This thesis introduces the current state health of the population of the Czech Republic, addressing mainly the steep increase in diseases of civilization. Diseases which are mainly the result of an inappropriate lifestyle. The aim of this work is the development of a business plan leading to the establishment of a company that will provide services in the field of nutrition counseling, physiotherapy, regeneration and physical activities. The theoretical part of this work defines the basic concepts and assumptions which are the building blocks of this thesis. Specifically, the tools for strategic management and business plan requirements. Furthermore, the theoretical part defines concepts such as health and a healthy lifestyle. The practical part of this thesis includes a research and analysis section which form the basis for the creation of the business plan. Methodologically, quantitative research was applied. A technically structured questionnaire was used to gain the required data. The analytical part then goes on to include an analysis of the general and business environments, or, to be precise, of the internal environment. This diploma thesis summarizes knowledge of significant genetically contitioned deseases occurring in border collies. There is described a total of 14 diseases, some with the location of causal mutation not yet known. Primary focus of this thesis is g.4411956_4411960delGTTT mutation of gene VPS13B causing Trapped Neuthrophil Syndrome (TNS), MDR1 gene's mutation AF045016.1: c.227_230delATAG associated with multidrug resistance (MDR1) and CUBN gene's mutation c.8392delC which causes intestinal malabsorption of cobalamin by another name ImerslundGräsbeck syndrome (IGS). A genotype analysis of 89 border collies with a proof of origin was performed. The DNA was extracted from buccal mucosal swabs, the isolation of DNA was performed by Chelex-100 from the native material. The analysis was proceeded by optimized PCR-RFLP method using restrictive MboI (MDR1) and Msl I (IGS) enzymes. There were detected 4 g.4411956_4411960delGTTT mutation vectors causing TNS. As for the MDR1 and IGS there wasn't detected any affected case.

Infectious diseases are a very hot topic nowadays in society. In recent years, there were several epidemics, infectious diseases (hepatitis A, Avian influenza, pandemic influenza A) and nobody knows when will the next "new" infection. At the outbreak of epidemics, there is great interest in the company to obtain information about the disease, but often also to unnecessary panic, because the media often publish incorrect information. It would be preferable, in order to improve public awareness and prevent the unnecessary spread of disease. Great emphasis should be given to prevent the disease - primarily on immunization, hygiene rules and principles of safe sex. Quality information should be mainly from teachers in teaching their pupils, as is clear from research books, infectious diseases are only mentioned in textbooks and the number of substantive information in them is missing. Cystic fibrosis and diseases associated with trinucleotide expansions are serious hereditary disease that serves in my Diploma thesis as role models suitable for teaching interesting topics of human genetics at secondary schools. By using appropriate methods of prenatal genetic treatment, it is possible to make a diagnosis of the developing fetus and to determine a corresponding prognosis in next prenatal and postnatal development in families at risk. The practical part of the thesis is devoted to the content analysis of biology schoolbooks for secondary schools and to the preparation of a prototype class of genetics at secondary schools. The aim of the content analysis of biology schoolbooks for secondary schools is the evaluation of various textbooks according to several preselected criteria. The aim of the presentation of the proposed prototype class was to determine, based on the responses obtained from the questionnaires, whether pupils of higher grades of secondary schools are interested in the subject matter of genetics, to find out what engaged their attention the most during the lessons or what they did not understand and what they considered as difficult. This work follows the studies focused on obesity, its global extent and its serious health and economic impact. At the same time, the work examines the unsuccessful fight of public policies which try to reverse growing prevalence of obesity. Nevertheless, the main goal of the thesis is a discussion about priorities of new orientation in the research on the causes of the obesity epidemic and effectiveness of precautions that were taken in many countries and international organizations with the main aim to stop the obesity and its consequences. The discussion leads to the more detailed description of the problems connected with obesity, for instance, environmental aspects, housing culture in relation to physical activities, lifestyle in relation to advertisement/marketing, psychological and socio- psychological aspects, combating stress, etc. My intention was to find out utility of contemporary research on obesity for setting of policy fighting against obesity.
