Effectiveness on color and COD of textile wastewater removing by biological material obtained from Cassia fistula seed

Nowadays, natural polymeric materials extracted from plants are the new alternatives for synthetic chemicals in water and wastewater treatment. The aim of this study is to evaluate the ability of Cassia fistula seed gum (CFG) as a coagulant aid with PAC in the treatment of textile wastewater. Jartest experiments were carried out to identify the optimal parameters of coagulation-flocculation for removing color and COD in synthesis wastewater containing Methyl blue and RB21 dyes, including pH, settling time, PAC dose, the optimal CFG dosage in comparing with the cationic polymer. After that, actual textile wastewater was treated by using PAC, PAC plus cationic polymer, and PAC plus CFG for evaluating the role of CFG. CFG supplementation has assisted the process effects at nearly 98% color, 85% COD for RB21 and 90% color, 70% COD for MB at the best dose of CFG 0.15 mL and 0.1 mL, respectively. The optimized parameters for the coagulation of real textile wastewater using PAC were pH = 6 and dose = 0.6 mL can removal 66% of color. By adding CFG to PAC, the efficient of treatment was increased about 70% even at the lower dosage of PAC and CFG (0.5 mL for each reagent). The yield of combining PAC and polymer was a little bit lower than PAC and CFG, for instant 68% color was decreased at the same condition. These achievements demonstrated a workable substitute of natural products such as Cassis fistula seed gum for synthetic chemical products in coagulation-flocculation process. / Hiện nay các loại vật liệu sinh học chiết xuất từ thực vật đang được nghiên cứu ứng dụng trong xử lý nước và nước thải thay cho các chất hóa học. Mục tiêu của nghiên cứu này là đánh giá hiệu quả của việc sử dụng gum được chiết xuất từ hạt cây Muồng Hoàng Yến (MHY) làm chất trợ keo tụ trong xử lý nước thải dệt nhuộm. Thí nghiệm Jartest được tiến hành nhằm xác định các điều kiện tối ưu cho quá trình xử lý nước thải tổng hợp chứa thuốc nhuộm Methyle Blue (MB) và RB21 bao gồm pH, thời gian lắng, liều PAC, liều gum MHY và liều polymer. Sau đó tiến hành xử lý nước thải thật với các điều kiện thích hợp đã xác định nhằm đánh giá vai trò của gum MHY. Gum MHY làm tăng hiệu quả của quá trình xử lý, đạt gần 98% đối với độ màu, 85% COD đối với RB21, 90% độ màu và 70% COD đối với MB với liều lượng tương ứng là 0,15 mL và 0,1 mL. Các thông số tối ưu cho quá trình xử lý trên mẫu nước thải thật là pH = 6, liều PAC = 0.6 mL có thể làm giảm 66% độ màu. Bổ sung gum MHY làm chất trợ keo tụ giúp gia tăng hiệu quả xử lý màu lên 70% dù với liều lượng rất thấp là 0,5 mL. Hiệu suất xử lý khi sử dụng kết hợp PAC và polymer thấp hơn trong trường hợp sử dụng PAC và gum MHY, cụ thể khoảng 68% độ màu được xử lý ở cùng một điều kiện. Những kết quả này cho thấy tiềm năng của việc sử dụng các vật liệu gum tự nhiên nhằm thay thế cho các hợp chất hóa học trong các quá trình keo tụ tạo bông để xử lý nước thải.

Pré-condicionamento em sementes de Cassia excelsa Schrad.

It could be used in landscape place and due to small size is used in street arborization, and could be recommended to recomposition of degradated areas. The aim of this work was to extend the knowlege about the ecophysiological behaviour, with the the study of priming in seed germination. For this purpuse, it was evaluated the effects of water and PEG solutions (-0,2; -0,4 and -0,6 MPa) in uptake process at 20ºC. The effects of the drying and storage after priming; the answer under water, thermic and salt stress after priming was also verified. Besides it, the priming effects in seed viability and vigor under optimal conditions (27ºC) were observed by eletrical conductivity, accelerated aging and submersion tests. In all the tests was used 4 replicates of 25 seeds, with daily evaluation, and considered germinated when radicle was > 2 mm long. According to the results of uptake curve of Cassia excelsa seeds was observed that with the decreased in the osmotic potential increase the time necessary to priming. At optimal temperature (27ºC) the answer of primed seeds was efficient if water or PEG solution at -0,2 MPa was used, and this answer is maintened after 15 days of storage at 5ºC, but the drying of the seeds overcome the positive effects of the pré-treatment. The priming in water or in PEG solutions increased germination percentage under water, salt and thermal stress, however, with restriction to germination rate. The electrical conductivity of the primed seeds in water or in PEG was reduced, constrated to the non-primed seeds, and increased in the dried seeds after the priming. The priming in water or PEG was efficiente to overcome the deteriorative process occurring in aging, in dehidrated and in moist seeds, after priming. The seed vigor of primed seeds was no affected by submersion in distilled water during diferents time periods. / Cassia excelsa Schrad. é uma espécie de porte arbóreo encontrada no cerrado e nas caatingas do nordeste brasileiro, conhecida também como cassia-do-nordeste. Pode ser empregada em paisagismo em geral e devido ao seu pequeno porte é utilizada na arborização urbana, podendo ser indicada para plantios associados a outras espécies destinados a recomposição de áreas degradadas (Figura 1). Considerando que o período compreendido entre a semeadura e o estabelecimento da planta é uma fase crucial no ciclo reprodutivo da planta e que o pré-condicionamento pode melhorar o desempenho germinativo das sementes, este trabalho tem por objetivo contribuir para o conhecimento do comportamento ecofisiológico desta espécie, a partir de estudos dos efeitos do pré-ondicionamento na germinação das sementes de Cassia excelsa. Para tanto, avaliou-se o efeito da água e soluções de PEG 6000 (a -0,2; -0,4 e -0,6 MPa) na curva de embebição a 20ºC; verificou-se o efeito da secagem e armazenamento após o condicionamento; avaliou-se a resposta sob condições de estresse hídrico, térmico e salino após o condicionamento; e por último, investigou-se o efeito do condicionamento osmótico na viabilidade e vigor das sementes, sob condições ótimas (27ºC) a partir do teste de condutividade elétrica, envelhecimento precoce e teste de exaustão. Utilizou-se em cada experimento 4 repetições de 25 sementes escarificadas, com avaliação diária, considerando-se sementes germinadas aquelas que apresentavam radícula > 2 mm. A partir curva de embebição das sementes de Cassia excelsa verificou-se que com a redução do potencial osmótico da solução aumenta o tempo necessário para o condicionamento osmótico. Em condição ótima de temperatura (27ºC) as sementes respondem positivamente ao condicionamento, se forem utilizadas água destilada ou soluções de PEG a -0,2 MPa, sendo mantida esta resposta durante o armazenamento por 15 dias a 5ºC, contudo, a secagem das sementes reverte os efeitos positivos obtidos pelo tratamento. Sob condições de déficit hídrico, sob temperaturas sub ou supra ótima e salinidade elevada houve um aumento na germinabilidade das sementes pré-condicionadas, tanto com água como em PEG, porém com restrição à velocidade de germinação. A partir do teste de condutividade verifica-se que as sementes condicionadas em água ou em PEG apresentaram redução na lixiviação de eletrólitos, comparado às sementes não condicionadas (controle), e as sementes que foram secas após o condicionamento apresentam maior lixiviação do que as úmidas. O précondicionamento com PEG ou água destilada foi eficiente em reverter o processo de deterioração que ocorre durante o envelhecimento precoce, tanto para as sementes que foram secas ou mantidas úmidas, após o condicionamento. O vigor das sementes não foi afetado pela submersão das sementes por 24, 48 e 72 horas após o pré-condicionamento em água ou em PEG.

They are commonly consumed fresh and their chemical composition and antioxidant capacity have never been studied. Extracts in acetone and methanol pulp, fruit peel and seed of fruits were obtained and analyzed by LC-MS leading to identify flavonoid glycosides, dimer flavonoid, phenolic acids and one organic acid present in the pulp, peel and fruit seed. The scavenger capacity and total phenolic content were also evaluated. The total phenolic of extracts of the edible part of marimari presented total phenolic content of 0.036 and 40.12 μg GAEq/100 g for methanolic and ketonic extracts respectively. The antioxidant activity evaluated by DPPH method revealed values of IC50 of 620.30 ± 0.01 and 364.94 ± 0.01 μg/ to mari-mari extracts pulp (methanolic and ketonic respectively). The best result was seeds pajurá extracted in methanol. Iron reduction capacity of the edible part of the fruit was 629.88 ± 0.01 and 634.46 ± 0.08 μmol FeSO4 g− 1 DW. Additionally, engeletin (1) and astilbin (2) were isolated and identified by RMN and comparison with literature. This is the first report about the chemical characterization and antioxidant activity from mari-mari and a contribution to studies from pajurá e pitomba fruits. / Mari-Mari (Cassia leiandra Benth), pajurá (Couepia bracteosa) e pitomba (Talisia esculenta) são frutas facilmente encontradas nos arredores da cidade e também na floresta amazônica. A composição química e informações sobre a capacidade antioxidante do mari-mari é desconhecida e do pajurá e pitomba pouco exploradas. Neste trabalho, foram produzidos extratos de polpa, cascas e sementes destes frutos utilizando acetona e metanol, para realização de análises por LC-MS para a identificação de compostos, especialmente da classe de flavonoides, quantificação do teor de fenólicos totais, avaliação da capacidade de sequestro de radicais DPPH e redução de Fe3+. Os resultados de quantificação dos fenólicos totais dos extratos da parte comestível de mari-mari apresentou teor de fenólicos totais de 0,036 e 40,12 mg GAEq/100 g de extrato seco nos extratos metanólico e cetônico, respectivamente. O extrato de polpa pajurá apresentou o maior teor de fenólicos no extrato metanólico. A atividade antioxidante avaliada pelo método de DPPH para os extratos metanólico e cetônico de polpa de mari-mari foi CS50 de 620,30 ± 0,01 e 364,94 ± 0,01 μg/mL respectivamente. Entre os frutos, o destaque foi para o extrato metanólico da amêndoa de pajurá (63,69 μg/mL ± 0,002). A capacidade de redução de Fe3+ da parte comestível do fruto foi 629,88 ± 0,01 e 634,46 ± 0,08 μmol FeSO4/g extrato seco. Além disso, engeletina (1) e astilbina (2) foram isolados e identificados por espectrometria de RMN e EM e comparação com a literatura. Este é o primeiro relato sobre a caracterização química e atividade antioxidante do fruto mari-mari e uma contribuição aos estudos de pajurá e pitomba.

Identification, characterization and quantification of the active and toxic compounds of two cinnamon species

M.Tech. / There are over 250 cinnamon species existing worldwide, and amongst them, 2 species Cinnamomum cassia and Cinnamomum zeylanicum, are commonly used all over the world as spices, fragrances in perfumes and as medicines. These two species are distinguished from each other by the presence and absence of certain compounds depending on the origin and distribution of the plant. Nevertheless, all cinnamon species contain essential oils and water soluble components and the composition of these components found in each species depends on the type of species involved. These components are made up of phenyl propanoids, terpenes, flavonoids and saponins. In general, the essential oil component contains the following compounds: cinnamaldehyde as a major constituent with its derivatives cinnamic acid, cinnamyl alcohol, ethyl cinnamate, cinnamyl acetate and 2-methoxy cinnamaldehyde; eugenol; linalool; coumarin; carvone; carvacrol and β-caryophyllene. Most of these compounds are abundantly found in the bark except in the case of eugenol which is found in either leaves or bark depending on the species involved. The other water soluble component is composed of a group of compounds such as tannin, chalcone, catechins and anthocyanidins. These compounds polymerize to form methyl hydroxy chalcone polymers (MHCP) which has been shown to play an important role in lowering blood sugar levels in Type ll diabetic individuals. It is said that MHCP has an ability to promote the phosphorylation process which in turn activates the beta-cells and thereby creating insulin activity that will then convert glucose into glycogen. Besides all health benefits of cinnamon, the plant contains a toxic compound, coumarin which impacts badly on animals resulting in death, and little information on its toxicity to human beings has been documented. Concerns arose about the possible presence of coumarin in these formulations, since these formulations are made from the cinnamon and the plant contains coumarin. This work has investigated the potential toxicity of coumarin from cinnamon powder and cinnamon formulation. The components from both cinnamon samples were extracted using various solvents according to their polarities and these compounds were screened on thin layer chromatography (TLC). The essential oil components were separated by column chromatography, and quantified by using high performance liquid chromatography (HPLC), all components (essential oil and water soluble) were identified by HPLC, and finally the characterization of the essential oil components was done with infrared (IR) and nuclear magnetic resonance (NMR) whereas those of the water soluble were characterized by using liquid chromatography coupled to a mass spectrometer (LC-MS).

Antimycobacterial evaluation, preliminary phytochemical and cytotoxicity studies of cassia petersiana

Thesis (M.Sc.(Microbiology)) -- University of Limpopo, 2022 / This study aimed to investigate antimycobacterial and cytotoxic compounds from Cassia petersiana. Cassia petersiana was selected for the current study based on its traditional use for treating tuberculosis (TB) symptoms. Extraction is an important step in the use of medicinal plants; hence, solvents of varying polarity were employed to extract a wide range of compounds where chloroform was the best extractant (67 mg). As there is no relation between the amount of plant material extracted and the bioactivity of the extracts, standard tests were used to determine the presence of different phytochemical constituents from Cassia petersiana and the total phenolic, flavonoid, and tannin contents were quantified using colorimetric assays. It was revealed that all the tested phytochemical constituents were present, and it was proven that phenolic compounds were the most abundant, followed by the tannins, while the flavonoids were the least among the common phytochemical constituents quantified. The phytochemical compounds were further profiled on thin-layer chromatography (TLC) and developed in BEA, CEF, and EMW solvent systems. Colourful compounds which indicated diverse phytochemicals were visualised with both vanillin-sulphuric acid and ultraviolet light on the phytochemical chromatograms and good separation of the compounds was from the BEA solvent system. The qualitative and quantitative antioxidant activity and antimycobacterial activity assays were used to evaluate the extracts from Cassia petersiana. Minimal antioxidant activity was observed on the qualitative antioxidant activity profile. These findings correlated with the minimal quantity of antioxidants from extracts of Cassia petersiana from the quantitative antioxidant assays; ferric reducing power and DPPH scavenging activity assays. Cassia petersiana extracts had bioactivity against Mycobacterium smegmatis as indicated by the lowest MIC value. The cell viability effects of the acetone crude extract from Cassia petersiana were evaluated against the tryptophan hydroxylase-1 (TPH-1) macrophage cells. Large scale extraction procedure was employed to extract a sufficient amount of plant material in preparation for the isolation of the bioactive compound. Bioassay-guided fractionation combined with column chromatography and TLC were used to isolate and purify the bioactive compound from the n-hexane extract of Cassia petersiana. The purified isolated compound was elucidated as β-sitosterol, which showed remarkable bioactivity against Mycobacterium smegmatis only on the TLC-bioautographic assay, while the quantitative antimycobacterial activity was higher xx with the MIC value of 2.5 mg/mL. Although β-sitosterol is known as a good antioxidant, it showed no antioxidant activity on the qualitative antioxidant activity assay. Therefore, further studies, including in vivo assay, are recommended on the isolated compound to evaluate its biological activities before consideration of its use in the development of alternative drugs.

Devido à necessidade de nematicidas mais eficientes e menos tóxicos para o homem e para o ambiente do que os disponíveis para o controle de Meloidogyne incognita, assim, neste trabalho objetivou-se estudar os óleos essenciais de Cinnamomum cassia e de Allium sativum, que foram descritos como ativos in vitro contra o nematóide Bursaphelenchus xylophilus. O óleo essencial de ambas as espécies foi obtido por hidrodestilação, dissolvidos em solução aquosa de Tween 80 a 0,01 g mL-1 e testado in vitro contra M. incognita. A 62 μg mL-1 as emulsões dos óleos foram mais ativas sobre ovos e juvenis de segundo estagio (J2) do que o Carbofuran a 173 μg mL-1. De acordo com a análise de cromatografia gasosa-espectrometria de massas, o constituinte majoritário de C. cassia é o (E)-cinamaldeído (83,3%), enquanto para o óleo de alho foram trisulfeto de dialila (66.7%) e dissulfeto de dialila (21.3%). Esses constituintes explicaram a atividade nematicida in vitro do óleo essencial de cada espécie de planta. A emulsão do óleo de C. cassia (500 μg mL-1), (E)-cinamaldeído (416 μg mL-1) e alho (250 μg mL-1) reduziram o numero de galhas e de ovos em plantas de soja e tomateiro, respectivamente, a valores estatisticamente iguais àquele obtido pelo Carbofuran (416 μg mL-1). Vapores dos óleos essenciais e do (E)-cinamaldeído foram ativos tanto quanto o nematicida fumigante Basamid usado nos ensaios in vitro contra M. incognita. A infectividade e a reprodução de M. incognita em plantas de soja e de tomate cultivadas em substrato infestado artificialmente com ovos do nematóide e tratado com 0,2 mL (alho), 1,0 mL (E)-cinamaldeído e 0,25 g de Basamid foram estatisiticamente iguais entre si na redução da população do nematoide. Esses resultados inequivocamente provam a atividade nematicida dos óleos essenciais e da substância (E)-cinamaldeído contra M. incognita, portanto, tanto o óleo quanto o (E)-cinamaldeído são muito promissores ao desenvolvimento de novos nematicidas fumigantes para o controle de nematóides em plantas de soja e de tomate. / Essential oils derived from secondary plant metabolism may have activities against root-knot nematodes, Meloidogyne incognita. Because nematicides that are more efficient and less toxic to humans and the environment than those available are desirable to control this pathogen, this work aimed at studying the essential oil of Cinnamomum cassia and Allium sativum, which were described as active in vitro against the nematode Bursaphelenchus xylophilus. The essential oils of both species were obtained by hydrodistillation, initially dissolved in aqueous solution of Tween 80 at 0.01 g mL-1 and tested in vitro against M. incognita. At 62 μg mL-1 the oil emulsions were more active on eggs and second stage juveniles (J2) than Carbofuran at 173 μg mL-1. According to gas chromatography-mass spectrometry analysis, the major constituent of C. cassia is (E) -cinnamaldehyde (83.3%), while for garlic oil were diallyl trisulfide (66.7%) and diallyl disulfide (21.3%). These constituents explained the in vitro nematicidal activity of the essential oil of each plant species. The emulsion of the C. cassia (500 μg mL-1), E)-cinnamaldehyde (416 μg mL-1) and garlic (250 μg mL -1) reduced the number of galls and eggs in roots soybean and tomatoes to values statistically equal to those obtained with Carbofuran at 415 μg mL-1. Vapors from the essential oils and (E)-cinnamaldehyde were active as much as the basamid fumigant nematicide used in the in vitro assays against M. incognita. Infectivity and reproduction of M. incognita in soybean and tomato plants grown on substrate artificially infested with nematode eggs and treated with 0.2 mL (garlic), 1.0 mL (E)-cinnamaldehyde and 0.25 g of Basamid were statistically equal among themselves in reducing the nematode population. Natural auxiliary coagulants - perspectives for the treatment of textile wastewater

Applying chemical coagulants and auxiliary coagulants in wastewater treatment has become more popular in Vietnam. Although the efficacy of chemical coagulants has been well recognized, there are disadvantages associated with the use of these products, such as the inefficiency at low temperatures, increasing the residual cation in solution, causing health problems and distribution water, relatively high cost, producing high volume of sludge. Thus, it is desirable to replace these chemical coagulants for products that do not generate such drawbacks, such as natural polymers. In this paper, the authors conducted experiments by using natural auxiliary coagulants extracted from seeds of Cassia fistula (gum MHY) and chemical polymer as auxiliary coagulation to treat textile wastewater with basic polluted parameters: pH = 9.0; COD = 800 mgO2/L, color = 750 Pt-Co. The Jartest experiment results showed that the process efficiency of chemical polymer and gum MHY is not so different, with the COD removal efficiencies of 60.3% and 59.7%; the color removal efficiencies of 87.3% and 87.1%; the SS removal efficiencies of 93.2% and 92.6%. There-fore, coagulants obtained from gum MHY can be applied as the alternatives for chemical polymer in the process of treating textile wastewater. / Các ứng dụng chất keo tụ và chất trợ keo tụ hóa học trong xử lý nước thải ngày càng trở nên phổ biến tại Việt Nam. Mặc dù có nhiều ghi nhận về hiệu quả xử lý của chất keo tụ hóa học, phương pháp xử lý này vẫn tồn tại một số nhược điểm như hiệu suất xử lý thấp ở nhiệt độ thấp, nước thải sau khi xử lý còn chứa nhiều hóa chất tiếp tục làm ô nhiễm nguồn tiếp nhận, chi phí xử lý cao và tạo ra nhiều bùn thải. Do đó việc tìm kiếm một phương án xử lý thay thế, chẳng hạn sử dụng polymer tự nhiên, có thể khắc phục những nhược điểm này là rất cần thiết. Nghiên cứu này tiến hành đánh giá hiệu quả sử dụng chất trợ keo tụ sinh học ly trích từ hạt trái Muồng Hoàng yến (Cassia fistula) và chất trợ keo tụ hóa học để xử lý nước thải dệt nhuộm có các thông số ô nhiễm cơ bản: pH = 9,0; COD = 800 mgO2/L, độ màu = 750 Pt-Co. Các thí nghiệm trên bộ Jartest cho thấy hiệu quả xử lý nước thải dệt nhuộm của chất trợ keo tụ gum Muồng Hoàng yến và chất trợ keo tụ hóa học không khác biệt có ý nghĩa với hiệu suất xử lý COD lần lượt là 60,3 và 59,7%; hiệu suất xử lý độ màu là 87,3 và 87,1%; xử lý SS là 93,2 và 92,6%. Kết quả nghiên cứu cho thấy gum hạt Muồng Hoàng yến có thể sử dụng làm chất trợ keo tụ thay thế chất trợ keo tụ hóa học trong xử lý ô nhiễm nước thải dệt nhuộm.

The ethno-ecological assessment of Cassia abbreviata Oliv. at Matsa village, Limpopo province, South Africa

MSc (Botany) / Department of Botany / Since time immemorial plants have traditionally been used for various purposes in many parts of the world including Vhembe district, Limpopo province of South Africa. The use of C. abbreviata plants at Matsa village of Vhembe district also contributes significantly to the livelihood of the community. However, little work has been made in the past to properly document and promote the knowledge. The purpose of the present study was to record and analyse the ethnobotanical knowledge of C. abbreviata within the people of Matsa village around Nzhelele in Vhembe district. Semi-structured questionnaires were administered on 50 informants from different households selected randomly to gather data regarding the local name of C. abbreviata, the uses and its availability. Significantly higher number of medicinal use was reported by elderly people including both men and women as compared to middle-aged, younger people. Other use categories included firewood, furniture and joinery production and shade for people in the area. Awareness on the importance of the species should therefore be created amongst the people, especially the young ones. Understanding the ethnobotanical knowledge of indigenous species within an area is crucial towards development of its management plan. / NRF
