiple Life History Stages

Oil spills in the marine environment can threaten vulnerable ecosystems that support ecologically and economically significant organisms, such as the eastern oyster (Crassostrea virginica), in coastal habitats. The use of chemical dispersant (Corexit 9500) was applied as a cleanup effort in response to the Deepwater Horizon blowout to minimize crude oil slicks, but also resulted in increased concentrations of polycyclic aromatic hydrocarbons in the water column. The effects of increased soluble fractions of crude oil and dispersant components may be harmful to marine organisms. This study aimed to investigate possible sublethal impacts to the eastern oyster at multiple life history stages in order to understand potential implications on performance at an organismal, population, and ecosystem levels. Specifically, this study addressed 1) veliger swimming, 2) pediveliger settlement rates, 3) pollutant induced larval inactivity and 4) adult clearance rates after acute exposures to relevant concentrations (10 – 100 µL L-1) of water accommodated fractions of crude oil (WAF) and with a combination of chemical dispersant (CEWAF). No significant differences were observed in any tested swimming kinematics between controls and WAF or CEWAF treatments after 24 hour exposures for early staged veligers at concentrations up to 100 µL L-1 WAF and CEWAF. However, settlements rates of competent pediveligers were significant decreased compared to control (52.1 % s.d. 1.66) rates at concentrations of 50 µL L-1 WAF (30.9% s.d. 6.16) and 10 (41.2 % s.d. 0.857) and 50 (22.0% s.d. 1.23) µL L-1 CEWAF. Later staged larvae also showed increased vulnerability to oil pollution given that a higher percentage of organisms were inactive (48.3% s.d. 4.80) compared to early staged larvae (12.7% s.d. 7.68 ) after initial exposure at 50 µL L-1 CEWAF. Based on this result, we assumed effects of oil pollution were not manifested until the later larval life history stage evident by metamorphosis failure during the complex settlement transformation that results in reduced spat and eventually reduced adult oysters. Adult oysters were also exposed to increasing concentrations of WAF and CEWAF for 24 hours and feeding experiments were conducted in both clean seawater and the same oiled seawater conditions as their initial exposure. Oysters fed in oiled seawater had decreased clearance rates, but oysters fed in clean water had increased clearance rates, suggesting feeding efficiency can be returned to control rates when moved to the presence of clean water. However, our long term study conducted in clean seawater suggested of the oysters exposed to crude oil only (9.31 L h-1 g-1 s.d. 2.04) are able to return to clearance rates comparable to controls (7.69 L h-1 g-1 s.d. 1.89) after the 33 day time period but oysters exposed to crude oil with a combination of chemical dispersant (2.12 L h-1 g-1 s.d. 1.08) were not. Decreased feeding efficiency can have negative impacts on water quality in estuarine ecosystems that support productive habitats. Understanding the impacts of crude oil, and crude oil with a combination of chemical dispersant on ecologically significant organisms can aid in future oil spill response decisions in order to minimize environmental impacts.

Avaliou-se a bioatividade de derivados do nim (Azadirachta indica A. Juss.) como componente de isca para adultos e como regulador de crescimento de Ceratitis capitata (Wied.) e Anastrepha fraterculus (Wied.), determinando-se, ainda, o efeito de concentrações subletais de extratos de nim e o efeito deterrente de oviposição do extrato metanólico de folhas sobre C. capitata. Utilizando o extrato aquoso de amêndoas de nim, estimou-se a CL50 para adultos e imaturos, cujos valores foram 7.522 e 1.368 ppm para os adultos e 13.028 e 9.390 ppm para os imaturos de A. fraterculus e C. capitata, respectivamente. Subsequentemente, o óleo de amêndoas, os extratos de folhas, de ramos e de amêndoas de nim em diferentes solventes (água, metanol, diclorometano e hexano) foram comparados com base nas CL50 obtidas para adultos e para imaturos. Posteriormente, foram estimadas concentrações subletais (CL15 = 39 ppm, CL30 = 225 ppm e CL45 = 888 ppm) do extrato aquoso de amêndoas para adultos de C. capitata e avaliouse nessas concentrações o efeito do extrato aquoso de amêndoas, de folhas em metanol e de ramos em diclorometano sobre a reprodução e a longevidade dessa espécie. A deterrência de oviposição do extrato metanólico de folhas de nim em diferentes concentrações (10.000, 18.000, 32.000 e 56.000 ppm) para C. capitata foi avaliada tratando bagas de uva, que foram expostas às fêmeas com chance e sem chance de escolha. Verificou-se que os derivados do nim na concentração de até 7.522 e 1.368 ppm não afetaram os adultos de A. fraterculus e C. capitata, respectivamente. Já para a fase imatura, os extratos de amêndoas e de ramos em diclorometano na concentração de 13.028 ppm causaram mortalidade de A. fraterculus e na concentração de 9.390 ppm o extrato de amêndoas em metanol e em diclorometano causaram mortalidade de C. capitata. Houve redução da fecundidade total e da fertilidade no 8º dia quando os insetos foram alimentados com extrato de ramos em diclorometano a 888 ppm. A longevidade de machos e fêmeas e o período de pré-oviposição não foram afetados pelos tratamentos. O extrato de folhas em metanol provocou deterrência de oviposição para C. capitata em situação de chance de escolha, porém, em confinamento, o referido extrato não afetou o comportamento dessa espécie. A experiência recente de C. capitata não alterou a sua resposta em relação a bagas de uva tratadas. Besides, the effect of sublethal concentrations of neem extracts and the oviposition deterrency of the methanolic extract from leaves on C. capitata was evaluated. The estimated LC50 of aqueous extract from seed kernels were 7,522 and 1,368 ppm for adults and 13,028 and 9,390 ppm for immatures of A. fraterculus and C. capitata, respectively. In addition, the oil from seed kernels and the extracts from leaves, branches and seed kernels in different solvents (water, methanol, dichloromethane and hexane) were compared based on the LC50 obtained for adults and immatures. Effects on Survival, Reproduction and Growth of Ceriodaphnia dubia following Single Episodic Exposure to Copper or Cadmium

Effects of episodic exposures have gained attention as the regulatory focus of the Clean Water Act has shifted away from continuous-flow effluents. Standardized laboratory toxicity tests require that exposure be held constant. However, this approach may not accurately predict organism responses in the field following episodic exposures such as those associated with rain-driven runoff events or accidental pollutant discharge. Using a modified version of the 7-day short-term chronic test recommended by the US Environmental Protection Agency, Ceriodaphnia dubia were exposed to copper or cadmium for durations ranging from 1 minute to 24 hours. In addition, adult reproductive recovery and effects on second generation individuals was assessed following select copper exposures. Finally, cadmium exposures were compared in reconstituted hard water (RHW) and municipal treated wastewater effluent (TWE). Following exposure, organisms were transferred to clean RHW or TWE and maintained for the remainder of the test. No- and lowest observed effect concentrations (NO- and LOECs) increased logarithmically with respect to logarithmic decreases in duration regardless of metal, endpoint or water type. Effective concentrations of cadmium however, were usually higher than those of copper, especially in TWE. LOECs for C. dubia survival following 24-hour and 5-minute exposures to copper were 116 and 417 µg/L, respectively. LOECs for fecundity were 58 and 374 µg/L, respectively. Neonate production of first generation adult C. dubia appeared to recover from pulsed copper exposure upon examination of individual broods. Cumulative mean neonate production however, showed almost no signs of recovery at exposure durations ≥3 hours. Pulse exposure to copper also resulted in diminished fecundity of unexposed second generation individuals. Such effects were pronounced following parental exposure for 24 hours but lacking after parental exposures ≤3 hours. LOECs for C. dubia survival following 24-hour and 5-minute exposures to cadmium in RHW were 44 and 9000 µg/L, respectively. LOECs for fecundity were 16 and 5000 µg/L, respectively. In TWE, LOECs for C. dubia survival were 83 and >10,000 µg/L, respectively. LOECs for fecundity in TWE were 48 and 7000 µg/L, respectively. Runoff pollution is site and event specific, however, data presented herein may be useful as a predictive tool under various conditions.

Subletální efekty insekticidů neonikotinoidů na migrační potenciál pavouků / Sublethal effects of the insecticides neonicotinoids on migration abilities of spiders

The purpose for using pesticides is to kill organisms that cause damage (so-called pests) on various crops and thus prevent possible crop losses. Their side effect is a negative impact on non-target organisms. The presented diploma thesis is focused on the effect of neonicotinoids on invertebrates particularly on spiders. Its main goal was to document their influence on behavioral parameters influencing dispersal abilities such as locomotion and tendency to spread by wind (so-called ballooning). The diploma thesis compares the influence of neonicotinoids on model species, Pardosa lugubris (Walckenaer, 1802) and Phylloneta impressa (L. Koch, 1881) with different modes of prey hunting and compares the influence on adult and nymphal stages. Pesticides Actara® 25 WG, Biscaya® 240 OD, Confidor® 200 OD and Mospilan® 20 SP were applied to the spiders under laboratory conditions and their effect was tested at different concentrations and different methods of application. The biggest impact on spiders had Confidor. It clearly had the most significant negative effect on mobility and a tendency to spider wind propagation. Orientador: Sergio Antonio de Bortoli / Resumo: O cultivo intensivo de Brassicaceae favorece o aparecimento de algumas pragas como Plutella xylostella (Linnaeus, 1758) (Lepidoptera: Plutellidae), a traça-das-crucíferas, cujos danos podem comprometer seriamente a produção. Esse inseto-praga pode ser controlado tanto com inseticidas sintéticos, como com biológicos, sendo, no entanto, usualmente utilizados os sintéticos em seu combate. A traça-das-crucíferas foi a primeira espécie de inseto a ser constatada resistência a inseticidas biológicos, com o uso indiscriminado de inseticidas sintéticos para o seu controle contribuindo também para a seleção de indivíduos tolerantes ou resistentes, além de causar a mortalidade de organismos não alvos. Sendo assim, este trabalho teve por objetivo avaliar a atividade inseticida e os efeitos subletais de duas toxinas lepidóptero-ativas de Bacillus thuringiensis, Cry1Ca e Cry2Ac, em três populações de traças-das-crucíferas, sendo uma delas suscetível (PC) e duas consideradas resistentes (Bt e NO-QA) a toxinas de B. thuringienis, em condições de laboratório. Foram realizados bioensaios de suscetibilidade com as três populações e as toxinas Cry1Ca e Cry2Ac, estimando-se a concentração letal (CL50) e os seus efeitos subletais. A toxina Cry2Ac não causou mortalidade nas lagartas das três populações, então os testes subletais foram realizados apenas com a toxina Cry1Ca que provocou mortalidade. (Resumo completo, clicar acesso eletrônico abaixo) / Abstract: Intensive brassics cultivation favors appearing of some pests as Plutella xylostella (Linnaeus, 1758) (Lepidoptera: Plutellidae), diamondback moth (DBM), whose damage can seriously compromise production. This insect-pest can be controlled either synthetic insecticides as biological insecticides, but synthetics are usually used to combat. Diamondback moth was the first insect species to be verified resistance to biological insecticides, with indiscriminate use of synthetic insecticides for its control, also contributing to selection of tolerant or resistant individuals, as well as causing mortality on non-target organisms. Therefore, the objective of this work was to evaluate insecticide activity and sublethal effects of two lepidopteran-active toxins of Bacillus thuringiensis, Cry1Ca and Cry2Ac, on three DBM populations, one of them susceptible (PC) and two considered as resistant (Bt and NO-QA) to B. thuringiens toxins under laboratory conditions. Susceptibility bioassays were performed on the three populations and toxins Cry1Ca and Cry2Ac, estimating lethal concentration (LC50) and its sublethal effects.
