La bactérie Pseudomonas aeruginosa est un pathogène opportuniste responsable de graves infections chez les personnes affaiblies immunitairement. Présentant des souches résistantes aux antibiotiques, une nouvelle approche thérapeutique est en cours de développement avec pour objectif l’inhibition des facteurs de virulence de la bactérie. Lors de son processus d’infection, le pathogène utilise les lectines pour reconnaître et se lier de manière spécifique aux glycoconjugués des cellules-hôtes en formant une interaction lectine/glycoconjugué. Plus particulièrement, la lectine PA-IL, spécifique du galactose, a été étudiée. A l’aide de glycomimétique, il semble possible de bloquer l’action de la lectine en créant une interaction lectine/glycomimétique. Pour développer cette approche, de nombreux glycocluster sont donc été élaborés et leur affinité avec la lectine PA-IL a été évaluée par plusieurs méthodes de caractérisation (SPR, HIA, ELLA, puce à sucre,…).Dans ce projet de thèse, nous avons cherché à visualiser par microscopie à force atomique (AFM) l’arrangement des complexes lectine PA-IL/glycocluster formés pour trois glycoclusters différents. Nous avons ainsi pu montrer l’influence du cœur du glycocluster et des bras-espaceurs sur l’arrangement des complexes. Suivant le glycocluster, l’arrangement prend la forme de filaments 1D,de structures dentelées avec des bras sinueux ou encore de larges structures compactes. Dans le cas des filaments, la résolution de nos images AFM nous a permis d’identifier les lectines à l’intérieur même de la structure filaire. Nous avons aussi démontré, en observant les lectines seules, l’existence d’une interaction lectine/lectine. De plus, des expériences ont été menées pour déterminer les conditions expérimentales appropriées à leur observation à l’air et en milieu liquide. / The bacterium P. aeruginosa is an opportunistic pathogen responsible for serious infections in immunocompromised patients. It also develops some strains resistant to antibiotics. A new approach is developed to inhibit virulence factors of the bacterium. During the process of infection, the pathogen uses lectins to recognize and bind specifically to glycoconjugates of the host cells forming alectin/glycoconjugate complex. Particularly, the lectin PA-IL, specific to galactose, was studied. Using glycomimetics, it seems possible to block the action of the lectin by creating lectin/glycomimetic interaction. To develop this approach, many glycoclusters were designed and their affinity with lectin PA-IL was evaluated by various characterization techniques (SPR, HIA, ELLA, microarrays,…).In this thesis project, we have tried to visualize by Atomic Force Microscopy (AFM) the arrangement of lectin PA-IL/glycocluster complexes with three different glycoclusters. Our results show the influence of the glycocluster core and the linker on the arrangement of complexes. Depending on glycocluster, the arrangement takes the form of 1D filaments, 2D "pinked" structures with sinuous branches or large compact structures. In the case of filaments, the resolution of AFM images allows us to identify lectins along the filament. We also demonstrated the existence of lectin/lectin interactions at high concentration of lectin. In addition, some experiments were performed to determine sample preparation techniques to observe lectins in air and in liquid.

La réalisation de fonctions de filtrage complexes nécessite une parfaite maîtrise du processus de dépôt ainsi qu’un contrôle précis et en temps réel de l’épaisseur optique des couches déposées. Au cours de ma thèse, consacrée au développement de nouvelles modalités de contrôle optique in situ, je me suis particulièrement intéressé à deux sujets différents, à savoir : - D’une part, la détermination de la dépendance spectrale des constantes optiques (indice de réfraction et coefficient d’extinction) de matériaux diélectriques. Un moyen possible pour effectuer cette détermination consiste à utiliser un système de contrôle optique large bande afin d’enregistrer les spectres de transmission de l’empilement au fur et à mesure de sa formation. En effet, l’évolution temporelle, à chaque longueur d’onde, de ces spectres de transmission contient des informations quantitatives liées aux constantes optiques que nous souhaitons déterminer. - D’autre part, la mesure en temps réel du coefficient de réflexion (r) d’un empilement, en amplitude et en phase, lors de son dépôt. En effet, les méthodes de contrôles optiques en intensité présentent des limitations que la connaissance de l’information de phase devrait permettre de contourner. Cette mesure est réalisée par interférométrie holographique digitale à faible cohérence sur un substrat éclairé par sa face arrière et dont la face avant est équipée d’un masque annulaire. Ceci donne accès aux information de phase et d’amplitude recherchées tout en s’affranchissant des vibrations générées par le fonctionnement de la machine de dépôt ainsi que du mouvement de rotation à 120 tours par minute qu’effectue le porte-substrat. / The realization of complex filtering functions requires a perfect mastering of the deposition process as well as an accurate real time monitoring of the optical thickness of the deposited layers. During my PhD thesis, devoted to the development of new methods of in situ optical monitoring, I was particularly interested in two different subjects, namely:- On the one hand, the determination of the spectral dependence of optical constants (refractive index and extinction coefficient) of dielectric materials. A possible way to achieve this determination consist in using a broadband optical monitoring system in order to record the transmission spectra, in real time, of the stack during its formation. Indeed, the temporal evolution, at each wavelength, of these transmission spectra provide quantitative information related to the optical constants that we wish to determine.- On the other hand, the real time measurement of the reflection coefficient (r) of a stack, in amplitude and phase, during its deposition. Indeed, the optical monitoring methods based on intensity proprieties present some limitations that the knowledge of phase information should overcome. This measurement is performed by low coherence digital holographic interferometry on a substrate illuminated by its rear face and whose front face is equipped with an annular mask. This gives access to desired phase and amplitude information while avoiding the parasitic influence of the substrate motions induced by the vibrations of the deposition machine, and the rotation of the substrate holder at 120 rounds per minute.

Neste trabalho avaliou-se a potencialidade do uso da sísmica rasa de reflexão multicomponente para investigação geológica-geotécnica. Foram abordados vários aspectos relacionados à aquisição dos dados sísmicos de reflexão multicomponente, com o objetivo de entender as vantagens e limitações do método para aplicação em investigações de subsuperfície rasa. Os ensaios de campo foram realizados em duas áreas, ambas em terrenos da Bacia Sedimentar de São Paulo, em área urbana da cidade de São Paulo. Para a interpretação dos dados sísmicos multicomponente foram investigados procedimentos para a inversão não-linear dos tempos de trânsito e das amplitudes. O testes realizados orientaram a escolha da aproximação não-hiperbólica mais apropriada para calculo dos tempos de trânsito visando à análise de velocidades do pacote acima do refletor. O estudo numérico desenvolvido para a inversão das amplitudes mostrou a viabilidade da estimativa das velocidades e densidades, acima e abaixo do refletor, empregando-se as equações de Zoeppritz para as ondas refletidas PP, PSv, SvP e SvSv, antes e depois do ângulo crítico. Dada a complexidade da inversão nao-linear das amplitudes, se fez necessário elaborar uma estratégia estocástica de otimização e desenvolver uma nova abordagem para análise da função objetivo multi-dimensional, garantindo confiabilidade ao resultado da inversão não-linear. Os resultados deste trabalho mostraram o potencial da sísmica de reflexão rasa multicomponente para caracterização geológica-geotécnica, possibilitando um melhor entendimento das camadas superficiais. / This thesis aims to evaluate the useful of the multicomponent seismic methods for shallow investigations, mainly its potential for the geotechnical and geological characterization of the nearsurface. Several aspects regarding the acquisition and processing data of multicomponent seismic data are discussed. They were based on data set acquired in the urban area of Sao Paulo city, Brazil. Two different areas were investigated. Both located in sedimentary terrains belonging to the Sao Paulo Sedimentary Basin. We present a non-linear travel time and seismic amplitude inversion scheme to quantitative interpretation of multicomponent seismic data. Several tests were performed to guide the choice of non-hyperbolic equation more suitable for travel time inversion aiming the velocity analysis above the reflector. A numerical experiment developed to solve the nonlinear inversion of seismic amplitudes showed the feasibility to estimate seismic interval velocities and layer densities above and below the reflector using the exact Zoeppritz equations for PP, PSv, SvP e SvSv reflected waves, before and after critical angle. Due to the apparent complexity of the nonlinear seismic amplitude inversion, it was necessary elaborate the strategy for stochastic optimization and develop a new approach to analyze the multi-dimensional objective function, with different implications for the accuracy and efficiency of the non-linear inversion. The study show the benefits of using the multicomponent seismic method for shallow geological-geotechnical characterization, improving the nearsurface understanding, once allows an integrated analyzes of a more complete record of the wave field.

[pt] Sistemas pneumáticos são equipamentos leves, baratos, limpos e de baixo risco, sendo apropriados para aplicações que necessitem de força e rapidez de resposta. Por outro lado, esse tipo de sistema apresenta restrições devido à principal característica do ar: sua compressibilidade confere efeitos não lineares ao sistema, desde um escoamento turbulento pelas válvulas de controle até sua atuação dentro do cilindro – a qual inclui alta sensibilidade ao atrito e volumes inativos durante o curso do pistão. Essas características particulares dificultam seu controle e posicionamento preciso e limitam sua aplicação, principalmente considerando seu emprego em um mecanismo tipo Plataforma de Stewart em escala reduzida. No presente trabalho apresenta-se a modelagem, simulação computacional e análise experimental do comportamento dinâmico de um sistema de atuação pneumático que inclui uma válvula de controle de vazão não convencional, composta de quatro válvulas proporcionais, e um atuador com haste simples de dupla ação. O objetivo deste trabalho é, baseado nos resultados experimentais, determinar as características desses componentes para desenvolver estratégias de controle em tempo real capazes de minimizar os efeitos das não linearidades típicas, visando sua utilização no mecanismo anteriormente mencionado. / [en] Pneumatic equipment is lightweight, cheap, clean and low-risk, being suitable for applications that require strength and high responsiveness. Nevertheless, this type of system has some limitations due to the air main feature: its compressibility results in nonlinear effects in the system, from the turbulent flow control valves to its performance inside the cylinder - which includes high sensitivity to friction and dead volumes during the stroke piston. These particular characteristics make its control and precise positioning difficult, limiting its application, especially when considered its use in a mechanism such as a Stewart Platform in a reduced scale. The present paper presents the modeling, computational simulation and experimental analysis of the dynamic behavior of a pneumatic actuation system that includes an unconventional flow control valve, consisting of four proportional valves, and a double acting single rod actuator. The final goal of this work is to, based on experimental results, determine the characteristics of these components in order to develop real-time control strategies which can minimize the effects of those typical nonlinearities for their use in the mechanism mentioned above.

Métodos: 48 pacientes (96 olhos) com GPAA assimétrico foram recrutados. Três medidas da PIO e da APO foram aferidas utilizando o TCD. Para o diagnóstico de assimetria eram necessárias uma diferença de perda de campo visual maior que 6 dB no índice “mean deviation” (MD), e uma diferença de 5 mmHg na PIO medida com o tonômetro de aplanação de Goldmann (TAG) entre o olho mais afetado e o contra-lateral. Todos os participantes se submeteram a um exame oftalmológico completo, incluindo paquimetria ultrassônica e ecobiometira. Os critérios de exclusão consistiram de: doenças ou cicatrizes corneanas, uso de medicação anti-glaucomatosa tópica ou sistêmica e cirurgia ocular prévia. Resultados: Não houve diferença com significância estatística (p = 0,142) entre o comprimento axial dos olhos do grupo melhor (22,95 +/- 0,91 mm) e pior (22,85 +/- 0,97 mm). Houve diferença estatisticamente significativa (p = 0,011) entre a espessura corneana central do grupo de olhos melhores (537,08 +/- 29,54 μm) e do grupo de olhos piores (534,40 +/- 29,87 μm). Os valores da APO do grupo de olhos melhores (3.32 +/- 1.14 mmHg) foram significativamente menores (p = 0,001) do que os obtidos no grupo de olhos piores (3,83 +/- 1,27 mmHg). Quando corrigimos as medidas de APO pela diferença de PIO entre os olhos houve uma perda da significância estatística entre os grupos (p = 0,996). Conclusão: A APO é semelhante entre os dois olhos de pacientes portadores de GPAA assimétrico com PIO assimétrica. De acordo com esses dados não há evidência de que a APO possa ter um papel no GPAA hipertensivo assimétrico / Aim: To evaluate ocular pulse amplitude (OPA) using the dynamic contour tonometer (DCT) in patients with asymmetric primary open-angle glaucoma (POAG) and asymmetric intra-ocular pressure (IOP). Methods: The participants consisted of 48 patients (96 eyes) with asymmetric POAG. Three measurements of IOP and OPA were taken using DCT. The diagnosis of asymmetry required a difference of glaucomatous visual field loss greater than 6 dB in the global index MD and a difference of 5 mmHg in IOP measured by Goldmann aplannation tonometry (GAT) between the more affected and the contra-lateral eye. All participants underwent full ophthalmologic clinical assessment including ultrasonic pachymetry and biometric measurements. Exclusion criteria were corneal diseases or scars, topical or systemic glaucomatous medications, and previous ocular surgery. Results: No difference (p = 0.142) was found between the axial length measurements of the better eyes group (22.95 +/- 0.91 mm) and worse eyes group (22.85 +/- 0.97 mm). There was a statistically significant difference (p = 0.011) between the central corneal thickness values of the better eyes group (537.08 +/- 29.54 μm) and worse eyes group (534.40 +/- 29.87 μm). The OPA values of the better eyes group (3.32 +/- 1.14 mmHg) were significantly lower (p = 0.001) than those obtained on worse eyes group (3.83 +/- 1.27 mmHg). When correcting the OPA readings by the IOP there was a loss of statistical difference between groups (p = 0.996). Conclusion: OPA is similar in both eyes of asymmetric hypertensive POAG patients with asymmetric IOP. According to this data there was no evidence that OPA could play a role in asymmetric hypertensive POAG

A investigaÃÃo à realizada, atravÃs de simulaÃÃes numÃricas, utilizando-se do mÃtodo de Runge Kutta de quarta ordem. Na primeira aplicaÃÃo, à analisada a possibilidade da realizaÃÃo de operaÃÃes lÃgicas pelo Acoplador Direcional NÃo-Linear (NLDC) Duplo SimÃtrico sem perda. Considerando a operaÃÃo das portas lÃgicas, foram utilizadas as quatro possÃveis combinaÃÃes para dois pulsos, nas fibras 1 e 2, modulados pela posiÃÃo temporal (PPM) nos nÃveis lÃgicos 0 ou 1. Nas duas aplicaÃÃes foram obtidas portas lÃgicas E e OU. / In this work, the performance study of a Symmetric Dual-Core Non-Linear Directional Coupler (NLDC), working with pulses of , in two different applications have been investigated: accomplishment of logical gates AND and OR, under Pulse Position Modulation (PPM); accomplishment of logical gates AND and OR, under Pulse Amplitude Modulation (PAM). The investigation is based in a numerical simulation study, using the fourth order Runge Kutta numerical method. In the first application, the possibility of the accomplishment of logical operations by Symmetric Dual-Core Non-Linear Directional Coupler (NLDC) without loss is analyzed. Considering the operation of the logical gates, the four possible combinations for two pulses, on the input fibers, modulated by the temporal PPM in the logical levels 0 or 1, were used. The effects of a variation in the coding parameter offset of the PPM modulation, that is, in the displacement of the input pulse relative to the reference time level, was investigated. In the second application, the symmetric dual-core NLDC is submitted to PAM modulation, using the four possible combinations for two pulses on the input fibers, were used, as well. The effects of a variation in the coding parameter offset of the PAM modulation relative to the reference amplitude, was investigated. The modulated output amplitude versus a phase difference between the input pulses , was also studied. On the two applications, logical gates AND and OR was observed.

The surface is free-standing and its primary purpose is to modify the phase distribution, or the phase and amplitude distribution of electromagnetic fields propagating through it: it is called phase shifting surface (PSS) in the first case, and phase and amplitude shifting surface (PASS) in the second case. For practical applications, the surface typically comprises three or four layers of metallic patterns spaced by dielectric layers. The patterns of the metallic layers are designed to locally alter the phase (and amplitude in the case of the PASS) of an incoming wave to a prescribed set of desired values for the outgoing wave. The PSS/PASS takes advantage of the reactive coupling by closely spacing of the metallic layers, which results in a larger phase shift range while keeping the structure significantly thin. The PSS concept is used to design components such as gratings and lens antennas which are presented in this document. The components are designed for an operating frequency of 30 GHz. The PSS phase grating gives high diffraction efficiency, even higher than a dielectric phase grating. Several types of lens antennas are also presented, which show comparable performance to that of a conventional dielectric plano-hyperbolic lens antenna with similar parameters. The PASS concept is used in a beam shaping application in which a flat-topped beam antenna is designed. This work demonstrates the potential for realising thin, lightweight and low-cost antennas at Ka band, in particular for substituting higher-gain antenna technologies such as conventional dielectric shaped lens antennas.
