Three species and two subspecies are synonymized: Rhopalurus amazonicus with R. laticauda, R. crassicauda with R. laticauda, R. acromelas with R. agamemnom, R. crassicauda paruensis with R. laticauda,R. pintoi kouruensis with R. pintoi. The genus present now seven valid species for South America. New data on distribution range and an identification key are presented. A comparative analysis of the morphology of male hemiespermatophore of 21 genera of the family Buthidae of Central and South America, Europe, Africa and Asia. Tityus stigmurus is a widely distributed scorpion species in Northeastern Brazil and known to cause severe human envenomations, inducing pain, hyposthesia, edema, erythema, paresthesia, headaches and vomiting. The present study uses a transcriptomic approach to characterize the molecular repertoire from the non-stimulated venom gland of Tityus stigmurus scorpion. A cDNA library was constructed and 540 clones were sequenced and grouped into 37 clusters, with more than one EST (expressed sequence tag) and 116 singlets. Forty-one percent of ESTs belong to recognized toxin-coding sequences, with antimicrobial toxins (AMP-like) the most abundant transcripts, followed by alfa KTx- like, beta KTx-like, beta NaTx-like and alfa NaTx-like. Our analysis indicated that 34% include other possible venom molecules , whose transcripts correspond to anionic peptides, hypothetical secreted peptides, metalloproteinases, cystein-rich peptides and lectins. Fifteen percent of ESTs are similar to cellular transcripts. Sequences without good matches corresponded to 11%. This investigation provides the first global view of cDNAs from Tityus stigmurus. This approach enables characterization of a large number of venom gland component molecules, which belong either to known or atypical types of venom peptides and proteins from the Buthidae family / No Brasil, acidentes com escorpi?es s?o considerados de import?ncia m?dicosanit?ria, n?o somente por sua incid?ncia, mas tamb?m pela potencialidade do veneno de algumas esp?cies em determinar quadros cl?nicos graves. Tityus stigmurus ? uma esp?cie de escorpi?o amplamente distribu?da na regi?o Nordeste do Brasil sendo conhecida por causar graves envenenamentos humanos, cujos sintomas incluem: dor, hipoestesia, edema, eritema, parestesia, cefal?ia e v?mitos. A composi??o qu?mica do veneno do escorpi?o T. stigmurus ainda n?o foi bem avaliada, havendo uma car?ncia na literatura de estudos com enfoque na elucida??o do repert?rio molecular dos componentes deste veneno. Neste trabalho, realizamos uma abordagem transcript?mica para caracterizar o repert?rio molecular da gl?ndula de veneno n?o-estimulada do escorpi?o Tityus stigmurus. Para tanto, uma biblioteca de cDNA foi constru?da e, ? partir dela, 540 genes foram clonados e agrupados em 37 clusters com mais de um EST (expressed sequence tag) e 116 singlets (somente um EST). Do total de transcritos, 41% pertencem a sequ?ncias de toxinas conhecidas, sendo os pept?deos antimicrobianos (AMPs) os transcritos mais abundantes, seguido por α-KTX, β- KTX, β-NaTx e α-NaTx, respectivamente. Nossa an?lise revelou ainda que 34% s?o "poss?veis toxinas", cujos transcritos correspondem a pept?deos ani?nicos, pept?deos hipot?ticos secretados, metaloproteases, pept?deos ricos em ciste?na e lectinas. Quinze por cento dos ESTs s?o semelhantes a transcritos celulares. Transcritos sem similaridade com outras sequ?ncias do GenBank, foram chamadas de no hit e correspondem a 11% do total. Este trabalho apresenta a primeira vis?o global de cDNAs das gl?ndulas venenosas do escorpi?o Tityus stigmurus. The scorpion Tityus serrulatus is the main responsible for more severe cases. Anti-scorpion serums are routinely produced by various institutions, despite their effectiveness, quality and action depends on how quickly treatment is started. Studies have been developed in the search for appropriate technologies to encapsulate and release recombinant or natives proteins capable of inducing antibody production. In this context, chitosan copolymer which can be obtained from the partial deacetylation of chitin or in some microorganisms and it is biocompatible and biodegradable has been widely used for this purpose. This study aimed to search for a system release from chitosan nanoparticles for peptide / protein of the venom of the scorpion T. serrulatus, able to provide a new model of immunization in animals, in order to obtain a potential novel polyclonal serum, anti-venom T. serrulatus. The chitosan nanoparticles were prepared by ionic gelation with polyanion tripolyphosphate (TPP). After standardizing the concentrations of TPP and chitosan was evaluated the efficiency of incorporation of bovine serum albumin (BSA) and scorpion venom, showed particle size compatible with the intended purpose. The particles showed adequate size around 200nm. The crosslinking was confirmed by absorption spectroscopy in the infrared. After verified the high encapsulation efficiency (EE) for acid bicincon?nico method (BCA) protein assay and the particle size distribution, the success of the technique was proven and the potential for in vivo application of nanoparticles. The experimental animals were vaccinated and the antibodies measured by ELISA / No Brasil, diversas esp?cies de escorpi?es s?o conhecidas por provocar acidentes que podem levar ao ?bito, sendo estas pertencentes principalmente ao g?nero Tityus. O escorpi?o Tityus serrulatus ? o principal respons?vel pelos casos de maior gravidade. Soros anti-escorpi?nicos s?o produzidos rotineiramente por diversas institui??es, e, apesar da sua efic?cia, a a??o depende da qualidade e da rapidez com que ? iniciado o tratamento. V?rios estudos t?m sido desenvolvidos na busca de tecnologias adaptadas para encapsular e liberar prote?nas nativas e/ou recombinantes capazes de induzir a produ??o de anticorpos. Neste contexto, a quitosana um copol?mero que pode ser obtida a partir da desacetila??o parcial da quitina ou em alguns microrganismos e por ser biocompat?vel e biodegrad?vel tem sido amplamente utilizada para esta finalidade. O presente trabalho teve como objetivo a busca de um sistema de libera??o a partir de nanopart?culas de quitosana para pept?deos/prote?nas do veneno do escorpi?o T. serrulatus, capaz de proporcionar um novo modelo de imuniza??o em animais, com a finalidade para potencial obten??o de um soro policlonal in?dito, anti-veneno de T. serrulatus. As nanopart?culas de quitosana foram preparadas por gelifica??o i?nica com tripolifosfato poliani?nico (TPP). Ap?s padronizar as concentra??es de TPP e de quitosana, foi avaliado a efici?ncia de incorpora??o de Albumina Soro Bovina (BSA) e o veneno do escorpi?o, apresentaram tamanho de part?cula compat?vel com a finalidade pretendida. As part?culas apresentaram tamanho adequado em torno de 200nm. A reticula??o foi comprovada por espectroscopia de absor??o na regi?o do infravermelho (IV).
