Quantifying the Lateral Bracing Provided by Standing Steam Roof Systems

One of the major challenges of engineering is finding the proper balance between economical and safe. Currently engineers at Nucor Corporation have ignored the additional lateral bracing provided by standing seam roofing systems to joists because of the lack of methods available to quantify the amount of bracing provided. Based on the results of testing performed herein, this bracing is significant, potentially resulting in excessively conservative designs and unnecessary costs. This project performed 26 tests with Vulcraft joists in a pressure box to investigate the effects of how many variables influence the lateral bracing provided to joists from standing seam roofing systems, including the variables joist length, panel gauge, clip height, thermal block presence, insulation thickness, and top chord size. Two methods were developed to account for this additional bracing: finite element computer modeling and an application of the Rayleigh-Ritz method called the Column-on-Elastic-Foundation Method. Variables influencing the pressure at failure, namely chord size and deck gauge, were those with the greatest effect on additional lateral bracing provided from standing seam roof systems. It was determined that higher roof stiffness values and higher failure pressures yield shorter effective lengths.

Le terme de CLIP-170 désigne la protéine de lien cytoplasmique de 170 KDa, isolée par Rickard et Kreis (1990). In vitro, elle constitue un lien statique entre les endosomes et les microtubules (MTs). En chacun des points où la CLIP-170 est localisée, elle se co-distribue avec le complexe moteur dynéine/dynactine (D/D). Les auteurs ont initialement établi un modèle de fonctionnement de concert de ces trois protagonistes : la CLIP-170 établirait le lien initial entre le cargo et le MT. Le complexe D/D serait recruté sur le cargo. Une fois le moteur associé au MT, le lien statique serait levé, rendant possible le mouvement. Ce modèle offrait une explication aux données d'immunolocalisation. Toutefois, le fonctionnement de concert de la CLIP-170 et du moteur moléculaire nécessitait que l'on puisse trouver une interaction entre les protagonistes. Les travaux présentés dans cette thèse apportent pour la première fois la preuve d'une interaction indirecte entre le complexe D/D et la CLIP-170. LIS1 est une protéine codée par le gène causal du syndrome de Miller-Dieker, une forme de lissencéphalie de type I. Elle sert d'adaptateur entre le domaine carboxy-terminal de la CLIP-170 et le complexe moteur dont elle régule l'activité. Nos résultats établissent que le domaine d'interaction de la CLIP-170 avec LIS1 est requis pour l'adressage de la première aux kinétochores prémétaphasiques. Il est nécessaire à l'adressage de LIS1 (et du complexe moteur) aux bouts (+) des MTs interphasiques. Nous présentons deux nouvelles approches permettant d'interférer avec le fonctionnement de la CLIP-170 tant en interphase qu'en mitose : la microinjection d'anticorps dirigés contre les domaines extrêmes de la protéine, ainsi que l'utilisation de siRNA dirigés contre l'ARNm la codant. Combinées aux techniques de suivi de protéines fluorescentes par vidéomicroscopie 3D développées au laboratoire, elles permettront d'interférer avec le fonctionnement de la CLIP-170 et de définir ainsi son rôle.

A comparison of double clip angle shear connections to shear tab connections in industrial applications

Kramer / In structural steel connection design, simple shear connections are one of the most common connection types utilized. The industry, especially from the side of the engineer, tends to lean toward using Double Clip Angle Connections as the default standard for simple shear connections. A double clip angle connection is a connection consisting of two angles transferring the shear forces from one member to the next either through bolts or welds. The design of Double Clip Angle Connections is efficient and the connections themselves are easy to fabricate. However, benefits to utilizing other types of shear connections exist. Many of these benefits are seen in the fabrication shop or during erection and construction. This is especially true of single shear plate or shear tab connections when applied to open structure design. Shear tab connections consist of a single plate that transfers the shear forces from one member to the next with bolts or with welds. The design of shear tab connections can be a more involved process than the design of double clip angles. Sometimes the shear plate or shear tab has to be longer than is typical. This is called an extended shear plate connection. These extended shear plates can bring other variables into the design that typically don’t occur with Double Clip Angle Connections such as bending of the plate or the need for multiple bolt columns. However, with proper planning and detailing, the benefits and savings experienced in the fabrication or construction phase may outweigh what can be seen as a more laborious design task. The purpose of this report is to identify the possible benefits achieved in using each of these connections, highlight the differences in the design approach for each, and use a study model to compare the outcome of using one connection over another in the design of a typical open structure. Double clip angles are typically the most efficient approach when speed of design and simplicity of fabrication are the desired outcomes. However, shear plate or shear tab connections have the potential to provide safer erection alternatives and materials savings if used in appropriate ways and with the right applications.

A molecular approach to genetic improvement of South African Angora goats

South Africa is considered the primary producer and most reliable source of high quality clip mohair in the world. The application of molecular technologies to improve mohair quality is relatively new to this industry. The aim of the study was to use a molecular approach to genetically improve South African Angora goats, with emphasis on mohair production. A reference population of Angora goats was firstly established consisting of twelve sire families with half-sib offspring (1067 individuals in total). The genetic variation of this population was evaluated using microsatellite markers and the average gene diversity was found to be above 60%. Ninety four microsatellite markers were then genotyped on the reference population, spanning 23 chromosomes (total length 1352cM) with an average marker interval of 23.0cM. This information was used to improve previously published goat linkage maps. Unmapped microsatellite markers were incorporated and previously published inter-chromosomal rearrangements between the goat and sheep genetic maps were confirmed or rejected. Nine new markers were mapped to the goat genome, and six chromosomes showed rearrangement when compared to the previous goat map. Four previously reported intra-chromosomal rearrangements were shown to be either population specific or mapping errors. Variance components and genetic parameters of mohair traits (FW, FD, CVFD, SDFD, CF, SF and SDA) were estimated; including the fibre diameter profile measured using OFDA technology that has not yet been included in genetic evaluations. Heritability estimates ranged between 0.14 (SDA) and 0.63 (CF). OFDA-measured traits should be considered for inclusion into the national breeding strategy. The reference population was lastly analysed to identify QTL associated with fleece traits. Eighteen putative QTL were identified for seven mohair traits on 13 chromosomes. Three putative QTL were detected for FW on CHI 2, 5 and 24 corresponding with KRT and KAP gene locations. Two QTL associated with mohair FD (on CHI 4 and 24) were detected. QTL contributions to variance ranged between 7.44% (CF) and 19.69% (SDA). This thesis presents experiments and numerical analysis of the cold-formed steel clip angle in three different limit states which are shear, compression, and combination of the screw connection. A previous cold-formed steel clip angle test program (which is Phase 1) developed design methods for clip angle. Therefore, the object of this thesis is to further investigate the behavior and design methods of loading-bearing cold-formed steel clip angles under different screw pattern. For each limit state, a test program was conducted to investigate the behavior, strength, and deflection of the clip angle. The test result were compared with previous CFS clip angle design method. Amending existing CFS clip angle method were developed by each of the four limit states studied in this project.

THE FUNCTIONAL ROLE OF RNA BINDING PROTEIN RBMS3 AS A TUMOR PROMOTER IN TRIPLE-NEGATIVE BREAST CANCER CELLS

RBMS3 belongs to the family of c-myc gene single-strand binding proteins (MSSPs) that play important roles in transcriptional regulation. Here, we show that RBMS3 functions as a tumor promoter in triple-negative breast cancer (TNBC), a highly aggressive BC subtype. Analysis of RBMS3 expression shows that RBMS3 is upregulated at both mRNA and protein levels in TNBC cells. Functionally, overexpression of RBMS3 increases cell migration, invasion and cancer stem cell (CSC) behaviors. Moreover, RBMS3 induces expression of epithelial-mesenchymal transition (EMT) and CSC markers. Conversely, loss of RBMS3 in TNBC BT549 cells inhibits cell proliferation, migration and mesenchymal phenotype. Correlation analysis shows RBMS3 is associated with TGF-β signaling. Mechanistically, RBMS3 interacts with Smad2, Smad3 and Smad4 mRNA and regulates the stability of these transcripts. Importantly, RBMS3 prevents TGF-β-induced cytostasis and apoptosis in premalignant cancer cells. Moreover, RBMS3 inversely correlates with expression of ESRPs, epithelial-specific splicing regulatory proteins that regulate morphogenesis-associated alternative splicing events. ESRPs appear to suppress EMT through distinct mechanisms: ESRP1 restricted cell migration, whereas ESRP2 prevented cell growth. RBMS3 significantly facilitates the EMT process when ESRPs are lost. Collectively, the studies within this dissertation identify RBMS3 as a positive regulator of EMT and breast cancer progression by regulating the TGF-β signaling pathway.
