Par?metros bioqu?micos e hormonais durante o ciclo reprodutivo de Vanellus chilensis (Aves, Charadriiformes)

Existe um n?mero grande de trabalhos sobre a avifauna urbana, mas nenhum utilizando a ecofisiologia como ferramenta de trabalho no Brasil. Este estudo teve como objetivo analisar em dois ambientes, um com caracter?sticas rurais e outro inserido dentro de um centro urbano, as varia??es dos n?veis plasm?ticos de: corticosterona, testosterona e metab?litos. Os animais foram sempre capturados ? noite e amostras de sangue e dados de muda foram coletados. Os metab?litos (glicose, lip?dios totais, triglicer?deos, colesterol, HDL, LDL, VLDL, glicerol, ?cido ?rico e prote?nas totais) foram quantificados no plasma utilizando-se espectrofotometria; a corticosterona e a testosterona determinadas por radioimunoensaio. Nos jovens da ?rea 1 verificamos um predom?nio de f?meas (2,7 f?meas: 1 macho); na ?rea 2 esta rela??o se inverte (2 machos: 1 f?mea), o que parece relacionar-se com os maiores n?veis de testosterona e menores de corticosterona observados nas f?meas (per?odo 1) na ?rea 2.Comparando-se as ?reas de captura, os maiores n?veis de corticosterona plasm?tica foram observados nos jovens da ?rea 2 sugerindo um maior estresse ambiental que pode estar relacionado com uma maior press?o de preda??o, e/ou pela intensa atividade e presen?a humana e/ou pela competi??o intra-espec?fica. Nos jovens, ao longo do estudo, nas duas localidades estudadas n?o observamos varia??es para os n?veis dos metab?litos plasm?ticos, com exce??o dos triglicer?deos dos jovens da ?rea 2. Contudo, ao compararmos a ?rea 1 com a ?rea 2 se observa um comportamento diferencial, ao longo do desenvolvimento dos jovens na concentra??o plasm?tica de triglicer?deos, HDL, VLDL e prote?nas totais; os demais par?metros analisados (glicose, lip?dios totais, colesterol, LDL, glicerol e ?cido ?rico) n?o apresentam um comportamento diferencial. Nos adultos houve diferen?a nos n?veis de corticosterona nas f?meas da ?rea 2, mas em ambos os sexos e localidades observamos uma tend?ncia de diminui??o dos n?veis hormonais ao longo do per?odo reprodutivo. Os resultados mostraram diferen?as nos n?veis de testosterona entre os machos e as f?meas da ?rea 1 quando comparadas ? ?rea 2 durante o per?odo de estudo; por?m, entre os per?odos da esta??o reprodutiva em cada localidade houve diferen?as somente nas f?meas.Na ?rea 1, o per?odo reprodutivo foi cerca de 2 vezes menor que na ?rea 2 e as f?meas apresentaram um aumento significativo de todos os metab?litos plasm?ticos, com exce??o do glicerol, j? os machos mostram um aumento da glicose, colesterol, HDL e LDL principalmente no per?odo 3. Na ?rea 1 observamos um aumento gradual dos n?veis de glicose no plasma atingindo valores m?ximos, tanto em machos como em f?meas, no per?odo 3, per?odo caracterizado pela presen?a de comportamentos de defesa e cuidado dos filhotes, sendo tamb?m, o per?odo que antecede a muda das penas de v?o. J? na ?rea 2 n?o foi observada varia??o nos n?veis de metab?litos plasm?ticos, exceto para a glicose em machos e triglicer?deos, glicerol e ?cido ?rico em ambos os sexos. Perfil que talvez possa ser explicado pela maior disponibilidade de alimento e/ou menor competi??o e/ou maior investimento na procura do alimento. Os resultados sugerem que Vanellus chilensis capturados na ?rea 2 apresentam um padr?o adaptativo em rela??o ? a??o humana e as rela??es bi?ticas e abi?ticas desta ?rea. Ao final deste trabalho pode-se concluir que houve diferen?a entre as duas ?reas de estudo, sendo os n?veis de corticosterona, de testosterona, de metab?litos plasm?ticos e a dura??o do per?odo reprodutivo influenciados pela a??o antr?pica.

Relações biogeográficas entre a avifauna de florestas de altitude no Nordeste do Brasil

No. of bitstreams: 1 arquivototal.pdf: 14079837 bytes, checksum: 75b0b495dcbefcccc93b45421a059bf2 (MD5) Previous issue date: 2014-11-21 / Coordenação de Aperfeiçoamento de Pessoal de Nível Superior - CAPES / A extensa área da região Nordeste do Brasil (1.540.827 km2) apresenta grandes variações no relevo, com altitudes inferiores aos 300 m na Depressão Sertaneja até áreas acima dos 1000m, como no Planalto da Borborema (Araujo et al. 1998). Essa variedade altitudinal é refletida na complexidade climática e conseqüentemente na variabilidade de seus tipos vegetacionais. Algumas dessas formações são áreas florestais, que cobrem desde porções litorâneas até áreas altas incluídas no domínio florístico-vegetacional da Caatinga. Os ambientes florestais de altitude encontrados na área da Caatinga podem estar relacionadas aos domínios da Floresta Atlântica, nos chamados Brejos de Altitude (Andrade-Lima, 1982), ou ainda das Florestas Neotropicais Estacionais Secas (FNES). As FNES são ecossistemas essencialmente dominados por árvores, com copa contínua ou quase contínua e na superfície do solo as ervas são os elementos de menor quantidade, quando comparadas a vegetação de savanas (Monney et al. 1995, Pennington et al. 2006). Segundo Gentry (1995) e Graham & Dilcher (1995), essas florestas secas ocorrem onde a precipitação é menor que 1600 mm/ano, com um período de no mínimo 5-6 meses recebendo menos que 100 mm. Rodal et al. (2008) demonstraram que, no nordeste do Brasil, as florestas secas estacionais estão localizadas onde o limite máximo de precipitação é 1000 mm e, no mínimo, oito meses do ano recebem menos 100 mm. Essas formações podem ser encontradas atualmente em margens de rios, como as matas secas do rio São Francisco e em áreas serranas, constituindo florestas de altitude em meio à vegetação arbustiva circundante, no domínio das caatingas. Nos estados de Pernambuco e Paraíba, o planalto da Borborema propicia a formação de microclimas com temperaturas mais baixas e maior precipitação que nas áreas adjacentes (Andrade- Lima, 1982). Os enclaves florestas em meio a Caatinga são considerados refúgios atuais de espécies dos ambientes florestais que teriam dominado o centro da região Nordeste em um período geológico mais úmido que o atual (Andrade-Lima, 1982). A teoria de refúgios pleistocênicos foi apresentada ainda nas décadas de 1960-1970 (eg. Haffer, 1969; Vanzolini & Williams, 1970; Brown e Ab Sáber, 1979) e estudos com a fauna mésica desses ambientes corroboraram com esta proposta (e.g. Mares et al. 1985). No entanto, análises de distribuição de táxons realizadas mais recentemente (eg. Santos et al. 2007, Roda et al. 2008, Carvalho 2013), bem como filogenias moleculares de alguns táxons relacionados a essas florestas, estão disponíveis apenas na última década e, portanto, outros sinais históricos que podem suportar, refutar ou complementar as histórias clássicas são possíveis de ser averiguados. Estudos comparativos da composição da avifauna associada aos ambientes florestais e estudos filogeográficos de espécies que ocorrem nas florestas de altitude no nordeste podem contribuir para explicar relações entre essas formações florestais. Visando testar a hipótese de múltiplas origens para as florestas de altitude utilizaremos três ferramentas: (1) análises da composição de comunidades de aves dependentes de ambientes florestais que ocorrem em diferentes florestas de altitude através de métodos cladísticos; (2) Análise do padrão de distribuição de espécies com ocorrência em florestas de altitude no nordeste setentrional, discutindo seus padrões de distribuição e histórias filogenéticas de taxa a elas relacionados; (3) e estudo filogeográfico de uma espécie de ave Passeriforme que ocorre nessas florestas. Para isso pretende-se nesse projeto: (a) verificar a composição da avifauna de florestas de altitude no nordeste brasileiro, através de amostragens de campo, visitas a museus e recuperação de dados publicados; (b) comparar a composição da avifauna, utilizando um misto da Análise de Parcimônia e Endemismo (PAE) e da Análise Cladística de Distribuição e Endemismo (CADE), e (c) obter filogegrafia de Arremon taciturnus, espécies de ave Passeriforme associada a tais ambientes florestados.

Revisão taxonômica de Dorisiana METCALF, 1952 (Hemiptera, Auchenorrhyncha, Cicadidae, Cicadinae, Fidicinini)

Cicadoidea is currently divided into Tettigarctidae and Cicadidae, and this into three subfamilies: Cicadinae, Cicadettinae and Tibicininae. Cicadinae has many neotropical tribes, Fidicinini, one of the most representative, is divided into Fidicinina and Guyalnina and has 16 genera. Into Fidicinina, Fidicina and Fidicinoides are the ones with highest number of described species and none of the other genera of the tribe has a taxomic revision, being absent publications that establish characters to distinguish them. Into Guyalnina, Dorisiana has economic importance as pest of coffee and also taxonomic problems, besides the lack of morphological characters needed for its determination, which is increased by the lack of a review covering all genera present in the tribe. The general goal of this work was execute the taxonomic revision of Dorisiana species improving the knowledge about the taxon, with the specific objective to analyze the morphology of the genus from deposited specimens of collections of national and international institutions; describe the morphological species, including photographs and ilustrations, mostly of the male genitalia; produce a pictorial dichotomous key to distinguish the Dorisiana species. Therefore, of the 16 species of the genus, previously known, four were synonymized, two new combinations and two new species were described, totaling 16 valid species, covered in the pictorial dichotomous key.From these results and studies that led to it, is perceived a great lack of group by works that delimit characters needed for your determination, for genus and species, which is also shared by others neotropical tribes. Starting of these assumptions suggested in the future more studies on characters for a cladistics analysis of the Dorisiana species and the continuity of the studies with the members of the tribe to establish reliable characters in recognition of evolutionary lineages. / Os cidadídeos são insetos conhecidos popularmente como cigarras, sendo facilmente reconhecidos no seu ambiente devido ao som emitido pelos machos. Cicadoidea se encontra atualmente dividida em Tettigarctidae e Cicadidae e, esta última em três subfamílias: Cicadinae, Cicadettinae e Tibicininae. Cicadinae comporta muitas tribos neotropicais, sendo uma das mais representativas Fidicinini, subdividida em Fidicinina e Guyalnina e contendo 16 gêneros. Em Fidicinina, Fidicina e Fidicinoides são os únicos que apresentam maior número de espécies descritas e nenhum dos demais gêneros da tribo possui uma revisão taxômica, sendo ausentes publicações que estabeleçam caracteres para distingui-los. Dentro de Guyalnina, Dorisiana apresenta importância econômica como praga do cafeeiro e também problemas taxonômicos, além da carência de caracteres morfológicos necessários para sua determinação, o que é acrescido pela falta de uma revisão que contemple todos os gêneros presentes na tribo. O objetivo geral desse trabalho foi realizar a revisão taxonômica das espécies de Dorisiana aprimorando os conhecimentos acerca do táxon, tendo como objetivos específicos analisar a morfologia das espécies do gênero a partir de exemplares depositados em coleções de instituições nacionais e estrangeiras; descrever as espécies morfologicamente, incluindo fotografias e desenhos, principalmente das peças da genitália do macho; produzir uma chave dicotômica pictórica para determinação das espécies. Para isso, a partir da análise dos exemplares encontrados nas coleções nacionais e pelo envio de fotografias de material das instituições estrangeiras, foi realizado o estudo morfológico dos espécimes e ilustrações das principais estruturas para a distinção das espécies de Dorisiana.Dessa forma, das 16 espécies do gênero anteriormente conhecidas, quatro foram sinonimizadas, duas novas combinações e duas novas espécies foram descritas, totalizando 16 válidas contempladas na chave dicotômica pictórica. A partir desses resultados, e dos estudos que levaram a ele, percebe-se uma grande carência do grupo por trabalhos que delimitem caracteres necessários para a sua determinação, tanto em nível de gênero quanto de espécie, o que também é compartilhado por outras tribos da região neotropical. Partindo-se desse presuposto, sugere-se no futuro o levantamento de mais caracteres objetivando uma análise cladística de Dorisiana e a continuidade dos estudos com representantes da tribo para se estabelecer caracteres confiáveis no reconhecimento de linhagens evolutivas.

Demersal communities in the Mediterranean Sea: a case study of Triglidae (Osteichthyes, Scorpaeniformes) on the conservation and sustainable use of marine resources

An appropriate management of fisheries resources can only be achieved with the continuous supply of information on the structure and biology of populations, in order to predict the temporal fluctuations. This study supports the importance of investigating the bio-ecology of increasingly exploited and poorly known species, such as gurnards (Osteichthyes, Triglidae) from Adriatic Sea (Mediterranean), to quantify their ecological role into marine community. It also focuses on investigate inter and intra-specific structuring factor of Adriatic population. These objectives were achieved by: 1) investigating aspects of the population dynamics; 2) studying the feeding biology through the examination of stomach contents; 3) using sagittal otoliths as potential marker of species life cycle; 4) getting preliminary data on mDNA phylogeny. Gurnards showed a specie-specific “critical size” coinciding with the start of sexual maturity, the tendency to migrate to greater depths, a change of diet from crustaceans to fish and an increase of variety of food items eaten. Distribution of prey items, predator size range and depth distribution were the main dimensions that influence the breadth of trophic niche and the relative difference amongst Adriatic gurnards. Several feeding preferences were individuated and a possible impact among bigger-size gurnards and other commercial fishes (anchovy, gadoids) and Crustacea (such as mantis prawn and shrimps) were to be necessary considered. Otolith studies showed that gurnard species have a very fast growth despite other results in other areas; intra-specific differences and the increase in the variability of otolith shape, sulcus acusticus shape, S:O ratios, sulcus acusticus external crystals arrangement were shown between juveniles and adults and were linked to growth (individual genetic factors) and to environmental conditions (e.g. depth and trophic niche distribution). In order to facilitate correct biological interpretation of data, molecular data were obtained for comparing morphological distance to genetic ones.

Identificazione di possibili unita' di stock di Anguilla anguilla mediante analisi microchimica e morfometrica degli otoliti / Identification of possible stock units of Anguilla anguilla through microchemical and morphometric otolith analyses

L’anguilla europea, è una specie eurialina catadroma con un complesso ciclo biologico: l’area di riproduzione, unica, si trova molto distante da quella di distribuzione. La specie necessita di una gestione dello stock a fini conservazionistici. Il problema è europeo: lo stock è unico, distribuito in Europa e nell’Africa settentrionale, si riproduce in Atlantico ed è panmittico. C’è preoccupazione per il declino del reclutamento e delle catture di adulti. Lo scopo del progetto è di individuare possibili unità di stock nella penisola italiana. La ricerca è basata sullo studio degli otoliti mediante analisi morfometrica e microchimica. I contorni degli otoliti sono sottoposti ad analisi ellittica di Fourier per individuare eventuali gruppi. Gli otoliti sono stati levigati per effettuare: letture d’età, indagini microstrutturali al SEM delle fasi larvali, analisi microchimiche LA-ICP-MS del nucleo, studiarne l’origine e valutare l’ambiente di sviluppo. Le indagini morfometriche mostrano evidenti pattern ontogenetici, ma non legati ocorrelati alla località, sesso o anno di nascita. Le indagini microstrutturali hanno evidenziano l’alto contenuto organico nucleare, un pattern comune di crescita ed eventi chiave delle fasi larvali, con una media di 212 anelli giornalieri. La microchimica rivela che le larve si sviluppano in acque salate fino alla metamorfosi, poi migrano verso acque meno salate. Le analisi su campioni nati nello stesso anno, evidenziano due gruppi: individui di rimonta naturale e individui di ripopolamento. I profili nucleo bordo evidenziano la permanenza a salinità intermedie degli adulti. L’attività di ricerca si è dimostrata proficua dal punto di vista tecnico con la messa a punto di protocolli innovativi e con forti ricadute sulla riduzione dei tempi e costi d’analisi. Il debole segnale di possibili unità di stock andrà verificato in futuro mediante analisi più dettagliate discriminando meglio la storia di ogni singolo individuo. / The European eel is euryhaline catadromous with a complex life cycle: spawning area, unique, is far from that of distribution. The species needs a stock management and its conservation. The problem is European: the stock is unique, distributed in Europe and North Africa, spawning in Atlantic Sea and is panmictic. There is concern about the decline of recruitment and catches of adults. The purpose of the project is to identify possible units of stock in the Italian peninsula. The research is based on the study of otoliths by morphometric and microchemical analyses . The otoliths contours are analyzed by elliptic Fourier function to discover any groups. Otoliths were polished to make: ageing, SEM microstructural investigations of larval stages, LA-ICP-MS microchemical analysis of the core to study the origin and along a transect core to edge to evaluate the development environment. Morphometric investigations show obvious ontogenetic pattern, but no location, gender or year of birth correlation. The microstructural investigations showed high organic content in the core, a common pattern of growth and key events of the larval stages, with an average of 212 daily rings. The microchemical reveals that the larvae develop into salt water until metamorphosis, then migrate to waters less salty. Analyses of samples born in the same year, showed two groups: wild and restocked. The profiles confirm that the adult stay in brackishwater. The development of innovative protocols and improved technics have guaranteed a strong reduction of time and cost of analysis. The weak signal of possible stock units will be further tested through more detailed analysis discriminating about the life history of each individual.

Transposable elements dynamics in taxa with different reproductive strategies or speciation rate

In recent years the advances in genomics allowed to understand the importance of Transposable Elements (TE) in the evolution of eukaryotic genomes. In this thesis I face two aspects of the TE impact on the in the animal kingdom. The first part is a comparison of the dynamics of the TE dynamics in three species of stick-insects of the Genus Bacillus. I produced three random genomic libraries of 200 Kbps for the three parental species of the taxon: a gonochoric population of Bacillus rossius (facultative parthenogenetic), Bacillus grandii (gonochoric) and Bacillus atticus (obligate parthenogenetic). The unisexual taxon Bacillus atticus does not shows dramatic differences in TE total content and activity with respect to Bacillus grandii and Bacillus rossius. This datum does not confirm the trend observed in other animal models in which unisexual taxa tend to repress the activity of TE to escape the extinction by accumulation of harmful mutations. In the second part I tried to add a contribute to the debate initiated in recent years about the possibility that a high TE content is linked to a high rate of speciation. I designed an evolutionary framework to establish the different rate of speciation among two or more taxa, then I compared TE dynamics considering the different rates of speciation. The species dataset comprises: 29 mammals, four birds, two fish and two insects. On the whole the majority of comparisons confirms the expected trend. In particular the amount of species analyzed in Mammalia allowed me to get a statistical support (p<0,05) of the fact that the TE activity of recently mobilized elements is positively related with the rate of speciation.

Characterization of mitochondrial genomes in bivalve species with doubly uniparental inheritance of mitochondria

Many bivalve species possess two distinct mtDNA lineages, called F and M, respectively inherited maternally and paternally: this system is called doubly uniparental inheritance (DUI). The main experimental project of my PhD was the quantification of the two mtDNAs during the development of the DUI species Ruditapes philippinarum, from early embryos to sub-adults, using Real-Time qPCR. I identified the time interval in which M mtDNA is lost from female individuals, while it is retained in males (which are heteroplasmic through all of their life cycle). The results also suggested absence of mtDNA replication during early embryogenesis, a process constituting a bottleneck that highly reduces the copy number of mtDNA molecules in cells of developing larvae. In males this bottleneck may produce cells homoplasmic for M mtDNA, and could be considered as a first step of the segregation of M in the male germ line. Another finding was the characterization, in young clams approaching the first reproductive season, of a significant boost in copy number of F mtDNA in females and of M in males. Given the age of animals in which this mtDNA-specific growth was observed, the finding could probably be the outcome of the first round of gonads and gametes production. Other lines of research included the characterization of the unassigned regions in mt genomes of DUI bivalves. These regions can harbor signals involved in the control of replication and/or transcription of the mtDNA molecule, as well as additional open reading frames (ORFs) not related to oxidative phosphorylation. These features in DUI species could be associated to the maintenance of separate inheritance routes for the two mtDNAs. Additional ORFs are also found in other animal mt genomes: I summarized the presence of gene duplications as a co-author in a review focusing on animal mt genomes with unusual gene content.

Mesenchymal stromal cell therapy for carti-lage regeneration: in vivo testing in a refined preclinical sheep model of chondral and os-teochondral lesions

Una de las patologías musculo-esqueléticas más prevalentes envuelve a las articulaciones y incluye la lesión del cartílago articular, siendo la rodilla una de las articulaciones más afectadas. Las lesiones del cartílago pueden ser divididas en dos tipos distintos: lesiones de espesor parcial (lesiones condrales) que no penetran el hueso subcondral adyacente, y lesiones de espesor total (lesiones osteocondrales) que penetran el hueso subcondral. En la actualidad no existe un tratamiento efectivo para las lesiones condrales u osteocondrales, a pesar de existir un gran esfuerzo de investigación en este sentido. La tendencia actual es desarrollar nuevos métodos celulares y basados en ingeniería tisular que puedan solucionar las limitaciones de las tecnologías existentes. Para este propósito, el uso de modelos animales es de extrema importancia, porque funcionan como una fuente de información única entre los estudios in vitro y la aplicación de la terapia en humanos. Diversos modelos animales son utilizados en el análisis de estrategias de reparación del cartílago, de pequeños a grandes animales. Los pequeños animales (roedores y conejos) están recomendados para estudios de mecanismo o prueba de concepto cuando datos sobre toxicidad, formulación, respuesta a dosis o seguridad son necesarios antes de otros estudios. No obstante, los grandes animales (perro, cerdo, cabra, oveja o caballo) son necesarios para una verdadera investigación traslacional cuyo objetivo sea obtener aprobación reguladora para el uso clínico en humanos. Este trabajo presenta el estudio de un modelo experimental animal para la evaluación de una abordaje de terapia celular en la regeneración del cartílago articular de la rodilla. El objetivo fue el desarrollo de un modelo animal refinado, para evaluar la eficacia y seguridad de terapias celulares utilizando células mesenquimales estromales con posible futura aplicación en la medicina humana, y establecer las bases para los ensayos clínicos. / One of the widest spread musculoskeletal diseases concerns the joints and involves the lesion of articular cartilage, being the knee one of the most affected joints. Cartilage lesions can be divided on two different types: partial-thickness cartilage lesions (chondral lesions) which do not penetrate the underlying subchondral bone, and full-thickness cartilage lesions (osteochondral lesions) which penentrates the subchondral bone. Actually there is no effective treatment for the chondral or osteochondral lesions, although a large investigation effort is being made in this issue. The present tendency is to develop new cell- and tissue-engineering-based methods that may overcome the size limitations of current technologies. For this purpose, the use of animal experimental models is of extreme importance, because they are a reliable source of information between the in vitro assays and the human therapy application. Several animal models are used for cartilage repair strategies testing, from small to large animal models. Small animal models (rodents or rabbits) are recommended for mechanism or proof of principle studies when data regarding toxicity, formulation, dose response or safety are needed before further pivotal studies. However large animal models (dog, pig, goat, sheep or horses) are necessary for truly translational research aimed at gaining regulatory approval for clinical use in humans. This work presents the study of an animal experimental model for the evaluation of the knee’s cartilage regeneration with cellular therapies approach. The aim was the development of a refined animal model to assess the efficacy and safety of mesenchymal stromal cellular therapies with a possible future application in human medicine, and settle the basis for the clinical trials.

Integrating geospatial methods into evolutionary biology and conservation: case studies on selected Western Palearctic herpetofauna

Many evolutionary processes are influenced by spatio-temporal environmental variation, including speciation, genetic divergence among populations, and evolutionary change in physiology, morphology and behaviour. However, despite the extensive environmental data available from Geographic Information Systems (GIS) most evolutionary biologists and conservationists have not taken advantage of this data until recently. The general objective of this thesis was to integrate and expand the use of geospatial methods in evolutionary biology (biogeography, phylogeography and systematics) and conservation research. This general objective was accomplished through three specific objectives spread over six chapters, which compromised both methodological developments and their application in a series of case studies on Western Palearctic herpetofauna. This thesis explored and successfully used a number of promising new geospatial methods in combination with more traditional molecular analyses. Such integrative approaches will ultimately allow us to better consider and examine the range of potential histories underlying both inter and intraspecific divergence patterns. / Molts processos evolutius estan influïts per variació ambiental-, incloent l’especiació, espai-temporal, divergència genètica entre poblacions, així com el canvi evolutiu en fisiologia, morfologia i comportament. No obstant això, malgrat l'àmplia quantitat de dades ambientals disponibles en els Sistemes d'Informació Geogràfica (SIG), la majoria dels biòlegs i conservacionistes no han aprofitat aquestes dades fins fa poc. L'objectiu general d'aquesta tesi és integrar i ampliar l'ús de mètodes geoespacials en biologia evolutiva (biogeografia, filogeografia i sistemàtica) i recerca de conservació. Aquest objectiu general es va aconseguir a través de tres objectius específics dividits en 6 capítols, els quals inclouen tant un desenvolupament metodològic i la seva aplicació en una sèrie de casos d'estudi espaciotemporals de herpetofauna paleárctica occidental. From Sanger to NGS: Detecting MHC (Major Histocompatibility Complex) Class II and OR (Olfactory Receptors) Genetic Variability in Italian Wolves (Canis Lupus) and relative Canids

In this PhD thesis I will describe different aspects of conservation genetics and genomics of two wild Canidae species, the wolf (Canis lupus) and the golden jackal (Canis aureus), through the study of two of the most variable gene families: the Major Histocompatibility Complex genes (MHC), and Olfactory Receptors genes (OR). In order to perform these studies both Sanger and next generation sequencing (NGS) DNA techniques have been used. The background of the thesis is described in the “General introduction” with phylogeny, classification and evolutionary ecology of the Canidae, with a focus on the species Canis lupus and its main conservation concerns in Italy. Moreover, I will introduce the importance to perform genetic studies as tools for wild-life conservation and management, with a description of the framework of the principal historical and currently used molecular markers that had driven to develop MHC and OR sequencing projects. The thesis is divided into two parts, “PART I – The MHC typing project” and “PART II – The OR genes typing project”. A total of four scientific papers (already published or under revision) will be introduced and illustrated as result of three years of PhD activities at ISPRA’s (Istituto Superiore per la Protezione e la Ricerca Ambientale), Laboratory of conservation genetics, in Ozzano dell’Emilia, and thankfully to a PhD fellowship granted by the Università di Bologna.
