Assessment of veld utilisation practices and veld condition in the Little Karoo

Thesis (MScConsEcol (Conservation Ecology and Entomology))--University of Stellenbosch, 2005. / The veld condition in the Little Karoo is in various states of degradation and grazing by domestic livestock is considered as the major anthropogenic force that changed the landscape. This region with its extremely rich plant species diversity and endemics, has supported small livestock for at least 2 000 years, and since colonization (250 years ago) been intensively used for the production of a variety of livestock. Ostrich production developed as the major source of income for this region. The first part of this study investigates the current veld management practices employed by livestock farmers in the Little Karoo region. Recommended veld management practices considered in this study are grazing rotation, moderate stocking rate control, moderate veld utilisation, separation of ecotopes, veld rehabilitation, controlling declared weeds and alien vegetation and regular assessment of veld condition. One hundred randomly selected farmers were personally interviewed by means of a structured questionnaire. Questions were grouped into the following categories: (a) demography of farmers, (b) ostrich farming, (c) perceptions and knowledge of farmers on farming practices, (d) grazing rotation, (e) stocking rate, (f) veld utilisation and veld assessment, (g) separation of ecotopes, (h) veld rehabilitation, (i) control of alien vegetation and (j) farmers’ knowledge on legislation. This was used to obtain information on the Little Karoo farming community, sizes of farms and camps, types of farming enterprises and on adoption of recommended veld management practices in the region. The main findings from this section are that relatively small farming units with few camps, poor separation of ecotopes and a low estimated grazing capacity, limit extensive livestock farming within the region. Perceptions of farmers on veld condition, grazing rotation, stocking rate, separation of ecotopes are fairly optimistic. As a result grazing capacities are overestimated and overstocking occurs within this region. The current stocking rate in ostrich camps (67.7% overstocked) and mammalian livestock camps (55.1% overstocked) is evidence that farmers overstock to compensate for these limiting factors in order to make a living from the land. The majority of farmers are well aware of the Articles in the Conservation of Agricultural Resources Act of 1983, which are applicable to veld management. Only more that 50% comply with this legislation by rehabilitating veld and 80% of them control invasive alien species on their farm. In the second part, veld assessments were done in randomly selected veld camps, using the multi-criterion, semi-subjective Quick Rangeland Health Assessment (QRHA) Method. Veld condition was significantly poorer closer to water or feeding points due to the piosphere effect caused by livestock. Veld condition in the Little Karoo can be related to altitude, vegetation types and land use. Therefore, the lowlying Little Succulent Karoo vegetation type is in a poorer condition compared to Spekboom Succulent Thicket and South and South-west Coast Renosterveld. Ostrich production on plains in the Little Succulent Karoo vegetation type is the main cause for the degradation of this vegetation type. It would seem as if historically high stocking rates cannot be ignored in explaining the current veld condition. A positive correlation between veld condition and the diversity of plant species (species density) were found, which highlights the importance of good veld management practices in sustainable agriculture. The third part tested whether all indicators in the QRHA method are equally sensitive and whether there is a positive correlation between the QRHA method and the Grazing Index Method. A significant positive linear correlation was found between the two methods. Cover was the least sensitive indicator of rangeland condition, and livestock induced disturbances (which include the indicators grazing intensity, disturbance indicators, soil health and species richness) were the most sensitive for Karoo veld assessment. A major benefit of the QRHA method is heuristic; therefore this method may have value in agricultural extension work.

Fynbos riparian biogeochemistry and invasive Australian acacias

Thesis (MScConEcol)--Stellenbosch University, 2012. / ENGLISH ABSTRACT: Riparian ecotones, transitional areas between upland terrestrial communities and aquatic ecosystems, are very dynamic and complex ecosystems with intrinsic ecological properties differing in spatial structure, function and temporal dynamics. Riparian habitats along rivers of the Mediterranean south-western Cape are sensitive to environmental change and particularly vulnerable to invasion by invasive alien plants (IAPs), especially nitrogen-fixing Acacia spp., and yet relatively little work has focused on how riparian ecosystems in this region respond to such stressors. The important roles that intact riparian vegetation play in maintaining ecosystem integrity and services have been increasingly highlighted as we acknowledge the degradation of these habitats. While the Working for Water (WfW) programme has been shown to be very successful in eradicating IAPs in riparian zones in the short-term, the extent to which riparian ecosystems recover following alien clearing activities remains poorly understood. The results presented in this study addressed several different aspects of riparian structure and function and acts as a steppingstone for guiding future research and management in riparian zones by adding to the evaluation of the success of clearing initiatives and restoration thereof. The aim of this study was to assess plant functional type (PFT) cover, soil physical and chemical properties, and selected biogeochemical processes in natural, Acacia- invaded and cleared riparian ecotones and associated non-riparian upland fynbos. Fieldwork was performed in mountain and foothill sections of six perennial river systems within the south-western Cape. Eleven sites of three categories were chosen: four natural sites (uninvaded); four moderate to highly invaded sites (predominantly A. mearnsii); and three cleared sites (a formerly invaded site that had been cleared more than 7 years prior to the study). Within each site, four to five replicate plots were established along each of three geomorphological zones (wet bank, dry bank, and upland fynbos). Seasonal soil samples were collected for a period of one year. Results from this study showed that PFT cover and composition, soil physical and chemical properties and rates of nitrogen (N) and phosphorus (P) mineralization differed amongst invasion status, between geomorphological zones and across seasons. Regarding most soil physical and chemical properties and indices N and P cycling, river floodplains (dry banks) were very similar to terrestrial uplands. Acacia spp. changes soil properties and affects plant functional attributes by i) enriching the system with N; ii) enhancing litter inputs; iii) altering soil physical properties; iv) changing the composition and reducing the cover of PFT; and v) enhancing P mineralization rates. Although measured soil physical and chemical properties and N and P mineralization rates were reduced to levels that were similar to or resembled the situation at natural areas, available inorganic N remained two times higher after more than seven years of clearance. Furthermore, cleared areas were characterized by sparse woody cover and a high cover of alien grasses. Correlations between soil silt and clay content and several soil properties measured in this and other studies indicates important linkages between soil texture and resource availability. Clearing Acacia spp. may initiate restoration of invaded riparian ecosystems, but changes in ecosystem function (e.g. elevated soil N availability) as a result of invasion may necessitate active restoration following the removal of the alien species. Active restoration under such conditions would be required to facilitate the restoration of cleared riparian communities. However, we still lack the mechanistic understanding around fynbos riparian recovery after clearing, as the success of restoration may depend on complex interaction and feedback cycles between plants and their physical environment. A greater comprehensive understanding of fynbos riparian ecological processes will not only improve the effectiveness of restoration initiatives by integrating science and management, but also advance the field of riparian ecology. / AFRIKAANSE OPSOMMING: Rivier oewerwal-areas, oorgang gebiede tussen aangrensende terrestriële gemeenskappe en akwatiese ekosisteme, is baie dinamiese en komplekse ekosisteme met intrinsieke ekologiese eienskappe wat verskil in struktuur, funksie (bv. biogeochemie siklusse) en temporale dinamika. Oewerhabitatte langs riviere van die Mediterreense suid-wes Kaap is sensitief vir omgewingsveranderinge en kwesbaar vir indringing deur uitheemse plante (bekend as “invasive alien plants” (IAPs)), veral stikstof-fiksering Acacia spp., en relatief min werk het nog gefokus op hoe ekosisteme in die streek reageer op sulke veranderinge in die omgewing. Die belangrike rol wat gesonde oewerwal plantegroei speel in die handhawing van ekosisteemdienste- en integriteit, is al hoe meer uitgelig soos ons die agteruitgang van hierdie habitat in ag neem. A comparison of standard scientific methods and pastoralists’ perceptions of vegetation responses to livestock exclusion in Namaqualand, South Africa

Thesis (MSc (Conservation Ecology and Entomology))--University of Stellenbosch, 2010. / ENGLISH SUMMARY: Protected areas do not always achieve the desired level of biodiversity conservation, while often reducing the welfare of indigenous communities by reducing availability of land for subsistence. Traditional agricultural landscapes are significant biodiversity refugia and can contribute meaningfully to conservation. Rangelands comprise one-third to one-half of the world’s terrestrial surface, providing livelihoods for around 220 million people, usually in a communal subsistence system. Colonial practices impinged on traditional land-use practices with far-reaching social and environmental impacts. This has resulted in management based on assumptions regarding vegetation dynamics and traditional lifestyles that are increasingly shown to be inaccurate. A comparison of a vegetation survey based on conventional scientific methods and a survey of the perceptions of pastoralists was undertaken to highlight differences and similarities between the two knowledge systems with the hope of providing guidelines for more sustainable land-use practices in the communal rangelands of Namaqualand, South Africa. Vegetation responses to removal of grazing pressure revealed complex interactions that do not correspond with the prevailing management paradigm. Rather than a predictive relationship between livestock and vegetation, environmental factors play a large role in determining plant composition, abundance and cover. Pastoralists’ perceptions reflected this complexity in rangeland resource dynamics. The impact of livestock on rangeland resource dynamics was perceived by herders to be secondary to a range of environmental and climatic factors. Both sets of results were at odds with the theories that currently govern management in this system. Studies in rangeland systems must take the complexity of the subject into account. Research into such socio-ecological systems must take a multiplicity of factors – social, environmental, economic, political and other – into account. Implications for management are that it is inappropriate to adhere strictly to the conventional, conservative strategies that are prescribed by conservation and agricultural authorities. Rather, a more flexible, opportunistic grazing strategy would allow the persistence of traditional subsistence livelihoods without serious negative consequences for biodiversity conservation. / AFRIKAANSR OPSOMMING: Die instelling van beskermde gebiede lewer nie altyd die gewenste vlak van biodiversiteitsbewaring, terwyl die welvaart van plaaslike gemeenskappe dikwels daaronder ly deur die afname in grond beskikbaar vir bestaanspraktyke. Morphological and molecular studies of tortricid moths of economic importance to the South African fruit industry

Thesis (PhD (Agric) (Conservation Ecology and Entomology))--University of Stellenbosch, 2005. / Six tortricid species are of major economic importance to the South African fruit industry. They are the codling moth Cydia pomonella, the oriental fruit moth Grapholita molesta, the false codling moth Thaumatotibia leucotreta, the macadamia nut borer T. batrachopa, the litchi moth Cryptophlebia peltastica and the carnation worm Epichoristodes acerbella. For phytosanitary purposes and to aid the management of population levels of the aforementioned species, their identities at species and population level were investigated using morphological and molecular genetic techniques. Morphological characteristics were used to distinguish and differentiate between the final instar larvae and pupae of the six species. For this purpose the morphology of the final instar larvae and pupae of the Afrotropical species T. leucotreta, T. batrachopa, Cr. peltastica and E. acerbella was described and illustrated using line drawings and scanning electron micrographs. Taxonomic characters found to be useful for the identification of the larvae were the presence and structure of the anal comb and the number and arrangement of crochets on the prolegs. The pupae could be distinguished based on the presence or absence of a distinct cremaster, the shape of the spiracle, the position of the setae on the anal rise, the structure of the mouthparts and the length of the procoxa in relation to that of the protarsus. These characters were used to develop keys to distinguish between the tortricid species occurring on tropical and subtropical fruit (T. leucotreta, T. batrachopa and Cr. peltastica) and deciduous fruit (E. acerbella, C. pomonella, G. molesta and T. leucotreta). At population level, molecular techniques were employed to compare geographic populations of each of the six species. Amplified fragment length polymorphism (AFLP) analysis with five selective primer pairs was used to investigate genetic diversity. In addition, host populations of species were compared where relevant. No evidence was found to suggest that populations from different hosts were genetically differentiated. However, geographic populations were found to be genetically distinct in each of the six species, with extensive genetic divergence apparent over local geographic scales and significantly high estimates of population differentiation ranging between Gst = 0.2625 and 0.3778. Factors influencing the genetic population structure of the six species were investigated by comparing the amount and distribution of genetic variation between oligophagous and polyphagous species as well as introduced and native species. Results indicated that host range and population history did not have a major effect on population genetic structure. It was therefore suggested that other factors such as limited dispersal were responsible for the extensive genetic divergence observed between geographic populations of each of the six tortricid species. These results should be incorporated into existing pest management programs and taken into consideration when designing new control strategies. This is the first report of its kind to identify, with a high level of accuracy, the aforementioned tortricids and the first to determine the population genetic structure of these species.

Climate change and vascular plant species interactions on sub-Antarctic Marion island

Thesis (PhD (Conservation Ecology and Entomology))--Stellenbosch University, 2008. / ENGLISH ABSTRACT: Shifts in species ranges are an important consequence of climate change, and can affect the composition, structure and functioning of ecosystems. Generally, in response to rising temperatures, species have increased their altitudinal and latitudinal distributions along their cooler boundaries, although there has been large variation between species. However, in addition to climatic factors, species range limits are also sensitive to biotic interactions. Therefore, interspecific interactions may be able to influence the impact of climate change on species distributions. In this thesis climate change, range shifts and spatial variation in plant-plant interactions are documented to examine the potential for biotic interactions to mediate climate-driven altitudinal range shifts on sub-Antarctic Marion Island. The climate on Marion Island changed considerably between 1949 and 2003, with significant trends in biologically-relevant parameters encompassing average climatic conditions, variability in climate parameters and extreme weather events. Combining these trends showed that while environmental conditions have ameliorated for some species, they have become more abiotically stressful for others (e.g. hygrophilous species). Concurrent with changes in the island’s climate, there have been rapid changes in the elevational distribution of Marion Island’s native vascular flora. On average species ranges expanded upslope (as predicted in response to the warming experienced on the island), although individual range expansion rates varied greatly. As a result of the idiosyncratic expansion rates, altitudinal patterns of species richness and community composition changed considerably. Therefore, both speciesand community-level changes occurred in the flora of Marion Island, demonstrating the community’s sensitivity to climate change. To determine the potential for interspecific interactions to have mediated these changes in species elevational ranges, spatial variation in the balance of positive and negative plant-plant interactions were examined. Increasing spatial association between four dominant species along an altitudinal severity gradient suggested that the intensity of facilitation, relative to the intensity of competition, is greater under more severe conditions. While, interaction strength varied between species pairs, the nature of relationship was consistent across the species. At a broader spatial scale, the performance of the grass Agrostis magellanica was compared in the presence and in the absence of the cushion plant Azorella selago along the entire altitudinal range of both species. The influence of the cushion plant on A. magellanica switched from negative to positive with increasing environmental severity, reaching a positive asymptote under extreme environmental conditions. Therefore, on Marion Island the spatial variation in the nature of biotic interactions is related to environmental severity, and facilitative interactions can be strong under extreme environmental conditions. These results show that positive biotic interactions are important at higher elevations on Marion Island, and could thus accelerate upslope range expansions in response to climate change. Nonetheless, competitive inhibition of upslope species movement, especially in areas of dense vegetation (i.e. mid or low altitudes), could have the opposite result, acting antagonistically with the effect of rising temperatures. This research highlights the importance of considering interspecific interactions when examining the biotic implications of climate change, both for affecting the rate at which species ranges change and the extent of species distributions. / AFRIKAANSE OPSOMMING: Die verskuiwing van spesies se verspreidingspatrone is ‘n belangrike gevolg van klimaatsverandering en kan die samestelling, struktuur en funksionering van ekosisteme affekteer. Die sensitiwiteit van Marion Eiland se flora ten opsigte van klimaatsverandering word deur hierdie verandering gedemonstreer. Om te bepaal of die interaksies tussen spesies verspreidingspatrone kan beïnvloed, is die geografiese variasie in die balans van positiewe en negatiewe interaksies ondersoek. Vier dominante vaatplante het meer geassosieerd geraak hoe hoër hulle bo seespieël voorgekom het. Dit is ‘n aanduiding dat die sterkte van die positiewe interaksies (fasilitering), relatief tot die sterkte van die negatiewe reaksie (kompetisie), groter onder uiterste omgewingstoestande is. Die sterkte van die interaksie het verskil tussen spesies pare, maar die verhouding tussen omgewingstoestande en die sterkte van die interaksie was dieselfde vir alle spesies. Op ‘n breër geografiese vlak is die opbrengs van die gras Agrostis magellanica in die teenwoordigheid en afwesigheid van die polsterplant Azorella selago op alle hoogtes waar die plante saam voorkom vergelyk. Die invloed van A. selago op A. magellanica was negatief op lae hoogtes bo seespieël, maar het meer positief geraak met ‘n toename in hoogte bo seespieël, tesame met omgewingshardheid. Die invloed van die polsterplant op die gras het ‘n positiewe asimptoot onder uiterste omgewingstoestande bereik. Dus, op Marion Eiland is die geografiese variasie in biotiese interaksies verwant aan omgewingstoestande, en positiewe interaksies kan selfs onder die uiterste omgewingstoestande sterk wees. Hierdie resultate wys dat positiewe biotiese interaksies tussen plante belangrik is by hoër hoogtes bo seespieël op Marion Eiland. Die interaksies kan dus die opwaartse verspreiding van spesies in reaksie op klimaatsverandering versnel. Nogtans kan negatiewe interaksies die teenoorgestelde effek hê aangesien kompetisie tussen plante, veral in plekke met digte plantegroei (d.w.s. lae of middel hoogtes bo seespieël) opwaartse verskuiwing van spesies kan verhinder. Plant response to habitat fragmentation : clues from species and functional diversity in three Cape lowland vegetation types of South Africa

With the need to prioritise conservation efforts, information on the conservation worthiness and management of these fragments is required. This study aimed to better understand how fragmentation affects the Cape lowland vegetation patterns and dynamics and more specifically, to determine if, and if so to what extent these fragments contribute to regional plant diversity and more importantly their functionality. The novel approach adopted focused on plant functional traits, which are better predictors of ecosystem response to global change than individual species. Species were sampled at four scales in four sites of decreasing sizes, including: a mainland and three fragments of three Cape lowland vegetation types i.e. Atlantis Sand Fynbos (ASF), Swartland Shale Renosterveld (SSR) and Langebaan Dune Strandveld (LDS). Traits such as dispersal, pollination, breeding mode and longevity were selected based on relevance to species’ and plantfunctional types’ (PFTs) responses to fragmentation. The findings revealed different effects on species richness and PFTs. The effect of reduced patch size on species richness was more evident in ASF where fragments below 600 ha had significantly fewer species than the mainland. This effect was not unequivocal in SSR and LDS due to several confounding factors (notably the grazing history of the sites). The SSR fragment grazed by indigenous herbivores had significantly more species than the ungrazed sites. Also, the largest LDS fragment grazed by livestock had significantly more species than the ungrazed mainland, indicating that grazing rather than fragment size influences species richness, although the smallest fragments of these two vegetation types had significantly fewer species than the larger fragments. Species turnover and complementarity were high for all three vegetation types, reflecting the degree of habitat heterogeneity and high contribution of beta diversity to overall gama diversity. The effect of reduced patch size was higher on PFT diversity than on PFT richness, with Langebaan Dune Strandveld where habitat fragmentation was more recent being the least affected of the three vegetation types. This indicates a degree of functional redundancy in the Cape lowlands, which is important for ecosystem resistance and resilience. The ASF mainland and the largest fragment had higher PFT diversity than the medium-sized and the smallest fragments; the mainland had also higher PFT diversity than all the fragments combined. Similarly, the smallest SSR fragment had significantly lower PFT diversity and richness than the other sites. The grazed SSR fragment had higher PFT richness and diversity than the ungrazed mainland and smallest fragment, indicating the role of grazing in maintaining renosterveld vegetation. The PFTs absent from the different sites were mostly short-distance dispersed dioecious and non-dioecious species, and some with highly specialised pollination systems. This suggests that dispersal and pollination are vital functional attributes for the persistence of the studied fragmented ecosystems. Habitat fragmentation effects plant community composition and ecological functions in the Cape lowlands, a conclusion supported also by the revealed significant trait-convergence and divergence assembly patterns. These communities result from various fragmentation filters that operate at different spatial-temporal scales and selecting species with suitable responses. All three vegetation types are susceptible to fragmentation, albeit at varying degree. The fragmentation effect was confounded by the sampling and temporal scales, the nature of disturbance regime, and the traitmediated differences in species’ response. The role of the surrounding matrix on fragment connectivity and gene flow appears to be of crucial importance, hence mitigation measures focusing on improving connectivity between patches, monitoring threatened taxa, and promoting dispersal and pollination have been recommended. / AFRIKAANSE OPSOMMING: Suid-Afrika se Kaapse laagland het dramaties verander en weggekwyn tot fragmente wat in matrikse van verskeie bedrywighede veranker is. Hierdie studie stel dit ten doel om beter begrip te vorm van hoe fragmentasie die plantegroeipatrone en -dinamiek in die Kaapse laagland raak, en meer bepaald om vas te stel óf, en indien wel, in watter mate, hierdie fragmente tot streeksplantdiversiteit en -funksionaliteit bydra. Die ongewone studiebenadering konsentreer op funksionele kenmerke van plante, wat beter aanwyser van ekosisteemreaksie op wêreldwye verandering is as individuele spesies. By 20 tot 30°C het temperatuurstygings die ontwikkeling van die insek laat versnel. By 35°C het die lewensduur van die volwasse motte afgeneem en is geen eiers gelê nie . Die valskodlingmot is deur middle van 'n kunsmatige metode geteel om genoegsame proefmateriaal te verkry. Verskeie verbeteringe is in die bestaande tegniek aangebring.

Characterisation of nematode symbiotic bacteria and the in vitro liquid culture of Heterorhabditis zealandica and Steinernema yirgalemense

Combined with their bacterial symbionts, these biocontrol agents have proven to be very effective against numerous pests. The nematodes belong to the families Steinernematidae and Heterorhabditidae, and are ideal to be used in, and integrated with, pest management systems. There is a dire need for new and innovative methods to control agricultural pests, as numerous pest insects have developed resistance against broad-spectrum insecticides. Together with the environmental impact of these insecticides and the safety aspect regarding humans and animals, the need to develop new technologies, including entomopathogenic nematodes for pest management, is high. In this study, the associated symbiotic bacteria of three entomopathogenic nematodes species were isolated, and the potential of two nematode species to be successfully mass cultured in liquid medium was evaluated. Regarding the symbiotic bacteria, results from the study showed that bacteria species from all three nematode species, Heterorhabditis noenieputensis, Steinernema khoisanae and Heterorhabditis zealandica, were novel. Heterorhabditis noenieputensis was isolated in the Mpumalanga province during a previous survey conducted in citrus orchards. The bacterium isolated from this nematode belongs to the genus Photorhabdus, and bear closest similarity (98.6%) to the type strain of P. luminescens subsp laumondii (TT01T). Photorhabdus luminescens subsp. noenieputensis subsp. nov., derives its name from the area where the nematode was sourced, namely the farm Springbokvlei, near the settlement Noenieput close to the Namibian border. Thus far, 85 Steinernema spp. have been described worldwide, including S. khoisanae which was isolated in the Western Cape province of South Africa. Four S. khoisanae strains, namely SF87, SF80, SF362 and 106-C, were used for characterisating the new bacteria from different localities in South Africa. Using the neighbor-joining method, all the strains were aligned with 97% homology to the 16S rRNA sequences of several Xenorhabdus- type strains, indicating that they belonged to the same genus. The multigene approach was used to distinguish between the Xenorhabdus spp. and partial recA, dnaN, gltX, gyrB and infB gene sequences of the various strains were analysed. The bacterium species was named Xenorhabdus khoisanae sp. nov. after the nematode from which it was isolated. The results showed that the third bacterium species, which was isolated from H. zealandica, was new. The sequence of the bacteria strain clustered with the type strains of P. temperata and P. asymbiotica, indicate that it belonged to the genus Photorhabdus. This is the first study to show that H. zealandica associates with a luminescent Photorhabdus species, rather than with the known non-luminescent P. temperata. The potential of H. zealandica and Steinernema yirgalemense mass culture in liquid was investigated. Results illustrated that H. zealandica and its P. luminescens symbiont can be successfully cultured in liquid. However, two generations occurred during the process time, instead of the desirable one-generation. The growth curve of the symbiotic bacteria during the process time was measured, in order to determine when the stationary phase was reached, with the results showing this to occur after 36 h. Therefore, the optimum amount of time required for inoculating the IJs and for aiding in maximum infective juvenile (IJ) recovery is 36 h for adding the nematodes post pre-culturing of the bacteria. Future research goals should be to increase the percentage recovery in liquid culture, which would increase the number of nematodes produced per ml, which would, therefore, reduce the processing time significantly. The results from mass culturing the second nematode species, S. yirgalemense, indicated an asynchronous nematode development in the first generation. Growth curves were performed with the symbiotic bacteria that showed the exponential phase of Xenorhabdus started after 15 h, and that, after 42 h, the stationary phase was reached, with an average of 51 × 107 cfu·ml-1. Bioassays were performed to compare the virulence between in vitro- and in vivo-produced nematodes, with the results showing that the in vitro-produced nematodes were significantly less virulent than were the nematodes produced in vivo. The success obtained with the production of S. yirgalemense in liquid culture can serve as the first step in the optimising and upscaling of the commercial production of nematodes in industrial fermenters. The last aim of the current study was to determine when Xenorhabdus reached the stationary phase, when it is grown in a 20-L fermenter, as this would be the optimum time at which to add the IJs of S. yirgalemense. Such characteristics as the effect of stationary phase conditions on the bacterial cell density and on the DO2 rate in the fermenter were investigated. The results showed that the stationary phase of Xenorhabdus was reached after 36 h at 30˚C, which took 6 h less than did the same procedures followed with the Xenorhabdus sp. cultured in Erlenmeyer flasks on orbital shakers. This is the first step toward the liquid mass culturing of S. yirgalemense in industrial-size fermenters. Data from this study indicated the optimum amount of time that is required for adding nematodes to the bacterial culture in the fermenter, and for ensuring the optimum recovery of IJs, as well as a subsequent high yield of nematodes within a minimum processing time. This is the first report of its kind to investigate comprehensively the successful liquid culture of two South African entomopathogenic nematode species for the sole purpose of evaluating potential commercialisation. Results emanating from this study could be used as groundwork in future, in combination with similar research such as culturing nematodes intensively in large fermenters. / AFRIKAANSE OPSOMMING: Entomopatogeniese nematodes het die potensiaal om as doeltreffende biologiese beheeragente teen sleutelplaaginsekte gebruik te word. Elke nematood werk interaktief met ‘n spesifieke bakterium. Entomopatogeniese nematodes, behorende tot die families Steinernematidae en Heterorhabditidae, is ideale kandidate vir gebruik in ‘n geïntegreerde plaagbestuurprogram. Tans is daar ʼn behoefte vir nuwe metodes vir die beheer van plaaginsekte, omdat meeste insekte reeds weerstand opgebou het teen bestaande plaagdoders. As gevolg van die negatiewe impak van plaagdoders op die omgewing, asook kommer oor veiligheid vir die mens en diere, is die ontwikkeling en gebruik van alternatiewe plaagbeheermiddels noodsaaklik. In die eerste deel van die studie word drie nuwe bakterie spesies geïsoleer en beskryf. Resultate van hierdie studie het aangetoon dat die bakterië spesies vanuit die nematode spesies, Heterorhabditis noenieputensis, Steinernema khoisanae, en Heterorhabditis zealandica, tot dusver onbeskryf was. Eersgenoemde, H. noenieputensis, is afkomstig van ʼn sitrusboord in die Mpumalanga Provinsie. Die bakterie hieruit geïsoleer behoort tot die genus Photorhabdus en is biologies verwant (98.6%) aan P. luminescens subsp laumondii (TT01T). Hierdie is die eerste studie met die bevinding dat H. zealandica ook met ʼn ander bakterie spesie geassosieer kan word buiten die normale P. temperata spesie. Die tweede deel van die studie gaan oor die teling van twee nematood spesies, H. zealandica en Steinernema yirgalemense, en hulle is geëvalueer vir hulle potensiaal om geteel te word in ʼn vloeibare medium. Die resultate het gewys dat H. zealandica met sy P. luminescens simbiont suksesvol in vloeistof aangeteel kan word, ten spyte van die feit dat daar twee generasies ontwikkel het, in plaas van die meer ideale enkel generasie. Die groeikurwe van die simbiotiese bakterie was gemonitor om te bepaal wanneer die stasionêre fase bereik word. Die resultate toon dat hierdie fase na 36 uur bereik was. Dus was die infektiewe nematode larwes eers na 36 uur tot die vloeibare medium waarin die bakterie geteel was bygevoeg. Navorsing in die toekoms moet dus gefokus wees om die persentasie herwinning van die infektiewe larwes te verhoog. Dit sal daartoe lei dat meer nematodes per ml geproduseer kan word en ook die prosesseringstyd van die nematodes verminder. ʼn Tweede nematode spesie, S. yirgalemense, was ook in vloeistof geteel. Hier het ʼn asinkroniese ontwikkeling in die eerste generasie plaasgevind wat problematies is. Groeikurwes is bepaal van die bakteriële simbiont en die resultate het gewys dat die groeifase van Xenorhabdus na 15 uur in aanvang geneem het en dat die stasionêre fase bereik was na 42 uur met ʼn gemiddelde van 51 × 107 selle·ml-1. Die virulensie van nematodes wat in vitro geteel is, is vergelyk met die virulensie van nematodes wat in vivo geteel is en die resultate het getoon dat die in vitro geteelde nematodes minder virulent was. Die teling van S. yirgalemense in vloeistof was oor die algemeen meer suksesvol as die teling van H. zealandica in dieselfde medium. Die doelwit van die laaste gedeelte van hierdie studie was om te bepaal wanneer Xenorhabdus die stasionêre fase bereik wanneer dit in ʼn 20-L fermenter gekweek word. Dit bepaal sodoende die optimale tyd wanneer die infektiewe larwes van S. yirgalemense bygevoeg behoort te word. Resultate het gewys dat die stasionêre fase van Xenorhabdus na 36 uur bereik was, wat 6 uur korter is as toe dit gekweek is in Erlenmeyer flesse. Hierdie studie is die eerste stap om die massa teling van S. yirgalemense in industriële fermenters suksesvol te bemeester. Die data wat verkry was, het aangedui wat die ideale tydsduur sal wees om die bakteriegetalle te vermeerder voordat die nematode bygevoeg word. Hierdie is die eerste studie wat die teling van twee Suid-Afrikaanse nematode spesies omvattend in vloeistof evalueer het. Die hoof doelwit is om die potensiaal van hierdie nematode spesies, met die oog op kommersiële gebruik, te meet. Die resultate van hierdie studie kan gekombineer word met toekomstige studies in hierdie spesifieke navorsingsveld.

Opportunities and challenges for mainstreaming ecosystem services in decision making

Thesis (PhD)--Stellenbosch University, 2014. / ENGLISH ABSTRACT: Ecosystem service research has grown significantly in recent years, highlighting links between ecosystem services and human well-being. Few studies are however, geared towards providing user-friendly, user-useful and user-relevant information suitable for implementing effective local management of areas that deliver ecosystem services, resulting in a gap between ecosystem service research and management. In response to this challenge, it has been argued that in order to safeguard the benefit flows provided by ecosystems, the concept of ecosystem services should be mainstreamed into land-use (and water-use) planning and management processes. As the conservation of ecosystem services is ultimately a social process operating in a social context, understanding the complexity of the research-management interface demands input from a range of stakeholders. Thus, mainstreaming the environment into decision making requires multi-stakeholder engagement processes that facilitate the co-production and exchange of knowledge. Accordingly, through the use of a transdisciplinary, mixed method approach, this dissertation contributes to a better understanding of some of the opportunities and challenges for mainstreaming ecosystem services in development planning at a local level, using a case study in the Eden District (Eden) of South Africa. Through an extensive stakeholder engagement process which explored what the information needs, responsibilities and capacities of decision makers in Eden are, both opportunities that facilitate the integration of ecosystem service information in decision making, and challenges impeding integration, have been identified. This research found that despite a history of ecosystem service research in Eden, there has been limited integration of the concept of ecosystem services into decision-making processes driving development. Insufficient capacity, limited resources, minimal proactive planning, a weak regulatory environment, entrenched disciplinary thinking and insufficient communication amongst diverse stakeholders on the benefits of using an ecosystem-based approach remain as challenges for the mainstreaming of ecosystem services in decision making. However, strategic opportunities for mainstreaming ecosystem services into decisions regarding current and future development trajectories were also found. It was found that the development, and nurturing of transdisciplinary learning networks that are problem driven, and action oriented, using a communities of practice model of engagement, can facilitate legitimate knowledge exchange processes. In this regard, the role of individual and institutional knowledge brokers was found to be critical. It was also found that the concept of risk was useful as a mainstreaming tool in bridging the gaps between different disciplines, and between science, policy and practice. A frame of risk enabled different disciplinary and knowledge communities to participate in joint activities and discussions during which information was co-produced and exchanged. Through this work, new relationships were built that facilitated both learning and action with regards to the importance of ecosystem services for mitigating risk. / AFRIKAANSE OPSOMMING: Ekosisteemdiens-navorsing het aansienlik gegroei in die afgelope jare, met ‘n klem op die verband tussen ekosisteemdienste en menslike welsyn. Min ondersoeke is egter gerig op die verskaffing van gebruikersvriendelike, -nuttige en -relevante inligting wat geskik is vir die implementering van doeltreffende plaaslike bestuur van die gebiede wat ekosisteemdienste lewer, en dit lei tot 'n gaping tussen ekosisteemdiens-navorsing en -bestuur. In reaksie op hierdie uitdaging word aangevoer dat ekosisteemdienste gehoofstroom moet word in prosesses ter beplanning en bestuur van grond- en watergebruik, ten einde die voordele wat deur ekosisteme gebied word, te beskerm. Omdat die behoud van ekosisteemdienste per slot van rekening `n sosiale proses is wat in `n sosiale konteks in werking is, word insette van 'n verskeidenheid van belanghebbendes vereis, om die ingewikkelde aard van die koppelvlak tussen navorsing en bestuur te verstaan. Maar strategiese geleenthede vir die hoofstroming van ekosisteemdienste in besluitneming in verband met huidige en toekomstige ontwikkelingsbane is ook aangetref. Daar is bevind dat, deur transdissiplinêre leernetwerke wat probleemgedrewe en aksie-georiënteerd is, te ontwikkel en koester met behulp van 'n praktykgemeenskappemodel van betrokkenheid, legitieme kennisuitruilingsprosesse gefasiliteer kan word. In hierdie verband is die rol van individuele en institusionele kennismakelaars as krities bevind. Daar is ook bevind dat die konsep van risiko nuttig is as 'n hoofstromings-instrument in die oorbrugging van die gaping tussen die verskillende dissiplines, en tussen wetenskap, beleid en praktyk. `n Raamwerk van risiko het verskillende dissiplinêre en kennisgemeenskappe in staat gestel om deel te neem aan gesamentlike aktiwiteite en besprekings, waartydens inligting saam geproduseer en uitgeruil is. Deur hierdie werk is nuwe verhoudings gevestig, wat beide leer en optrede rakende die belang van ekosisteemdienste vir risikotempering gefasiliteer het.
