Expressão de fatores de proliferação e antiapoptóticos em carcinomas basocelulares

Pode se manifestar como lesão nodular, superficial, esclerodermiforme, micronodular e fibroepitelioma de Pinkus. O CBC é um carcinoma de baixa mortalidade, porém com elevada morbidade devido ao potencial destrutivo local e às elevadas taxas de reincidência. As diferenças evolutivas dos diversos tipos de carcinomas basocelulares não são bem definidas. A análise de fatores de proliferação e apoptose pode subsidiar o conhecimento sobre a fisiopatologia e as diferenças evolutivas dessas lesões. A literatura revela a escassez de investigações sobre a expressão diferencial de fatores de proliferação (Ki-67) e relacionados à apoptose (p53, survivina e NF-kB-p105) dos diferentes subtipos de carcinomas basocelulares e ainda tumores recidivados. O objetivo desse estudo é avaliar a expressão de marcadores de proliferação celular e apoptose em carcinomas basocelulares dos tipos nodular, superficial, esclerodermiforme e tumores recidivados. Foram selecionados subtipos histológicos únicos, como forma de tentar compreender o comportamento dos CBCs individualmente. As lâminas desses espécimes foram submetidas ao protocolo de marcação imuno-histoquímica. (Resumo completo, clicar acesso eletrônico abaixo) / Abstract: Basal cell carcinoma (BCC) is the most common malignant neoplasm among men and its incidence is increasing in our country. It can manifest as nodular, superficial, sclerodermiform, micronodular and Pinkus fibroepithelioma lesions. BCC is a lowmortality carcinoma, but with high morbidity due to local destructive potential and high rates of recurrence. The evolutionary differences of the various types of basal cell carcinomas are not well defined. The analysis of proliferation and apoptosis factors may support knowledge about the pathophysiology and evolutionary differences of these lesions. The literature reveals the scarcity of investigations on the differential expression of proliferation factors (Ki-67) and related apoptosis (p53, survivin and NF-kB-p105) of the different subtypes of basal cell carcinomas and recurrent tumors. The aim of this study is to evaluate the expression of markers of cell proliferation and apoptosis in basal cell carcinomas of the nodular, superficial, sclerodermiform and recurrent tumors. Unique histological subtypes were selected as a way to attempt to understand the behavior of individual BCCs. The slides of these specimens were submitted to the immunohistochemical labeling protocol. A total of 100 samples were studied, being 50 normal epidermal units and the others distributed between each tumor subtype. The marking pattern of each marker on the different tissue types was evaluated from generalized linear models (GLMs) followed by Sidak post-hoc test, when necessary. Statiscally significant differences (p<0,01) in the immunostaining of different tissue types were found for the Ki-67, p53 and survivin markers, but not for the p105 marker (p=0,21). Of this group, acetyl-salicylic acid, or aspirin (ASA) has proved to be the most potent in each case. Although hydrolysis, the first step in biotransformation of aspirin takes place very rapidly, many workers feel that the greater effectiveness is due to a unique action of the intact molecule; however, others disagree. In this study, the effects of aspirin and in vitro were compared in order to obtain a greater amount of information relating to the problem.

Cell Proliferation and Hepatocarcinogenesis in Rat Initiated by Diethylnitrosamine and Promoted by Phenobarbital: Potential Roles of Early DNA Damage and Liver Metallothionein Expression

Cell proliferation plays an important role in multistage chemical carcinogenesis. Again, several reports demonstrated that upregulation of metallothionein (MT) expression is associated with increased cell proliferation that may contribute to the pathogenesis of preneoplastic phenotype to frank malignancy. In this study, we evaluated the roles of early DNA damage, altered expressions of liver MT and Ki-67 nuclear antigen, and altered hepatic levels of zinc (Zn) and copper (Cu) on cell proliferation and the progression of hepatocarcinogenesis through premalignant, late premalignant and malignant transformation phases in male Sprague-Dawley rats. We have further studied the association between MT expression and cell proliferation in hepatocarcinogenesis. There was substantial induction of DNA single-strand breaks (SSBs) (P < 0.001) and development of hepatocellular premalignant lesions along with significant decrease in hepatic levels of Zn and increase in Cu content following a single, necrogenic, intraperitoneal (i.p.) injection (200 mg/Kg body weight) of diethylnitrosamine (DEN) at week 4 of the experimental protocol. Moreover, DEN + phenobarbital (PB)-treatment significantly elevated MT-, Ki-67-, and BrdU-immunoexpressions along with their immunolabeling indices. Furthermore, positive correlations between MT- and Ki-67- labeling (P = 0.0006) at various time intervals, as well as, between MT immunoreactivity and 5'-bromo-2'-deoxyuridine-labeling index (BrdU-LI) (P = 0.0007) indicate that, MT expression might be associated with Ki-67 expression and cell proliferation thereby. The study suggests that DEN treatment may lead to alteration of Zn and Cu levels resulting in early DNA damage along with elevation of MT expression that may ultimately lead to hepatic cell proliferation. The results thus provide evidence in support of the role of MT as a potential positive regulator of cell growth during the early stages of hepatocellular transformation in rats.

The Sheddase Activity of ADAM10/ADAM17 on CXCL16 Increases Proliferation and Survival of Colorectal Cancer Cells

CXCL16 is an interferon-inducible chemokine of the CXC-subfamily and functions as an adhesion molecule, when membrane bound, and a chemoattractant when soluble. Upregulation of cell associated CXCL16 (cCXCL16) in colorectal cancer is associated with increased tumor infiltrating lymphocytes and good prognosis. ADAM10 and ADAM17 are metalloproteinases responsible for cleaving CXCL16, releasing soluble CXCL16 (sCXCL16) and contributing to proliferation and migration of mesangial cells, in kidney inflammatory disease. We hypothesize that cCXCL16 is a substrate for ADAM10 and ADAM17 cleavage in colorectal cancer, releasing sCXCL16, which mediates cell proliferation. To this end, we first identified CXCL16 in the human colon carcinoma cell line, RKO, by immunohistochemistry. cCXCL16 was found in the membrane, cytoplasm and nucleus. We treated RKO, in vitro, with an inflammatory cytokine mix containing 1.4 nM rhIFN[gamma], 2.0 nM rhTNF[alpha] and 2.0 nM rhIL1[beta] to increase the cleavage of cCXCL16 to sCXCL16. Overnight incubation with the cytokine mix significantly (P=.004) increased the release of sCXCL16 compared to normal conditions. To confirm that a metalloproteinase is responsible for the cleavage of cCXCL16, we used a broad spectrum metalloproteinase inhibitor, GM6001, in combination with inflammatory stimulation, in cell culture. We assayed the supernatant using ELISA for sCXCL16. GM6001 at 100 [mu]M decreased sCXCL16 to levels indistinguishable from the background. Using siRNA, we knocked down the expression of ADAM10 and ADAM17, independently, to determine if the activity of each on cCXCL16 was mediated by inflammatory stimulation. It was shown that ADAM10 constitutively cleaved cCXCL16, and ADAM17 cleavage activity was induced by inflammatory stimulation. To determine if sCXCL16 increased colorectal cancer cell (CRC) proliferation through ligand-receptor binding, we treated cells with a range of rhCXCL16 from 3.125-100 ng/mL. rhCXCL16 did not increase RKO proliferation at doses up to 100 ng/mL. We used GM6001, to inhibit the cleavage of cCXCL16 into sCXCL16 then performed an ATPase assay and 6 day cell cycle analysis, under inflammatory stimulation. Increased cleavage of cCXCL16 induced by inflammatory stimulation with the cytokine mix containing 1.4 nM rhIFN[gamma], 2.0 nM rhTNF[alpha] and 2.0 nM rhIL1[beta], increased RKO proliferation and reduced apoptosis. We conclude that ADAM10 and ADAM17 cleavage of cCXCL16 to sCXCL16 is increased by ADAM17 activation with inflammatory stimulation. The cleavage of the extracellular portion from cCXCL16 is associated with increased proliferation and decreased apoptosis of colorectal cancer cells.

Comparative study of homologous structures in closely related species allows the identification of changes in gene regulatory mechanisms and their impact on the evolution of developmental processes. Nematodes, the invertebrate roundworms, are well suited for such studies, especially the Caenorhabditis briggsae and its famous cousin C. elegans. These two worms diverged from a common ancestor roughly 30 million years ago, yet appear morphologically almost identical. My Ph.D. thesis has focused on a set of nuclear factors in C. briggsae that negatively regulate cell proliferation to generate the hermaphrodite-specific mating and egg-laying organ, i.e., vulva. To this end, I have taken a two-pronged approach: one, developing resources to facilitate genetic and genomic studies in this species, and two, characterizing the roles of a novel class of genes and known repressors of vulval development. My work has uncovered substantial differences in the underlying genetic networks that regulate vulva formation in C. briggsae and C. elegans. / Thesis / Candidate in Philosophy
