Cargo inspection by means of electromagnetic radiation

This study is based on a detailed literature review of the approaches that are being used for inspecting cargos. It was concluded that especially X-rays are suitable for precise inspection of cargos due to high-quality output images. The images captured with X-Ray scanning methods are sufficient for discriminating various materials. Methods for the inspection of cargos with the help of electromagnetic radiation at radio frequencies are also discussed.

Fatigue analysis and reconstruction of helicopter load spectra

Helicopter load histories applied to notched metal samples are taken as examples, and their fatigue lives are predicted by using a simplified version of the local strain approach. This simplified method requires an input load history in the form of the rain-flow matrix and places bounds on the fatigue life. A peak-valley reconstructed history is generated based on the standard spectrum Helix. A second history studied is a more irregular one based on actual flight data. lt is used to generate three reconstructed histories based on three principles: peak-valley, to-from, and rain-flow. Emphasis is given to the rain-flow reconstruction method, and different reconstruction methods based on rain-flow cycle counting are presented. Life predictions are presented for all of the above cases, and the comparison with test data and other considerations suggest that the most promising reconstruction approach is one based on rain-flow cycle counting. Finally, a method is presented which reconstructs a history with the same rain-flow cycles and also the same distribution of relative time increments between adjacent peaks and valleys. This reconstructed history gives the same fatigue life as the original history. / Ph. D.

Cargo Cycles in Urban Freight Transport : Obstacles and facilitating factors for utilising cargo cycles in urban freight transport in Stockholm, Sweden

Cargo cycles can play a considerable role in mitigating the negative impacts of urban freight transport while still ensure that the material needs of the city are fulfilled. Their small size, lower operating costs, smaller carbon footprint, lack of tailpipe emissions, and manoeuvrability in congested areas are considerable advantages over traditional urban freight vehicles. However, the advantages of cargo cycles are not inherent in every urban environment and under all conditions. This problematics is in general insufficiently researched and more context specific knowledge is needed. Thus, this study investigates the obstacles and facilitating factors for utilising cargo cycles in urban freight transport in Stockholm, Sweden. To investigate these context specifications, case study approach was employed and consisted of qualitative content analysis, semi-structured interviews, and observations. This study argues that in Stockholm, contradictory forces affect the utilisation of cargo cycles in urban freight transport. On one hand, the facilitating factors are mostly associated with measures of sustainable urban freight transport and sustainable development in general: reaction to traffic situation; strategic orientation of the city; public-private partnership; and bicycle infrastructure. On the other hand, path dependence of the city connected to traditional urban freight vehicles (vans) symbolises obstacles: the absence of direct planning for cargo cycles; lack of recognition; and inconsistency of bicycle infrastructure.

Este trabalho está relacionado coma implantação de adequados terminais intermodais para granéis sólidos nas margens das hidrovias, de modo a aumentar a participação do modal fluvial na matriz de transporte do país. Apresenta-se, inicialmente, um panorama da distribuição modal de transportes em que se enfatiza a deficiência dos terminais fluviais existentes como umfator responsável por esta distribuição. É estabelecido, então, como objetivo do trabalho o desenvolvimento do projeto de umterminal intermodal para a hidrovia Tietê-Paraná. O primeiro passo para o projeto é um estudo de demandas e de capacidade da via onde se define: o tipo de carga, quantidade de carga movimentada atualmente e a projeção de demanda, bem como a distribuição da movimentação ao longo do ano. E, emseguida, estima-se a capacidade de movimentação da via. Para desenvolver o projeto é feita uma compilação de diretrizes propostas para dimensionamento de terminais e de seus subsistemas (hidroviário, ferroviário, rodoviário, de armazenagem e de movimentação). Além disto, são analisados os critérios para avaliação do terminal. Antes de desenvolver o projeto, faz-se uma análise crítica dos terminais existentes na hidrovia, assinalando-se suas falhas. Para o dimensionamento do terminal são formuladas diversas configurações que envolvem combinações de diferentes taxas de movimentação e diferentes capacidades de armazenagem. Utilizando a técnica de simulação, com o emprego do software ARENA, obtém-se o desempenho destas configurações em termos de tempo de permanência para cada modal, que mede o nível de serviço oferecido pelo terminal. As alternativas geradas são, então, avaliadas em função do nível de serviço oferecido e do valor presente líquido do investimento, o que conduza escolha de melhor solução. / This thesis is related to the implementation of suitable intermodal terminals for solid bulk cargo with the purpose of increasing the inland waterway participation in the country transportation matrix . Firstly, it is presented a general view of the modal transportation distribution, mainly based on the highway modal, in which the deficiency of the existing inland terminals is stressed as a factor for this distribution. It is specified, therefore, as the purpose of this thesis, the development of the project of an intermodal terminal for the Tietê-Paraná waterway. As the first step of the study, it is performed an analysis of cargo demand as well as of the waterway capacity, in which it is defined: the type of cargo, the amount of cargo transported and its distribution along the year. It is also estimated the cargo demand for the future which remains below the waterway capacity. In order to prepare the project development, it is done a compilation of procedures recommended for multimodal terminals and their components (waterway, railway and highway ends, storage systems and cargo handling equipments). Besides, some criteria to evaluate the terminal performance are presented. A critical analysis of the waterway existing terminals is then presented and their drawbacks are pointed out. For the terminal project, some configurations, which involve combinations of different handling rates and storage capacities , are formulated. The performance evaluation of each option , in terms of vehicle stay time at the terminal, is done by probabilistic simulation technique, employing the commercial software ARENA. The generated options are then compared in terms of service level and net present value and the best configuration is then selected.

Protoporphyrie érythropoïétique : thérapie génique non intégrative par oligonucléotide antisens adressé par peptides bifonctionnels RTf1-CPP / Erythropoietic protoporphyria : non-integrative gene therapy by antisens oligonucleotide addressed by TFR1-CPP bifunctional peptides

La protoporphyrie érythropoïétique (PPE) est une maladie héréditaire rare caractérisée par un déficit en activité FECH responsable d’une accumulation de PPIX. Elle se manifeste par une photosensibilité très invalidante. Il n’existe pas de traitement efficace pour la PPE. 95 % des malades présentent un allèle FECH hypomorphe (c.315-48C) en trans d'une mutation FECH délétère, ce qui entraine une diminution de l'activité FECH résiduelle dans les érythroblastes en dessous d'un seuil critique d'environ 35 % de l'activité normale. L’allèle hypomorphe (c.315-48C) favorise l'utilisation d'un site cryptique d'épissage situé en -63 de l’intron 3 générant un ARNm FECH incluant une partie de l’intron 3 et possédant un codon stop prématuré. L’ARN est alors dégradé par NMD pendant sa maturation. Nous avons déjà identifié un oligonucléotide antisens (ASO-V1) qui redirige l'épissage vers le site accepteur physiologique de l’intron 3 et augmente la production d’ARN FECH WT. Nous avons développé par ce travail une nouvelle stratégie d’adressage d’ASO-V1 en utilisant des peptides ciblant le récepteur de la transferrine (RTf1) qui est exprimé à un niveau très élevé dans les progéniteurs érythroïdes en différenciation concomitamment à la FECH. Nous avons développé des peptides bifonctionnels à partir des séquences peptidiques ciblant le RTf1 tout en les couplant à des séquences Cell Penetrating Peptide (CPP) qui facilitent la sortie de l’ASO-V1 de la vésicule endosomale. Après la transfection des lignées lymphoblastoïdes de malades PPE par différents nanocomplexes RTf1-CPP/ASO-V1, nous avons pu montrer que plusieurs des peptides bifonctionnels utilisés permettaient une redirection efficace et prolongée de l’épissage cryptique vers l’épissage physiologique exon3-exon4 et que cela permettait une correction des taux d’ARN FECH WT. Nous avons ensuite testé l’effet des nanocomplexes RTf1-CPP/ASO-V1, ex vivo, dans les progéniteurs érythroïdes en différenciation de différents sujets atteints de PPE et nous sommes arrivés à augmenter l’ARN FECH WT et diminuer significativement l’accumulation de la PPIX dans ces cellules par rapport à celles transfectées par des nanocomplexes RTf1-CPP/ASO-Mock. La prochaine étape de notre étude serait d’apporter la preuve de concept, in vivo, dans un modèle murin humanisé de PPE après l'administration de nanocomplexes RTf1-CPP/ASOV1 / Erythropoietic protoporphyria (EPP) is a rare hereditary disease characterized by a deficiency in FECH activity responsible for the accumulation of PPIX. EPP is manifested by a very disabling photosensitivity. There is no effective treatment for EPP. 95% of the patients present a hypomorphic FECH allele (c.315-48C) in trans of a deleterious FECH mutation, resulting in a decrease in residual FECH activity in erythroblasts below a critical threshold of about 35% of normal activity. The hypomorphic allele (c.315-48C) promotes the use of a cryptic splicing site located at -63 of the intron 3 generating a FECH mRNA including a part of the intron 3 and possessing a premature stop codon. The RNA is then degraded by NMD during its maturation. We have previously identified an antisense oligonucleotide (ASO-V1) that redirects splicing to the physiological acceptor site of intron 3 and increases the production of WT FECH mRNA. Here, we developed a new ASO-V1 addressing strategy using transferrin receptor (TRf1) targeted peptides. TfR1 is expressed at a very high level in differentiating erythroid progenitors concomitantly with FECH. We developed bifunctional peptides from peptide sequences targeting TfR1 while coupling them to Cell Penetrating Peptide (CPP) sequences that facilitate the release of ASO-V1 from the endosomal vesicle. We transfected the lymphoblastoid cell lines from EPP patients by different TfR1-CPP/ASO-V1 nanocomplexes and we demonstated that several of the bifunctional peptides allowed an efficient and prolonged redirection of the cryptic splicing towards the exon3-exon4 physiological splicing and the correction of the WT FECH mRNA levels. Then, we tested the effect of TfR1-CPP/ASO-V1 nanocomplexes, ex vivo, in differentiating erythroid progenitors of different EPP subjects and we were able to increase WT FECH mRNA and decrease significantly the accumulation of the PPIX in these cells compared to those transfected by TfR1-CPP/ASO-Scr nanocomplexes. The next step of our study would be to provide a proof of concept, in vivo, in a humanized murine model of EPP after the administration of TfR1-CPP/ASOV-1 nanocomplexes

Pedaling towards a sustainable future: lessons Learned from Cargo Bike Pool Operators in Sweden

This study examines the impact and potential of cargo bikes as socio-economically and ecologically beneficial modes of transportation. It focuses on the perspectives of cargo bike pool members and the challenges faced by pool operators. Three interviews were conducted with cargo bike pool operators in Sweden to understand the challenges and considerations involved in establishing and operating a pooling service, as well as their opinions on its financial future. Additionally, a user survey targeting pool members from one of the interviewed operators was conducted in 2020. The survey investigated user barriers, motivations, opinions towards the pooling service, and its effect on user mobility habits before and after joining. Although the acceptance and utilization of these services depend on factors such as proximity, pricing, availability, quality, and convenience of the service. The findings show that pooling services increase member interest in owning and using cargo bikes as a replacement for cars. However, concerns regarding the marketability of cargo-bike pools alone are raised in this study, thus, to enhance the attractiveness of cargo-bike pools, this study suggests incorporating additional modes of transportation, offering supplementary value propositions, and ensuring a user-friendly experience. On the operator side, this study highlights the importance of stakeholder collaboration in maximizing the potential of cargo bike pools for sustainable transportation enabling resource sharing, and marketing efforts, and improving the overall service by leveraging financial, spatial, and logistical benefits. Finally, this study finds that the current state of cargo bike pools necessitates collaboration and subsidies from municipalities and stakeholders. Suggesting innovations in business models and increased marketing strategies that capture a broader demographic through an enhanced value proposition that includes other forms of modality and benefits in their services, are vital for the growth of cargo biking through pooling systems.  The research provides insights for prospective operators in establishing and operating cargo bike pools, emphasizing considerations related to the physical layout, financial aspects, marketing, and value proposition of the service and has broader implications in how to encourage more sustainable mobility practices.
