Synthesis and Characterization of Nanoporous Copolymers with Potential Gas Storage ApplicationsZhou, Xu 10 October 2013 (has links)
Nanoporous organic polymers, including hypercrosslinked polymers (HCPs), covalent organic frameworks (COFs), polymers of intrinsic microporosity (PIMs), and conjugated microporous polymers (CMPs) etc., are considered good candidates for potential gas storage and gas separation applications.
Porosities and surface areas of a series of semirigid alternating copolymers, which contained tert-butyl carboxylate-functionalized stilbene or tert-butyl carboxylate-functionalized styrene, and maleic anhydride or tert-butyl carboxylate-functionalized phenyl maleimide, were investigated using nitrogen sorption/desorption isotherms at 77 K and molecular simulations. These alternating copolymers were found to have Brunauer-Emmett-Teller (BET) surface areas in the range of 20-40 m2/g. Surface areas of these alternating copolymers increased as the steric crowding of the polymer backbone increased, which was the result of introducing extra phenyl rings and/or N-phenyl substituent maleimide units. Surface areas were found to increase as the persistence length increased.
A series of HCPs containing functionalized stilbene and N-substituted phenyl maleimide were synthesized via free radical suspension polymerization. The incorporation of these functionalized, chain stiffening, Tg enhancing comonomers raised the Tgs of precursor polymers before they were crosslinked. Surface areas of these HCPs, obtained from nitrogen adsorption/desorption isotherms at 77 K, were up to 1058 m2/g. However, the surface areas of these HCPs were systematically lower than the controls. The high rigidity of the polymer backbone, which was the result of incorporating Tg enhancing comonomer, likely affected the chain mobility of the precursor polymer, decreased the efficiency of post-crosslinking reactions, and thus resulted in lower surface areas.
Amine-functionalized styrene/stilbene polymers were prepared via free radical polymerization or post-modification. Amine-containing silica-based sorbents were prepared using the impregnation method. Sorption of CO2 by these materials was tested using TGA and compared with control samples. Both high amine content and certain levels of surface area were found to be important for a sorbent to achieve high CO2 uptake. Highest CO2 uptake (12 wt%) under our testing condition in these materials was achieved by an amine-containing silica sorbent. / Ph. D.

Cette étude porte sur la décontamination de matrices textiles en milieu CO2 dense (CO2 liquide ou CO2 supercritique). Elle s'inscrit dans le cadre de la décontamination des textiles utilisés dans l'industrie nucléaire. Le CO2 dense est proposé comme alternative au milieu aqueux utilisé dans le procédé actuel et qui génère une importante quantité d'effluents aqueux contaminés nécessitant un post-traitement. Le contaminant étudié est le cobalt qui peut se présenter sous forme ionique ou particulaire. L'extraction du cobalt en milieu CO2 dense est assurée par un additif : un tensioactif macromoléculaire CO2-phile/CO2-phobe complexant. Plusieurs familles d'additifs ont été synthétisées par polymérisation radicalaire contrôlée : des copolymères à gradient comportant des motifs CO2-philes, siliciés ou fluorés, et des motifs CO2-phobes complexants de types acétoacétoxys, diéthylphosphonates ou acides phosphoniques. Le comportement de ces copolymères dans le CO2 dense a été évalué grâce à la détermination des diagrammes de phases copolymère-CO2 (par la mesure du point de trouble) et grâce à l'étude de leur autoorganisation dans le CO2 dense (par diffusion de neutrons aux petits angles). Les copolymères fluorés se sont avérés être les plus avantageux en termes de solubilité. Néanmoins, les copolymères siliciés présentent une solubilité compatible avec le procédé et ils constituent donc une alternative intéressante pour éviter la présence de fluor gênant pour le conditionnement des déchets nucléaires. L'étude de la complexation du cobalt par les copolymères (par spectrométrie UV-visible et par torche à plasma couplée à un spectromètre d'émission atomique) a permis d'établir des relations entre le type de motif complexant et l'affinité avec le cobalt. La solubilité dans le CO2 dense de ces complexes copolymères-cobalt est comparable à celle des copolymères seuls. De plus, l'étude de l'auto-organisation en milieu CO2 dense a révélé un faible taux d'agrégation des complexes copolymères-cobalt. Enfin, les copolymères synthétisés ont été mis en oeuvre dans les procédés de décontamination particulaire et ionique. Dans le cas du procédé de décontamination ionique, l'emploi du copolymère à gradient poly(acrylate de 1,1,2,2-tétrahydroperfluorodécyle-co-diacide vinylbenzylphosphonique) a permis d'atteindre environ 70% de décontamination grâce à la formation d'une microémulsion d'eau dans le CO2 dense. L'efficacité du procédé dedécontamination a été portée à 97% grâce à l'emploi de pyridine comme tiers additif. / This study is about textile decontamination in dense CO2 (liquid CO2 or supercritical CO2). The study is carried out in the framework of decontamination of textile used in the nuclear industry. The dense CO2 offers an alternative to aqueous medium used in the current process which generates a huge quantity of contaminated aqueous effluent requiring a post-treatment. Cobalt is the targeted contamination and can be found as ionic species or particles. The cobalt extraction in dense CO2 is achieved with an additive : a complexing CO2-philic/CO2-phobic macromolecular surfactant. Several types of additives were synthesized by controlled free radical polymerization : gradient copolymers made with CO2-philic groups (silicone-based or fluorinated moieties) and CO2-phobic complexing groups (acetoacetoxy, diethylphosphonate or phosphonic acid moieties). The copolymer behavior in dense CO2 was determined by phase diagram measurements (cloud point method) and their self-assembly in dense CO2 was investigated by small angle neutron scattering. The fluorinated copolymers were found advantageous in terms of solubility. Nevertheless, the silicone-based copolymers showed solubilities which are compatible with the process, therefore they are a good alternative to avoid fluorinated compounds which are unwanted in the conditioning of nuclear wastes. The study of cobalt complexation by the copolymers (UV-vis spectroscopy and inductively coupled plasma-mass spectroscopy) established relations between the type of complexing group and the affinity with the cobalt. The solubility of copolymer-cobalt complexes in dense CO2 is similar to those of copolymers. Moreover, the self-assembly study of the complex revealed a low aggregation. Finally, the synthesized copolymers were used in particle or ionic decontamination processes. In the case of ionic decontamination process, a rate of 70% of decontamination was reached with the use of gradient copolymer poly(1,1,2,2-tetrahydroperfluorodecyle acrylate-covinylbenzylphosphonic diacid) which allowed the formation of water-in-CO2 microemulsion. The efficiency of the decontamination process was even improved up to 97% with the addition of pyridine in the process.

O comportamento eletroquímico do Nitrofural (NFZ) foi estudado em eletrodos de pasta de carbono, carbono vítreo e diamante altamente dopado com boro (EDADB) em tampão Britton-Robinson (BR) usando a voltametria cíclica. Os melhores resultados foram obtidos em EDADB, no qual foi possível estabilizar o radical aniônico (R-NO2•‾) em meio predominantemente aquoso, como também estudar a interação entre o R-NO2•‾ a glutationa (GSH), a L-cisteína (Cis) e o O2. Somente na superfície do EDADB, o NFZ foi diretamente reduzido à amina derivada (R-NH2) no intervalo de 2,02 ≤ pH ≤ 4,03 num processo envolvendo seis (6,0 ± 0,4) elétrons e seis prótons. Na faixa de 7,04 ≤ pH ≤ 12 e meio predominantemente aquoso, a etapa de redução desmembrou-se em seus dois componentes: a redução do NFZ ao nitro radical aniônico (R-NO2•‾) e posterior redução deste ao derivado hidroxilamínico (R-NHOH) em processos envolvendo um e três (3,1 ± 0,1) elétrons respectivamente. No sentido anódico da varredura dos voltamogramas cíclicos registrados em pH 8,06, observou-se a oxidação da hidroxilamina ao derivado nitroso (R-NO), num processo envolvendo 2 (1,7 ± 0,2) elétrons e 2 prótons. Além disto, um novo pico de oxidação não relatado na literatura em qualquer material de eletrodo foi detectado em pH ≤ 8,06, que foi observado não importando se o NFZ havia sido previamente reduzido ou não. O cálculo do número de elétrons, n, envolvido em cada etapa eletroquímica foi satisfatoriamente estimado usando-se a equação de Randles-Sevcik. Na presença de [Tiol] ≤ 3,7 x 10-2 mol L-1, o NFZ é reduzido ao aduto R-NO-Tiol em um processo eletroquímico envolvendo dois elétrons e dois prótons. Por outro lado, o O2 atua como um aceptor de R-NO2•‾ e a constante de equilíbrio para a transferência monoeletrônica do R-NO2•‾ para o oxigênio, kO2, é 60. O processo é catalítico e pode ser utilizado para a determinação analítica de NFZ na faixa de 9,9 x 10-7 ≤ [NFZ] ≤ 1,1 x 10-5 mol L-1 em pH 8,06, com sensibilidade amperométrica de 2,2 x 104 µA L mol-1 cm-2 e limite de detecção de 3,4 x 10-7 mol L-1. Os parâmetros analíticos foram similares aqueles obtidos em pH 4,0 usando a redução direta do NFZ à respectiva amina derivada em um processo envolvendo seis elétrons e seis prótons. A caracterização do processo de redução total do NFZ em meio aquoso e velocidades de varredura relativamente baixas, 100 mV s-1, somente foi possível devido às características superficiais intrínsecas do EDADB, o qual estabiliza o radical livre, R-NO2•‾, permitindo trabalhar numa ampla janela de potencial, sem haver perda do sinal de oxidação do R-NO2•‾. / The electrochemical behaviour of nitrofural (NFZ) at carbon paste, glassy carbon and highly boron doped diamond (HBDD) electrodes was studied in Britton- Robinson (BR) buffer using cyclic voltammetry. The best results were obtained at HBDD electrodes, in which it was possible to stabilize the radical anion (R-NO2•‾) in predominantly aqueous medium as well as to study the interaction between the radical anion and glutathione (GSH), L-cysteine (Cys), and O2. Only at HBDD surface, NFZ was directly reduced to the amine derivative (R-NH2) in the pH range of 2.02 to 4.03 in a process involving six (6.0 ± 0.4) electrons and six protons. In the range of pH 7.04 to 12 and, predominantly aqueous medium, the reduction step split into its two components: the reduction of NFZ to the nitro radical anion (R-NO2•‾) and reduction of R-NO2•‾ to hydroxylamine derivative (R--NHOH) in processes involving one and three (3.1 ± 0.1) electrons respectively. On the anodic scan of the cyclic voltammograms and at pH 8.06, it was observed the oxidation of the hydroxylamine to the nitroso derivative (R-NO), in a process involving 2 (1.7 ± 0.2) electrons and 2 protons. In addition and unreported in the literature on any electrode material, was the detection of a new oxidation peak at pH > 8.06, which was observed regardless of whether NFZ had been previously reduced or not. The calculation of n, number of electrons, involved in each electrochemical step was satisfactorily accomplished using the Randles-Sevcik equation. In presence of [Thiol] ≤ 3.7 x 10-2 mol L-1 NFZ is directly reduced to R-NO---Thiol adducts in an electrochemical process involving two electrons and two protons. On the other side, O2 acts as a R-NO2•‾ scavenger and the equilibrium constant for the electron transfer from nitro radical to oxygen, kO2, is 60. The process is catalytic and can be used to the analytical determination of NFZ in the range of 9.9 x 10-7 ≤ [NFZ] ≤ 1.1 x 10-5 mol L-1 at pH 8.0, with amperometric sensitivity of 2.2 x 104 µA L mol-1 cm-2 and detection limit of 3.4 x 10-7 mol L-1. The analytical parameters were similar to those obtained at pH 4.03 using the direct reduction of NFZ to the respective amine derivative in a process involving six electrons and six protons. The characterization of NFZ global reduction process in aqueous medium and at relative low scan rate, 100 mVs-1, was only possible due the intrinsic superficial characteristics of the HBDD electrode, which stabilize the R-NO2•‾ free radical, allowing to work in a large potential window, without losing the R-NO2•‾ oxidation signal.

La Résonance Magnétique Nucléaire RMN DOSY (Diffusion Ordered SpectroscopY) 1H est une technique permettant de mesurer les coefficients de diffusion des espèces en solution. Elle permet ainsi d’obtenir des informations structurales de par sa dimension RMN conventionnelle mais également des informations physico-chimiques telles que la taille de la (macro)molécule ou encore la formation d’agrégats. L’objectif de ces travaux a été d’examiner comment les méthodes de RMN (RMN 1D ,2D et DOSY), permettent de fournir des informations sur la caractérisation des matériaux polymères. La première partie de ce travail souligne l’intérêt de la RMN pour la caractérisation de la microstructure de quelques matériaux polymères en s’attardant plus particulièrement sur l’exploration de leurs extrémités de chaînes. La RMN s’est en effet révélée très performante pour établir le lien entre extrémité de chaîne et méthode de polymérisation appliquée. Le deuxième volet de ces travaux concerne l’étude de quelques architectures complexes (type dibloc et tribloc) et la mise en évidence du caractère contrôlé d’une copolymérisation radicalaire grâce à la RMN diffusionnelle. Nous avons également vu comment déterminer la masse moyenne en poids (Mw) par une simple mesure du coefficient d’autodiffusion. Finalement, l’ensemble des travaux présentés dans ce manuscrit a été valorisé grâce à l’étude des interactions par RMN diffusionnelle. Plus particulièrement la RMN DOSY 1H a permis d’observer des phénomènes de micellisation, d’agrégation, d’encapsulation et de relargage de molécules de principe actif. / Diffusion Ordered Spectroscopic Nuclear Magnetic Resonance (DOSY NMR 1H) makes it possible to determine physicochemical data such diffusion coefficients. While typical NMR data on the structure of molecules can be obtained, this technique also permits determinations of the physicochemical characteristics, such as molecular sizes or aggregate formation. The objective of this work was to examine how various NMR methodologies, specifically, 1 and 2 dimensional DOSY NMR, can be applied to the characterization of polymers. The first part of the manuscript relates the NMR characterization of some polymeric materials with a special interest in chain-end groups. In particular, we demonstrate the relationships between chain-ends and the polymerization methods employed. In a second part, the potential of the DOSY technique is used to determine controlled features of a radical polymerization resulting in di- and triblock architectures. Molecular weights are also determined by self-diffusion coefficient measurements. The results presented herein demonstrate that diffusional NMR can be effectively employed to understand interactions in polymeric structures. Indeed, DOSY 1H-NMR provides new interesting results concerning micellization, aggregation, encapsulation phenomena and the release of active molecules.

Electrolytes polymères gélifiés pour microbatteries au lithium / Gel polymer electrolytes for lithium microbatteriesChaudoy, Victor 15 November 2016 (has links)
Au cours de cette thèse, un nouvel électrolyte polymère gel pour la réalisation de microbatteries au lithium a été développé. Le gel a été préparé par « confinement » d’une phase de N-propyl-N-méthylpyrrolidinium bis(fluorosulfonyl)imide (P13FSI) et de LiTFSI dans un réseau semi-interpénétré (sRip) de polymère (PVdFHFP/ réseau de POE). L’électrolyte gel a tout d’abord été optimisé et étudié en termes de propriétés physicochimiques et de transport ionique en fonction de sa composition. Ensuite, des batteries Li/LiNi1/3Mn1/3Co1/3O2 ont été assemblées en utilisant l’électrolyte sRip. Les performances ont par ailleurs été comparées aux systèmes de références utilisant l’électrolyte à base de POE ou de PVdF-HFP. Outre ses propriétés améliorées par rapport au PVdF-HFP et au réseau de POE (propriétés mécaniques, confinement), l’électrolyte sRip est compatible avec le procédé de dépôt de l’électrode négative en lithium par évaporation sous vide. L’électrolyte sRip optimisé a donc été utilisé pour fabriquer une nouvelle génération de microbatteries en s’affranchissant de l’électrolyte céramique, le LiPON, afin d’abaisser la résistance interne. Les microbatteries Li/sRip gel/LiCoO2 délivrent une capacité nominale stable de 850 μAh à C sur 100 cycles à 25°C. / In this thesis, a new polymer gel electrolyte was prepared and optimized for Li based microbatteries. The gel consisted of an ionic liquid based phase (P13FSI/LiTFSI) confined in a semi-interpenetrating polymers (sIPN) network (PVdF-HFP/crosslinked PEO). sIPN electrolytes were prepared and optimized according to the PVdFHFP/ crosslinked PEO ratio and the liquid phase fraction. Furthermore, the sIPN electrolyte was used as an electrolyte in Li/LiNi1/3Mn1/3Co1/3O2 battery. The performances of the battery (specific capacity, efficiency, cyclability) were determined and compared to batteries using a crosslinked PEO or PVdF-HFP based gel. Such a thin and stable sIPN electrolyte film enabled the preparation of Li based microbatteries using thermal evaporation deposition of lithium directly conducted on the sIPN electrolyte film. This assembly (Li/sIPN) was therefore used to prepare a LiCoO2/sIPN gel/Li quasi solid-state microbattery. This microbattery showed a stable nominal capacity of 850 μAh for over 100 cycles of charge and discharge under 1 C rate at 25°C.

Réactions de carboalcénylation d'ène-carbamates et d'énamides : recherche de nouveaux processus radicalaires sans étain / Radical carbo-alkenylation reactions of ene-carbamates and enamides : research of new tin-free radical processesPoittevin, Clément 19 December 2014 (has links)
Les réactions multicomposants radicalaires sont d’une grande efficacité synthétique et s’avèrent en phase avec les principes de la chimie dite "verte". Ce manuscrit s’articule autour de trois axes : l’étude de la réaction de carbo-alcénylation radicalaire d’oléfines azotées riches en électrons, la valorisation des structures obtenues en se dirigeant vers la synthèse de molécules cycliques ou polycycliques complexes, et pour finir la mise au point de nouveaux médiateurs radicalaires sans étain. Dans un premiers temps, sur la base de travaux antérieurs de notre laboratoire, la réaction de carbo-alcénylation radicalaire a été étendue avec succès à l’utilisation d’énamides et d’ène-carbamates. Cette étude a permis de montrer la forte réactivité de ces oléfines vis-à-vis de radicaux électrophiles ainsi que l’importante force motrice que constitue la formation du radical amidoyle intermédiaire.Parmi tous les énamides et ène-carbamates testés, seules les structures cycliques ont permis d’obtenir une excellente diastéréosélectivité en faveur des produits trans. Dans un second temps, une valorisation des structures obtenues lors de la réaction de carbo-alcénylation a été réalisée par la mise au point de processus de cyclisations radicalaires et ioniques. Des cyclisations par activation via des énamines ont permis d’obtenir diastéréosélectivement des squelettes aza-décalones, présents notamment dans les alcaloïdes de la famille des lycopodiums. Un processus de double cyclisations de type 5-exo-trig/6-exo-trig, totalement diastéréosélectif, a également été établi donnant accès à des composés tri- et tétracycliques diversement substitués présents dans d’autres types d’alcaloïdes.Enfin, le développement de nouveaux médiateurs radicalaires silylés non réducteurs, comme substituts aux diétains, a été réalisé. L’intérêt s’est porté sur divers disilanes, thiosilanes, borosilanes et borothiosilanes, précurseurs de radicaux silyles permettant de promouvoir la propagation de chaînes radicalaires. Ces médiateurs ont été testés dans des réactions de vinylation directes de bromures d’alkyle ou dans des processus de carbo-alcénylation à trois composants. Dans le cas des thiosilanes et borosilanes des résultats encourageants et prometteurs ont été obtenus. / Multicomponent radical reactions have high synthetic efficiency and are in line with "green"chemistry concepts. This manuscript focuses on three topics: the study of the radical carboalkenylation reactions on electrons-rich olefins, the further elaboration of the structures directed towards the synthesis of complex cyclic or polycyclic molecules, and finally the development of newtin-free radical mediators.In the first part, based on previous work from our laboratory, the radical carbo-alkenylation reaction was successfully extended to enamides and ene-carbamates. This study demonstrated the high reactivity of these olefins towards electrophilic radical species and also the major driving force which constitutes the formation of the intermediate amidoyl radical. Amongst all enamides and enecarbamates tested, only the cyclic compounds led to a good diastereocontrol in the favor of transproducts.In a second part, the structures obtained in the carbo-alkenylation reaction were further elaborated through the development of radical and ionic cyclization processes. Activation via enamines allowed the diastereoselective formation of aza-decalin backbones, presents in the lycopodium alkaloid family. A totally diastereoselective double 5-exo-trig/6-exo-trig cyclization process was also achieved,generating various substituted tri- and tetracyclic compounds present in other classes of alkaloids. Finally, the development of new non-reductive free-radical silylated mediators as ditin surrogates was achieved. The research focused on various disilanes, thiosilanes, borosilanes and borothiosilanes,precursors of silyl radicals sustaining the radical chain. These mediators were tested in directvinylation reactions of alkyl bromides or in three components carbo-alkenylation processes. Forthiosilanes and borosilanes encouraging and promising results were obtained.

Nouvelles réactions radicalaires multicomposants : carbo-arylation, carbo-oximation, carbo-alcénylation d'oléfinesOvadia, Benjamin 19 December 2014 (has links)
L’objectif de ces travaux de thèse a consisté en le développement de nouveaux accepteurs sulfonylés pour l’élaboration de processus multicomposants radicalaires originaux. De nouvelles réactions de carbo-arylation, carbo-oximation et carbo-alcénylation d’oléfines ont ainsi été mises au point. Nous avons, dans un premier temps, étudié la faisabilité de nouvelles réactions de carbo-hétéroarylation basées sur deux modes d’activation des hétérocycles. Cette méthodologie s’est toutefois révélée être un réel défi en raison de la non-complémentarité des polarités du système, entrainant la formation de plusieurs réactions secondaires et des rendements assez faibles.Dans une seconde partie nous avons étudié le diastéréocontrôle lors de réactions de carbo-oximation d’allylsilanes et d’esters chiraux. Lors de ces processus, les produits de carbo-oximation sont obtenus avec de bons niveaux de diastéréocontrôle mais avec des rendements modestes en raison de l’encombrement stérique et de l’apparition de réactions secondaires. La configuration relative des diastéréoisomères majoritaires n’a pas encore pu être établie avec certitude, mais des modèles d’état de transition peuvent toutefois être proposés afin de prédire la stéréochimie la plus favorable.Dans le but de nous affranchir de l’utilisation d’une quantité stoechiométrique de diétain nous avons également développé de nouveaux accepteurs bi-fonctionnels capables d’agir à la fois comme accepteurs de radicaux nucléophiles et comme sources de radicaux électrophiles via l’α-scission du radical alkylsulfonyle généré. Nous avons mis au point une méthode de préparation de ces accepteurs extrêmement simple et efficace permettant l’accès à une large série de composés.Dans une dernière partie nous avons étendu les résultats obtenus lors des réactions de carbo-alcénylation en préparant de nouveaux accepteurs halogénés et sulfonylés très appauvris en électron par la présence de groupements électroattracteurs. Nous avons finalement réalisé un premier test concluant de carbo-alcénylation à partir d’un vinylsulfoxyde permettant d’envisager la réalisation de ces réactions en version énantiosélective à partir de vinylsulfoxydes énantioenrichis. / This thesis consists in the development of new sulfonylated acceptors for the elaboration of original free-radical multicomponent processes. Thereby, we have developed new free-radical carbo-arylation, carbo-oximation and carbo-alkenylation processes onto olefins.We have first studied the feasibility of new free-radical carbo-heteroarylation based on two different way of activation. This process proved to be very challenging due to the mismatch polarity between the different components, leading to the formation of side reactions and therefore relatively poor yield.In a second part, we examined the diastereocontrol arising from carbo-oximation of chiral allylsilanes and allylic esters. Carbo-oximation products are obtained with high level of selectivity, albeit in modest yields, due to steric hindrance and the formation of side-products. The relative configuration of the major diastereoisomer could not be established, but transition state models may be proposed to predict the most favorable stereochemistry. In order to overcome the use of stoechiometric amount of tin, we have developed new bi-functional reagents which can act both as a trap for nucleophilic radicals as well as a source of electrophilic radical via α-scission of generated alkylsulfonyl radicals. We have developed a very simple and efficient method for the preparation of these acceptors allowing an access to a wide range of compounds.In the final part, we extended the results obtained in the carbo-alkenylation reactions by preparing new activated halogenated and sulfonylated acceptors containing electron-withdrawing groups. We finally carried out a successful test on a carbo-alkenylation reaction using a vinylsulfoxide, offering an entry toward the development of such reactions in an enantioselective series starting from enantioenriched vinylsulfoxides.

Delayed Cell Death after Traumatic Brain Injury : Role of Reactive Oxygen SpeciesClausen, Fredrik January 2004 (has links)
<p>Traumatic brain injury (TBI) is a leading cause of death and disability TBI survivors often suffer from severe disturbances of cognition, memory and emotions. Improving the treatment is of great importance, but as of yet no specific neuroprotective treatment has been found. After TBI there are changes in ion homeostasis and protein regulation, causing generation of reactive oxygen species (ROS). Overproduction of ROS can lead to damage cellmembranes, proteins and DNA and secondary cell death. In the present thesis experimental TBI in rats were used to study the effects of the ROS scavengers α-phenyl-N-tert-butyl-nitrone (PBN) and 2-sulfophenyl-N-tert-butyl-nitrone (S-PBN) on morphology, function, intracellular signalling and apoptosis. </p><p>Posttreatment with PBN and S-PBN resulted in attenuation of tissue loss after TBI and S-PBN improved cognitive function evaluated in the Morris water maze (MWM). Pretreatment with PBN protected hippocampal morphology, which correlated to better MWM-performance after TBI.</p><p>To detect ROS-generation in vivo, a method using 4-hydroxybenzoic acid (4-HBA) microdialysis in the injured cortex was refined. 4-HBA reacts with ROS to form 3,4-DHBA, which can be quantified using HPLC, revealing that ROS-formation was increased for 90 minutes after TBI. It was possible to attenuate the formation significantly with PBN and S-PBN treatment. </p><p>The activation of extracellular signal-regulated kinase (ERK) is generally considered beneficial for cell survival. However, persistent ERK activation was found in the injured cortex after TBI, coinciding with apoptosis-like cell death 24 h after injury. Pretreatment with the MEK-inhibitor U0126 and S-PBN significantly decreased ERK activation and reduced apoptosis-like cell death. Posttreatment with U0126 or S-PBN showed robust protection of cortical tissue.</p><p>To conclude: ROS-mediated mechanisms play an important role in secondary cell death following TBI. The observed effects of ROS in intracellular signalling may be important for defining new targets for neuroprotective intervention.</p>
