Aplikace pokročilých přístrojových technik pro analýzu polutantů životního prostředí

Increasing of human life comfort and safety requires development and application of new chemical substances, which, unfortunately, cannot be completely removed by WWTPs. Thus, these compounds, their metabolites and TPs formed during human/animals/bacteria metabolism, wastewater treatment and natural environmental processes are tend to contaminate all the environmental compartments. Appearance of advanced analytical instrumentation and techniques enable to perform research on emerging environment contaminants fate. This thesis was focused on application of high resolution mass spectrometer and new generation of triple quadrupole for analysis of the environmental pollutants. All studies were mainly devoted to such a group of contaminants as pharmaceuticals because of their high consumption, frequent occurrence in all environmental matrices and potential to cause adverse effects for non-targeted organism due to their biological activity. The first part of the thesis is related to method development for identification and quantification of metabolites in the different tissues of rainbow trout exposed to diltiazem. As a result, 17 diltiazem metabolites were tentatively identified by HRMS and spectral interpretation software. The complex metabolic pathway in metabolic phase I for diltiazem and tissue specific bioconcentration in fish were proposed in the study. The second part of the work is mainly focused on investigation of pharmaceuticals fate in the soil environment. The transformation of several pharmaceuticals, such as atenolol, metoprolol, carbamazepine, sulfamethoxazole, trimethoprim, and clindamycin in 13 different soils was studied under laboratory conditions. The parent compounds were analyzed in all studied soils and dissipation curves were plotted. Finally, 15 known and new metabolites were detected and identified. Moreover, identity of 6 of them was confirmed by available analytical standards. It was shown that almost the same metabolites profile was observed in all soils, but formed amount and degradation rate of all detected metabolites were soil type dependent. Several parent compounds and metabolites were shown to be persistent in the soils. Consequently, pharmaceuticals can be transported via ground water to plant. Further fate of pharmaceuticals must be studied as it can have adverse effect on crop consumers. The third part of the dissertation dealt with an important issue of pharmaceuticals transport between different aquatic environmental compartments. Water, sediments and fish from a pond affected with TWW from a local WWTP were analyzed within this project. 18 pharmaceuticals and 7 their metabolites were found in all studied matrices. Solid water distribution coefficients for sediments and bioaccumulation factors for fish liver were obtained in this study under real conditions. Seasonal variations in all studied parameters were investigated for one year period. This study help to extend a limited available information on fate of wide range of compound, especially metabolites in the aquatic environment affected by treated wastewater.

Abstract/Summary The discovery of a cadaver or skeletal remains (hereinafter the "Remains") raises a number of questions, one of which is the matter of the identity of the remains. The answer to this particular question can be found through forensic identification and utilization of one of the methods thereof. The aim of my thesis is to provide a brief overview of the methods which are currently most commonly used for the purposes of identifying Remains and introduce the reader to their basics, options and limitations. In order to ensure a better understanding of the subject, I have included a section concerning the Remains and the post-mortem changes thereof. In this thesis, I will focus only on those parts of scientific fields, which are relevant for identification of the Remains, and, similarly with respect to information systems, I target only the information being collected and analyzed by such systems in cases of Remains whose identity is not know. In this thesis, I have chosen to proceed from general terms, division of forensic identification, activities being carried out upon the discovery of the Remains, to the individual methods of identification, information system and practical case studies. Title of diploma thesis: The analysis of Psilocybe constituents II. The aim of this thesis was to analyse Psilocybe constituents, to plot the places of occurence of chosen Psilocybe brands and also to determinate their water content. The thesis deals with 22 samples that originate in 10 habitats in Slovakia and Moravian- Silesian region. Accuracy of brand specification was supervised by a member of mycological society. Psilocybe serbica var. bohemica was identified in 2, whereas Psilocybe semilanceata in 8 habitats. The average content of H20 in samples was 87,07 % ± 10,91 %. All of the samples were analysed by LC-MS/MS (LIT) method. The qualitative analysis of determinated substances showed the presence of psilocine (PSC) and psilocybine (PSB) in all samples. The results of quantitative analysis (expressed in percentage of dry matter) are similar in particular habitats, but there is a significant difference between two specific brands of Psilocybe mushrooms. The content of determinated alkaloids in P. bohemica was: PSC 0,001-0,011 % a PSB 0,01-0,07%. P. semilanceata contained 0,0005 - 0,011 % PSC a 0,074 - 0,763% PSB....

Cílem této diplomové práce je zanalyzovat současný stav identifikace obyvatel v České republice a poté navrhnout novou strukturu bezvýznamového identifikátoru obyvatel. Součástí této práce jsou také možné varianty, jak nejlépe využít tento nový identifikátor obyvatel při elektronické komunikaci. Z možných variant je pak vybrána ta nejvýhodnější varianta z hlediska rychlosti a náročnosti zavádění či finančních nákladů. Návrh řešení vychází ze získané teorie a z vypracované detailní analýzy týkající se identifikace obyvatel a elektronické komunikace. Byly vypracovány také odhady finančních nákladů a časový harmonogram zavádění nového identifikátoru obyvatel v České republice, díky kterému čtenář získá představu o časové náročnosti implementace navrhovaného řešení. Tato práce může sloužit jako důležitý zdroj informací při návrhu identifikátoru obyvatel v České republice.

An existing and successful methods are being described in the thesis and pre-processing of fingerprint images is being realized. Automatic fingerprint identification algorithm based on distance of minutiae from the core, core type and orientation field of the fingerprint is proposed. The proposed algorithm was tested on a database of fingerprint images and was statistically analysed.

This thesis explores the optimizing of the methods relevant to the cultivation, isolation, and identification of individual yeast strains from selected matrices, using the molecular biology methods. Grape berries, vine-leaves and soil from vineyard have been used as matrices. As yeasts are an integral part of fermentation processes, they are used for making of wine, the organoleptic charakteristics of which they influence. For identification of yeast strains the PCR-RFLP method has been used. Specifity of yeast DNA sequences of certain species has been used for this analysis. These spacers have been amplificated through the use of PCR and consequently they have been subjected to a restrictive analysis with specific restrictive endonucleases. These fragments have been detectioned through horizontal electrophoresis. In the literature background research section the basic informations about viticulture, yeasts, their cultivation and separation, PCR, restrictive analysis and horizontal electrophoresis is presented.
