Thesis (MScAgric)--Stellenbosch University, 2014. / ENGLISH ABSTRACT: In South Africa, canola (Brassica napus L.) is produced under short day conditions during winter months. These conditions, together with high fertiliser application levels required to maximize grain yields, often result in tall growing bulky crops which are prone to lodging. This will especially be true if canola production is expanded to irrigated areas. Plant growth regulators (PGRs) have successfully been used to reduce canola plant height and lodging under experimental conditions in Australia and are worldwide commercially used to reduce plant height and lodging in winter cereals such as wheat and barley. The primary objective of this study was to determine the effect of anti-lodging PGRs on the agronomic and quality characteristics of commercial canola cultivars under South African conditions. This study was conducted under field conditions at three research farms, as well as controlled glasshouse conditions at Welgevallen Research Farm, situated in the Western Cape Province of South Africa. Foliar treatments consisted of a control (untreated) and four PGRs; CeCeCe® 750 (chlormequat chloride), Moddus® 250 EC (trinexapac-ethyl), Primo MAXX® (trinexapac-ethyl), and Kelpak®, applied either individually or in combination with wetting agent at budding stage (growth stage 3.1) of canola. Whilst glasshouse trials were conducted with spring canola cultivars “Hyola 555TT” and “43C80”, field trials were done with Hyola 555TT only. Monitoring and measuring various plant parameters during different growth stages of canola, the morphological and physiological impact of PGR-treatments on growth and development were determined. Though Primo MAXX® tends to reduce plant height in all trials; reductions were only significant during one of the glasshouse trials. Fortunately, compared to the control, none of the PGRs significantly reduced the leaf area, number of flowers or number of pods plant-1 during this study, while Primo MAXX® and Moddus® 250 EC tend to increase the grain yield under field conditions. This study indicates that PGRs can possibly be used to improve lodging resistance and yield of canola. Identifying the most effective PGRs on specific cultivars, the results of the study will contribute to the knowledge of using PGRs in canola to reduce lodging and improve grain yields in South Africa. / AFRIKAANSE OPSOMMING: In Suid-Afrika word canola (Brassica napus L) gedurende die wintermaande onder kort-dagtoestande verbou. Hierdie verbouingstoestande, tesame met verhoogde toediening van bemesting vir maksimum opbrengs, lei dikwels tot welige, hooggroeiende plantegroei, wat „n geneigdheid toon om om te val (lodge). Hierdie probleem vererger indien canola onder besproeiingstoestande gekweek sou word. Plantgroeireguleerders (PGRs) is reeds met sukses onder eksperimentele toestande in Australië gebruik om die planthoogte en dus omval (lodging) van canola te beperk. Plantgroeireguleerders word reeds kommersieel gebruik om planthoogte en omval van wintergraangewasse, soos koring en gars te verminder. Die hoofdoel van hierdie studie was om die effek van omval-teenwerkende PGRs op die agronomiese en kwaliteitseienskappe van kommersiële canola-kultivars onder Suid-Afrikaanse groeitoestande te bepaal. Hierdie studie is uitgevoer d.m.v. veldproewe op drie navorsingsplase, sowel as gekontrolleerde glashuisproewe te Welgevallen Navorsingsplaas, geleë in die Wes-Kaapprovinsie van Suid-Afrika. Verskeie plantparameters is gemonitor en gemeet gedurende die verskillende groeistadia van canola, waartydens en die morfologiese en fisiologiese impak van PGR-toedienings op die groei- en ontwikkeling van canola bepaal is. Alhoewel Primo MAXX® neig om die planthoogte in al die proewe te verkort, het dit planthoogte slegs beduidend verkort in een van die glashuisproewe. Geen van die PGRs het in vergelyking met die kontrole, die blaar-oppervlakte, aantal blomme of aantal peule plant-1 beduidend verminder gedurende die studie. Carne Bovina Reestruturada com Óleo de Canola e Antioxidante: desenvolvimento e atributos sensoriais / Restructured beef with canola oil and antioxidant: development and sensory attributes

As indústrias do setor da carne têm buscado meios para agregar valor através da adição de ingredientes benéficos à saúde e pela utilização de cortes de baixo valor comercial. A produção de carnes reestruturadas e o uso de óleos vegetais em substituição a gordura animal são estratégicas tecnológicas, criando produtos mais adaptados as necessidades do consumidor em termos de conveniência, uniformidade, tamanho de porção, composição, fácil preparação e alimentação saudável. Portanto, objetivou-se avaliar as características quantitativas e qualitativas e a vida útil de bifes reestruturados desenvolvidos com músculo Triceps brachii (miolo da paleta), utilizando a enzima transglutaminase, antioxidante e adição de óleo de canola, de acordo com os tratamentos: (1) controle, (2) adição de 5% de óleo de canola, (3) adição de eritorbato de sódio e (4) adição de 5% óleo de canola + eritorbato de sódio. A carne foi cortada e processada com 1% de NaCl, 0,3% de tripolifosfato de sódio, 1% de enzima transglutaminase e 10% de gordura da carne. Ainda no misturador foram adicionados 5% de óleo de canola (2 e 4) e 0,05% de antioxidante (3 e 4). Os bifes foram embalados á vácuo individualmente e armazenados congelados a -18°C por até 120 dias. Foram analisados: composição centesimal, pH, perdas durante o descongelamento e cozimento, análise instrumental da cor, teor de colesterol, oxidação lipídica, textura e análise sensorial Os valores de pH foram maiores (P<0,05) aos 120 dias de armazenamento congelado, independente dos tratamentos. A adição de óleo de canola afetou (P<0,05) a composição centesimal dos bifes, com exceção do teor de cinzas que não se alterou (P>0,05). A maior perda de cozimento foi encontrada nos bifes reestruturados formulados com antioxidante mais óleo de canola (23,66%), diferindo (P<0,05) do bife reestruturado somente com antioxidante que apresentou a menor perda (16,34%), porém não deferiram dos tratamentos controle e com óleo de canola. Não houve diferença (P>0,05) para oxidação lipídica entre os tratamentos aos 0 e 30 dias de armazenamento, já aos 60, 90 e 120 dias, houve diferença (P<0,05), onde os tratamentos sem adição de antioxidante tiveram os maiores resultados para TBARS. O tratamento com canola (2) também diferiu do tratamento controle (1) apresentando menores valores de TBARS aos 90 e 120 dias de armazenamento. A adição do óleo de canola e do antioxidante afetou (P<0,05) o teor de colesterol na carne reestruturada crua, aumentou a luminosidade (L*) e intensidade do amarelo (b*), mas não influenciou a intensidade de vermelho (a*). Os tratamentos com adição de óleo de canola apresentaram os menores valores de dureza. Os bifes reestruturados com adição de óleo de canola e eritorbato de sódio possuem propriedades físico-químicas e sensoriais aceitáveis, podendo ser comercializado como um produto de preparo rápido e possivelmente com maior valor agregado. / The meat sector industries are looking for ways to add value by adding ingredients beneficial to health and by use the cuts of low commercial value. The restructured meat production and use vegetable oils in place of animal fat are strategic technology, creating products more adapted to the needs of the consumer in terms of convenience, uniformity, portion size, composition, easy preparation and healthy eating. Therefore, the objective was to evaluate the quantitative and qualitative characteristics and shelf life of the restructured beef developed with Triceps brachii, using the enzyme transglutaminase, antioxidant and addition of canola oil, according to the following treatments: (1) control, (2) added 5% canola oil, (3) added of sodium erythorbate and (4) added of 5% canola oil + sodium erythorbate. The meat was cut and processed with 1% NaCl, 0.3% sodium tripolyphosphate, 1% transglutaminase enzyme and 10% beef fat. Also in the mixer was added 5% canola oil (2 and 4) and 0.05% of antioxidant (3 and 4). The steaks were vacuum packaged individually and stored frozen at -18 °C up to 120 days. In the final product were analyzed: chemical composition, pH, losses during thawing and cooking, instrumental analysis of color, cholesterol, lipid oxidation, texture and sensory analysis. The pH values were higher (P <0.05) at 120 days of frozen storage for all treatments. The addition of canola oil affected (P <0.05) the chemical composition of restructured steaks, with the exception of ash content did not change (P> 0.05). The greatest loss of cooking has been found in restructured steaks formulated with antioxidant more canola oil (23.66%) differing (P <0.05) of the restructured steak with only antioxidant that showed the smallest loss (16.34%), but not differed the control and canola oil treatments. There were no difference (P> 0.05) for lipid oxidation between treatments at 0 and 30 days of storage, but at 60, 90 and 120 days, there were a difference (P <0.05), where treatments without antioxidant addition had the greatest results for TBARS. Treatment with canola (2) also differed from the control treatment (1), exhibited lower TBARS values at 90 and 120 days of storage. The addition of canola oil and antioxidant affected (P <0.05) cholesterol content in raw restructured beef increased the lightness (L *) and intensity of yellow (b *), but did not influence the intensity of red ( a *). The treatments with the addition of canola oil had the lowest hardness values. The steaks restructured with the addition of canola oil and sodium erythorbate have physicochemical and sensory properties acceptable and can be marketed as a product of rapid preparation and possibly with greater added value.

O óleo de canola é considerado um alimento saudável, e também é utilizado na produção de biodiesel. Além disso, a canola pode ser considerada boa opção no esquema de rotação de culturas. O nitrogênio é um dos nutrientes mais exigidos pela planta para aumento na produtividade, enquanto que o enxofre é essencial para proporcionar qualidade às sementes. Assim, objetivou-se avaliar os efeitos de doses de nitrogênio e enxofre na produtividade, nos teores de S-SO4 no solo, nas concentrações de N e S nas folhas e nos teores de óleo e proteína bruta nos grãos de canola. O experimento foi realizado no município de Uberaba, Minas Gerais, na área experimental do Instituto Federal do Triângulo Mineiro - IFTM, em Latossolo Vermelho distrófico, textura média, no ano de 2010. Utilizou-se o híbrido Hyola 401 em delineamento em blocos ao acaso com quatro repetições e 20 tratamentos em esquema fatorial 5 x 4, sendo cinco doses de N (0, 60, 100, 140 e 180 kg ha-1) e quatro doses de S (0, 15, 30 e 60 kg ha-1). A adubação nitrogenada e sulfatada aumentou a produtividade de grãos de canola, sem alterar significativamente os teores de óleo e proteína bruta. Doses superiores a 140 kg ha-1 de N e 15 kg ha-1 de S proporcionaram produtividades de grãos acima de 1100 kg ha-1. Concentrações superiores a 52 g kg-1 de N nas folhas proporcionaram produtividades de grãos superiores a 1000 kg ha-1. Os maiores teores de S foram observados na camada de 0,15-0,30 m, indicando mobilidade do ânion sulfato. As menores produtividades de grãos foram observadas quando os teores de S no solo nessa camada eram menores que 4 mg dm-3. Molecular strategies for increasing seed oil content in canola

Previous research has shown that microsomal DGAT activity from cultures of Brassica napus was stimulated by human acylation stimulating protein (ASP) and bovine serum albumin (BSA). Genetic constructs were engineered to facilitate the expression of ASP or BSA in developing seeds to test the stimulatory effect of these proteins at the site of TAG formation. As well, genetic constructs were designed to produce a truncation of the BSA polypeptide in an attempt to localize the portion fo the macromolecule responsible for stimulation of DGAT activity. An oleosin promoter was used for seed specific expression and to express the polypeptides at a precisely cooridnated time when oil was accumulating in the developing seeds. Lipid analysis coordinated time when oil was accumlating in the developing seeds. Lipid analysis was performed on the seeds of transgenic plants designed to cytosolically express these mammalian proteins and the seeds of control plants. The first generation data revealed that the total lipid within the mature seeds of ASP and BSA plants was not significantly different from the total lipid of negative control plants using both gravimetric and low resolution-nuclear magnetic resonance methods of analysis. The seeds from ASP 8 and ASP 10 plants, however, did produce significantly more lipid on a per seed basis as compared to negative control plants. The levels of the fatty acid composition for total acyl lipids were measured in these first generation transgenic plants. ASP 3 had significantly lower levels of linoleic acid, ASP 14 had significantly lower levels of a-linolenic acid and BSA 11 had significantly higher levels of both of these fatty acids in comparison to negative control plants. / xix, 245 leaves : ill. ; 28 cm.

The meat was cut and processed with 1% NaCl, 0.3% sodium tripolyphosphate, 1% transglutaminase enzyme and 10% beef fat. Also in the mixer was added 5% canola oil (2 and 4) and 0.05% of antioxidant (3 and 4). The steaks were vacuum packaged individually and stored frozen at -18 °C up to 120 days. In the final product were analyzed: chemical composition, pH, losses during thawing and cooking, instrumental analysis of color, cholesterol, lipid oxidation, texture and sensory analysis. The pH values were higher (P <0.05) at 120 days of frozen storage for all treatments. The addition of canola oil affected (P <0.05) the chemical composition of restructured steaks, with the exception of ash content did not change (P> 0.05). The greatest loss of cooking has been found in restructured steaks formulated with antioxidant more canola oil (23.66%) differing (P <0.05) of the restructured steak with only antioxidant that showed the smallest loss (16.34%), but not differed the control and canola oil treatments. There were no difference (P> 0.05) for lipid oxidation between treatments at 0 and 30 days of storage, but at 60, 90 and 120 days, there were a difference (P <0.05), where treatments without antioxidant addition had the greatest results for TBARS. Treatment with canola (2) also differed from the control treatment (1), exhibited lower TBARS values at 90 and 120 days of storage. The addition of canola oil and antioxidant affected (P <0.05) cholesterol content in raw restructured beef increased the lightness (L *) and intensity of yellow (b *), but did not influence the intensity of red ( a *). The treatments with the addition of canola oil had the lowest hardness values. The steaks restructured with the addition of canola oil and sodium erythorbate have physicochemical and sensory properties acceptable and can be marketed as a product of rapid preparation and possibly with greater added value.

MODELO DE GERENCIAMENTO DOS SERVIÇOS DE DISTRIBUIÇÃO FÍSICA NUMA AGROINDÚSTRIA

In a globalized and competitive world, logistics is receiving a lot of attention from business managers because of its importance concerning the fidelity of customers and their participation in the total costs of the company. Therefore, this paper aims to develop a model to the management of the services of physical distribution based on the six sigma strategy in an agroindustry that produces canola oil and it is located in the northwest of the state of Rio Grande do Sul, Brazil. The data were collected by using a questionnaire which refers to the level of satisfaction and the importance of the services offered by the company concerning the customers attendance; the kind of ordering, invoice service and payment; logistic of transports and the information given. In order to analyze the data it was used the descriptive statistics and the techniques of analyses Cluster hierarchic and Cluster k-means. The six customers who buy unmanufactured canola oil from this company took part on this study. The results showed that the customers are satisfied with most of the services offered by the company. It was also observed that the customers demonstrated a lot of satisfaction referring to the kind of ordering, invoice service and payment. However, the clients consider the attendance service as being the most important feature of the company. Based on these results, a model to the management of the services of physical distribution based on the six sigma strategy was proposed with the purpose of guaranteeing the quality of services offered. / Em um mercado globalizado e competitivo a logística começa a receber maior atenção por parte dos gestores empresariais, devido à sua importância quanto a fidelização de clientes e sua participação nos custos totais da empresa. Por isso, este trabalho tem por objetivo propor um modelo para o gerenciamento dos serviços de distribuição física, com base na estratégia seis sigma, em uma agroindústria produtora de óleo bruto de canola situada na região noroeste do Estado do Rio Grande do Sul. Os dados foram coletados por meio de um questionário, referentes ao grau de satisfação e importância dos serviços prestados pela empresa quanto ao atendimento; tomada de pedido, faturamento e pagamento; logística de transportes e informações prestadas. Para a análise dos dados utilizou-se a estatística descritiva e a técnica da análise de Cluster hierárquico e Cluster k-means. Fizeram parte deste estudo os seis clienteadquirem o óleo bruto de canola da empresa estudada. Os resultados mostraram que os clientes estão satisfeitos com a maioria dos serviços prestados pela empresa. Observou-se, também, que os clientes estão mais satisfeitos com os serviços referentes à tomada de pedidos, faturamento e pagamento. Porém, quanto ao grau de importância, eles consideram mais importante os serviços prestados no atendimento. Com base nestes resultados foi proposto um modelo de gerenciamento para os serviços de distribuição física baseado na estratégia seis sigma, visando garantir a qualidade dos serviços prestados.
