Crescimento relativo e composição do ganho de tecidos na carcaça de bubalinos mediterrâneos jovens abatidos com diferentes pesos /

Orientador: André Mendes Jorge / Banca: Renata Bonini Pardo / Banca: Heraldo César Gonçalves / Resumo: Com o objetivo de avaliar a composição do ganho e as curvas de crescimento de componentes corporais, foram utilizados 32 bubalinos da raça Mediterrâneo, recriados em pastagem nativa do litoral norte de São Paulo, não castrados, com idade média de 14 meses e peso vivo médio inicial de 330 Kg. Os animais foram divididos aleatoriamente em 5 grupos experimentais (grupo I, grupo II, grupo III e grupo IV), de sete animais em cada grupo e um grupo referência de 4 animais (grupo AB). Foi fornecida a mesma dieta ad libitum a todos os animais, duas vezes por dia. O abate foi precedido de jejum de sólidos de 16 horas, permanecendo livre acesso à água. O grupo controle (grupo AB) foi abatido logo após o período de adaptação de 30 dias e os demais grupos foram pesados a cada 28 dias até atingirem o peso pré estabelecido para o abate de 450 (grupo I), 480 (grupo II), 510 (grupo III) e 540 kg (grupoIV). Após o abate, as carcaças foram resfriadas por 18 horas a -5oC e a 9o, 10o e 11o costelas da meia carcaça esquerda foram submetidas à secção HH para verificar a proporção de ossos, músculo e gordura na carcaça. Para predição dos conteúdos de músculo, tecido adiposo e ossos da carcaça dos animais no corpo vazio, adotou-se a equação de regressão do logaritmo do conteúdo destes componentes no corpo vazio, em função do logaritmo do peso do corpo vazio - PCVZ (ARC, 1980). Derivando-se as equações acima, obtiveram-se as equações de predição da participação dos componentes corporais no ganho de 1 kg de peso de corpo vazio (GPCVZ). Na carcaça, o tecido adiposo teve maior impulso de crescimento em idade mais tardia, os tecidos ósseo e muscular tiveram maior impulso para crescimento em idade mais precoce. A carcaça apresentou alometria com valores bem próximos a 1, refletindo o seu desenvolvimento foi proporcionalmente igual ao desenvolvimento do PCVZ. / Abstract: The present study was performed to evaluate the relative growth of tissues in carcass of young buffaloes. Thirty and two Mediterranean non-castrated buffalo males, with an average age of 14 months and an initial living Weight of approximately 330 kilos, raised on native pasture on north litoral of Sao Paulo State, were randomly distributed in five experimental groups (I, II, III, IV) with 7 animals each and another four animals were considered as control group (AB). All of them received the same experimental diet ad libitum, twice daily. Animals were kept under solid fasting 16 hours before slaughter with free access to water. The experiment did not have a pre-defined duration, control group (AB) was slaughtered after an adaptation period of 30 days and the other groups were weighted every 28 days until they reached the pre-established weights of 450 (group I), 480 (group II), 510 (group III) and 540 kg (group IV). They were weighted every 28 days and as an animal approached the expected weight the measurement was performed in smaller intervals. After slaughter, each carcass was divided into two halves using a chainsaw, were individually weighted and then chilled for approximately 18 hours under - 5oC. Then the left half was submitted to a transversal section that included the 9a, 10a and 11a ribs in which the HH section was performed to a further and proportional analysis of muscle, adipose and bone tissues. The following regression formula of the logarithm of carcass and tissues (muscle, adipose and bone) Weights in function of the logarithm of empty body Weight. In the carcass, adipose tissue presented a late growth while bone and muscular tissues developed in an earlier age. The carcass presented alometric coeficient with approach value 1, reflecting its development was proportionally the same to the development of the PCVZ. / Mestre

The impact of rhizoma chuanxiong in fetal bone development

Background and purpose: Rhizoma Chuanxiong (CX), the dry rhizome of Ligusticum chuanxiong Hort., is a commonly used Chinese herbal medicine to treat gynecological diseases. So far, more than 60 chemical components have been identified from CX such as volatile oils (ligustilide, etc.), phenolic acids (ferulic acid, etc.) and alkaloids (chuanxiongzine, etc.). These components in CX are the basis of its wide pharmacodynamic actions including estrogen-like, progesterone-like and anti-coagulant/anti- platelet effects. In our recent survey based on previous published clinical trials, CX was ranked as one of the top 20 herbs commonly used for anti-miscarriages amongst Chinese pregnant women. However, CX should be used with caution during pregnancy as its property of 2invigorating blood circulation and removing blood stagnation3. Despite its wide applications, the safe dosage of CX in pregnant women remains unclear with no records found in the current Chinese Pharmacopoeia or other guidelines. Thus, verification regarding the impacts of CX preparations and its components in embryonic development is urgently required. In view of the limited experimental evidence that is currently available to assess the safety of CX, this project aims to (1) identify the general impacts of CX aqueous extract in maternal function and fetal development with an in vivo mouse model; and to (2) investigate the adverse impacts and underlying mechanisms of CX aqueous extract in fetal bone development with a biomarker assay and metabolomics analysis.;Concusion: CX aqueous extract at a low dosage of 2 g/kg/day (equals to the daily dosage of human adults) did not cause adverse effect in pregnant mice, and it suggested that this dosage of CX preparations should be safe for pregnant women. Our data demonstrated that high dosage and long-term use of CX aqueous extract might result in embryonic toxicities including fetal bone malformations for the first time. As the CX aqueous extract in this study was not contaminated by pesticide residues and heavy metals, the adverse impacts of CX aqueous extract should be considered as a result of its intrinsic components in the herb. Furthermore, CX aqueous extract might significantly down-regulate biomarkers related to bone formation and metabolism during osteogenesis. It is therefore valuable to establish a practical approach to systematically assess the safety of CX and other herbal medicines.;Method: Referred to the guidelines of WHO, the Chinese Pharmacopoeia and the Hong Kong Chinese Materia Medica Standards, CX aqueous extract was prepared, and its reference marker (ligustilide and ferulic acid) were quantitatively authenticated by HPLC analysis. LC/MS fingerprint analysis was performed for the quality control purposes. In addition, pesticide residues and heavy metals found in CX aqueous extract were examined using GC-MS and ICP-MS analysis. In the Segment II study as per FDA and OECD guidelines, pregnant mice were randomly assigned into 6 groups (n=18 per group): i.e. mice were orally administrated with distilled water as the negative controls (Group 1); or CX aqueous extract of 2, 16, 24 and 32 g/kg/day respectively from the gestation day (GD)6 to 16(Group 2, 3, 4 and 5); or vitamin A (200,000 IU) on GD7, 9 and 11 as the positive controls (Group 6). All mice were sacrificed to assess maternal and fetal parameters on the GD18. In the mechanistic study, the expressions of biomarkers related to fetal bone development including PICP, ICTP, B-ALP, BGP, Gdf-5, BMPs, BMP-6, BMP-8, BMP-11, IL-4, IL-4r, IL-10 and IL-10r in fetal tissue samples of the Group 1 and 5 (32 g/kg/day, n=18) were measured using ELISA analyses on GD16. Meanwhile, the metabolites of two-group samples were also analyzed by the UHD Accurate-Mass Q- TOF LC/MS, and profiling data was further analyzed by specific software. During statistical analysis, measurement data from G1, 2, 3 4 and 5 groups were analyzed using one-way ANOVA(SPSS software, version 16.0). LSD test in Post hoc method was applied to compare differences between every two groups. Pearsons x 2 - test was used to analyze category data from G1, 2, 3, 4 and 5 groups, and Fishers exact test was applied to compare differences between different groups. The student t-test was also used to compare differences between G1 and G6 groups in animal studies as well as G1 and G5 groups using ELISA or metabolomics results. An intragroup difference with a p-value less than 0.05 was considered as statistical significant.;Resutt:(1) There was no statistical significant difference in maternal and fetal parameters found between the Group 1 and 2 (p> 0.05). Determination of Hydroxyproline in Bone Collagen: Potential Application as a Biomarker for Bone Diseases

Hydroxyproline (Hyp), a non-proteinogenic amino acid is a component of the organic material in bone. It has been used for 14C-dating of bone and the measurement of Hyp could be used as a biomarker in bone metabolism. Hydroxyproline is a component of collagen, the main structural protein in bone. The analyses of 14C in collagen and Hyp in human bones may provide timing information about bone processes and diseases, such as osteoarthritis and osteoporosis. The analysis of Hyp in bones (e.g., the determination of Hyp content) primarily relies on a spectrometric technique, liquid chromatography-mass spectrometry (LC-MS), and the determination of 14C content requires accelerator mass spectrometry (AMS). Moreover, to obtain these materials from bone requires the successful extraction of collagen and thr separation of Hyp from the collagen. This study aims at comparing methods for extracting collagen from bone, which do not destroy the Hyp. These methods include the use of either NaOH, KOH or HCl in one stage of the extraction process and separating sufficient Hyp for 14C analysis. This will provide information to determine whether Hyp can be used as a biomarker for bone diseases like osteoarthritis and osteoporosis. A preliminary 14C AMS analysis on collagen extracted by the NaOH method was carried out on human bones previously analyzed for forensic purposes. This demonstrated the ability of this technique to provide recent (post 1950) timing information. The collagen extractions by three different methods were first conducted on modern chicken bone, and the results showed that KOH method is the best bone collagen extraction method, yielding a largest quantity of Hyp. The KOH method was then employed to extract collagen from cow bone as a test of a more human-like (mammalian) material. As this was successful, collagen was extracted from diseased human bone fragments, obtained from the Ottawa Hospital. The data revealed that Hyp was successfully obtained from these bones. The study demonstrates that the extraction as well as the separation methods (preparative HPLC) can provide sufficient Hyp from bones for 14C AMS analysis. This will lead to future studies of Hyp in bone turnover, which may lead to its use as a novel biomarker for bone diseases such as osteoarthritis and osteoporosis.

Os resultados mostraram que: a nanotopografia é capaz de aumentar o contato osso-implante (BIC) mesmo na presença da inalação intermitente da fumaça de cigarro, e foi capaz de reduzir, porem não totalmente, seus efeitos deletérios e uma menor formação óssea; A nanotopografia de titânio pode ter papel importante no processo de mineralização da matriz extracelular e na modulação da expressão de citocinas; E que a incorporação de LN-1 a superfície de titânio com nanotopografia demonstrou favorecer uma maior expressão de importantes genes envolvidos na cascata da osseointegração. / The aims of this study were to evaluate the influence of nanotopography of titanium surfaces in the process of osteogenesis by means of histological assessment of bone-implant contact, in rats, under the effect of forced inhalation of cigarette smoke; To assess the osteogenesis process and the expression of cytokines in primary cell culture on titanium disks with nanotopography; And evaluate morphological and molecular the effect of incorporation of laminin-1 (LN-1) surfaces with nanotopography, in rabbits. Implants and titanium disks were specially produced and submitted to different methods for obtaining surfaces with turned, micro and nanotopography. The surfaces have been properly characterized as its topography, morphology and chemistry. The results showed that: nanotopography is able to increase bone-implant contact (BIC) even in the presence of intermittent inhalation of cigarette smoke, and was able to reduce, although not entirely, their harmful effects and a lower bone formation; Nanotopography may play a important role in mineralization process and in cytokine expression.; And the incorporation of LN-1 on titanium surface with nanotopography favor a higher expression of important genes involved in the cascade of osseointegration.

Materiais e Métodos: Trinta pacientes com indicação para reabilitação por meio de implantes instalados na região posterior de mandíbula foram divididos em dois grupos, seguindo os critérios de inclusão e exclusão previamente estabelecidos. O primeiro grupo não apresentava alterações sistêmicas (GSA) e não fazia uso de qualquer medicação e, o segundo grupo foi composto por pacientes hipertensos diagnosticados e medicados por antagonistas do SRAA (GAS). Durante o procedimento cirúrgico para instalação dos implantes osseointegráveis com superfície texturizada, foram coletados blocos ósseos por meio de biópsia com broca trefina de 3,0mm de diâmetro nos locais de instalação dos implantes. As biópsias coletadas foram separadas para avaliação por microtomografia computadorizada, sendo avaliados os seguintes parâmetros: volume ósseo (BV/TV), percentual de volume ósseo (BV/TV), espessura de trabéculas (Tb.Th), número de trabéculas (Tb.N), separação entre as trabéculas (Tb.S) e porosidade total (Po-tot). Também foi realizada a análise dos cortes histológicos corados por hematoxilina e eosina, bem como a avaliação imunoistoquímica de proteínas que caracterizam as células da linhagem osteoblástica: Runx2, Osteopontina e Osteocalcina. Resultados: Para todos os parâmetros analisados (BV, BV/TV, Tb.Th, Tb.N, Tb.S e Po-tot), houve similaridade na comparação entre os grupos GAS e GSA (p<0,05, teste t). As imunomarcações para osteopontina e osteocalcina mostraram-se semelhantes nos dois grupos. Vale destacar importante marcação observada no grupo GAS para o fator de transcrição Runt 2 (Runx2), em comparação a marcação da mesma proteína no grupo GSA. Além disso, a biologia do tecido ósseo do ponto de vista histológico foi semelhante nos dois grupos. Conclusões: Foi possível concluir que indivíduos hipertensos tratados com antagonistas do SRAA apresentam grande similaridade óssea microestrutural e também celular/protéica aos indivíduos normotensos. / Objectives: This study aimed to evaluate the morphometric characteristics, histological and immunohistochemical bone tissue collected from patients with hypertension offset by the use of drugs antagonists of the renin angiotensin system. Material and Methods: 30 patients referred for rehabilitation with implants placed in the posterior mandible were divided into two groups, following the criteria of inclusion and exclusion previously established. The first group showed no systemic changes (GSA) and did not use any medication and the second group was composed of diagnosed hypertensive patients treated by RAAS (GAS) antagonists. During the procedure for installation of dental implants with textured surface, they were collected bone blocks through biopsy trephine drill 3.0mm diameter implants in the installation sites. The collected biopsies were separated fo evaluatio by microtomography, being evaluated the following parameters: bone volume (BV / TV), trabecular thickness(Tb.Th), trabecular number (Tb.N), separation of the trabecular (Tb.S) and total porosity (Po-tot). Also the analysis of histological sections stained with hematoxylin and eosin staining was performed as well a the immunohistochemical evaluation of proteins that characterize the cells of osteoblast lineage: Runx2, osteopontin and osteocalcin. Results: For all parameters (BV, BV / TV, Tb.Th, Tb.N, Tb.S and E-tot) were similar in the comparison between GAS and GSA (p> 0.05, t test). The biology of bone tissue to analyze by immunohistochemistry presented similar results in both groups, with markings scores in ordinal qualitativ analysis for transcription factor related to Runt 2 (Runx2) GSA (light) and GAS (moderate), osteopontin (OP) GSA (mild to moderate) and GAS (moderate to intense), osteocalcin (OC) GSA (intense) and GAS (moderate). Furthermore, the biology of bone tissue was histologically similar in both groups. Conclusions: Therefore, it was concluded that hypertensive subjects treated with renin-angiotensin system antagonists have great bone microstructural similarity and also cell/protein to normotensive individuals.

Foram selecionadas 30 crianças de ambos os sexos, com faixa etária entre 9 e 14 anos para o sexo feminino e, entre 11 e 16 anos para o sexo masculino. De cada criança foram obtidas duas radiografias, uma pela técnica convencional e outra digital direta para a posterior análise comparativa das imagens. O filme e o sensor foram posicionados e centralizados na região da articulação metacarpofalangeana do polegar esquerdo, de forma que o centro do sensor ou filme coincidisse com a posição da articulação metacarpofalangeana. As radiografias convencionais e digitais diretas foram analisadas por uma Odontopediatra, procurando identificar no osso sesamóide: ausência (A), calcificação inicial (CI) e calcificação presente (CP). Na falange proximal do polegar esquerdo, foram identificados os estágios de capeamento epifisário a= epífise menor que a largura da diáfise, b= epífise igual à largura da diáfise, c= estágio de capeamento, d= fusão da epífise com a diáfise. As observações foram submetidas a análise estatística chegando-se as seguintes conclusões: 1 - A ossificação do osso sesamóide mostrou a melhor correlação entre os dois tipos de radiografias analisadas 2 - O observador apresentou melhor índice de coincidência nos fatores analisados na radiografia digital, sendo sua maior dificuldade o reconhecimento do estágio do início da calcificação do osso sesamóide na radiografia convencional. / Abstract: The aim of this study was to evaluate the analysis of bone maturation in the simplified radiographic technique thirty children, age from 9 to 14 years old for female and from 11 to 16 for male took part of this study. From each child were obtained two radiographies, using conventional and digital techniques. The film and the sensor were positioned centralized in the area of the metacarpophalangeal joint of the left Thumb. The images were analyzed by a Pediatric Dentist. Calibrated to identify in the sesamoid bone: absence (A), initial calcification (CI) and present calcification (CP). In the proximal phalange of the left thumb were identified the epiphyseal changes: a - Ephiphysis equal in width to diaphysis, b= Appearance of sesamoid of the thumb, c= Capping of epiphysis, d= Fusion of epiphysis. The results shouled that the sesamoid bone avaliation had the best correlation in both radiographies : Conventional and Digital. The observer was more reliable when analysed digital X rays. / Mestre

Orientador: Carlos Ducatti / Banca: Ariel Antonio Mendes / Banca: Marcelo Zacharias Moreira / Banca: José Roberto Sartori / Banca: Fabiana Ribeiro Caldara / Resumo:O estudo objetivou rastrear a inclusão de farinha de carne e ossos bovina (FCOB) em dietas de poedeiras comerciais, por meio da análise dos ovos e suas frações (gema e albúmen), pela técnica dos isótopos estáveis do carbono e nitrogênio, assim como avaliar o índice analítico mínimo detectável. Foram utilizadas 240 galinhas poedeiras da linhagem Shaver White de 73 semanas de idade, em delineamento experimental inteiramente casualizado, com cinco tratamentos e seis repetições. Os tratamentos consistiram de níveis de inclusão (0; 1,5; 3,0; 4,5 e 6,0%) de FCOB na dieta a base de milho e farelo de soja. No 35º dia foram tomados aleatoriamente 24 ovos por tratamento, dos quais 12 serviram para amostragem de gema e albúmen e os outros 12 para ovo (gema + albúmen). Os resultados isotópicos foram submetidos à análise multivariada de variância e a partir das matrizes de erro, com 95% de confiança, foram determinadas elipses para identificar as diferenças entre os tratamentos experimentais (dietas com FCOB) e o grupo controle (0% de FCOB). No ovo e na gema a partir do par isotópico do tratamento com inclusão de 3,0% de FCOB houve diferenciação do par do tratamento controle, no albúmen a diferenciação ocorreu a partir do tratamento com 1,5% de FCOB. Conclui-se que, a técnica dos isótopos estáveis é capaz de rastrear a FCOB, utilizada na alimentação de poedeiras, no produto final, em nível mínimo de inclusão de 1,5% no albúmen e 3,0% no ovo e na gema. / Abstract: The aim of this study was to trace the inclusion of bovine meat and bones meal (BMBM) in diets of laying hens analyzing eggs and theirs fractions (yolk and albumen), by carbon (δ13C) and nitrogen (δ15N) stable isotopes, as well as to evaluate the detectable analytical minimal index. Two hundred forty (240) Shaver White laying hens aging 73 weeks never fed with animal origin ingredients were randomly distributed in five treatments and fed with a corn and soybean based diet (control) and four increasing levels (0; 1.5; 3.0; 4.5 and 6.0%) of BMBM. At the 35th day, 24 eggs per treatment were randomly collect, twelve for yolk and albumen sampling and twelve for egg (yolk + albumen) sampling. The isotopic results were analyzed in a multivariate analysis of variance. Through an error matrix (95% confidence) the ellipses were determined to identify the differences among the treatments (diets BMBM inclusion) from the control group (0% BMBM group). It was possible to detect BMBM inclusion through the isotopic pair of yolk and egg at 3.0% of inclusion. In the albumen it was possible to detect the 1.5% BMBM inclusion. In summary, the stable isotopes technique is able to trace BMBM in laying hens feed, in the final product at a minimal level of inclusion of 1.5% in the albumen and 3.0% in the egg and yolk. / Doutor

Increased CKIP-1 suppresses Smad-dependent BMP signaling to inhibit bone formation during aging

Emerging evidence indicates that the dysregulation of protein ubiquitination plays a crucial role in aging-associated diseases. Smad-dependent canonical BMP signaling pathway is indispensable for osteoblastic bone formation, which could be disrupted by the ubiquitination and subsequent proteasomal degradation of Smad1/5, the key molecules for BMP signaling transduction. However, whether the dysregulation of Smad1/5 ubiquitination and disrupted BMP signaling pathway are responsible for the age-related bone formation reduction is still underexplored. Casein kinase-2 interacting protein-1 (CKIP-1), also known as Pleckstrin homology domain-containing family O member 1 (PLEKHO1), is a previously identified ubiquitination-related molecule that could specifically target the linker region between the WW domains of Smurf1 to promote the ubiquitination of Smad1/5. Here, we found an age-related increase in the expression of CKIP-1 in bone specimens from either fractured patients or aging rodents, which was associated with the age-related reduction in Smad-dependent BMP signaling and bone formation. By genetic approach, we demonstrated that loss of Ckip-1 in osteoblasts could promote the Smad-dependent BMP signaling and alleviated the age-related bone formation reduction. In addition, osteoblast-specific Smad1 overexpression had beneficial effect on bone formation during aging, which could be counteracted after overexpressing Ckip-1 within osteoblasts. By pharmacological approach, we showed that osteoblast-targeted CKIP-1 siRNA treatment could enhance Smad-dependent BMP signaling and promote bone formation in aging rodents. Taken together, it suggests that the increased CKIP-1 could suppress Smad-dependent BMP signaling to inhibit bone formation during aging, indicating the translational potential of targeting CKIP-1 in osteoblast as a novel bone anabolic strategy for reversing established osteoporosis during aging.
