This diploma thesis is concerned with determination anthocyanins dyes in elderberry (Sambucus nigra L.). In theoretical part there is described general characterization of the plant, its chemical composition, short view of elderberry breeding and possibilities of usage in food and medical industry. Bigger attention is paid to anthocyanins dyes. There is described their characterization, occurrence, factors that have an impact on their stability. Biosynhesis of the pigments and metabolism, benefit effects and their usage in food chemistry are then shortly sumarized. Methods of determination of anthocyanins are discused too. Next chapter deals with high performance liquid chromatography. At the end of theoretical part, there is sumarized findings of anthocyan dyes analysis by expert scientists in various matrix. In experimental part, total of 16 species of elderberry were analyzed. These species are from research and breeding institute of pomology in Holovousy. The dyes were identification and quantification by high performace liquid chromatography coupled with a diode array detector. Detection wavelenght was 520 nm. Gradient of mobile phase was optimilized because of shortening analyses time and resolution improvement. Four dyes were identified by HPLC: cyanidin-3-sambubioside, cyanidin-3-glukoside, cyanidin-3,5-diglukoside a cyanidin-3-sambubioside-5-glukoside. Total amount of anthocyanins ranged from 33627167,0 to 5525284,0 mAU•min-1. The most anthocyanins dyes were determined in Mammut species (33627167,0 mAU•min-1), the lowest amount was measured in Aurea species (5525284,0 mAU•min-1). The most abundant anthocyanin in all species of eldeberry fruit except Haschebrg and Aurea species was cyanidin-3-sambubioside. The concentrations of cynidin-3-glucoside in all of the berries were quantified using regression equation of calibration curve. Mammut species had the most amount of this anthocyan (0,486 mg•ml-1) and the lowest amount had Aurea species (0,033 mg•ml-1).

This work deals with the elemental analysis of different elderberry (Sambucus nigra L.) cultivars. In the theoretical part the botanical characteristics of elderberry, including its extension and places of occurrence, the possibilities of collecting the various parts of the elderberry and its use in food and cosmetics is described. A large part is devoted to the occurrence and characteristics of minerals in food, and to the methods that are suitable for elemental analysis and decomposition of biological material. The aim of the experimental part was to analyze different cultivars of elderberry and juice made from this cultivars, and to determine concentration of selected elements (Zn, Mn, Fe, Mg, Cu, P, Ca, Na and K) using ICP–OES. The total amount of 24 cultivars was analysed from which 6 varieties were harvested in two different seasons in 2011 and 2013. Measured concentrations of elements in different varieties of elderberry were mutually compared using statistical methods. The differences between the content of elements in fruits and juice, obtained by pressing the fruit, and the effect of seasonal conditions on the content of elements in the fruit juice from different elderberry cultivars were studied.

The purpose of this work has been to describe some techniques which are normally used for cluster data analysis process of unsupervised learning. The thesis consists of two parts. The first part of thesis has been focused on some algorithms theory describing advantages and disadvantages of each discussed method and validation of clusters quality. There are many ways how to estimate and compute clustering quality based on internal and external knowledge which is mentioned in this part. A good technique of clustering quality validation is one of the most important parts in cluster analysis. The second part of thesis deals with implementation of different clustering techniques and programs on real datasets and their comparison with true dataset partitioning and published related work.

The thesis solves the processing of a project documentation of a veterinary clinic in Nachod. The project and attachments are processed in accordance with current laws, regulations and standards. The Veterinary Clinic is located on parcels č.122 / 3 and 122/4, which are parts of the cadastral municipality Nachod. Near the land under the adjoining local roads, there are utilities routed. In the building of veterinary clinic, there will be 2 surgeries, 2 operating rooms, laboratory, pharmacy, grocery breeders and editorial office. Veterinary Clinic is a two-storey building with a rectangular ground plan with dimensions of approximately 40 x 19 meters. The building has 3 main entrances. Admission to the clinic and store are situated on the northwestern side. Entry into the pharmacy and the editorial office, which is located on the 2nd floor, are on the southeast side. The master thesis deals with the historical development of humanitarian aid against the backdrop of transformations of the world order. Firstly, the thesis scrutinizes the differences between the classical humanitarianism of the Red Cross and the so-called new humanitarianism, which emerged during the Nigerian civil war (1967-1970). Secondly, the thesis demonstrates that although the new humanitarianism tends to be presented in an opposition to and as a more historically advanced form of the classic humanitarianism, when examined in a more detailed manner, the two humanitarian approaches face similar dilemmas in relation to political power. The overcoming of the "end of history" era, which gave rise and prominence to the new humanitarianism, opened discussion about the alternatives to the future of the world order. This shift allows us to formulate the main argument of the thesis - the evolution of the humanitarianism should not be seen as teleological and linear but as a continuous process in which both the classic and the new humanitarianism need to balance between strict legalism which can lead to moral failure and exaggerated moralism which can have consequences contradicting the basic principles of humanitarian action. The aim of the diploma thesis ,,The Availability of Social and Consecutive Services for the Homeless in the Klatovy District" is to determinate the real availability of services in the Klatovy district and the awareness of people without shelter about the possibilities of assistance. Theoretical part explains the terms exclusion, poverty and homelessness (identification, causes, consequences). The theoretical part in the second chapter discusses the possibilities of social work with people without shelter. There are defined participants who engaged in problems of homelessness at the beginning. The work defines the social services, social benefits and the process of reintegration subsequently. The theoretical part presents some housing models - Housing Ready, Housing First and Social Housing in the end. Practical part is the most important part of the thesis. There is the social demographic analysis of the Klatovy region in the beginning. The main section contains research work. The results of the research indicate that the services are available for people from the Klatovy region more than for other candidates. This diploma thesis called ČT sport newsroom without an editor-in-chief. Where are its limits? focuses on the leadership of sports newsroom, which operated without editor-in-chief between the years 2013 and 2019. In January 2020 the sports department was reorganized thus there is the editor-in-chief at the head of the organization since then, who manages the organization in terms of journalism. The researcher paid attention to the analysis of both possible structures. In the theoretical part, the author describes possible organizational structures according to which the organization can be structured. She focuses on media routines and history and the presence of the sports newsroom, but she is mostly targeting the hierarchy of the department. The empirical part is based on 14 in-depth interviews with workers of the newsroom, who describe how the sports department works and point out the positives and negatives of the system. The research took place in two phases. The first one before the restructuring of the department, the second one a few months after. The result is, how the sports newsroom according to its employees worked in this kind of atypical structure.

In my diploma thesis I focus on women who work in the sex business and their careers. The aim of the work is to present relevant theories and current knowledge about the life careers of sex workers, expanded by my own research. The work is dividing sex business according to the place of provisioning of services as well as according to the trends of this profession in our country and abroad. It describes present social and health risks in the historical perspective, career in the sex business, motivation to do such a job, and also mentioning an organization that works with women from the sex business. The work concerns only women who provide sexual services for a fee and are over 18 years of age. In my work I focus on biographies of women who provide paid sexual services from their point of view, and how they view their lives. I also adapted a selected sample of respondents to this work and addressed both existing clients of ROZKOŠ bez RIZIKA, z. s., including those who no longer work in the sex business. I used the method of structured interviews, through which it was possible to get a basic picture of the interviewee's family, personal and work situation. This diploma thesis, called "Life's Career of Female Sex Workers - Clients of Organisation Bliss without Risk", concentrates on the topic of prostitution and on the particular period of Bliss without Risk clients' life. The theoretic part includes an explanation of terms of the prostitution scene as well as it mentions and considers new words of this branch such as a sex work or a sex worker. The diploma work tackles the problem of what is very specific for the difficulties of the prostitution branch (heath, social, law or criminal difficulties). It mostly concentrates on the problematic of on sex workers committed violence. The coping strategies being used by women in sex business are also described. The practical part includes the phone research based on information collected from the Bliss without Risk's ex-clients.
