L’optimisation du statut tissulaire en acides gras polyinsaturés à longues chaines n-3 (AGPILC n-3) fait partie des objectifs actuels en nutrition humaine. En effet, de nombreux effets bénéfiques sur la santé humaine ont été démontrés pour ces différents AGPILC n-3. Néanmoins, la bioconversion de ces dérivés à partir de leur précurseur alimentaire, l’acide α-linolénique, est limitée chez l’Homme et ne permet pas de subvenir aux besoins. Parmi cette famille d’acides, l’acide docosahexaénoïque (DHA) et de l’acide eicosapentaénoïque (EPA) ont été largement étudiés à l’inverse de ceux de l’acide docosapentaénoïque n-3 (DPA). Le premier objectif de ces travaux a été d’étudier l’impact d’une supplémentation nutritionnelle en DPA sur le métabolisme des acides gras et le métabolisme lipidique chez le rat sain. Pour cela, une méthode de purification du DPA à haute pureté et en grande quantité par chromatographie liquide a tout d’abord été mise en place. Ce résultat participe à faciliter la recherche future des effets in vivo du DPA dont la disponibilité commerciale est faible. Après sa supplémentation nutritionnelle, le DPA a été incorporé dans de multiples tissus et a impacté les compositions tissulaires en AGPILC n-3 de certains organes spécifiques comme le foie, le cœur, le poumon, la rate et le rein. Ainsi, la supplémentation en DPA pourrait être une source d’EPA, de DPA, et dans une moindre mesure de DHA. Enfin, comparé à une supplémentation en EPA ou en DHA, le DPA a été l’AGPILC n-3 le plus puissant pour améliorer la triglycéridémie et la cholestérolémie des animaux. Ces informations participeront certainement à une prise en compte plus importante du DPA tant dans les études nutritionnelles ainsi que dans l’installation des futures recommandations nutritionnelles. Le deuxième axe de recherche a porté sur la capacité d’une incorporation partielle de lipides laitiers dans le régime à augmenter le statut tissulaire en AGPI-LC n-3. L’incorporation de lipides laitiers a augmenté le statut en DHA cérébral et rétinien et le statut en DPA dans les autres tissus étudiés. De plus, l’induction de la conversion des AGPILC n-3 à partir de leur précurseur par les lipides laitiers a été complémentaire avec la supplémentation en DPA pour augmenter le statut tissulaire en AGPI-LC n-3. Ces résultats s’insèrent dans un contexte de nutrition infantile, période pendant laquelle l’incorporation des AGPI-LC n-3 est maximale dans les tissus nerveux. Ainsi, ces résultats apportent des données intéressantes tant pour les entreprises que pour les professionnels de santé pour recommander à la consommation des formules infantiles contenant des lipides laitiers. / The optimization of tissue status in n-3 long-chain polyunsaturated fatty acids (n-3 LCPUFA) is one of the current goals in human nutrition. Indeed, many beneficial effects on human health have been demonstrated for these different n-3 LCPUFA. However, the bioconversion of these derivatives from their food precursor, α-linolenic acid, is limited in humans and it does not meet the needs of the organism. Among this family of fatty acids, docosahexaenoic acid (DHA) and eicosapentaenoic acid (EPA) have been widely studied in contrast to those of n-3 docosapentaenoic acid (DPA). The first objective of this work was to study the impact of dietary supplementation with DPA on fatty acid metabolism and lipid metabolism in healthy rats. For this, a method for purifying DPA with high purity and in large quantities by liquid chromatography was first established. This result helps future research of the in vivo effects of DPA, poorly commercially available. After its nutritional supplementation, DPA was incorporated into multiple tissues and it has affected the n-3 LCPUFA tissue compositions of specific organs such as the liver, heart, lung, spleen and the kidney. Thus, supplementation with DPA could be a source of EPA, DPA, and to a lesser extent DHA. Finally, compared to supplementation with EPA or DHA, DPA was the most potent n-3 LCPUFA to improve triglyceridemia and cholesterolemia in animals. This information will certainly contribute to a greater consideration of DPA both in nutritional studies and in the direction of future nutritional recommendations. The second area of ​​research focused on the ability of a partial incorporation of dairy lipids in the diet to increase the tissue status in n-3 LCPUFA. Incorporation of dairy lipids in the diet increased DHA status in brain and retina and DPA status in the other studied tissues. In addition, the induction of conversion of n-3 LCPUFA from their precursor by dairy lipid was complementary with the DPA supplementation to increase n-3 LCPUFA status in tissues. These results fit into a context of infant nutrition, during which the incorporation of n-3 LCPUFA is maximal in nervous tissues. Thus, these results provide interesting data for both companies and health professionals to recommend infant formulas containing dairy lipids for consumption.

Eco-valorisation de la plante Kniphofia uvaria : de la plante à la galénique / Eco-valuation of the Kniphofia uvaria plant : from the plant to the galenic form

À l’heure où l’intégration des enjeux environnementaux dans le développement de procédés éco-efficients joue un rôle essentiel dans le moteur de l’innovation responsable, la chimie verte est devenue l’un des sujets de préoccupation majeure. Ainsi, le développement de nouveaux procédés éco-respectueux pour la production d’ingrédients naturels issus de matières premières végétales renouvelables est devenu une démarche incontournable dans le modèle de recherche. L’objectif de cette thèse a consisté au développement d’une stratégie d’éco-valorisation innovante employant les fluides sub/supercritiques pour l’extraction, la caractérisation, la production et l’imprégnation sur support cosmétique de produits naturels d’origine végétale. Pour cela, nous avons utilisé comme modèle végétal : les graines oléagineuses de la plante Kniphofia uvaria, sélectionnée pour des applications cosmétiques grâce à ses propriétés bioactives antioxydantes et anti-âge. Dans un premier temps, le développement de méthodes complémentaires en SFC ainsi que le développement du couplage SFC-MS a été réalisé à l’aide de la source APCI afin d’identifier les molécules responsables des activités bioactives des graines de Kniphofia uvaria. Ainsi, le développement d’un système hybride (U)HPLC/SFC-HRMS a été réalisé afin de mettre en place ce couplage. Des optimisations en termes de proportion et nature de solvant make-up ainsi qu’un travail au niveau des paramètres SFC et MS ont été faits afin de d’améliorer la sensibilité et la spécificité des analyses lipidiques. Dans un second temps, nous nous sommes attachés au développement d’une stratégie d’enrichissement en composés bioactifs à l’aide des méthodes : SFE et CPC. Ainsi, en SFE, des optimisations en termes de température, pression, nature/proportions de co-solvant dans le fluide ont été réalisées alors qu’en CPC, des optimisations au niveau de l’injection ont été faites. Des conditions optimales pour le fractionnement sélectif des anthraquinones et des triglycérides ont été déterminées en SFE et CPC. Dans un dernier temps, ce travail a consisté à développer un couplage en-ligne pour extraire et imprégner sélectivement sur silice cosmétique : les anthraquinones. Le développement et l’optimisation de ce procédé en-ligne ont été réalisés à l’échelle du laboratoire et ont démontré la faisabilité de ce couplage ainsi qu’un intérêt certain pour l’obtention de produits naturels sous une première forme galénique, destinée à une future incorporation dans la formulation de cosmétiques. / Nowadays, green chemistry is a great challenge. It seeks innovation in the development of eco-efficient processes. The production of natural products from renewable materials by these new environmentally friendly processes is more and more used. The aim of this Ph.D thesis is to develop an eco-valuation strategy to extract, characterize, produce and impregnate natural products onto a cosmetic support using sub/supercritical fluids. Consequently, we used oleaginous plant seeds from Kniphofia uvaria as a plant model, which was selected for its interesting cosmetic properties such as antioxidant or anti-ageing. Firstly, the SFC-MS hyphenation with the APCI as an ionization source was developed to screen bioactive molecules; responsible of cosmetic properties. This coupling was performed by the hybrid combination of (U)HPLC/SFC-HRMS. Various optimizations in terms of the solvent make-up (nature and proportion), modulation with SFC and MS parameters were carried out in order to improve sensitivity and selectivity of lipid analysis. Secondly, an enrichment strategy to concentrate bioactive compounds in the final extract was developed by SFE and CPC. Thus, in SFE, experimental parameters (temperature, pressure, nature/proportion of the modifier in the CO2 fluid) were optimized while in CPC, the injection optimization was realized. Methods for the selective fractionation of anthraquinones and triglycerides were obtained in CPC and SFE. Finally, an on-line sub/supercritical extraction-impregnation process was developed to extract and for simultaneously impregnating anthraquinones onto a cosmetic silica. Development and optimization of this process was realized on a laboratory scale. Consequently, this study demonstrated the feasibility of this concept and it presents a great interest to provide natural products as a galenic form, which could be used in the cosmetic formulation.

Neste contexto objetivou-se, com este trabalho, a elaboração de misturas especiais de óleos vegetais para fins alimentícios, com base nos óleos de amendoim, gergelim, maracujá e soja. Os óleos de amendoim e gergelim como alternativa por serem culturas produzidas no Brasil, constituem uma fonte de ácidos graxos e antioxidantes tão pouco explorados na alimentação dos brasileiros, tal como o óleo de maracujá como co-produto, descartado pela indústria de suco e ainda o óleo de soja cuja matéria-prima abundante, tornam o produto acessível aos consumidores de todas as classes sociais. Os óleos de amendoim e gergelim foram extraídos de sementes das cultivares BRS-Havana e BRS-Seda, respectivamente, cedidas pela Embrapa Algodão enquanto o óleo de maracujá foi adquirido no comércio de São Paulo e o de soja no de Campina Grande. As misturas foram elaboradas com base em uma matriz de planejamento experimental e, em seguida, submetidas às análises de composição de ácidos graxos e ensaios físico-químicas (umidade, índice de acidez, índice de iodo, índice de refração, densidade e viscosidade) segundo a metodologia do IAL (2008). As medidas obtidas foram utilizadas para estudar e otimizar a elaboração das misturas de óleos vegetais e como variáveis para predição por espectrometria no infravermelho próximo (NIR) foram utilizados os mínimos quadrados parciais (PLS) como metodologia de calibração multivariada. Os resultados foram submetidos à análise de variância (ANOVA), à análise de variância multivariada (MANOVA), à análise de correlação canónica e à análise de correlação de Pearson, pelo software SAS 9.1.3; na análise de componentes principais (PCA) e regressão por mínimos quadrados parciais (PLS) utilizou-se o software Uncrambler 9.8®. Concluiu-se que a mistura equivalente a 40% de óleo de amendoim, 20% de óleo de gergelim, 20% de óleo de maracujá e 20% de óleo de soja apresentou-se como a melhor mistura para fins alimentares, com base na composição de ácidos graxos. Os métodos de predição por NIR foram eficazes para a estimativa não destrutiva, rápida, de baixo custo e direta, dos ácidos graxos palmítico, esteárico, oleico, linoleico, araquídico e linolênico e para as medidas físico-químicas de densidade, índice de refração, viscosidade, índice de acidez e índice de iodo. / Brazil has great potential to produce vegetable oils as a source of raw materiais for food and energy. But in the last five years has been the largest importer of olive oil in the worid due to its health benefits and wide divulgation. In this context, the aim of this work was the development of special blends of vegetable oils as an alternative to olive oils based on peanut, sesame, soy and passion fruit. The peanut and sesame oils because they are a source of fatty acids and antioxidants in the diet of little explored in Brazil, passion fruit oil as co-product of the juice industry, and soybean oil abundant raw material which makes the product available to consumers of ali social classes. The peanut and sesame oils were extracted from seeds of BRS-Seda and BRS-Havana, while passion fruit oil was purchased commercially of city São Paulo-SP and trade soybean in the city of Campina Grande-PB. The mixtures were prepared based on an array of experimental design and then submitted to analysis of fatty acid composition and physico-chemical (moisture, acid value, iodine value, refractive index, density and viscosity). Moreover, were used as reference variables for prediction via infrared spectroscopy (NIR) using the partial least squares (PLS) and multivariate calibration methodology. The results were subjected to analysis of variance (ANOVA), multivariate analysis of variance (MANOVA), canonical correlation analysis and Pearson correlation analysis by SAS 9.1.3 software, the principal component analysis (PCA) and Partial least squares regression (PLS) was used Uncrambler 9.8 ® softwarlt was concluded that the mixture equivalent to 40% peanut oil, 20% sesame oil, 20% passion fruit oil, and 20% soybean oil was presented as the best mix for food composition based on fatty acids. The prediction by NIR methods were effective for estimating non-destructive, rapid, inexpensive and direct fatty acids palmitic, stearic, oleie, linoleic, linolenic and arachidic and physico-chemical measurements of density, refractive index, viscosity, acid value and iodine value.

Sustainability Evaluation of Transformer Insulating Liquid against UN SDGs

To achieve a shared blueprint for peace and prosperity for people and the planet, the United Nations has defined the 2030 Agenda for Sustainable Development, including economic, social, and environmental sustainability as three pillars. This thesis work is aimed at building a sustainability evaluation framework to investigate the product, which in this study is transformer insulating liquid. What’s more, this study focuses on the sustainability evaluation of the extraction step of the raw material which is crude oil and vegetable oil for insulating liquids. The evaluation method is based on the triangular fuzzy number variables, which represent the ambiguity of linguistic variables. Meanwhile, the sustainable development goals are discussed and developed into feasible evaluation criteria. According to the defined criteria, the performance of each indicator is taken from published literature, reports, and official organization. After that, the sustainability index of each oil is calculated and compared, thus a general sustainable result is given. The result shows that mineral oil is more sustainable than vegetable oil only in economic sustainability, while in other aspects including the overall aspect vegetable oil is more sustainable than mineral oil. Among that, mineral oil performs most weakly in environmental sustainability, while vegetable oil is in economic sustainability. Furthermore, the framework designed is expected to be used to evaluate the entire life cycle of a product and can be referred to in future supply chain improvement, since more indexes can be developed based on the mathematics fuzzy number calculation principle. / För att uppnå en gemensam plan för fred och välstånd för människor och planeten har FN definierat 2030-agendan för hållbar utveckling, inklusive ekonomisk, social och miljömässig hållbarhet, som tre pelare. Detta examensarbete syftar till att bygga ett ramverk för hållbarhetsutvärdering för att undersöka produkten, som i denna studie är transformatorisoleringsvätska. Dessutom fokuserar denna studie på hållbarhetsutvärderingen av utvinningssteget av råmaterialet som är råolja och vegetabilisk olja för isolerande vätskor. Utvärderingsmetoden är baserad på de triangulära luddiga talvariablerna, som representerar tvetydigheten hos språkliga variabler. Samtidigt diskuteras målen för hållbar utveckling och utvecklas till genomförbara utvärderingskriterier. Enligt de definierade kriterierna är prestandan för varje indikator hämtad från publicerad litteratur, rapporter och officiell organisation. Därefter beräknas och jämförs hållbarhetsindexet för varje olja, vilket ger ett generellt hållbart resultat. Resultatet visar att mineralolja är mer hållbar än vegetabilisk olja endast i ekonomisk hållbarhet, medan i andra aspekter inklusive den övergripande aspekten är vegetabilisk olja mer hållbar än mineralolja. Bland annat presterar mineralolja sämst i miljömässig hållbarhet, medan vegetabilisk olja har ekonomisk hållbarhet. Dessutom förväntas det utformade ramverket användas för att utvärdera hela livscykeln för en produkt och kan hänvisas till i framtida förbättringar av försörjningskedjan, eftersom fler index kan utvecklas baserat på matematikens luddiga talberäkningsprincip.

Experimental Study of the Fuel Effect on Diffusion Combustion and Soot Formation under Diesel Engine-Like Conditions

[ES] Las emisiones de CO2 en el sector transporte se han incrementado considerablemente durante los últimos años debido al desarrollo económico mundial. El crecimiento de las flotas de transporte, junto con otros factores, ha contribuido al desequilibrio del ciclo de carbono del planeta. Es por ello que el CO2 se considera un gas de efecto invernadero de origen antropogénico que debe ser reducido para evitar el calentamiento global. Las estrategias para reducir el CO2 en el sector transporte están enfocadas a la electrificación y al uso de combustibles neutros o de bajo impacto al ambiente. Sin embargo, una efectiva implementación de esta última requiere un profundo entendimiento de la combustión con tales combustibles. En la presente tesis doctoral, se ha caracterizado experimentalmente la combustión de diferentes tipos de combustibles, entre ellos, algunos de bajo impacto en emisiones de CO2 como lo son el Aceite Vegetal Hidrotratado (HVO) y dos éteres de oximetileno (OME1 y OMEx).Además, por su potencial en la reducción de contaminantes se han evaluado mezclas de diésel y gasolina y de HVO y Gas Licuado de Petróleo (LPG), lo que requirió adecuar el sistema de inyección para evitar la evaporación a lo largo de la línea. Todos estos combustibles y mezclas han sido inyectados con una tobera mono-orificio y han sido evaluados mediante técnicas de visualización a alta velocidad bajo diferentes condiciones termodinámicas típicas de un motor de encendido por compresión operando en condiciones de combustión a baja temperatura, en instalaciones con accesos ópticos. Se ha analizado el efecto de las propiedades físico químicas de estos combustibles y mezclas sobre los parámetros característicos de un chorro como lo son la longitud líquida y la penetración de vapor. La combustión ha sido evaluada mediante la caracterización del tiempo de retraso, de la liberación de calor y la longitud del despegue de la llama, que viene condicionada por el proceso de mezcla. Igualmente, el estudio de la formación de hollín en función de las propiedades del combustible y de las características del proceso de mezcla, representa un aporte importante de esta tesis. En adición a los beneficios en reducción de CO2 que brindan los combustibles y mezclas utilizados en este estudio, estos también redujeron la formación de hollín en la cámara de combustión, destacándose entre ellos los combustibles oxigenados, especialmente el OMEx que además de no formar hollín, fue el de mayor reactividad en todas las condiciones de operación evaluadas. / [CA] Les emissions de CO2 en el sector transport s'han incrementat considerablement durant els últims anys a causa del desenvolupament econòmic mundial. El creixement de les flotes de transport, juntament amb altres factors, ha contribuït al desequilibri del cicle de carboni del planeta. És per això, que el CO2 es considera un gas d'efecte hivernacle d'origen antropogènic que ha de ser reduït per evitar l'escalfament global. Les estratègies per reduir el CO2 dins el sector transport, estan enfocades a l'electrificació i a l'ús de combustibles neutres o de baix impacte ambiental. No obstant això, una efectiva implementació d'aquesta última, requereix un profund coneixement del procés de combustió d'aquests combustibles. En la present tesi doctoral, s'ha caracteritzat experimentalment la combustió de diferents tipus de combustibles, entre ells, alguns de baix impacte en emissions de CO2 com són l'Oli Vegetal Hidrotratat (HVO) i dos èters de oximetileno (OME1 i OMEx) .A més , degut al seu alt potencial en la reducció de contaminants, s'han avaluat mescles de dièsel i gasolina, i de HVO i Gas Liquat de Petroli (LPG), el que va requerir adequar el sistema d'injecció per evitar l'evaporació al llarg de la línia. Tots aquests combustibles i mescles han estat injectats amb una tovera mono-orifici i han estat avaluats mitjançant tècniques de visualització a alta velocitat a través dels accessos òptics del que disposa la instal·lació. Les diferents condicions termodinàmiques utilitzades son típiques d'un motor d'encesa per compressió operant en condicions de combustió a baixa temperatura. S'ha analitzat l'efecte de les propietats fisicoquímiques d'aquests combustibles i de les mescles sobre els paràmetres característics d'un raig com són la longitud líquida i la penetració de vapor. La combustió ha estat avaluada mitjançant la caracterització del temps de retard, de l'alliberació de calor i de la longitud de l'enlairament de la flama que ve condicionada pel procés de mescla. A més, l'estudi de la formació de sutge en funció de les propietats del combustible i de les característiques del procés de mescla, representa una aportació important d'aquesta tesi evidenciant que a més dels beneficis en reducció de CO2 que brinden tots aquests combustibles i mescles, també varen reduir la formació de sutge a la cambra de combustió, destacant-se entre ells els combustibles oxigenats, especialment el OMEx, que a més de no formar sutge, va ser el de major reactivitat en totes les condicions d'operació avaluades. / [EN] CO2 emissions in the transport sector have increased considerably in recent years due to global economic development. The growth of transport fleets, along with other factors, has contributed to the imbalance of the planet's carbon cycle. For that, CO2 is considered a greenhouse gas from anthropogenic origin that must be reduced to avoid global warming. Strategies to reduce CO2 in the transport sector are focused on electrification and the use of neutral fuels or those with a low impact on the environment. However, an effective implementation of the latter requires a deep understanding of the combustion with those fuels. In this doctoral thesis, the combustion of different types of fuels has been experimentally characterized, including some with low impact on CO2 emissions such as Hydrotreated Vegetable Oil (HVO) and two oxymethylene ethers (OME1 and OMEx). Furthermore, due to their potential in reducing pollutants, blends of diesel and gasoline and HVO and Liquefied Petroleum Gas (LPG) have also been evaluated, which required adapting the injection system to avoid evaporation along the injection line. All these fuels and blends have been injected with a single-hole nozzle and they have been evaluated using high speed visualization techniques under different thermodynamic conditions typical of a compression ignition engine operating under low-temperature combustion conditions in installations with optical accesses. The effect of the physical-chemical properties of these fuels and blends on the characteristic parameters of a jet, such as the liquid length and the vapor penetration, has been analyzed. Combustion has been evaluated by characterizing the ignition delay, the heat release and the flame Lift-off length that is conditioned by the mixing process.
