Estudo genético quantitativo de uma população de ovelhas Santa Inês / Quantitative genetic study of a population Santa Ines sheep breedAguirre Riofrio, Edgar Lenin 05 August 2016 (has links)
É de grande importância estudar o potencial genético da raça Santa Inês, que cada vez mais está ocupando espaço, devido a sua alta resistência e capacidade de adaptação aos diversos ambientes. A informação consistiu de 37.735 dados de pedigree, 11.851 registros fenotípicos de cordeiros que foram usados para o estudo das características de desenvolvimento ponderal e 6.566 registros fenotípicos de 2.211 ovelhas, usados para a avaliação das características de eficiência reprodutiva. O período de análise foi de 12 anos (2003-2014), e as informações pertencentes a 33 rebanhos distribuídos em dez estados brasileiros. O banco de dados foi gerido através do programa Microsoft Visual FoxPro, a significância dos efeitos fixos a incluir nos modelos foi desenvolvida pelo programa estatístico R. Estimação dos componentes de variância, parâmetros e valores genéticos foram obtidos pelo método DFREML, utilizando o Programa MTDFREML em análise unicaracterística empregando o modelo animal. Foram avaliadas dez características ponderais: peso ao nascer (PN), peso ao desmama com 60 dias de idade (PD60), peso aos 180 dias de idade (P180), peso aos 270 dias de idade (P270), ganho diário de peso ao desmame (GPD), ganho diário de peso entre 60 e 180 dias (GPD180), ganho diário de peso entre 180 e 270 dias (GP180-270), ganho diário de peso entre 60 e 270 dias (GPD270), musculosidade da perna (MP) e presença de lã na pele (PL). Sete características reprodutivas: idade ao primeiro parto (IPP), intervalo de partos (IP), sobrevivência dos cordeiros ao desmame (SBV), número de cordeiros vivos ao parto/ovelha (NCP), número de cordeiros ao desmame/ovelha (NCD), peso total dos cordeiros ao parto/ovelha (PTCP) e peso total dos cordeiros ao desmame/ovelha (PTCD). Nas características de peso obteve-se um incremento nos valores dos componentes de variância conforme a idade do animal aumenta, enquanto que o efeito materno e as correlações genéticas direta-materna foram significativas no início e logo declinaram no pós-desmame, a influência do ambiente permanente materno (pe2) foi de magnitude alta (PN: 0,57, P60: 0,60, P180: 0,54 e GPD60: 0,48), provavelmente pelo sistema extensivo de pastagem que tem as mães, a herdabilidade total nas características avaliadas são de alta magnitude (PN: 0.38, PD60: 0.29, P180: 0.43, P270: 0.49, GPD: 0.48, GPD180: 0.43, GP180-270: 0.43, GPD270: 0.44, PL: 0.28, MP: 0.35), e as estimativas de repetitividade no desempenho das ovelhas (tm), são de alta intensidade para PN: 0.82, PD60: 0.8, P180: 0.7 e GPD: 0.61. Nas características reprodutivas encontrou-se uma notória variabilidade fenotípica e de ambiente permanente, as herdabilidades diretas foram 0.13, 0.04, 0.01, 0.12, 0.03, 0.16 e 0.18 para IPP, IP, SBV, NCP, NCD, PTCP e PTCD, respectivamente, o efeito do ambiente permanente foi de alta magnitude e variou de 0.79 a 0.97, enquanto que a grande variância ambiental e escassa variância residual das características conduzem a uma repetibilidade alta (entre 0.92 e 0.98). O ganho genético anual apresenta uma tendência irregular e de ligeiro incremento para as características ponderais, enquanto que as características reprodutivas apresentam uma tendência ligeiramente negativa em todas elas, exceto para IPP e SBV. Os resultados obtidos levam à conclusão que a seleção direta para características pre-desmama, levaram à seleção indireta para habilidade materna e, a seleção massal baseada sobre qualquer característica pós-desmama vai ter um rápido progresso genético, também as mudanças nas condições ambientais dos rebanhos, além da seleção direta para PTCP e PTCD, avaliadas ao primeiro parto, podem ser usadas como critério de seleção para a diminuição do intervalo geracional e a melhora da produtividade das ovelhas Santa Inês. / Studying genetic parameters and genetic changes in the Santa Ines sheep breed is important, as it is the most prevalent breed in Brazil and can adapt to many environments. This study included genetic data obtained from 37,735 pedigree records, of 11,851 performance records of lambs were used for the study of growth traits and 6,566 data of 2,211 registers of ewes were available to evaluate the reproductive traits. These data were collected over a period of 12 years (2003-2014) from 33 flocks in 10 Brazilian States. The database was kept and maintained using Microsoft Visual FoxPro 9.0 and the significance of fixed effects to include into the models was performed for the statistic program R. Variance components, genetic parameters and breeding values were estimated by the implementation of single-trait animal models via DFREML method, using MTDFREML software. Traits for growth as: birth weight (BW), weaning weight at 60 days of age (WW), weight at 180 days of age (W180), weight at 270 days of age (W270), average daily weight gain from birth at weaning (DGW), average daily weight gain from weaning to 6 months (DGWS), average daily weight gain from 6 months to 9 months (DGSN), average daily weight gain from weaning to 9 months (DGWN), the presence of hair in fur (HF) and leg muscularity (LM) were assessed. Reproductive traits of age at first lambing (AFL), lambing interval (LI), survivability (SU), litter size at birth (LSB), litter size at weaned (LSW), total litter weight at birth (TLWB) and total litter weight at weaning (TLWW) were also assessed. Variance components values increased in weight traits when lambs grew with age, while the maternal effect and genetic maternal-direct correlations were firstly significant which faded for post-weaning ages. Maternal permanent environment was of high magnitude (BW: 0.57, WW: 0.60, W180: 0.54 and DGW: 0.48), probably due to specific features involved in extensive livestock production systems where sheep were bred. High values were estimated for total heritability with respect to BW (0.38), WW (0.29), W180 (0.43), W270 (0.49), DGW (0.48), DGWS (0.43), DGSN (0.43), DGWN (0.44), HF (0.28), and LM (0.35), while the estimation of the repeatability of ewe performance was high for BW (0.82), WW (0.8), W180 (0.7) and DGW (0.61). For the reproductive traits, it was found pronounced phenotypic and permanent environment variances. The direct heritability estimates were 0.13, 0.04, 0.01, 0.12, 0.03, 0.16 and 0.18 for AFL, LI, SU, LSB, LSW, TLWB and TLWW, respectively. The estimated fractions of variance due to the permanent environmental effects in any traits were high magnitude (0.79 to 0.97), while the little residual variance that have the traits leading to a higher repeatability (0.92 to 0.98). The genetic trends observed for growth traits analyzed in this study were irregular and of slight increase, while by the reproductive traits has a slightly negative trend in all of them except for AFL and SU. Results obtained in the present study allow the conclusion that direct selection for pre-weaning traits conducted to an indirect selection for maternal ability. At the same time, mass selection based on post-weaning related traits would have faster results. Also, changes on environmental conditions of the flock, as well as the direct selection for TLWB and TLWW evaluated at first lambing, should be used as selection criteria for aiming reduction the generational interval and general improving the productivity of Santa Ines sheep.

Phenotypic and genetic evaluation of Fraser strain Arctic charr (Salvelinus alpinus) in brackish and freshwaterChiasson, Marcia 08 April 2013 (has links)
I examined phenotypic and genetic variation in growth traits in 30 families of commercial Fraser strain Arctic charr (Salvelinus alpinus) reared in freshwater (FRW) and brackish water (BRW) in Eastern Canada. I detected family by treatment interactions for all traits [body weight (BW), condition factor (K) and specific growth rate (SGR)] across all measurement dates and growth intervals, however, mean family BW in FRW was correlated phenotypically with BRW BW. In addition, FRW fish showed significantly greater survival than those transferred to BRW and fish which survived until the conclusion of the experiment were significantly heavier in BW at the baseline assessment than their full-sibs that died. These observations suggest that BW in FRW and BW in BRW should be analyzed as separate but correlated traits in Arctic charr breeding programs. I then tested the potential for genetic improvement in this species by calculating genetic parameters for BW and K, and tested if previously identified quantitative trait loci (QTL) for these traits were detectable across the broodstock. QTL with experiment-wide and chromosome-wide significance for body size and condition factor were detected on multiple linkage groups. Heritability for BW and K was moderate in FRW (0.29-0.38) but lower in BRW (0.14-0.17). Genetic correlations for BW across environments were positive and moderate (0.33-0.67), however equivalent K correlations were weaker (0.24-0.37). This information was then used to predict the rate of genetic change following one generation of selection for BW using phenotypic selection and genomic methodologies including marker-only selection and marker assisted selection. The greatest response in the rate of genetic change was achieved by selecting only from families in which significant BW QTL had been identified. As such, marker assisted selection showed the greatest gain in genetic response with 5.4% in FRW and 4.3% in BRW. These results have applications to commercial aquaculture as the Canadian aquaculture industry is attempting to diversify with alternative species. Such genetic improvement strategies will aid in developing a strain of Arctic charr characterised by increased BW. / Funding provided through the NSERC Strategic grants program. The effects of the synthetic strigolactone GR24 on Arabidopsis thaliana callus cultureMdodana, Ntombizanele Thobela 12 1900 (has links)
Thesis (MSc)--Stellenbosch University, 2012. / ENGLISH ABSTRACT: Plant growth promoting substances (PGPS) are emerging as useful tools in the
investigation of important plant growth traits. Two PGPS, smoke-water derived from
burning plant material and a synthetic strigolactone analogue, GR24, have been
reported to regulate a wide variety of developmental and growth processes in plants.
These PGPS are beginning to receive considerable attention in the area of improving
plant biomass yield and production. Variation in growth between plants is a major
impediment towards the complete understanding of the intrinsic processes that
control biomass production. Callus cultures of the model plant Arabidopsis thaliana
could overcome some of these hindrances. However, the suitability of these callus
cultures as a model system for plant biomass production must be established first.
This study aimed at using A. thaliana callus cultures as a platform to study the plant
growth promoting activities of smoke-water and GR24.
The first part of this study was conducted to develop an optimal protocol for inducing
A. thaliana callus formation. Wild-type A. thaliana Col-O, as well as strigolactone
deficient and insensitive mutants (max1-1, max2-1, max2-2, max3-9 and max4-1)
were cultured for callus induction. Hypocotyl and leaf explants were cultured onto
MS media supplemented with different hormone concentrations of 2,4-D and kinetin
(2:2 mg/L 2,4-D:kinetin and 0.5:0.05 mg/L 2,4-D:kinetin). Both media proved suitable
for callus induction of all genotypes, with max1-1 showing the highest efficiency
(83.33% and 92.22%) of callus induction. Calli were then used as a platform for
future investigations into the effects of smoke-water and GR24. Secondly, this study examined the effects of smoke-water and GR24 on wild-type A.
thaliana Col-O callus. Basic physiological studies were conducted to determine if
these two compounds would positively affect callus growth, as was shown in
previous studies using whole plants. Calli cultivated on MS media containing the two
different hormone concentrations were transferred onto the same fresh MS medium,
supplemented with either smoke-water or GR24. Growth promotion by smoke-water
and GR24 in calli was characterized by a significantly increased mass (biomass).
Calli were additionally transferred onto MS medium containing either auxin only or kinetin only and supplemented with GR24 or smoke-water. In the auxin only system,
increased mass was recorded for both GR24 and smoke-water treatments, while
these two compounds seemed to reduce growth in the kinetin only system. The
positive growth stimulatory effect observed for the auxin only system could be
attributed to the synergistic relationship between auxin and strigolactones, whilst the
reduced mass in the latter system could be due to the antagonistic interaction
between strigolactones and cytokinins.
Finally, this study has discovered a dual role of strigolactones in biomass
accumulation and adventitious root formation for Arabidopsis thaliana callus. On an
auxin- and cytokinin-free MS medium supplemented with GR24, calli of Arabidopsis
thaliana strigolactone deficient mutants (max1-1 and max4-1) and the wild-type Col-
O, but not the strigolactone response mutant (max2-2), showed enhanced biomass
accumulation. In addition to this, the max4-1 mutant and wild-type Col-O
demonstrated enhanced adventitious rooting, which was not apparent in max2-2.
Together these data suggested that the biomass accumulation and the adventitious
rooting activities of GR24 in Arabidopsis thaliana calli are controlled in a MAX2-
dependent manner. The interaction between strigolactone, auxin and cytokinin
signalling pathways in regulating these responses appears to be complex. Gene
expression profiling showed regulation of stress-related genes such as B-box
transcription factors, CALCINEURIN B-LIKE and RAP4.2 Genes encoding hormones
associated with stress (ABA, ethylene) and defence mechanisms (JA) were upregulated.
Expression of stress related genes indicated clues on some kind of stress
mediation that might be involved during the regulation of the rhizogenic response.
Conversely, smoke-water treatment could not enhance the biomass of the calli and
nor could it induce adventitious rooting in the absence of auxin and cytokinin. This
observation strongly emphasized the distinct roles of these two compounds, as well
as the importance of the interaction and ratio of auxin and cytokinin in callus growth.
This study has demonstrated a novel role of strigolactones in plant growth and
development, i.e. enhancement of biomass production in callus cultures. Secondly
the enhanced adventitious rooting ability is in agreement with recently published
literature on the role of strigolactones in regulating root architecture. In vitro callus
production is advantageous to plant sciences. It creates an opportunity for increasing plant material for cultivation and offers the use of cell cultures that accurately mimic
specific growth responses. It could greatly contribute to the study of intricate
regulatory and signalling pathways responsible for growth and development in
plants. Because the regulation of plant biomass production is very complex and the
molecular mechanisms underlying the process remain elusive, it is of paramount
importance that further work be done in order to gain more in-depth insights and
understanding of this aspect and subsequently improve efficiency and returns when
applying biotechnology tools on commercially important crop plants. / AFRIKAANSE OPSOMMING: Verbindings wat plantgroei bevorder (PGBV) het as nuttige alternatief ontstaan om
plant groei te ondersoek. Rook-water, afkomstig van verbrande plant material, en ‘n
sintetiese strigolaktoon analoog, GR24, wat ‘n α, β-onversadigde furanoon
funksionele groep in gemeen het, is vir die regulering van ‘n wye verskeidenheid
ontwikkelings- en groei prosesse in plante verantwoordelik. Tans ontvang hierdie
PGBVs aansienlik aandag in die area van die verbetering van plant biomassa
opbrengs en -produksie. Die variasie in groei tussen plante is ‘n groot hindernis om
die intrinsieke prosesse wat biomass produksie beheer, volledige te verstaan. Deur
gebruik te maak van kallus kulture van die model plant Arabidopsis thaliana kan van
hierdie hindernisse oorkom word. Tog moet die geskiktheid van kallus kulture as ‘n
model sisteem vir plant groei biomass produksie eers gevestig word. Die doel van
hierdie studie was om A. thaliana kallus kulture as ‘n platform vir die studie van die
plantgroei bevorderingsaktiwiteite van rook-water en GR24 te gebruik.

Laastens het hierdie studie het ‘n gelyktydige rol van strigolaktone vir biomassa
akkumulasie en bywortelvorming in Arabidopsis thaliana kallus ontdek. Kallus van
A. thaliana strigolaktoon afwesige mutante (max1-1 en max4-1) en die wilde tipe

uitsonderlike rol van die twee verbindings, asook die belang van die interaksie en
verhouding van ouksien en sitokinine vir die groei van kallus. Hierdie studie toon op ‘n nuwe rol van strigolaktoon in plant groei en ontwikkeling,
d.w.s die verhoogde biomassa produksie in kallus kulture. Tweedens, die
verhoogde bywortelvormingsvermoë is in ooreenstemming met literatuur wat
onlangs gepubliseer is i.v.m die rol van strigolaktone in die regulering van wortel
argitektuur. Die in vitro produksie van kallus is voordelig in plant wetenskappe. Dit
skep ‘n geleentheid vir die vermeerdering van plant materiaal vir kultivering en bied
die gebruik van selkulture wat spesifieke groei reaksies op ‘n merkwaardige wyse
akkuraat namaak. Dit kan grootliks bydra tot die studie van die delikate
regulatoriese en sein transduksie paaie wat vir groei en ontwikkeling van plante
verantwoordelik is. Aangesien die regulering van plant biomassa produksie baie
kompleks is en die molekulêre meganismes vir die proses onbekend bly is dit van
grootskaalse belang dat meer werk gedoen word om ‘n meer in diepte insig en
kennis van die aspekte en gevolglike verbetering van effektiwiteit en wins te kry deur
die toepassing van biotegnologiese metodes op die gewas plante wat van
kommersiêle belang is.

However, most of these studies use information related to the body weight on specific ages or average weight gain. Thus, it becomes interesting to evaluate all the growth information with the focus on longitudinal data simultaneously. This study aims to use random regression models (RRM) in order to detect QTL (quantitative trait loci) for growth traits in swine from a population F2 Piau x Commercial. To this end, it was considered a RRM with random polygenic effects, permanent environment and QTL, being the covariance matrix associated with the latter known as IBD (identical-by-descent). The presence of a significant QTL was found by likelihood ratio test considering the model described above as being the complete and this same without the QTL effect as null model. Comparisons between these models were made at the positions of the markers (6 microsatellite markers) and at positions between markers. In the position with greater evidence of QTL effect was calculated the heritability and the genetic polygenic and QTL values for body weight throughout the growth period studied. The results showed a significant effect of QTL at position 65 of chromosome 7. / Muitos estudos são voltados para a busca de QTL (locos de características quantitativas) que afetam características de crescimento em suínos, sendo que a quase totalidade tem encontrado QTL com efeitos significativos. No entanto, a maioria destes estudos utiliza informações referentes a peso corporal em idades específicas ou ganho de peso médio. Dessa forma, torna-se interessante avaliar simultaneamente todas as informações de crescimento sob o enfoque de dados longitudinais. O presente trabalho tem como objetivo utilizar modelos de regressão aleatória (MRA) com o intuito de detectar QTL para características de crescimento em suínos de uma população F2 Piau x Comercial. Para tanto, considerou-se MRAs com efeitos aleatórios poligênico, de ambiente permanente e de QTL, sendo a matriz de covariância associada a este último denominada de IBD (identical-by-descent). A presença de um QTL com efeito significativo foi verificada mediante teste de razão de verossimilhanças considerando o modelo descrito como sendo o completo e o mesmo sem o efeito de QTL como sendo o modelo nulo. As comparações entre estes modelos foram realizadas nas posições dos marcadores (6 marcadores microssatélites) e nas posições entre marcadores. A informação consistiu de 37.735 dados de pedigree, 11.851 registros fenotípicos de cordeiros que foram usados para o estudo das características de desenvolvimento ponderal e 6.566 registros fenotípicos de 2.211 ovelhas, usados para a avaliação das características de eficiência reprodutiva. O período de análise foi de 12 anos (2003-2014), e as informações pertencentes a 33 rebanhos distribuídos em dez estados brasileiros. O banco de dados foi gerido através do programa Microsoft Visual FoxPro, a significância dos efeitos fixos a incluir nos modelos foi desenvolvida pelo programa estatístico R. Estimação dos componentes de variância, parâmetros e valores genéticos foram obtidos pelo método DFREML, utilizando o Programa MTDFREML em análise unicaracterística empregando o modelo animal. Foram avaliadas dez características ponderais: peso ao nascer (PN), peso ao desmama com 60 dias de idade (PD60), peso aos 180 dias de idade (P180), peso aos 270 dias de idade (P270), ganho diário de peso ao desmame (GPD), ganho diário de peso entre 60 e 180 dias (GPD180), ganho diário de peso entre 180 e 270 dias (GP180-270), ganho diário de peso entre 60 e 270 dias (GPD270), musculosidade da perna (MP) e presença de lã na pele (PL). Sete características reprodutivas: idade ao primeiro parto (IPP), intervalo de partos (IP), sobrevivência dos cordeiros ao desmame (SBV), número de cordeiros vivos ao parto/ovelha (NCP), número de cordeiros ao desmame/ovelha (NCD), peso total dos cordeiros ao parto/ovelha (PTCP) e peso total dos cordeiros ao desmame/ovelha (PTCD). Nas características de peso obteve-se um incremento nos valores dos componentes de variância conforme a idade do animal aumenta, enquanto que o efeito materno e as correlações genéticas direta-materna foram significativas no início e logo declinaram no pós-desmame, a influência do ambiente permanente materno (pe2) foi de magnitude alta (PN: 0,57, P60: 0,60, P180: 0,54 e GPD60: 0,48), provavelmente pelo sistema extensivo de pastagem que tem as mães, a herdabilidade total nas características avaliadas são de alta magnitude (PN: 0.38, PD60: 0.29, P180: 0.43, P270: 0.49, GPD: 0.48, GPD180: 0.43, GP180-270: 0.43, GPD270: 0.44, PL: 0.28, MP: 0.35), e as estimativas de repetitividade no desempenho das ovelhas (tm), são de alta intensidade para PN: 0.82, PD60: 0.8, P180: 0.7 e GPD: 0.61. Nas características reprodutivas encontrou-se uma notória variabilidade fenotípica e de ambiente permanente, as herdabilidades diretas foram 0.13, 0.04, 0.01, 0.12, 0.03, 0.16 e 0.18 para IPP, IP, SBV, NCP, NCD, PTCP e PTCD, respectivamente, o efeito do ambiente permanente foi de alta magnitude e variou de 0.79 a 0.97, enquanto que a grande variância ambiental e escassa variância residual das características conduzem a uma repetibilidade alta (entre 0.92 e 0.98). O ganho genético anual apresenta uma tendência irregular e de ligeiro incremento para as características ponderais, enquanto que as características reprodutivas apresentam uma tendência ligeiramente negativa em todas elas, exceto para IPP e SBV. Os resultados obtidos levam à conclusão que a seleção direta para características pre-desmama, levaram à seleção indireta para habilidade materna e, a seleção massal baseada sobre qualquer característica pós-desmama vai ter um rápido progresso genético, também as mudanças nas condições ambientais dos rebanhos, além da seleção direta para PTCP e PTCD, avaliadas ao primeiro parto, podem ser usadas como critério de seleção para a diminuição do intervalo geracional e a melhora da produtividade das ovelhas Santa Inês. / Studying genetic parameters and genetic changes in the Santa Ines sheep breed is important, as it is the most prevalent breed in Brazil and can adapt to many environments. This study included genetic data obtained from 37,735 pedigree records, of 11,851 performance records of lambs were used for the study of growth traits and 6,566 data of 2,211 registers of ewes were available to evaluate the reproductive traits. These data were collected over a period of 12 years (2003-2014) from 33 flocks in 10 Brazilian States. The database was kept and maintained using Microsoft Visual FoxPro 9.0 and the significance of fixed effects to include into the models was performed for the statistic program R. Variance components, genetic parameters and breeding values were estimated by the implementation of single-trait animal models via DFREML method, using MTDFREML software. Traits for growth as: birth weight (BW), weaning weight at 60 days of age (WW), weight at 180 days of age (W180), weight at 270 days of age (W270), average daily weight gain from birth at weaning (DGW), average daily weight gain from weaning to 6 months (DGWS), average daily weight gain from 6 months to 9 months (DGSN), average daily weight gain from weaning to 9 months (DGWN), the presence of hair in fur (HF) and leg muscularity (LM) were assessed. Reproductive traits of age at first lambing (AFL), lambing interval (LI), survivability (SU), litter size at birth (LSB), litter size at weaned (LSW), total litter weight at birth (TLWB) and total litter weight at weaning (TLWW) were also assessed. Variance components values increased in weight traits when lambs grew with age, while the maternal effect and genetic maternal-direct correlations were firstly significant which faded for post-weaning ages. Maternal permanent environment was of high magnitude (BW: 0.57, WW: 0.60, W180: 0.54 and DGW: 0.48), probably due to specific features involved in extensive livestock production systems where sheep were bred. High values were estimated for total heritability with respect to BW (0.38), WW (0.29), W180 (0.43), W270 (0.49), DGW (0.48), DGWS (0.43), DGSN (0.43), DGWN (0.44), HF (0.28), and LM (0.35), while the estimation of the repeatability of ewe performance was high for BW (0.82), WW (0.8), W180 (0.7) and DGW (0.61). For the reproductive traits, it was found pronounced phenotypic and permanent environment variances. The direct heritability estimates were 0.13, 0.04, 0.01, 0.12, 0.03, 0.16 and 0.18 for AFL, LI, SU, LSB, LSW, TLWB and TLWW, respectively. The estimated fractions of variance due to the permanent environmental effects in any traits were high magnitude (0.79 to 0.97), while the little residual variance that have the traits leading to a higher repeatability (0.92 to 0.98). The genetic trends observed for growth traits analyzed in this study were irregular and of slight increase, while by the reproductive traits has a slightly negative trend in all of them except for AFL and SU. Results obtained in the present study allow the conclusion that direct selection for pre-weaning traits conducted to an indirect selection for maternal ability. At the same time, mass selection based on post-weaning related traits would have faster results. Also, changes on environmental conditions of the flock, as well as the direct selection for TLWB and TLWW evaluated at first lambing, should be used as selection criteria for aiming reduction the generational interval and general improving the productivity of Santa Ines sheep.

Previous issue date: 2012-12-27 / The goals of this thesis were to verify the conservation genetic status of the Segureño Spanish sheep breed and to evaluate the principal factors involved in the determination of growth traits in this breed. The data used n this study was provided by the Asociación Nacional de Criadores de Ovino Segureño – ANCOS, located in Granada province, Spain. The information about genetic variability and population structure were obtained using genealogical records from 37.459 animals registered between the years 1984 – 2007. To evaluate the inbreeding effects on growth traits, weight measurements taken between the years 1992 – 2007 were considered. The traits evaluated were: birth weight (BW), weight at 30 (W30), 45 (W45) and 75 (W75) days of age and also the average daily gains between 0-45 (ADG0-45), 0-75 (ADG0-75) and 45-75 (ADG45-75) days of age. The parameters used to evaluate the genetic diversity were: effective number of founders , effective number of ancestors , effective population size , inbreeding coefficient , average relatedness coefficient , generation interval , and the fixation indexes . To analyze the effect of the inbreeding on growth traits, were considered the individual inbreeding estimates and the environmental effects of sex, year-season-herd, birth type and age of the ewe. For the herds studied, a significant loss of genetic variability was not observed. The fixation indexes suggest that there is no population subdivision within the breed and the inbreeding level can be considered satisfactory for growth traits. Nucleus herds were not observed which may have contributed for the absence of within breed substructure. Among the environmental factors considered, the contemporary group (formed by individuals born in the same year-season-herd), the birth type and the sex of the lamb were the most important factors affecting the weights and average weight gains studied. / Objetivou-se, com esse trabalho, verificar o estado de conservação genética da raça espanhola de ovinos Segureño e avaliar os principais fatores envolvidos na determinação das características de crescimento em cordeiros da raça. Os dados usados neste estudo são provenientes da Asociación Nacional de Criadores de Ovino Segureño- ANCOS, sediada na província de Granada-Espanha. As informações sobre a variabilidade genética e a estrutura populacional foram obtidas utilizando as informações genealógicas de 37.459 animais registrados entre os anos de 1984 a 2007. Para avaliação dos efeitos da endogamia sobre o crescimento dos cordeiros, foram utilizadas as informações de pesagens de cordeiros, entre os anos de 1992 a 2007, e avaliados os pesos ao nascer (PN), aos 30 dias (P30), aos 45 dias (P45) e aos 75 dias (P75) de idade, além dos ganhos médios medidos entre 0-45, 0-75, e 45-75 dias de idade. Os parâmetros utilizados na avaliação da diversidade genética foram: número efetivo de fundadores , número efetivo de ancestrais , tamanho efetivo (Ne), coeficiente de consanguinidade (F), coeficiente de parentesco médio (AR), intervalo de gerações (IG) e índices de fixação ou estatística-F . Para análise dos efeitos dos níveis de endogamia sobre as características de crescimento, foram considerados os valores de endogamia individual e os fatores ambientais de sexo, ano-estação-rebanho, tipo de nascimento e idade da ovelha. Não foram observadas perdas de variabilidade genética significativas nos rebanhos estudados. Os índices de fixação indicam que não está ocorrendo subdivisão dentro da raça e os níveis de endogamia para as características de crescimento são considerados satisfatórios. Não foram identificados rebanhos núcleo, fato que pode ter contribuído para ausência de subestruturação dentro da raça. Entre os fatores ambientais considerados, o grupo de contemporâneos, formado por animais nascidos no mesmo ano-estação-rebanho, o tipo de nascimento e o sexo do cordeiro foram os que mais influenciaram os pesos e os ganhos médios de peso estudados.

A mixed model evaluation for growth and reproduction parameters in a SA mutton merino flockHartman, Carina 24 January 2003 (has links)
Records of the La Rochelle S.A. Mutton Merino stud were analysed to determine the importance of possible contributing non-genetic sources of variation on growth and reproductive traits. Growth traits analysed were pre-weaning weight (42dW), weaning weight (100dW), 205d weight, ADG's and Kleiber Ratio's (KR) for both 42d and 100d. All the included effects, i.e. year, sex, birth status, season, age of dam and the year x sire interaction, were highly significant (P<0.001) for the growth traits. Reproduction traits measured were mean number of lambs born per ewe per year, mean Kg lamb weaned per ewe per year, percentage twins and fecundity. Year was the only significant effect (P<0.001) for all four reproduction traits. Data of records between 1974 and 1995 were used to estimate additive direct and additive maternal heritabilities for seven growth traits. The estimates of the direct heritabilities varied between 0.166 for 42dW and 0.513 for 205dW. The estimates of the maternal heritabilities were within the 0.042 (42dW) to 0.370 (100dADG) range. Estimates of the covariance between the direct additive and maternal additive components were generally negative and of a substantial size (-0.215 to -0.674). Additive direct and additive maternal heritabilities for four reproductive traits were also estimated. The estimates for direct additive heritabilities varied between 0.039 for fecundity and 0.177 for Kg lamb weaned per ewe per year. Estimates for the maternal additive heritabilities were correspondingly low, varying from 0.007 for lambs born per ewe per year to 0.197 for percentage twins. Estimates of covariance between the additive direct and additive maternal components were negative. Genetic trends of the flock were studied. The main objective of the breeder was to improve the reproductive efficiency of the flock, whilst decreasing the incidence of very large lambs. Selection pressure on reproductive performance hasn't yet attained the desired effect as three of the four reproductive traits measured, showed very little or no improvement. O banco de dados utilizado nas análises constituiu-se de 39.290 animais no arquivo de pedigree e de 24.353 animais no arquivo de produção. Os parâmetros populacionais foram obtidos por meio dos softwares POPREP (GROENEVELD et al., 2009) e ENDOG v 3.2 (GUTIÉRREZ e GOYACHE, 2005). As estatísticas descritivas e os parâmetros genéticos para cada característica estudada foram estimadas por meio de quatro análises hepta-característica utilizando o programa VCE-6 (KOVAC & GROENEVELD, 2003) sob modelo animal completo. As tendências genéticas e fenotípicas foram obtidas pela regressão dos valores genéticos e fenotípicos respectivamente sobre o ano de nascimento dos animais e os coeficientes da regressão foram estimados por meio do método de quadrados mínimos. Apesar da endogamia média do rebanho ser considerada moderada verificou-se alta porcentagem de indivíduos endogâmicos e reduzido tamanho efetivo populacional. Além disso, o incremento contínuo de endogamia ao longo dos anos demonstra a necessidade de intervenção na seleção dos indivíduos para reprodução, de modo que, problemas futuros possam ser evitados. Os coeficientes de herdabilidade direta foram estimados em 0,24, 0,31, 0,31, 0,21, 0,19, 0,30, 0,41, 0,19 e 0,17 respectivamente para peso ao nascimento (PN), peso aos 120 dias de idade (P120), peso à desmama (PD), peso ao ano (PES12), ganho de peso pós desmama, de 205 aos 550 dias (GP345), ganho de peso na prova à pasto da ABCZ, em 224 dias (GP224), perímetro escrotal (PE), temperamento (TEMP), e repelência (REP) e indicam que a variabilidade genética aditiva existente é suficiente para a obtenção do ganho genético em resposta à seleção desde que sejam realizados ajustes, tanto nas estratégias de seleção adotadas para a obtenção de maiores ganhos nos valores genéticos, como também nos fatores ambientais que possam possibilitar ao máximo, a expressão do potencial genético dos animais. / The present study had as objective to evaluate the population structure, to estimate parameters (coefficients of heritability and correlation) and to determine the phenotypic and genetic trends for growth and adaptability traits in Nellore cattle, Lemgruber line. The database used in the analysis consisted of 39,290 animals in the pedigree and 24,353 animals in the production file. The population parameters were obtained from the software POPREP (Groeneveld et al., 2009) and ENDOG v 3.2 (Gutierrez and GOYACHE, 2005). The descriptive statistics and genetic parameters for each characteristic studied were estimated by four seven-traits analysis using the program VCE-6 (Kovac & GROENEVELD, 2003) which uses the animal model. The phenotypic and genetic trends were obtained by means of phenotypic and genetic values respectively on the year of birth of the animals and the regression coefficients were estimated by the method of least squares. Although the average inbreeding of the herd being considered moderate, there was high percentage of inbred individuals and small effective population size. Furthermore, the continuous increase in inbreeding over the years demonstrates the need for intervention in the selection of individuals for reproduction, so that future problems can be avoided. Direct heritability coefficients were estimated as 0.24, 0.31, 0.31, 0.21, 0.19, 0.30, 0.41, 0.19 and 0.17 respectively for birth weight (PN), weight 120 days of age (P120), weaning weight (PD), weight at 12 months of age (PES12), weight gain after weaning from 205 to 550 days (GP345), weight gain during the pasture test of ABCZ in 224 days (GP224), scrotal circumference (PE), docility (TEMP) and repellency (REP) and indicate that the additive genetic variability is sufficient to obtain genetic gain in response to selection as far as adjustments in selection strategies are adopted to achieve larger gains in breeding values, and environmental factors that may allow the expression of the genetic potential of the animals.

Stock improvement of giant freshwater prawn (Macrobrachium rosenbergii) in Vietnam: Experimental evaluations of crossbreeding,the impact of domestication on genetic diversity and candidate genesThanh Nguyen Unknown Date (has links)
Aquaculture plays an important role in economic development and food security in many countries in the world. World aquaculture production in 2006 was 51.7 million tonnes with an estimated value of US$ 78.8 billion (FAO, 2009). World production will need to increase however by 30-40 million tonnes from its current production level by 2030 to meet growing global demand for fish. In this context, aquaculture in Vietnam has developed rapidly over the past decade and the fisheries sector ranked fourth in terms of export value in 2008 (Vietnamnet, 2008). Total fisheries production in Vietnam in 2007 was 4.149 million tonnes, of which fisheries production from catch and aquaculture were 2.064 and 2.085 million tonnes, respectively. A variety of aquatic species are cultured in Vietnam, but shrimps (mainly Black Tiger shrimp Penaeus monodon, and Pacific white shrimp Litopenaeus vannamei) and ‘tra’ or ‘basa’ catfish are the most common species used in aquaculture. The giant freshwater prawn (GFP), Macrobrachium rosenbergii, is one of the most important crustacean species in inland aquaculture in many countries across the world where this species is either native or exotic. GFP is suitable for culture in a variety of farming systems, including monoculture or polyculture in ponds, pens, and integrated or rotational rice-prawn culture models. The GFP industry worldwide relies totally on wild or unimproved stocks, a practice that threatens the long-term sustainability of GFP farming due to low productivity and vulnerability of farmed stocks to disease. The current status of GFP aquaculture highlights the need for initiation of a systematic stock improvement program for the species to improve economically important traits. Large-scale selective breeding programs have been instigated for some finfish, salmonids and GIFT tilapia for example, and some selective breeding trials have been conducted on crustacean species, namely marine penaeid shrimp and freshwater crayfish. Examples of selective breeding programs on aquatic species have demonstrated that significant genetic gains can be achieved for growth rates with gains of around 10-20% per generation. While a selective breeding program is an option for GFP stock improvement, an alternative approach to improving GFP productivity, potentially with more immediate effect and one that is less expensive, is crossbreeding which may produce heterosis or hybrid vigour in crossbred offspring. Therefore, a crossbreeding strategy was trialed in the current study as a starting point for a stock improvement program for the GFP industry in Vietnam. The current study assessed the growth performance of three GFP strains (two wild Vietnamese strains from the Dong Nai and Mekong rivers, and a single domesticated Hawaiian strain) and their reciprocal crosses in a complete 3x3 diallel cross, i.e. three purebred and six crossbred strains. The diallel cross was carried out over two consecutive generations (G1 and G2). Juveniles for the experiments were produced using single-pair matings. Juveniles from each strain combination were stocked into three replicate hapas for 15 weeks. Growth data (body weight, carapace length, standard length) from the G1 and G2 were pooled for all subsequent analyses as there was no effect of generation on growth traits. Results showed that the Hawaiian strain performed best among purebred strains, and crosses with the Dong Nai or Mekong strains as dams and the Hawaiian strain as sires grew significantly faster than did the purebred Dong Nai or Mekong strains. These results suggest potential for heterosis among some crosses. Growth data were analyzed in depth by partitioning the strain combination (cross) effect into three components: strain additive genetic effects, heterotic effects, and strain reciprocal effects. Strain additive genetic and reciprocal effects were significant sources of variation for all growth traits measured. Strain additive genetic effects were highest for the Hawaiian strain and lowest for the Mekong strain for all growth traits. Reciprocal effects influenced negatively on growth rate of crosses with the Hawaiian (H) strain as dams and the Dong Nai (D) or Mekong (M) as sires compared with their reciprocal crosses (DH and MH). Heterotic effects for all growth traits were small and not significantly different from zero (P > 0.05). These results indicate that a crossbreeding approach based on the strains evaluated here provides only limited potential for improving growth rates based simply on heterotic outcomes and that a likely more productive option would be to trial artificial selection on a diverse synthetic stock. The current study also employed genetic markers (microsatellites) to characterize levels and patterns of genetic diversity in three purebred strains of GFP that originated from the diallel cross above. All three purebred strains showed relative high levels of genetic diversity in terms of allele number and individual heterozygosity across the six marker loci screened. Levels of genetic diversity present in the three purebred strains combined into a single stock were compared with that from a combination of three wild river stocks to assess the impact of domestication on genetic diversity of a ‘synthetic’ population. Results demonstrated that there was no significant loss of genetic diversity in the three purebred strains combined compared with a reference set containing the three wild populations. Therefore, a synthetic population formed from these purebred strains successfully captured the majority of genetic variation present in the wild broodstock. This synthetic population provides a potential stock for a future selective breeding program for GFP in Vietnam. The current study was also the first attempt to identify single nucleotide polymorphisms (SNPs) in key growth genes in GFP. Two key candidate genes were targeted, actin and crustacean hyperglycemic hormone (CHH), that are potentially linked to growth performance in GFP. The study screened SNPs in GFP females only, because growth performance of GFP males is influenced strongly by social rank. The study identified four SNPs in intron 3 of the CHH gene that were significantly correlated with individual body weight at harvest, while no SNPs detected in the actin gene were associated with growth traits in GFP. This finding however, needs to be confirmed using larger sample sizes and other GFP lines. The current study has produced important basic knowledge relevant to implementation of an effective stock improvement program for GFP in Vietnam. Results indicate that a selective breeding strategy rather than a crossbreeding approach is likely to be the best strategy for improving GFP culture stocks in Vietnam. In addition, the study demonstrates that application of modern molecular genetic technologies can be efficient in developing a genetically diverse, synthetic population for stock improvement and for identifying potential markers correlated with important commercial traits in GFP. Integration of DNA techniques with traditional breeding practices can facilitate GFP stock improvement in Vietnam and accelerate the industry development when improved lines are available. Some limitations of the current study and recommendations for further work are discussed.

Os coeficientes de herdabilidade direta foram estimados em 0,24, 0,31, 0,31, 0,21, 0,19, 0,30, 0,41, 0,19 e 0,17 respectivamente para peso ao nascimento (PN), peso aos 120 dias de idade (P120), peso à desmama (PD), peso ao ano (PES12), ganho de peso pós desmama, de 205 aos 550 dias (GP345), ganho de peso na prova à pasto da ABCZ, em 224 dias (GP224), perímetro escrotal (PE), temperamento (TEMP), e repelência (REP) e indicam que a variabilidade genética aditiva existente é suficiente para a obtenção do ganho genético em resposta à seleção desde que sejam realizados ajustes, tanto nas estratégias de seleção adotadas para a obtenção de maiores ganhos nos valores genéticos, como também nos fatores ambientais que possam possibilitar ao máximo, a expressão do potencial genético dos animais. / The present study had as objective to evaluate the population structure, to estimate parameters (coefficients of heritability and correlation) and to determine the phenotypic and genetic trends for growth and adaptability traits in Nellore cattle, Lemgruber line. The database used in the analysis consisted of 39,290 animals in the pedigree and 24,353 animals in the production file. The population parameters were obtained from the software POPREP (Groeneveld et al., 2009) and ENDOG v 3.2 (Gutierrez and GOYACHE, 2005). The descriptive statistics and genetic parameters for each characteristic studied were estimated by four seven-traits analysis using the program VCE-6 (Kovac & GROENEVELD, 2003) which uses the animal model. The phenotypic and genetic trends were obtained by means of phenotypic and genetic values respectively on the year of birth of the animals and the regression coefficients were estimated by the method of least squares. Although the average inbreeding of the herd being considered moderate, there was high percentage of inbred individuals and small effective population size. Furthermore, the continuous increase in inbreeding over the years demonstrates the need for intervention in the selection of individuals for reproduction, so that future problems can be avoided. Direct heritability coefficients were estimated as 0.24, 0.31, 0.31, 0.21, 0.19, 0.30, 0.41, 0.19 and 0.17 respectively for birth weight (PN), weight 120 days of age (P120), weaning weight (PD), weight at 12 months of age (PES12), weight gain after weaning from 205 to 550 days (GP345), weight gain during the pasture test of ABCZ in 224 days (GP224), scrotal circumference (PE), docility (TEMP) and repellency (REP) and indicate that the additive genetic variability is sufficient to obtain genetic gain in response to selection as far as adjustments in selection strategies are adopted to achieve larger gains in breeding values, and environmental factors that may allow the expression of the genetic potential of the animals.
