Interakce hyaluronanu s tenzidem CAE / Interaction of hyaluronan with surfactant CAE

The diploma thesis is focused on the study of interactions between hyaluronan of various molecular weights with CAE surfactant. This surfactant does not have the exact composition and there is not known their molecular weight. One of the main parameters needed to describe the interactions between surfactant and hyaluronan, there is the determination of critical micellar concentration. The value of critical micellar concentration of CAE is not known yet. All measurements were made for solutions in aqueous solution and 0.15 M NaCl. The determination of the molecular weight of this surfactant was performed by the technique SEC-MALS-dRI. High resolution ultrasonic spectroscopy was chosen to determine the critical micellar concentration and tensiometry was chosen as a complementary method. The interactions of CAE surfactant with polysaccharide of hyaluronan were showed by high resolution ultrasonic spectroscopy and densitometry. Compressibility was calculated from ultrasonic velocity and density.

Mechanical Evaluation Methods for Polymer and Composite Systems

This dissertation describes the development and application of various mechanical characterization techniques to four types of polymer composite materials. The composite nature of these materials ranges from molecular to macro-scale, as do the size scales probed by the techniques chosen. The two main goals of this work are to evaluate the suitability of existing characterization methods to new composite materials (and augment the methods as needed), and to use these methods to determine optimal composite system parameters to maximize the desired mechanical response. Chapter 2 employs nondestructive ultrasonic spectroscopy for characterizing the stiffness response of both micron-scale woven composites and macro-scale glass-polymer-glass laminates. Both traditional wavespeed measurement as well as aspects of resonant ultrasonic spectroscopy are applied to determine material parameters. The laminates are also examined in Chapter 3, which utilizes both large-scale and small-scale quasi-static and dynamic puncture tests to elucidate the size- and rate-dependence of dynamic behavior. Because of limitations encountered with these methods, a smaller-scale, more fundamental test is developed and applied which focuses solely on the deformation and delamination of the polymer. These two processes, which account for the vast majority of energy absorbed during a puncture event, can be evaluated in terms of self-similar process zone propagation process models. Identifying and optimizing the relevant model parameters can promote the design of systems with maximum energy absorption. Exploratory work on nanocomposite systems is presented in Chapter 4. The polymer matrix from the laminated systems of the previous chapter is used to produce nanosilica composites. A range of techniques are employed to determine the level of dispersion and the mechanical reinforcement provided. The final project presented investigates copolycarbonates, or molecular composites, that have been developed to lessen the detrimental effect of aging on mechanical properties. Mechanical and thermal measurements can elucidate the effect of structure, specifically molecular mobility, on susceptibility to physical aging. The differences in molecular mobility contribute to differences in energy absorption by plastic deformation and damage, which is required for material toughness. Thus, understanding the molecular structure allows for determination of an optimal structure or copolymer concentration to maximize fracture toughness.

Produkce, charakterizace a využití biomasy různého původu / Production, Characterization and Utilization of the Biomass from Various Sources

Úprava biomasy je jedním z nejdůležitějších problémů v moderních přírodních vědách, protože je základní kategorií týkající se zemědělství, potravinářství, ekologie, zpracování odpadu a biotechnologie. Ať už živočišného, rostlinného nebo mikrobiálního původu, biomasa představuje obrovský zdroj surovin jako potravin, čistých chemikálií, bioaktivních molekul atd., jejichž izolace, charakterizace a formulace může vést k zajímavým novým produktům určeným pro lidskou spotřebu, nebo jako nový materiál v biomedicíně. Předložená studie byla zaměřena na výzkum dvou druhů biomasy - kuřecí kůže jako zdroje kolagenu t a biomasy mikrořasy Chlorella sorokiniana obohacené selenomethioninem (SeMet). V první části práce byl z kuřecí kůže izolován, identifikován a charakterizován kolagen typu I. Molekulární vlastnosti kuřecího kolagenu byly analyzovány a srovnány s jinými kolageny z živočišných kůží. Pro molekulární charakterizaci kolagenu byla použita viskosimetrie a ultrazvuková spektroskopie. Ultrazvukovou spektroskopií bylo zjištěno, že disagregace a zkapalňování hovězího kolagenu začíná při teplotě 40 °C, zatímco u kuřecího kolagenu začíná až při 50 °C. Viskosimetrie dále potvrdila vyšší tepelnou stabilitu kolagenu z kuřecí kůže, jeho denaturační teplota byla 50 °C, což je rovněž o deset stupňů více než u hovězího kolagenu. Kuřecí kolagen obsahuje dvakrát vyšší množství lysinu, což poskytuje tepelnou stabilitu kolagenu. Na základě získaných výsledků lze říci, že vzhledem ke své vysoké tepelné stabilitě a vhodnému aminokyselinovému složení, kuřecí kůže může být použita jako alternativní zdroj kolagenu typu I s aplikacemi v potravinářském průmyslu a biomedicíně. Druhá část práce byla zaměřena na obohacení biomasy zelené mikrořasy C. sorokiniana selenomethioninem. Experimentální část byla provedena v Laboratoři biotechnologie řas na Univerzitě Huelva ve Španělsku. Cílem první části experimentů bylo studovat vliv selenu na životaschopnost řas, morfologii buněk a akumulaci SeMet v biomase mikrořasy kultivované v dávkových kulturách. Subletální koncentrace Se v živném médiu, 40 mgL-1 (212 M), snížila rychlost růstu o 25 % ve srovnání s kontrolní kulturou. Hodnota EC50 45 mgL-1 (238,2 M) byla stanovena pro selenan. Ultrastrukturální studie ukazovaly na strukturální změny chloroplastu (granulární stroma, redukce thylakoid). Elektroforéza proteinů z biomasy mikrořasy ukazuje, že Se ovlivňuje expresi genu enzymu Rubisco. C. sorokiniana byla schopna akumulovat až 140 mgkg-1 SeMet během 120 h kultivace. Další část experimentální práce byla zaměřena na obohacování biomasy mikrořasy C. sorokiniana selenomethioninem během kontinuální kultivace s použitím 2,2 L bioreaktoru v kultivačním médiu s přídavkem koncentrace selenu v rozmezí od 5 do 50 mgL-1. C. sorokiniana rostla stejně ve všech testovaných koncentracích selenu kromě koncentrace 50 mgL1, která byla již po krátké době kultivace letální. Během kontinuální kultivace se 40 mgL-1 selenu, bylo získáno maximálně 246 gL-1 selenomethioninu denně. Výsledky ukazují, že kultivace v dávkových kulturách a dlouhodobá kontinuální kultivace mikrořasy C. sorokiniana pro získaní biomasy obohacené SeMet je možná pečlivým výběrem podmínek kultivace a subletálních koncentrací selenu v živném médiu.

Studium interakcí hyaluronan-aminokyseliny / Study of interactions of hyaluronan-amino acids

The master´s thesis deals with the study of the interaction between the polysaccharide hyaluronan of diffrerent molecular weights with the amino acids arginine, lysine, arginine hydrochloride and 6-aminocaproic acid. They are expected interaction between carboxyl groups of hyaluronan and amino groups of amino acids. These interactions were investigated by using ultrasonic spectroscopy, DLS, measuring pH and conductivity. Obtained results were compared with sodium polystyrene sulfonate. With ultrasonic spectroscopy was observed a change of concentration inkrement for titration of amino acid to water or polymers solutions especially for high molecular weight hyaluronan and for NaPSS in combination with 6AKK in concentration range of added amino acid 0–30 mM. The size of this change could mean a degree of interaction between polymers and amino acids. This theory has not been confirmed by other methods. By pH and conductivity measurements interations between arginine and low molecular weight hyaluronan and NaPSS were only confirmed. There was no possibility to make unequivocal conclusions from determination of particle size and zeta potential by DLS. Overall, the issue of the interaction of amino acids with polyanions was proved above expectations complex and will be appropriate to further expand the observations made in this thesis.

Interakce hyaluronanu s DNA / The interactions of hyaluronan and DNA

This diploma thesis deals with the study of possible interactions between hyaluronan (HA) and plasmid DNA (pDNA). Plasmid DNA was isolated from E. coli JM109 (pUC19) and resuspended in TE buffer as well as high molecular weight hyaluronan. Individual samples of pDNA, HA and pDNA-HA were characterized by gel electrophoresis, CD spectroscopy and high resolution ultrasonic spectroscopy. Agarose gel electrophoresis examined the effect of the addition of hyaluronan to plasmid DNA on the migration of samples to the positive electrode. Structural changes in pDNA-HA samples were examined using CD spectroscopy. Individual CD spectra describes the dependence of the difference in absorption coefficients for left-hand and right-handed elliptic polarized light at wavelength. High resolution ultrasonic spectroscopy has been used to study interactions. It is an analytical method based on ultrasonic velocity and attenuation. We classify this technique as a non-destructive method because the passing waves do not affect the structure of the analyzed sample.

Vliv "zaplňovacích" činidel na interakce DNA-tenzid / Effect of crowding agents on DNA-surfactant interaction

Tato diplomová práce se zabývá vlivem zaplňovacích činidel na interakce v systému DNA-tenzid. DNA o velikosti 4017 párů bází byla připravena polymerázovou řetězovou reakcí, jako templát byl použit plasmid pSB-E1g. Polyetylen glykol (PEG) byl použit jako zaplňovací činidlo a jeho vliv na DNA-tenzid interakce byl zkoumán experimenty založenými na fluorescenci a gelové elektroforéze. Také byl studován vliv iontové síly za použití NaBr na interakce DNA-tenzid za použití zaplňovacího činidla. Data byla vyhodnocena a evaluována v této práci. V úvahu byl brán i možný vliv polyetylen glykolu na kritickou micelarní koncentraci (CMC) tenzidu, bylo provedeno měření CMC pomocí ultrazvuku s vysokým rozlišením, avšak nebyl zjištěn žádný značný vliv zaplňovacího činidla na CMC tenzidu. Část této práce bude zahrnuta v publikaci s anglickým názvem Combined role of macromolecular crowding and cationic surfactant in efficient DNA condensation.

Teplotní závislost karagenanu a hyaluronanu / Temperature dependence of carrageenan and hyaluronan solutions

This diploma thesis focuses on the study of temperature dependence of -carrageen, hyaluronan of low molecular weight (250–450 kDa) and hyaluronan of high molecular weight (1 500–1 750 kDa) in aqueous solutions. The description of temperature depending changes in properties of these substance is based on rheological method. This work concentrates on the characterization of viscoelasticity through measurement of the storage (elastic) and loss (viscous) modulus using -carrageen and hyaluronan solutions of different molecular weights and different concentrations depending on temperature (20–50 °C, alternatively, in carrageen, 20–80 °C). The temperature dependence was further measured using an uncommon technique called high resolution ultrasonic spectroscopy (HR-US) which measured relative velocity of selected concentrations of -carrageen and hyaluronan of high molecular weight depending on the temperature of heating and cooling (20–80 °C). The measurements of temperature dependence were carried out for varied oscillation frequencies of rheometer sensor/frequency of ultrasonic waves. Knowledge of temperature dependence of the investigated substances may be applied in drug chemistry, primarily with respect to degradation due to high temperatures. The measured data gives information about sol-gel temperature and gel-sol temperature of carrageen.

Studium hydrofobních domén v huminových kyselinách / Study of Hydrophobic Domains in Humic Acids

Fyzikálně-chemická povaha hydrofobních domén huminových kyselin byla studována z několika hledisek. K objasnění významu fluorescenčních spekter byly vzorky podrobeny sekvenční frakcionaci, která pomohla k částečnému objasnění vlivu vodorozpustných složek, volných a vázaných lipidů na optické vlastnosti huminových kyselin. Výsledky naznačily, že fluorescenční píky tradičně přiřazované superpozici jednotlivých struktur jsou spíše důsledkem agregačních vlastností huminových molekul tvořících vlivem hydrofobního efektu zdánlivě vysoce aromatické struktury. Dále pak bylo zjištěno, že na optických vlastnostech huminových kyselin mají podíl i molekuly, které nemají primárně fluoroforní nebo chromoforní vlastnosti. Tento pohled je v souladu s teorií supramolekulárního uspořádání huminových kyselin. Dále byly studovány agregace, konformační chování a termodynamická stabilita huminových kyselin pomocí metody vysoce rozlišovací ultrazvukové spektroskopie. Le thème central de cette thèse est l'étude des résonances pour le problème intérieur en élastodynamique (géométries elliptique et sphéroïdale), et pour le problème de diffusion en acoustique (géométrie elliptique). On s'intéresse en particulier à la levée de dégénérescence des résonances liée à la brisure de symétrie de l'objet lors de la transition du disque circulaire vers le disque elliptique (2D), et de la sphère vers le sphéroïde (3D). Ce phénomène est étudié et interprété d'un point de vue théorique en prenant en compte les symétries de l'objet à l'aide de la théorie des groupes. Cette approche est complétée par une modélisation numérique et une partie expérimentale. En 2D, nous étudions le problème intérieur pour un disque elliptique élastique (étude des modes résonants) et le problème de la diffusion acoustique par des cylindres elliptiques élastiques. Ils sont traités à partir du formalisme modal combiné à la théorie des groupes dans le contexte vectoriel de l'élastodynamique. La levée de dégénérescence est observée théoriquement mais aussi expérimentalement en diffusion. La méthode simplifie considérablement le traitement numérique des problèmes étudiés, fournit une classification des résonances selon les 4 représentations irréductibles du groupe de symétrie C2v (associé à la géométrie elliptique) et donne une interprétation physique de la levée de dégénérescence en termes de brisure de symétrie. Une partie expérimentale en spectroscopie ultrasonore vient compléter l'étude théorique du problème de diffusion. Une série d'expériences en cuve est menée dans le cas de cylindres elliptiques de différentes excentricités en aluminium immergés dans l'eau, dans la bande de fréquence 0 ≤ kr ≤ 50, où kr est le nombre d'onde réduit dans le fluide. Les résultats expérimentaux présentent un très bon accord avec les résultats théoriques, la levée de dégénérescence est observée expérimentalement sur des fonctions de forme et mise en évidence sur des diagrammes angulaires. Le problème intérieur en 3D est traité expérimentalement à partir de la génération et la détection optiques d'ondes élastiques. Une série d'expérimentations sur des objets tridimensionnels (sphère, sphéroïdes oblates et prolates de différentes excentricités) en aluminium est réalisée. Ils sont mis en vibration par impacts laser et les mesures de vitesse et de fréquence s'effectuent par vibrométrie laser. On réalise ainsi une comparaison qualitative entre la théorie 2D et l'expérience 3D. Les mesures sont menées à la fois dans les domaines temporel et fréquentiel pour mettre en évidence la levée de dégénérescence d'une part, et l'onde de Rayleigh qui se propage sur la surface des objets d'autre part. Nous identifions deux trajets pour cette onde en géométrie sphéroïdale, l'un circulaire et l'autre elliptique.Enfin, dans le cadre des problèmes intérieurs 2D et 3D, on donne une interprétation en termes de rayons à travers la dualité entre le spectre des résonances et le spectre des longueurs des orbites périodiques (OPs), avec la mise en évidence du phénomène de conversion de mode et l'identification de l'onde de Rayleigh. Un phénomène, nouveau à notre connaissance, vient s'ajouter au phénomène de bifurcation de certaines orbites. Au cours de la déformation vers le disque elliptique, les orbites avec conversion de mode du disque circulaire se séparent en deux orbites dont les longueurs sont associées aux trajets minimal et maximal qu'elles parcourent. Cette observation s'interprète comme une conséquence du théorème de Fermat. Dans le cas du sphéroïde, on retrouve les orbites du disque circulaire dans le plan équatorial et celles du disque elliptique dans le plan méridien. Nous mettons également en évidence les pics associés aux deux trajets parcourus par l'onde de Rayleigh sur le spectre des OPs. / Resonances for the interior problem in elastodynamics and the acoustic scattering problem are considered in elliptical and spheroidal geometries. Interest is focused on the splitting up of resonances which occurs when the symmetry is broken in the transition from the circular disc to the elliptical one (2D), and from the sphere to the spheroid (3D). From the theoretical point of view, this physical phenomenon is studied and interpreted taking into account the symmetries of the object with the help of group theory. This approach is completed by a numerical modeling and an experimental part. As far as the two dimensional problems are concerned, the interior problem for an elliptical elastic disc (study of resonant modes) and the acoustic scattering problem for infinite elliptical elastic cylinders are studied combining modal formalism and group theory in the vectorial context of elastodynamics. The splitting up of resonances due to the circular symmetry breaking is not only theoretically observed but also experimentally for the scattering problem. The method significantly simplifies the numerical treatment of the problems studied, provides a full classification of resonances over the 4 irreducible representations of the symmetry group C2v (associated with the elliptical geometry) and gives a physical interpretation of the splitting up in terms of symmetry breaking of the symmetry group O(2) (invariance under rotation). An experimental part based on ultrasonic spectroscopy complements the theoretical study of the scattering problem. A series of tank experiments is carried out in the case of aluminum elliptical cylinders immersed in water, in the frequency range 0 ≤ kr ≤ 50, where kr is the reduced wave number in the fluid. The experimental results provide a very good agreement with the theoretical ones, the splitting up is observed on experimental form functions and the split resonant modes are identified on angular diagrams. The interior problem in 3D is studied by means of an experimental approach based on the optical generation and detection of elastic waves. A series of experiments is performed on three-dimensional objects in aluminium. These objects (sphere, prolate and oblate spheroids of various eccentricity) are excited by laser impacts, and the velocity and frequency measurements are carried out by laser vibrometry. Theory and experiments are qualitatively compared. The measurements are performed in both the frequency and time domains to highlight the splitting up of resonances on one hand, and the Rayleigh wave propagating on the surface of the objects on the other hand. We emphasize two paths for this surface wave in spheroidal geometry: a circular one in the equatorial plane and an elliptical one in the meridian plane. Finally, in the context of the interior problems in 2D and 3D, a physical interpretation of resonances in terms of geometrical paths is provided. Mode conversion is highlighted and the Rayleigh wave is identified on the periodic orbits lengths spectrum.In addition to the bifurcations of some periodic orbits, a phenomenon, new to our knowledge, appears. The orbits with mode conversion of the circular disc split in two orbits when the transition to the elliptic disc occurs. The lengths of these orbits are associated with the minimal and maximal travel paths. This observation is interpreted from Fermat's theorem.For the spheroid, orbits of the circular disc and those of the elliptical disc are recovered in the equatorial and meridian planes respectively. We also emphasize the peaks associated with the travel paths of Rayleigh wave in spheroidal geometry appearing on the periodic orbits spectrum.

Zu den wenigen Methoden, die sich für eine prozessnahe Charakterisierung von dispersen Stoffsystemen bezüglich der Partikelgrößenverteilung eignen, zählt die Schalldämpfungsspektroskopie. Dennoch ist ihr Einsatz in der industriellen Praxis auf einen überschaubaren Bereich an Messaufgaben und Stoffsystemen beschränkt. Insbesondere findet sie kaum Einsatz für die Partikelgrößenanalyse von submikronen Emulsionen. Die vorliegende Arbeit macht es sich deshalb zum Ziel, ausgehend von grundsätzlichen Überlegungen die Eignung der Schalldämpfungsspektroskopie zur prozessnahen Charakterisierung des Dispersitätszustandes von submikronen Emulsionen zu bewerten und zu verbessern. Schwerpunkte der Arbeit sind die modellhafte Beschreibung des Schalldämpfungsverhaltens von hoch konzentrierten Partikelsystemen im Submikrometerbereich, die Abschätzung des Einflusses von Emulgatoren auf die Schalldämpfung, die Erkennbarkeit von Einzelheiten der Partikelgrößenverteilung, die Sensitivität berechneter Partikelgrößenverteilungen bezüglich der Partikelkonzentration und der verschiedenen Stoffeigenschaften sowie die Auswertung von Schalldämpfungsspektren bei Unkenntnis dieser Modellparameter. Die angesprochenen Aspekte werden am Beispiel typischer Charakterisierungsaufgaben diskutiert. / The ultrasonic spectroscopy is one of few measurement methods applicable to the online characterization of disperse systems with regard to particle size distribution. However, its application in industrial practise is restricted to a rather low number of measurement tasks and material systems. In particular it is hardly used for the particle size analysis of submicron emulsions. This thesis therefore aims to the evaluation of the ultrasonic spectroscopy regarding its applicability to the online characterization of submicron emulsions. Main issues of the thesis are the modelling of sound attenuation in dense particle systems, the estimation on the influence that surfactant layers exert on sound attenuation, the resolution at which size distribution can be measured, the sensitivity of calculated size distributions to model parameters (particle concentration and material properties) and the analysis of measured attenuation spectra by unknown model parameters. The meanining of these issues are illustrated for typical characterization tasks.
