O timo é o órgão linfóide primário responsável pelo estabelecimento inicial de um repertório funcional de células T. A via de sinalização Notch é essencial para o desenvolvimento de células T a partir de células-tronco hematopoiéticas, e a distribuição de seus receptores e ligantes no timo humano ainda é desconhecida. A expressão de AIRE é crucial para a seleção de um repertório de receptores de linfócitos T (TCR) sem autorreatividade. Neste estudo, analisamos o padrão de expressão de AIRE e a distribuição de Notch em timos pacientes com cardiopatias congênitas, parte dos quais com síndrome de Down. Descrevemos a localização intratímica e os tipos celulares capazes de expressar os diferentes receptores e ligantes Notch. A expressão de AIRE em células epiteliais medulares foi significantemente reduzida em timos de crianças com síndrome de Down, deficiência esta que pode explicar a alta incidência de doenças autoimunes nesta cromossomopatia. / The thymus is a primary lymphoid organ which is essential for the initial establishment of a functional repertoire of T cells. Notch signaling is crucial for T-cell lineage development from hematopoietic stem cells; however, distribution of Notch ligands and receptors in human thymus is still unknown. AIRE is crucial for the selection of a T-cell-receptor (TCR) repertoire purged of self-reactive specificities. In this study, we analyzed the expression patterns of AIRE and Notch in human thymuses from children with congenital cardiopathies that undergo heart surgery, part of whom with Down syndrome. We described the intra-thymic localization and the cell types that express Notch receptors and ligands. AIRE expression in medullary epithelial cells is significantly decreased in Down syndrome patients. This deficiency could explain higher incidence of autoimmune disease in Down syndrome.

O timo secreta substâncias promotoras da sobrevivência de linfócitos e hormônios ligados à dinâmica do citoesqueleto. Esses agentes são produzidos em grande quantidade pelo animal recém-nato, e apresentam efeito homeostático sobre a população de linfócitos T circulantes, por toda a vida. Na sepse, a atrofia dos órgãos linfóides, o surgimento de acúmulos de actina intersticial, e a diminuição dos níveis de fibronectina plasmática guardam relação com a mortalidade. O trabalho aqui descrito procurou detectar o efeito do sobrenadante de cultura de timo de rato Wistar neonato, saudável, sobre a mortalidade e as expressões simultâneas de actina e fibronectina em timos de ratos Wistar adultos submetidos a sepse peritoneal. Foi constatada uma diminuição na expressão de actina e de fibronectina no interstício de timos de ratos Wistar adultos sépticos, tratados com a aplicação intraperitoneal do sobrenadante de explante de timo de ratos neonatos. Tal efeito provocou um aumento da sobrevivência semelhante ao tratamento com meio de cultura apenas ( sem explante), contudo houve uma notável melhora dos sinais clínicos . Não houve qualquer efeito anti-inflamatório, com o tratamento com o sobrenadante, sobre inflamação periférica em animais saudáveis submetidos a injeção intramuscular de glutaraldeído, o que afastou a hipótese de influência sobre a enzima tranglutaminase tecidual, ativada em estados inflamatórios , e responsável pela aglomeração intersticial, da actina originada de células lesadas. O autor concluiu que os efeitos observados pelo tratamento com sobrenadante de cultura de timo de animal recém-nato, podem ser atribuídos a estimulação de mecanismos promotores da integridade celular, frente aos estresses químicos vigentes no meio interno, durante a sepse. O autor sugere que a observação simultânea de actina e fibronectina no timo pode auxiliar nos estudos de tratamentos experimentais da sepse. / The response of septic adult Wistar rats submitted to treatment with supernatant from the culture of thymus of healthy newly-born Wistar rats were observed. Although there has not been significant reduction in mortality compared relation to treatment with culture medium only (without explanted) , it was possible to observe a rapid improvement in the clinical conditions and a lower agglomeration of actin and fibronectin in the thymus of treated animals. We did not observe any anti-inflammatory effect of the supernatant in animals with paw inflammation exclusively, which averted the modulation of the Tissue Transglutaminase Enzyme, agglomerator of the actin released by diseased cells. The author concludes that the treament of sepsis with supernatant stimulated survival in thymus, and suggests that the immuno-hystochemical study of actin and fibronectin in the thymus isa useful method in the analysis of experimental treatments of sepsis.

Quorum Sensing Inhibitory Activities of Various Folk-Medicinal Plants and the Thyme-tetracycline Effect.

Pseudomonas aeruginosa is an opportunistic, nosocomial pathogen for which antibiotic resistance and biofilm development is common. Quorum sensing communication is known to be a major controlling factor in virulence gene expression, biofilm development, antibiotic resistance factors, and specifically MexAB-OprM multi-drug efflux pump expression in P.aeruginosa. MexAB-OprM efflux pumps contribute to antibiotic resistance of tetracycline and other antibiotics in pseudomonads and other organisms. P.aeruginosa infections are problematic in cystic fibrosis and burn patients; it is also the number one causative agent of respiratory infections for intensive care unit patients. Present day antibiotics are losing the battle against these infections. In theory, quorum sensing inhibitors (QSI) reduce pathogencity of the organism; making it less virulent, thus allowing either the host immune system to clear the infection or use of a QSI in combination with an antibiotic to clear more persistent pathogens. For these reasons two alternative modes of treatment were explored in this study: quorum sensing inhibition by folk-medicinal plant extracts and an example of combination drug therapy, the “thyme-tetracycline effect”. Fifty folk-medicinal plant extracts were screened for potential anti-quorum sensing activity using two quorum sensing inhibition (QSI) reporter strains, Pseudomonas aeruginosa QSIS2 and Chromobacterium violaceum 12725. These were used to test specifically for C4-C6 and C12 HSL quorum sensing inhibition. Of the fifty plants tested, thirty plant families were represented. Eleven plant extracts (basil, chaparral, clove, cranberry, oregano, pomegranate, rosemary, sage, sassafras, thyme and witch hazel) showed C4 HSL quorum sensing inhibition as determined by both assays. Interestingly, five of the plants were from the Lamiaceae family. Thymus vulgaris (thyme), also from the Lamiaceae family, was chosen for further assessment. Previous research has shown that thyme extract can synergistically augment tetracycline activity against tetracycline-resistant Pseudomonas aeruginos, creating the “thyme-tetracycline effect.” Disc diffusion assay, thin layer chromatography (TLC), and TLC bioassay techniques were used to show that thymol is the active component in the thyme extract that augments tetracycline activity against resistant Pseudomonas. This study also showed that thymol is a potent C4 HSL quorum sensing inhibitor. The collective data suggests a potential mode of action for the thyme-tetracycline effect: thymol appears to prevent MexAB-OprM efflux pump gene expression. By blocking MexAB-OprM expression, tetracycline antibiotic accumulation can occur within the cell, thus allowing cellular damage.

Identificación y organización de los componentes celulares del timo de lubina (Dicentrarchus labrax)

Se realizó el estudio histológico del timo de lubina, con el fin de conocer la arquitectura del microambiente tímico y las relaciones estructurales de sus diferentes componentes. Las células linfoides sufren procesos de muerte celular programada por apoptosis, no descrita previamente en timo de peces; indicando la existencia de procesos de selección negativa durante su diferenciación intratímica. Existen células interdigitadas, no descritas previamente en peces. Hay áreas mielopoyéticas, con actividad eritropoyética y granulopoyética heterófila. La fragilidad de la barrera epitelial, que separa la cavidad branquial del parénquima tímico, puede verse incrementada por la presencia de macrófagos intraepiteliales y macrófagos con inmunoglobulinas de superficie. El transito de células sanguíneas a través de la capa de células epitelio-reticulares limitantes y de capilares sanguíneos indican la carencia de barrera hematotímica. Estos datos sugieren la existencia de procesos antígen o dependiente y la persistencia de funciones propias de órgano linfoide secundario en timo de lubina. / The histological study of the sea bass thymus was performed to know the architecture of the thymic microenvironment and the structural relationships of its different components. The lymphoid cells suffer processes of programmed cell death by apoptosis, not described previously in fish thymus, indicating the existence of negative selection processes during its intrathymic differentiation. There are interdigitating cells, which has not described before in fish. Exist myelopoiesis areas, with erythropoietic and heterophilic granulopoietic activity. The fragility of the epithelial barrier, that separates the branchial cavity from the thymic parenchyma, is increased by the presence of intraepithelial macrophages and macrophages with surface immunoglobulins. The traffic of blood cells across the layer of limiting reticular epithelial cells and blood capillaries indicate the lack of hemato-thymic barrier. This information suggests the existence of antigen dependent process and the p ersistence of own functions of secondary lymphoid organ in thymus of sea bass.

LE ROLE DE L'AUTOANTIGENE DANS LES MALADIES AUTO-IMMUNES : ETUDE DE LA DESMOGLEINE 1 AU COURS DES PEMPHIGUS

Les pemphigus sont des maladies auto-immunes spécifiques d'organe qui affectent la peau et les muqueuses. Ils sont caractérisés par la production d'autoanticorps pathogènes dirigés contre des protéines du desmosome, plus particulièrement, les desmogléines qui permettent l'adhésion entre eux des kératinocytes de l'épiderme. Au cours des pemphigus superficiels (PS), la réponse auto-immune est dirigée contre la desmogléine 1 (Dsg1). Chez les malades atteints de PS, des lymphocytes T autoréactifs vis à vis de la Dsg1 sont présents et interviennent dans la production d'autoanticorps anti-Dsg1. Cet autoantigène est aussi la cible de la réponse auto-immune au cours d'autres formes de pemphigus tels que les pemphigus vulgaire et paranéoplasique et par conséquent, semble jouer un rôle clé dans ces maladies. Depuis plus d'une décennie, le rôle de l'autoantigène lui-même dans l'initiation, la propagation et la pérennisation de la réponse auto-immune a été conforté par de nombreux arguments expérimentaux. En nous appuyant sur ce concept, nous avons entrepris d'étudier l'intervention de la Dsg1 dans les mécanismes physiopathologiques qui concourent au développement des pemphigus. Dans un premier temps, nous avons démontré l'expression dans l'épiderme humain, d'une isoforme tronquée de la Dsg1 générée par épissage alternatif des transcrits DSG1. Cette Dsg1 soluble est porteuse d'une séquence peptidique spécifique qui se fixe avec une forte affinité à certaines molécules HLA de classe II de susceptibilité au PS en particulier, à la molécule DRB1*0102. Ce peptide est par ailleurs capable d'induire la prolifération de cellules mononuclées du sang périphérique chez 50% des malades atteints de la forme sporadique de PS. Ces patients expriment des allèles HLA de classe II associés à la maladie et deux d'entre eux sont porteurs de la molécule DRB1*0102. Ainsi, la modification de la Dsg1 par épissage alternatif pourrait-elle intervenir dans la rupture de la tolérance au niveau du compartiment T chez les individus prédisposés génétiquement par l'expression de certains allèles HLA de classe II et in fine, conduire à l'initiation d'une réponse auto-immune B dirigée contre la Dsg1. En second lieu, nous avons observé une diversification de la réponse anticorps chez des souris normales immunisées avec la région extracellulaire recombinante de la Dsg1. Les animaux immunisés produisent non seulement des IgG dirigées contre la Dsg1 mais aussi, contre d'autres protéines de l'épiderme. Nous avons dérivé cinq anticorps monoclonaux à partir des splénocytes isolés de ces souris et montré que trois d'entre eux sont dirigés spécifiquement contre la Dsg1. Les deux autres, 10A1 et CK1, ne réagissent pas avec la Dsg1 mais reconnaissent des protéines épidermiques de plus haut poids moléculaire, compatible avec ceux des autoantigènes de la famille des plakines spécifiquement reconnus par les anticorps au cours du pemphigus paranéoplasique, e.g. l'envoplakine et la périplakine. Grâce à une analyse protéomique ciblée combinant l'immunocriblage d'une carte protéique 2D d'épiderme humain et la spectrométrie de masse MALDI-ToF, nous avons montré que la protéine cible du 10A1 est l'envoplakine, et que le CK1 reconnaît à la fois l'envoplakine et la périplakine. Ainsi, en accord ce modèle expérimental murin, la réponse B anti-Dsg1 pourrait-elle gouverner la réponse vis à vis d'autres protéines du desmosome en particulier, les plakines, mimant de ce fait la diversité de la réponse auto-immune B observée au cours du pemphigus paranéoplasique. Enfin, nous démontrons par des analyses en PCR que l'ARNm de la Dsg1 est exprimé dans le thymus humain normal, que son expression augmente avec l'âge et de ce fait, que l'absence de l'expression thymique de cette autoantigène ne constitue pas l'origine de la rupture de la tolérance qui concoure au développement des PS. Nos résultats mettent en exergue le rôle la Dsg1 dans le processus auto-immun au cours des pemphigus, d'une part, à la phase d'initiation, avec l'intervention de cet autoantigène modifié par épissage alternatif dans la réponse lymphocytaire T et d'autre part, à la phase de propagation, avec la diversification de la réponse anticorps vis à vis d'autres antigènes desmosomiaux induite chez des souris normales immunisées avec cet autoantigène.

Evidence of a thymic abnormality in relapsing-remitting multiple sclerosis

The peripheral naive CD4 T cell pool is homeostatically regulated through a balance of thymic production, delivery of survival signals and homeostatic proliferation. CD4 recent thymic emigrants (RTEs) have a high T cell receptor excision circle (TREC) content and express high levels of CD31. We report premature thymic involution in RRMS, initiated by reduced numbers of naive CD4 T cells and various naive CD4 T cell subsets in peripheral blood. Further, CXCR4, a receptor involved in emigration from the thymus, and CD127 and Bcl-2 (survival signals) are upregulated in various naive CD4 T cell subsets in RRMS. As a compensatory process, naive CD4 T cells undergo homeostatic proliferation. This proliferation is a form of peripheral positive selection through self-MHC/self-antigen interaction and thus can contribute to the expansion of autoreactive T cells and predispose to development of RRMS.

O efeito do hormônio do crescimento na transmigração de timócitos in vitro / The effect of growth hormone on transendothelial of thymocytes in vitro

Transendothelial migration is a key process in lymphocyte trafficking. It is known that growth hormone (GH) modulates intrathymic T cell migration and that directly or indirectly influences the biology of endothelial cells and thymocytes. In this context, we evaluated the effect of GH on the transendothelial migration of thymocytes, in the presence or absence the chemokine CXCL12. For the study, we applied the murine thymic endothelial cell line (tEnd.1) and thymocytes freshly isolated from male C57BL/6 mice aged 4-6 weeks. Through in vitro studies of transendothelial migration of thymocytes, and analyses by flow cytometry, we observed a statistically significant reduction in the numbers of transmigrated thymocytes, mainly seen for CD4+CD8+ cells, across endothelial barriers treated with GH [100 ng/ml], for 8 h. Importantly, this effect was reverted when thymocytes were treated with GH [100 ng/ml] for 1 h. Similar effects were noted in transmigration through joint action of GH and CXCL12. Despite the fact that all the CD4/CD8-defined subsets have been affected, we observed a statistically significant increase in the number of transmigrated thymocytes, mainly of CD4+CD8+ and CD4+CD8- cells, when they were treated with GH and allowed to migrate in response to CXCL12, when compared to untreated controls in response to CXCL12. We demonstrated, by real-time PCR, that GH at 100 ng/ml did not modulate the expression of VCAM-1 and CXCL12 by tEnd.1 cells treated for 8 h. Furthermore, we found by cytofluorimetric assay that GH alone did not change the membrane expression of VLA-4 and CXCR4 receptor in transmigrated thymocytes. Through joint action of GH and CXCL12, was observed increase in expression of VLA-4 on CD4-CD8+ cells from the group of GH-treated thymocytes, when compared to groups in which endothelial cells were treated. As for expression of CXCR4, we found that the GH/CXCL12 decreased the expression of this receptor on transmigrated thymocytes, mainly in CD4+CD8+ cells from the group in which the thymocytes were treated. In conclusion, our results reinforce that GH influence the thymus physiology and modulate the biology of endothelial cells and thymocytes, with differential effects in the process of endothelial transmigration of murine thymocytes. / Fundação de Amparo a Pesquisa do Estado de Alagoas / A transmigração endotelial é um processo essencial no trânsito de linfócitos. É sabido que o hormônio do crescimento (GH) está envolvido na migração intratímica de células T, e que influencia direta ou indiretamente a biologia das células endoteliais e dos timócitos. Neste contexto, avaliamos a ação do GH na migração transendotelial de timócitos, frente ou não à quimiocina CXCL12. Para o estudo, utilizamos a linhagem endotelial tEnd.1 e timócitos recém isolados de camundongos machos C57BL/6 de 4-6 semanas. Através do ensaio de transmigração endotelial in vitro de timócitos, e análise por citometria de fluxo, verificamos uma redução estatisticamente significativa no número de timócitos transmigrados, principalmente nas subpopulações CD4+CD8+, frente a barreiras endoteliais tratadas com GH [100 ng/ml], por 8 horas. Este efeito foi revertido quando os timócitos foram tratados com GH [100 ng/ml], por 1 hora. Efeitos similares na transmigração foram notados pela ação do GH frente a CXCL12. Apesar de todas as subpopulações CD4/CD8 terem sido afetadas, observamos um aumento estatisticamente significativo no número de timócitos transmigrados, principalmente das células CD4+CD8+ e CD4+CD8-, quando foram tratados com GH e colocados para migrar em presença da CXCL12, em relação ao grupo controle não tratado e em presença da quimiocina. Demonstramos, ainda, por RT-PCR, que o GH na concentração de 100 ng/ml, não modulou a expressão de VCAM-1 e CXCL12 por células tEnd.1 tratadas, por 8 horas. Além disso, verificamos por análise citofluorimétrica, que o GH sozinho não alterou a expressão dos receptores VLA-4 e CXCR4 nos timócitos transmigrados. Pela ação conjunta do GH e CXCL12, foi registrado aumento na expressão de VLA-4 sobre células CD4-CD8+ no grupo experimental em que os timócitos foram tratados com GH, quando comparamos aos grupos onde as células endoteliais foram tratadas. Quanto à expressão de CXCR4, verificamos que o GH/CXCL12 diminuiu a expressão deste receptor sobre timócitos transmigrados, principalmente em células CD4+CD8+, nos grupos experimentais em que os timócitos foram tratados. Em conclusão, nossos dados fortalecem o postulado de que o GH influencia a fisiologia do timo, e modula a biologia das células endoteliais e dos timócitos, com efeitos diferenciais no processo de transmigração endotelial de timócitos murinos.

Avaliação da perda linfocitária do timo de frangos de corte utilizando sistema de análise digital de depleção linfóide (ADDL)

O timo é um dos órgãos linfoides mais acometidos pelas patologias imunodepressoras, o qual geralmente é avaliado quanto ao grau de perda linfocitária a partir de técnicas histológicas subjetivas. Este trabalho teve como propósito a adaptação e aplicação do sistema ADDL (Análise Digital de Depleção Linfóide) em timos, a fim de tornar sua análise mais precisa. Primeiramente, realizou-se um estudo piloto, no qual foram utilizadas amostras de timo do trabalho desenvolvido por Camillotti (2011), porém constatou-se um número insatisfatório de lobos tímicos para os três escores de depleção linfocitária adotados, assim como dificuldade na categorização com apenas 3 graus de lesão. Optou-se, então, pela indução de imunodepressão através da utilização de um glicocorticóide, e pela classificação dos timos em cinco escores (1 a 5). Para tanto, foram utilizados 70 aves de um dia de idade, sendo que aos 21 dias de idade, foram separadas 15 aves para composição do grupo controle e 55 para o grupo tratado. Foi empregado o protocolo de tratamento com dexametasona adaptado de Kong, et al. (2002). Durante as coletas, de cada uma das aves, ambas as cadeias tímicas eram pesadas, sendo selecionados seis lobos para o processamento histológico de rotina, análise quanto ao grau de depleção linfocitária por dois avaliadores (A e B) e submissão a todas as etapas de processamento pelo sistema ADDL. Foram construídas ao final, cinco RNAs (Redes Neurais Artificiais): duas tendo em vista o escore por lobo (escore do avaliador A; escore do avaliador B) e três considerando o escore por ave (moda dos escores de 6 lobos/ave do avaliador A; moda dos escores de 6 lobos/ave do avaliador B, moda conjunta dos escores dos avaliadores A + B). A fim de relacionar o peso relativo (PR) do timo ao escore digital de depleção, foi aplicado o teste de correlação de Spearman, seguido de análise de regressão, e para evidenciar diferenças entre os escores atribuídos pelos avaliadores, utilizou o teste de Wilcoxon-Mann-Whitney. Constatou-se que o tratamento com dexametasona permitiu a aquisição de timos com escores entre 1 a 5. Houve diferença significativa entre as leituras realizadas pelos avaliadores considerando o lobo e também entre a moda de escores dos dois avaliadores, assim como entre a moda do avaliador B e a moda conjunta (p<0,005). Foi observado que as duas RNAs construídas por lobo obtiveram um percentual de 50% de classificações corretas. As RNAs construídas utilizando a moda mostraram um percentual de acerto de 86,57% para o avaliador A, 80,60% para avaliador B e 94,03% para a rede construída a partir da moda conjunta dos avaliadores. Paralelamente, observou-se ρ= -0,77 e R2=0,58 entre as variáveis “PR” e “Escore digital de depleção linfocitária”. Em conclusão, foi possível a aplicação do sistema ADDL para timos, sendo a RNA treinada com a moda conjunta dos escores dos avaliadores A e B, o modelo que alcançou melhor desempenho. Portanto, o uso de ferramentas de análise de imagem em conjunto com as RNAs, torna possível uma avaliação do sistema imune da ave adotando critérios objetivos para as análises histológicas quanto a perda linfocitária. / The thymus is a lymphoid organs most affected by immunosuppressive diseases, which is usually assessed on the degree of lymphocyte loss from subjective histological techniques. This work aimed to the adaptation and application of the system DALD (Digital Analysis of Lymphoid Depletion) in the thymus in order to make your analysis more accurate. First,a pilot study was performed, in which samples from the thymus of the work of Camillotti (2011) were used, but it was found a number of poor thymic lobes for the three scores lymphocyte depletion adopted, as well as difficulty with categorization only three degrees of injury.
