Repression of the blood endothelial marker CD146 by the homeobox gene PROX1

CD146 is a cell adhesion molecule that has been shown to regulate cell adhesion, migration and proliferation of different cell types. It is highly expressed in blood endothelial cells (BECs), but is only lowly expressed in lymphatic endothelial cells (LECs). The PROX1 homeobox gene is a master regulator of lymphangiogenesis and its expression is necessary and sufficient to drive venous endothelial cells into a LEC phenotype. The highly permeable nature of the lymphatic vessels may partially derive from PROX1 mediated repression of CD146 transcription. We hypothesize that PROX1 promotes lymphatic differentiation by repressing CD146 transcription. In gain of function studies, Human Umbilical Vein Endothelial Cells (HUVECs) were infected with adenoviruses encoding EGFP, wild type PROX1 (AdProx1) or a Homeo-Prospero domain deleted version of PROX1 (AdHDPD), which cannot bind DNA. In order to knockdown PROX1, LECs were transfected with PROX1 specific siRNA. When compared to EGFP infected HUVECs, AdProx1 infected HUVECs had decreased CD146 expression both at protein and mRNA levels. In contrast, AdHDPD infected HUVECs had increased levels of CD146 expression. In support of a role for PROX1 in repressing CD146, PROX1 siRNA transfected LECs express higher levels of CD146 as compared to mock transfected LECs or LECs transfected with control siRNA. Based on these results, we predict that CD146 expression is kept at basal levels by an unknown repressor bound to the CD146 promoter. By interacting with this unknown repressor, PROX1 further represses CD146 expression. On the other hand, the DNA binding-deficient ΔHDPD version of PROX1 binds the unknown repressor and sequesters it from the CD146 promoter, thereby relieving the repression of CD146 expression in ECs. Different levels of CD146 expression between BECs and LECs might reflect the structural and functional differences between blood and lymphatic vessels. Since CD146 plays a critical role in EC adhesion, regulation of CD146 expression in ECs might be one of the key factors regulating vessel permeability.

Fate and Death through a Daimonic Lens

This thesis is concerned with the ancient Greek conceptualization of fate and death, as explored through the figure of the daimon in literature from Homer and Hesiod to Plato and Euripides. Filling a gap in scholarship, I elucidate the spectrum of meaning inherent in the word daimon, and how it shifts over time. From the Archaic to the Classical period the word daimon is found as a synonym for theos, “god”, as a vocative address, or in reference to “fate” and the generalized “will of heaven.” At the same time, a particular group of divine personifications, including Thanatos, Moira, Ker, and Erinys are counted as daimones. We also find the term used to designate unnamed but individuated lesser divinities, guardian spirits, and demonic possessors, and even as the divine aspect of the self. In the early Archaic poets these latter categories are only nascent. The individuated daimon becomes the focus of the lyric poets and pre-Socratic philosophers; in the later pre-Socratics the daimon begins to be internalized, moving from possessive spirit to psychic force. Tragedy meanwhile focuses on the daimon as a force of retribution, as curse or afflicting demon. It is Plato who explores and expands upon all of these categories, crystallizing the notion of the internalized daimon, as reconceived in the context of his philosophical eschatology. Chapters 1 and 2 provide surveys of the word daimon diachronically in each of these genres, mapping the expanding continuum of meaning. What to control when cancer comes? : the relationship of multidimensional health locus of control, fate control and subjective well-being among Chinese cancer patients

Cancer, the most common cause of death in Hong King, poses marked psychological impacts through treatment into cancer survivorship. Literature indicated that internal health locus of control was associated with better psychosocial adjustment (Wang et al., 2013). Meanwhile, fatalistic view was related to avoidant coping and poor psychological adjustment (Chan, 2000). However, fatalism might have a different meaning for the Chinese (Ho et al., 2003). The present study examined the relationship of health locus of control, fate Control and subjective well-being among Chinese cancer patients. Ninety-nine cancer patients were assessed with Multidimensional Health Locus of Control Scale, Fate Control Scale from Social Axiom Survey and Functional Assessment of Cancer Therapy Scale-General during active treatment (Time 1) and with WHO-5 Well-Being Index at a six-month follow-up (Time 2). At Time 1, results indicated positive correlation between powerful others health locus of control and functional well-being. At Time 2, internal health locus of control was positively correlated with WHO-5 while social well-being at Time 1 was positively correlated with WHO-5. Determining Fate and Transport Parameters for Nitroglycerine, 2,4-Dinitrotoluene, 2,6-Dinitrotoluene and Nitroguanidine in Soils

During military operations, a small fraction of propellant mass is not consumed during firing and is deposited onto the ground surface. Soluble propellant constituents can be released from particulate residues into the environment. The release of uncombusted propellant residues and transformation products results in soil, surface water, and ultimately groundwater contamination. It is important to study the potential for off-site migration of munitions and to provide qualitative data that helps better understand transit of these propellant constituents. Propellant constituents of interest for this study were nitroglycerine (NG), 2,4-dinitrotoluine (2,4-DNT), 2,6-dinitrotoluine (2,6-DNT), and nitroguanidine (NQ). The goal of this work is to determine fate and transport parameters, partition/distribution coefficients and transformation rates for these compounds in three soils that represent a range of geographic locations and soil properties. This supports a companion study that looks at dissolution of constituents from fired and unfired solid propellant formulations and their transport in soil.
