The content of this thesis is the design of an upholstered armchair. The starting point of the design is the theoretical part, which primarily contains general requirements for dimensions, construction and safety. The practical part of the thesis discusses the design procedure of the product with regard to the information of the theoretical part. The procedure described is supplemented with drawings and visualizations. Subsequently, the production process is recorded with the description of the whole construction process. The final draft is presented using photographs of a 1:1 model and visualizations of the element in the interior. In conclusion, the entire element is evaluated in terms of use in practice.

Bacterial bodies (colonies) can develop complex patterns of color and structure. These patterns may arise as a result of both colony-autonomous processes (self-patterning) and environmental influences, including those generated by neighbor bodies. We have studied the interplay of intra-colony signaling (self-patterning) and inter-colony influences in related clones of Serratia rubidaea on rich media. We show that the mutual influencing of colonies, present in a common morphospace, is communicated by at least two putative signals. Leishmania of the subgenus Viannia are protozoan parasites transmitted by phlebotomine sandflies (Diptera: Phlebotominae). They occur in tropical and subtropical areas in South America, where they cause cutaneous and mucocutaneous leishmaniasis. In this thesis, we studied developmental pattern of Viannia group and factors affecting its development within the sand fly gut. First, we investigated Leishmania braziliensis development within the Lutzomyia longipalpis digestive tract. Using GFP-labeled strain we demonstrated peripylar development: promastigotes escaped from the endoperitrophic space, colonized the hindgut and then migrated anteriorly. Four morphological forms were found within the Lu. longipalpis digestive tract: elongated nectomonads, short nectomonads, metacyclic promastigotes and paramastigotes. Furthermore, using the histological methods we demonstrated parasite attachment in pylorus region, while there were only free promastigotes in the midgut; neither form was found attached to the midgut epithelium. The next part was devoted to the effect of temperature on Viannia in Lu. longipalpis. We compared development of two closely related species L. peruviana and L. braziliensis at 20 řC and 26 řC. Úloha oxidu dusnatého (NO) v průběhu embryonálního vývoje pokožky drápatky vodní / The role of nitric oxide (NO) during Xenopus laevis embryonic epidermis development

Nitric oxide (NO) is an interesting molecule, which is involved in many important biological processes such as vasodilatation, neurotransmission, immune response and cell proliferation. This work presents the crucial role of NO during the Xenopus laevis embryonic epidermis development. The outer layer of the embryonic epidermis is composed of 4 cell types (small secretory cells, multi-ciliated cells, ionocytes and goblet cells). Embryonic epidermis composition reflects specialized epithelia such as a respiratory epithelium of mammals. Therefore, Xenopus embryonic epidermis has become a suitable model for the study of human mucosal and mucociliary epithelium and their defects. I found that NO is mainly produced in ionocytes and multi-ciliated cells by using molecular and immunohistochemical approaches. The study of molecular and cellular phenotype changes in embryos with inhibited NO production revealed the necessity of this molecule for correct formation and function of the mucociliary epithelium. NO inhibition caused reduction of specialized epidermal cell types (small secretory cells, ionocytes and multi-ciliated cells) and structural changes in multi-ciliated cells. The diploma thesis deals with adolescents who grew up in a dysfunctional family environment. The content of the literary-review part of the diploma thesis will be devoted to dysfunctional family systems in which the basic needs of the child are not met. It will also address the key areas of development for understanding the child's experience within individual ages of development up to adolescence. Attention will also be paid to coping strategies and child support resources during development. The research survey will consider the qualitative design, which will take place through an interview with the help of adolescents. The target group will be adolescents who grew up in a dysfunctional family environment. The aim of this research will be to describe the experience of adolescents in dysfunctional family systems. Thanks to the research part, the elaboration of this diploma thesis can be a tool for understanding the feelings and experiences of children who grew up in this environment. It can thus be applied in working with adolescents, but also with the parents of these children. At the same time, it can be a source for the prevention of mental decompensation and the possible development of more serious mental problems. Emphasis is placed mainly on the contradictory development in both countries, as the previously flourishing economy of Zimbabwe almost collapsed in the last decade, while the once poor Botswana gradually became Africa's miracle and role model for the optimal development policy. This thesis also seeks to propose measures that would help the distressed Zimbabwean economy to overcome this crisis and return on the growth path once again.
