Resistance of Zymoseptoria tritici populations to some active ingredients of fungicides

During the different stages of its development, wheat is possible infected by many plant pathogens. One of the most common is Zymoseptoria tritici, the causal agent of the disease called Septoria Tritici Blotch (STB). The most common strategy in the Z. tritici disease management is treating plants with fungicides. Unfortunately, this plant pathogen as many others evolved resistance towards some of the most commonly used fungicidal classes. The aim of this study was to detect the resistance of Z. tritici to strobilurin fungicide azoxystrobin. During the April 2017, around 300 plant samples were collected from 11 geographical regions in the Czech Republic. From those plant samples, 52 monosporic Z. tritici isolates were obtained and tested for the presence of the resistance. The fungicide resistance was measured and detected by laboratory agar dilution biotest and molecular methods as a CAPS marker (Cleaved Amplified Polymorphic Sequences) and qPCR (Quantitative Polymerase Chain Reaction). By agar dilution biotest, resistance to azoxystrobin was confirmed in 54% of total 52 analysed isolates. By CAPS marker analysis, the presence of G143A mutant allele of the mitochondrial cytochrome b gene, linked to the fungicide resistance, was confirmed in all selected isolates marked as resistant in the biotest. The DNA of one infected leaf sample collected from the field marked as 17Zt212 was isolated and tested by the qPCR method. In the field sample (17Zt212), the 4% of the Z. tritici population was found to be resistant to fungicide azoxystrobin.

Genetická diverzita lipana podhorního (Thymallus thymallus) a její vztah ke stavu jeho populací v ČR / Genetic diversity of European greyling (Thymallus thymallus) in the Czech Republic based on mitochondrial DNA variation

Goal of this Thesis is the comparison of the Grayling˘s genetic diversity. The complete mitochondrial DNA (mtDNA) control region (1083 base pairs) and gene for cytochrom B (1076 base pairs) were sequenced in 147 European grayling (Thymallus thymallus), from 21 populations in Czech Republic, Slovakia and Bosnia. In samples, 17 haplotypes were detected with average haplotype diversity Hd = 0,5480 and genic differentiation (FST) from 0,001 to the 0.9341. The most common haplotype TH1 was detected in 92 samples. An inferred intraspecific phylogenetic tree revealed three well-separated clades. One clade was represented by Danubian haplotypes from Slovakia. Second highly divergent clade contained individuals from river Vrbas in Bosnia, and the last biggest clade included haplotypes of all three sea basins from Czech Republic. We suppose that the gained results will be later utilized to the choice of suitable conservation measures that will lead to stabilization of the state of ancestral population of this ever more rare species in our region.

Die vorliegende Arbeit beschäftigt sich mit Cbs1p und Cbs2p, zwei spezifische Translationsaktivatoren der COB-mRNA. Im Mittelpunkt standen sowohl die weitere molekularbiologische und biochemische Charakterisierung von Cbs1p als auch ein Screening von Interaktionskandidaten, die mit Cbs1p und/oder Cbs2p physikalisch wechselwirken könnten. Cbs1p liegt als peripheres Membranprotein fest mit der inneren Mitochondrienmembran matrixseitig assoziiert vor. Dabei spielen möglicherweise hydrophobe und/oder Protein-Protein-Wechselwirkungen mit integralen Membranproteinen eine essentielle Rolle bei der Membranverankerung von Cbs1p. Durch die Identifizierug von atmungsdefekten Cbs1p-Mutanten, deren Mutationen in Bereichen mit Homologie zu RNA-bindenden Proteinen liegt, verstärken sich die Hinweise zur Beteiligung von Cbs1p an der direkten physikalischen Wechselwirkung mit dem 5´-leader der COB-mRNA. Darüber hinaus konnte gezeigt werden,, dass die abspaltbare Präsequenz nicht notwendig für einen mitochondrialen Import ist. Die Ergebnisse präzisieren und erweitern das vorliegende Modell der Wirkungsweise der Translationsaktivatoren Cbs1p und Cbs2p (Michaelis, 1991). Aufgrund der Membranverankerung von Cbs1p wird auch die gebundene COB-mRNA in räumlicher Nähe zur Membran gebracht. Darüber hinaus definiert Cbs1p damit möglicherweise auch den Ort der Insertion des nascierenden Apocytochrom b in die Membran. Cbs2p vermittelt die Bindung zur kleinen Untereinheit der mitochondrialen Ribosomen und könnte seinerseits ebenfalls in Interaktionen mit Untereinheiten des bc1-Komplexes involviert sein.

Evolutionary relationships between Barbus species have been already studied in the past, but most of the studies were based only on mitochondrial DNA, there is no detailed study based on nuclear markers. This is mainly due to the fact that Barbus species are tetraploid. The western part of Greece has been studied insufficiently, and moreover a greater diversity of freshwater ichthyofauna is expected here. Greece is one of the hotspots of biodiversity. Greek river system is characterized by a significant proportion of endemic species of freshwater fishes. In this work evolutionary relationships between Barbus species were studied, with focus on diversity of Greek lineages. The geographical distribution of Barbus species has been also revised. In total, 192 individuals were analyzed, 156 individuals were from 40 Greek localities and another 36 Barbus species were from other areas of distribution of the genus. Dataset was supplemented with avaible published sequences. For the phylogenetic analysis mitochondrial gene cytochrom b, 1st intron of ribosomal protein gene S7 and 2nd intron of gene beta-actin were used. Krasová polje východní Hercegoviny z perspektivy ryby rodu Telestes / Telestes perspective on the karstic poljes of eastern Herzegovina

The Dinaric karst region in the Balkans is one of the hot spots of biodiversity with a high number of endemic freshwater fish. The major cause of the high degree of endemism are numerous paleoclimatic and geological events that have affected the formation of the entire area. Flows of karst poljes represent a specific ecosystem to which some fishes have adapted. The poljes have been periodically flooded and these floods may activate different underground channels, which lead to interconnection of poljes. These fish species spend a part of the year in surface water and a part in groundwater and they may use the underground flows between poljes for migration. The aim of this thesis is to evaluate the genetic variability of two endemic fish species of the genus Telestes living in karst poljes in the region of eastern Herzegovina, and to detect a possible current gene flow between populations of these species due to the connection of underground flows. For this purpose, the DNA from 102 individuals from five different localities was isolated. The mitochondrial marker cytochrom b and 16 microsatellite loci were used for the analyses.
