Maintaining genomic integrity is crucial for cell survival. In Escherichia coli, Rec-Amediated homologous recombination (HR) plays an essential role in the repair of DNA double-strand breaks (DSB) and the SOS response through a series of highly dynamic interactions with the chromosome. A greater understanding of the mechanism of homologous recombination requires quantitative analysis of genomic studies in live cells. The aim of this thesis was to investigate the dynamics of the RecA-DNA interactions in vivo following the induction of a site-specific DSB in the chromosome of E. coli. This DSB is caused by the cleavage of a DNA hairpin by the hairpin-specific endonuclease, SbcCD. The DNA hairpin is formed only on the lagging strand template of replication by a 246 bp-interrupted palindrome. As a result cleavage only occurs on one sister chromosome, leaving one unbroken chromosome to serve as a template for repair by HR. Here, this system has been used as a basis to develop a method that combines chromatin immunoprecipitation with quantitative PCR (ChIP-qPCR) and next-generation sequencing (ChIP-Seq) to quantify RecA protein binding during the active repair of a single chromosomal DSB. This study reports that DSB-dependent RecA binding is stimulated in response to the eight base DNA sequence Chi (5’-GCTGGTGG-3’). Increasing the number of Chi sites close to the DSB stimulates more RecA loading to DNA, with ChIP-Seq analysis also revealing a role for subsequent Chi sites in RecA binding during DSBR. If the Chi sites close to the DSB are removed then Chi-dependent RecA binding to DNA can be observed at distances greater than 100 kb from the DSB, suggesting that these subsequent Chi sites can be engaged in DSBR. Through collaboration, these in vivo data were combined with stochastic modeling to determine that, in vivo, Chi is recognised by the RecBCD complex with an efficiency of 20- 35%. The genomic analysis also revealed two unexpected aspects of RecA protein binding. First, ChIP-Seq analyses identified that following a DSB at lacZ there is RecA enrichment detected in the terminus region of the E. coli chromosome. This RecA binding is Chi-dependent, indicating a role for HR. Second, DSB-independent binding was observed at the RNA encoding genes dispersed throughout the chromosome. A temporal analysis of RecA dynamics was also performed. These analyses revealed that RecA binding to DNA near the DSB is extremely dynamic, cycling between periods of high RecA enrichment and periods of low RecA enrichment. This is the first in vivo study of DSB-dependent RecA-DNA distribution and dynamics in recombination proficient E. coli cells.

Syftet med detta examensarbete var att beräkna vad det kostar att ha UPS-kraft (Uninterruptible Power Supply). Arbetet utfördes för Västfastigheter på Södra Älvsborgs Sjukhus (SÄS) i Borås. Studien utfördes på en UPS i drift i byggnad 7. UPS: en i denna byggnad har 10 kVA kapacitet och är av dubbelkonverterande typ. Batterier av Ni-Cd (Nickel-Kadmium) typ används till backup med UPS: en. UPS av dubbelkonverterande typ är känd för att skapa mycket övertoner, men är fördelaktig för dess skyddande förmåga mot spänningsspikar.Mätningar har gjorts för att beräkna kostnadsberäkningarna. Transações bancárias, negociações na bolsa de valores e compras pelo computador ocorrem em qualquer horário e as empresas precisam garantir a disponibilidade de seus serviços sempre. Para garantir o fornecimento ininterrupto de energia para estes sistemas de informações são utilizados grupos motorgerador (GMG) e uninterruptible power supply (UPS). Quando há dificuldade de acoplamento entre os dois equipamentos, na prática, os projetistas superdimensionam o GMG em relação ao UPS sem embasamento teórico para esta ação. Este estudo propõe apresentar a correlação entre os dados de placa desses equipamentos e o distúrbio de tensão entre eles para proporcionar uma maior segurança no dimensionamento de sistemas com UPS e GMG. A metodologia desse estudo propõe o uso de Redes Neurais Artificiais (RNA s) para descrever esta correlação e transformada wavelet para destacar os distúrbios medidos e auxiliar na convergência da RNA em seu treinamento. Os resultados obtidos com as redes desenvolvidas neste trabalho sugerem que o menor distúrbio no sinal, obtido devido ao acoplamento de UPS e GMG, ocorrerá se as potências dos equipamentos (dados de placa) forem próximas, independente do nível de carregamento do sistema. À medida que esta relação se distancia do valor unitário, a perturbação aumenta e um UPS com potência menor que o GMG contribui mais com a distorção do sinal do que ao contrário

The reliability of monetary policy as an economic stabilisation tool depends on the understanding of the empirical effects of policy intervention on macroeconomic aggregates. Since investigating the interdependencies between macroeconomic variables necessarily involves studying their interactions over time, time series analysis is an important tool. This thesis sets out to examine the presence and effects of nonstationarity in the form of a structural break in a basic VAR of four endogenous variables. Specifically, the transmission of a monetary policy shock on the macroeconomic aggregate of 11 Euro Area countries is estimated for the period 1999–2017, employing variables based on previous studies. A Quandt-Andrews breakpoint test is used to identify the break date, and a comparison is made between the periods. This study finds support for the presence of a break in the regression estimate from the breakpoint test, although the reults from the IRFs cannot be shown to be statistically significant, nor to be bias-free.

DNA double strand break (DSB) repair is critical to maintain genomic integrity and cell viability. DSBs can occur during the course of cell cycle during replication or transcription, or by exogenous agents such as chemicals or ionizing radiation. For my thesis, I studied homologous recombination (HR), which has two sub-pathways: Homology Directed Repair (HDR) and Single Strand Annealing (SSA). HDR involves strand invasion of a homologous template to prime DNA synthesis; SSA involves annealing of homologous segments flanking a DSB. Background data showed that depletion of XAB2 protein by RNA interference reduced both HDR and SSA events. XAB2 protein contains 15 tetratricopeptide repeat (TPR) motifs, which likely enable protein-protein interactions. While XAB2 is speculated to have a role in transcription coupled repair and pre-mRNA splicing, its role in HR pathway is uncertain. The overall hypothesis for my thesis is that XAB2 mediates a specific step of HR (5’-3’ end resection), and the TPR motifs present in XAB2 enable the protein to function in a complex during HR. By using an end resection assay and cell biology analysis, I found that XAB2 is essential for 5’ – 3’ end resection, an intermediate step common to both HDR and SSA pathways. With a functional complementation assay I developed, I have shown that specific TPR regions are critical for XAB2 functions in HR. Overall, my research demonstrates that XAB2 protein has a key role in the 5’-3’ end resection step of HR, and its function in HR requires specific sets of its TPR regions.

Hunddagiskooperativet kommer att drivas som en ekonomisk förening för att ha möjlighet att i framtiden kunna vara vinstdrivande.</p><p>En enkätundersökning genomfördes för att se vad potentiella kunder efterfrågar av ett hunddagis och vad som skulle kunna göra att kunderna väljer detta dagis framför andra. Until recently, research has been scarce in the field of physical modelling of dam breaching. Over the past few years, teams from the University of Ottawa, Canada, Delft University of Technology, Netherlands, and HR Wallingford, United Kingdom have worked on several physical models to help determine how various dam breaching characteristics vary due to changes in dam geometry and geotechnical properties. The purpose of this project is to use these new experimental data sets to compare and validate the applicability range of two existing pieces of software, MIKE11-DB and BREACH developed by the Danish Hydraulic Institute and National Weather Service, respectively. Several breaching characteristics such as the outflow hydrograph, peak flow, lag time, breaching time, breach width, and water level are considered in the present study. A sensitivity analysis is also performed on the model’s main input parameters and their sensitivity and performance is ranked accordingly.
