This master's thesis addresses TED, the global phenomenon dedicated to spreading revolutionary ideas worldwide. The aim of this thesis is to find and show correlations between interesting ideas from TED talks - under the tags 'education' and 'teaching', and their potential implementations among the Czech society. Through monographic method I investigate the most significant attributes/trends of the current crisis of the educational systems worldwide and their impact on a teacher - their role as a whole and the perception and acceptance of the goals of their work. These issues are addressed in a discourse about the changes in educational systems in 21st century worldwide and in the Czech Republic. It also explores the interconnections between problems in education and the state of society, its values and philosophical superstructures. One of the sections shows reader a compilation of categorize analyses of individual talks from TED based on their topic and content. The final part revolves around the most common topics from the talks that have direct connection to the current state of the Czech educational system which have been identified through educational research. The trend to take a stand in campaigns has been identified as an upcoming phenomenon and the term brand activism has been discovered in the context. One of the challenges that brands are facing is to keep the core identity and at the same time communicate responsible positions that consumers demand without getting inauthentic. Since little or no research has been done from the brand's perspective the study has found a research gap that could be filled by examining how their brand identity and political or social messages affect their campaigns. The purpose of the study is to look at three brands and their campaigns in relation to their brand identity and their role within the campaigns. Furthermore, the study investigates if there are any connections between brands taking a stand and brand activism. To achieve the aim of this research, a qualitative method has been conducted based on semi-structured interviews with three types of brands and an expert in communication. The results showed that brand activism in some cases has been used as a tool for brands to challenge structures and cause a dialogue, but the result also identified how brands manage to stay authentic while communicating societal issues.

The object of the study is urban planning - architectural design strategy of conversion and revitalization of the factory PEARL 01 in Usti nad Orlici. The area of the site is about 3 hectares, an area eight times larger than the actual main square in the town. Most of the land is owned by the city. Current status of the area does not match the aesthetic or functional demands placed on this space, which is intended for expansion of the city center, whose current capacity does not match the needs of even the size of the city. Textile industry and Perla itself played an important role in the history of the region and influenced the lives of most local citizens. Therefore, the proposal seeks to retain as much of the original structure as possivle and with them the charm and spirit of the original factory and its buildings. The area is cleaned up, breathes and becomes in the first time in history a part of the urban space, and not a big barrier in its center.

Stanovení pravosti a falšování medu iontově výměnnou chromatografií / Determination of honey authenticity and adulteration by anion exchange chromatography

Cieľom tejto diplomovej práce je stanoviť kvalitu a zloženie medu, taktiež stanoviť jeho botanický pôvod na základe profilu sacharidov v mede. Med je prírodný produkt vysokej kvality a vďaka značnej spotrebe a nemalej trhovej hodnote je tiež terčom na falšovanie. S cieľom identifikovať podvody a cudzorodé látky, pochádzajúce z cukrových sirupov, boli analyzované oligosacharidy a polysacharidy na báze maltodextrínov. Teoretická časť popisuje chemické zloženie medu a hlavné princípy aplikovanej inštrumentálnej techniky. Poskytuje literárny prehľad existujúcich analytických metód na stanovenie sacharidov prítomných v mede a na odhalenie jeho falšovania. Boli definované hlavné body falšovania a autenticity, vrátane legislatívnych aspektov a opisu bežných druhov falšovania. Experimentálna časť obsahuje postupy, ktoré vedú k vývoju a optimalizácii chromatografických podmienok a parametrov elektrochemického detektora na stanovenie sacharidov a maltodextrínov (oligosacharidov a polysacharidov) vo vzorkách medu. Použitá analytická technika bola vysoko účinná aniónovo-výmenná chromatografia (HPAEC) spojená s pulzným amperometrickým detektorom (PAD). Vyvinutá a optimalizovaná chromatografická metóda bola taktiež validovaná z hľadiska linearity, dynamického rozsahu, analytických limitov, presnosti a správnosti. Na záver bola táto vypracovaná metóda úspešne zhodnotená analýzou autentických a falšovaných vzoriek medu rôzneho botanického pôvodu pochádzajúcich z členských štátov EÚ.

Studium autenticity koření a kořenících přípravků / Study of the authenticity of spices and condiments

This disertation deals with geographical authentification of different types of spices and spice preparations. Investigated spice species were garlic (Allium sativum), ground pepper (Capsicum anuum), and dried carrot (Daucus carota). Theoretical part of the disertation describes main qualitative parameters of the examined species and production technology, means of food adulteration and statistic methods used in chemometrics. The aim of this dissertation was to verify the hypothesis whether it is possible to use targeted analytical techniques commonly used in quality control of spices and condiments, and statistical processing of measured data to distinguish samples of spices and condiments of different geographical origin. The use of non-targeted analysis was also investigated. Samples of garlic and ground pepper were used for targeted analysis. The examined parameters were dry matter and moisture of the sample, ash content, total phenolic content according to Folin-Ciocaulteu, carbohydrate content, alliin concentration, total nitrogen content, total color pigment (ASTA), pH of aqueous extract, total fat content, and concentration of selected elements (Ca, Cu, Fe, K, Mn, Mg, Na, P, Zn). The instrumental techniques used were molecular absorption spectrometry, inductively coupled plasma optical emission spectrometry and high performance liquid chromatography. The obtained data were statistically processed by analysis of variance (ANOVA) and principal component analysis (PCA). Using statistical analysis significant differences between samples that came from more distant areas were found. However, samples from closer areas could not be distinguished. The researched hypothesis could not be unequivocally confirmed or refuted. Metabolic fingerprint of carrot samples was determined using non-targeted analysis. Metabolic analysis was performed using the tandem LC-TOF-MS technique. The data were processed by recursive peak extraction (BRE) and subsequently uvaluated with PCA. The samples were divided into clusters according to their origin. Targeted and non-targeted techniques have great potential in verifying the geographical authenticity of different types of spices. However, the main condition is consistent and sufficient sampling, guaranteed information on the origin of the sample and obtaining a sufficient amount of input data for statistical analysis.

Využití analytických technik a chemometrie k autentifikaci vín / Application of analytical techniques and chemometry for authentification of wine

This PhD. thesis is focused on application of different analytical techniques for determination of elemental composition of Moravian wine combined with analysis of selected polyphenol compounds. Two spectrometric techniques were used for elemental analysis of wine – Mass spectrometry and Optical emission spectrometry with inductively coupled plasma (ICP-MS and ICP-OES). Analysis of organic constituents of wine was evaluated by High performance liquid chromatograph with diode array detector (HPLC-DAD). Data from analysis of wine were used for construction of mathematical models for determining geographical origin of wine and for determining type of varietal wine. Data handling was performed by usage of multivariate statistical methods (cluster analysis, principal component analysis and canonical discriminant analysis). For the discrimination of wine by the geographical origin, two separate models for white and red wine were constructed. Effectiveness of discrimination of Moravian wine (with knowledge about origin) into 4 wine regions was 100 % successful for white wine and 96,43 % for red wine samples. Those models were used for cross validation test of unknown samples with result 95 % successfully classified samples of white wine and 82,15% of red wine. Besides that, mathematical model for discrimination and classification of varietal wine was made. Müller Thurgau, Riesling and Gruner Veltliner were discriminated by the usage of 8 elemental parameters. Discrimination success rate of known samples was evaluated by canonical discriminant analysis with 95,83% of correctly classified known samples and 70,83% of correctly classified unknown varietal wine (cross validation test). In addition to these part of research, impact of different grape production systems on elemental composition of wine and Vitis Vinifera was studied. The core of the study was to find the connections between the type of agriculture and chemical properties of wine and Vitis Vinifera. Two agriculture systems were assessed – Organic agriculture and Integrated pest management. . Results of this study showed that quantity and manner of pesticide application is very important factor affecting the final elemental composition of the grapevine and wine, especially the content of copper, zinc and manganese. That knowledge can be used in the future for creation of classification model of wine according to grape production system. For comparison of radical scavenging activity, electron paramagnetic resonance spectroscopy (EPR) with DPPH method was used. In the case of this study, the influence of vine growing under different farming systems on the total antioxidant activity of wine was found to be insignificant.

UNTARGETED METABOLOMIC FINGERPRINTING FOR AUTHENTICITY AND TRACEABILITY OF FOODS

La globalizzazione del mercato agroalimentare ha determinato una crescente attenzione da parte dei consumatori verso i prodotti alimentari, non solo in termini di qualità e di sicurezza, ma anche di origine geografica. Infatti, il territorio d’origine ha un forte impatto sull’alimento, dovuto alle condizioni pedoclimatiche che ne determinano le caratteristiche. Poiché non esistono dei metodi analitici di routine per l’autenticazione della provenienza geografica, lo scopo del progetto di ricerca è quello di determinare l’origine geografica e l’autenticità degli alimenti mediante profiling dei composti fenolici e steroli, grazie all’applicazione di tecnologie omiche, tecniche statistiche e chemometriche. La componente fenolica e/o steroli dei campioni, viene analizzata tramite cromatografia liquida (UHPLC) accoppiata ad uno spettrometro di massa (Q-TOF-MS). I dati così ottenuti, vengono elaborati mediante statistica multivariata. L’applicazione combinata di avanzate tecnologie omiche e tecniche statistiche chemometriche ha portato come risultato l’effettiva identificazione della provenienza geografica e autenticità di numerose matrici alimentari. I dati ottenuti dimostrano che i metaboliti secondari possiedono proprietà discriminanti. L’approccio di metabolomica UHPLC/Q-TOF-MS combinato a una statistica multivariata risulta essere adeguato per identificare potenziali markers. Il lavoro attuale è focalizzato sulla ricerca di nuovi metaboliti che, insieme a fenoli e steroli possano confermare la potenza di questo approccio. / Nowadays, food traceability is a growing consumer interest worldwide. Food traceability could be considered a fundamental tool for ensuring safety and high quality of food. Food quality is based not only on the safety and integrity of food, but also on the authenticity, the genuineness of the raw material and the geographical origin. The aim of the work was to investigate the potential of untargeted metabolomics to ensure the authenticity and traceability of foods. Secondary metabolites, like polyphenols and sterols, could be conveniently used to meet this goal due to their chemical diversity and their responses to environmental stimuli. Samples were analyzed through UHPLC-ESI/QTOF-MS. The obtained data were subjected to multivariate statistical analysis. The obtained results showed that secondary metabolites can be efficiently used for authenticity and traceability purposes, with regards to cultivars and geographical origin. These information confirm the role of environmental factors in shaping the actual profile of secondary metabolites in plant foods. The markers found could be used for a target quantification method, a less expensive and less sophisticated analysis, in order to provide an efficient tool that could help to guarantee food quality on routine basis.

Since 2003 the Swedish Armed Forces uses Developmental Leadership as their formal leadership model. The model is characterised by the leader acting as a role model and providing inspiration, motivation and personal consideration. The model stresses the importance authenticity plays in achieving the motivational aspects of leadership. The current literature on authenticity shows almost solely positive effects of the application of authenticity. However, there are organizational factors that seem to impede the non-commissioned officers’ ability to freely express their authentic self. Therefore, this study has aimed to deeper investigate the possible discrepancy between the application of authenticity in theory in relation to practice. To achieve this, we strive to answer the question as to how non-commissioned officers experience the practical application of authenticity in military contexts. A qualitative semistructured interview guide was chosen as the research method. The participants consisted of six non-commissioned officers in troop leading positions within the Swedish Armed Forces. Through a thematic analysis the data resulted in three main themes with a total of eleven subcategories. The study concludes that the results support the Developmental Leadership model´s claim that environmental factors affect the way leaders can exert Developmental Leadership. In addition, the study also contribute to a clearer picture of how these factors specifically affect leaders ability to act authentically. / Försvarsmaktens formella ledarskapsmodell Utvecklande Ledarskap förordar autenticitet hos ledare inom myndigheten. Teorier inom autenticitet visar på närmast enkom positiva effekter. Dock finns det organisatoriska faktorer som inverkar och begränsar specialistofficerares möjligheter till autentiskt handlande. Syftet med denna studie var att undersöka hur specialistofficerare upplever tillämpningen av autenticitet i sin yrkesroll. Som undersökningsmetod valdes kvalitativa semistrukturerade intervjuer och urvalet bestod av sex aktivt tjänstgörande specialistofficerare i truppförande positioner inom Försvarsmakten. Den data som framkom analyserades genom en tematisk analys och resulterade i tre teman med elva underliggande kategorier. Resultaten stödjer teorin om hur omgivningskarakteristika påverkar ledares möjlighet att utöva utvecklande ledarskap men tillför en tydligare bild av hur dessa faktorer specifikt påverkar ledares autenticitet.

In the very center of my research is so called "dynamic cognitional structure" presented by Canadian thinker, Bernard Lonergan. The purpose of the study is to analyze his epistemology in the context of the "generalized empirical method" and to investigate the relevance of this method for contemporary theology. The thesis also focuses on the question if theological epistemology could be based on intellectual knowledge only, or how far it requires authentic spiritual life of an existential subject. Lo sviluppo sostenibile è oggi uno dei temi più attuali e, allo stesso tempo, una sfida per le aziende. La società, a livello mondiale, deve affrontare, ora più che mai, problemi sia di rilevanza sociale, come una popolazione in costante aumento, sia di tipo ambientale, come il cambiamento climatico. La costante perdita dell’ozonosfera, la distruzione della biodiversità, la sovrappopolazione, l’overtourism, sono solo alcune delle problematiche che sottolineano l’urgente bisogno di individui che siano capaci di far fronte a tali questioni grazie ad un comportamento imprenditoriale sostenibile. Seppur le serie conseguenze del degrado ambientale siano note da tempo, la situazione attuale ci richiede ora di riconoscere e, soprattutto, di ammettere, che le risorse, specialmente quelle naturali, non sono infinite. Il comportamento imprenditoriale con un orientamento spiccatamente green, costituisce una parziale soluzione all’esaurirsi di tali risorse. A tale scopo, la tesi si propone di indagare i drivers dell’imprenditoria sostenibile, al fine di risolvere il degrado ambientale e raggiungere uno sviluppo sostenibile. Attraverso un’analisi della environmental passion, dell’environmental bricolage e dell’authenticity, la tesi pone le fondamenta per lo sviluppo di futuri studi capaci di far riflettere su tali tematiche. / Sustainable development is perhaps the most prominent topic of our time and challenge for businesses. Today, we are facing both social issues, such as an ever-increasing population, and environmental issues, like climate change. We are thus suffering from ozone depletion, destruction of biodiversity, loss of living species, overpopulation, overtourism, and the need of individuals who are able to deal with these challenges through their entrepreneurial behaviour is more than urgent. The serious consequences of environmental degradation have been pointed out for decades, and the current situation requires us to recognize that resources, especially our natural ones, are finite. The thesis focuses on the drivers of sustainable entrepreneurial behaviours aimed at resolving environmental degradation and achieving sustainable development. Through an exploration of environmental passion, environmental bricolage and authenticity, the thesis sets a foundation for developing future thought-provoking studies.
