Créer des surfaces spécifiques apparaît comme un enjeu majeur de la chimie actuelle. La formation de monocouches auto-assemblées (SAMs) à partir de dérivés soufrés est l’une des stratégies pour modifier des surfaces d’or. A ce jour, le mécanisme de formation des SAMs sur or n’est toujours pas élucidé.Une nouvelle approche de formation de SAMs par voie électrochimique a récemment été développée, avec la possibilité de suivre in-situ en temps réel la chimisorption de dérivés soufrés, notamment avec un cycle dithiolane. Contrairement aux méthodes standards d’adsorption par simple immersion, cette approche par polarisation de l’électrode tend à faciliter la chimisorption (cinétique, reproductibilité …). L’excellente résolution temporelle du suivi cinétique conjuguée à une bonne reproductibilité met en évidence un mécanisme d’adsorption via un phénomène de nucléation-croissance. Déjà identifié pour la formation de SAMs en phase gazeuse, il n’a jamais été clairement établi en phase liquide. L’utilisation de cette méthode a également permis l'élaboration de monocouches mixtes binaires de manière contrôlée et prédictible. En se basant sur les constantes d’affinité, nous avons pu réaliser des SAMs dont les proportions en surface reflétaient celles en solution. Ce résultat majeur laisse entrevoir des applications en particulier pour l’étude de systèmes enzymatiques sur surface.Enfin, afin de mieux cerner les facteurs clés régissant le mécanisme de formation, une étude systématique en fonction de la nature du groupe d’ancrage a permis de souligner le rôle crucial de celui-ci concernant aussi bien la cinétique d’adsorption que la stabilité de la monocouche. / Create specific surfaces appears as a major issue in chemistry. The formation of self-assembled monolayers (SAMs) using sulfur derivatives is one of the most commonly used strategy to modify gold surfaces. Nowadays, the formation mechanism of self-assembled monolayers (SAMs) on gold is still investigated.A new electrochemical approach for SAMs formation has recently been developed allowing an in situ and real time monitoring of the chemisorption of various sulfur compounds including dithiolane derivatives. Unlike standard adsorption methods which consist in a simple immersion, this approach tends to facilitate the chemisorption by polarizing the electrode. The high temporal resolution coupled to this good reproducibility highlight an adsorption mechanism which proceeds by a nucleation-growth process. This type of mechanism, already identified for the SAMs formation in the gas phase, has never been reported in the liquid phase.In addition, this method allows the preparation of binary mixed monolayers in a controlled and predictable manner. Knowing the affinity constants, it is possible to produce SAMs whose surface proportions reflect those in solution.Finally, to better understand the key factors governing the SAMs formation mechanism, a systematic study depending on the nature of the anchoring group has highlighted a significant difference between the different anchoring groups for the kinetics as well as for the stability of the monolayer.

AÃÃo da terapia fotodinÃmica antimicrobiana sobre biofilmes orais crescidos in vitro e in situ / The Photodynamic Antimicrobial Chemotherapy effect on in vitro and in situ biofilms

A Terapia FotodinÃmica Antimicrobiana (TFDA) apresenta-se como uma opÃÃo alternativa ao tratamento clÃssico, promovendo a morte bacteriana por meio da fotossensibilizaÃÃo dos componentes microbianos. Este estudo verificou a aÃÃo antimicrobiana da terapia fotodinÃmica sobre biofilmes orais produzidos in vitro e in situ utilizando um diodo emissor de luz (LED) associado ao fotossensibilizador azul de orto-toluidina (TBO). No estudo in vitro, biofilmes de Streptococcus mutans UA159, foram formados sobre discos de hidroxiapatita utilizando um modelo de banhos de cultura e submetidos à TFDA apÃs 5 dias. Para o estudo in situ, vinte e um voluntÃrios foram previamente selecionados para utilizar dispositivos intra-orais palatinos contendo 8 blocos de dentina humana durante 7 dias. SoluÃÃo de sacarose 10% foi gotejada sobre os blocos dentais 8 vezes ao dia. O biofilme formado em um dos lados do dispositivo recebeu tratamento da TFDA, e o lado oposto serviu como grupo controle. O material coletado passou por um processo de disrupÃÃo para a dispersÃo das cÃlulas e diluiÃÃo em sÃrie decimal de 10-1 a 10-4. Em ambos os experimentos, meios de cultura especÃficos para o crescimento de estreptococos totais e estreptococos do grupo mutans foram inoculados e incubados em condiÃÃes ideais para o crescimento desses microrganismos. ReduÃÃes significativas acima de 99,99% (p<0.05) foram observadas na viabilidade das colÃnias de S. mutans UA159 quando expostos ao TBO e LED no estudo in vitro. Entretanto, nos biofilmes formados in situ e submetidos Ãs mesmas condiÃÃes experimentais, nÃo foram verificadas diferenÃas estatisticamente significantes (p&#8805;0.05) da contagem microbiana quando comparadas ao grupo controle. Portanto, podemos concluir que a TFDA foi efetiva na reduÃÃo microbiolÃgica de S. mutans UA159 crescidos em modelo de formaÃÃo de biofilme in vitro, mas, pouco efetiva sobre biofilmes de estreptococos orais formados in situ. / The treatment of diseases caused by oral biofilms involves mechanical removal and use of antibiotics and antiseptics which can lead to problems of bacterial resistance. Photodynamic antimicrobial chemotherapy (PACT) represents an alternative option to conventional treatment, promoting bacterial killing by photo-sensitization of microbial components. This study assessed the antimicrobial action of photodynamic therapy on oral biofilms produced in vitro and in situ using a light emitting diode (LED) associated with the photosensitizer toluidine blue O (TBO). Biofilms of Streptococcus mutans UA159 were grown on hydroxyapatite discs immersed in bathing culture and submitted to PACT after 5 days. For the in situ study, twenty-one volunteers were previously selected to use intra-oral palatal appliances containing eight blocks of human dentin during 7 days. Sucrose solution (10%) was dripped onto the dental blocks 8 times a day. The biofilm formed on one side of the device received treatment (PACT), and the opposite side acted as control. The collected material has gone through a process of disruption to the dispersion of cells and diluted in decimal series (10-1 to 10-4). In both experiments, specific culture media for the growth of total streptococci and mutans streptococci were inoculated and incubated under optimal conditions for growth of these microorganisms. Significant reductions in excess of 99.99% (p<0.05) were observed in the viability of colonies of S. mutans UA159 when exposed to TBO and LED on in vitro study. However, the biofilms formed in situ and subjected to the same experimental conditions showed no statistically significant differences (p&#8805;0.05) in the microbiological counting when compared with control group. Therefore, we conclude that PACT was effective in microbiological reduction of S. mutans UA159 grown in an in vitro biofilm model, but very little effective on oral streptococci biofilms produced in situ.

Dans des contextes semi-arides et fortement anthropisés la caractérisation et la quantification des transferts de polluants dans les eaux de surface et vers la mer est importante à appréhender pour aider à une meilleure gestion de la ressource en eau dans un contexte de changements environnementaux. Sur le Nord-africain notamment la difficulté d’obtenir des données en quantité et qualité suffisante est difficile. La présente étude s’intéresse aux mécanismes de transfert des nitrates et des sédiments à l’échelle du bassin versant de la Tafna (7200 km2) et des plus grands fleuves de l'Afrique du Nord par une approche couplant les mesures in situ et le modèle SWAT (Soil and Water Assessment Tool). La partie in situ a permis de quantifier la quantité de nitrates apportée par les différents exutoires des sous bassins de la Tafna. Elle a aussi mis en évidence la contribution du bassin versant à l’eutrophisation de la Méditerranée tout en mesurant les quantités de nitrates acheminées vers les eaux marines. Les résultats ont mis en évidence les facteurs qui influent sur les flux et les concentrations en nitrates et notamment l’occupation du sol et les barrages ont une grande influence sur les flux de nitrates dans la Tafna. Les stations d’étude ont été classées selon un gradient d’eutrophisation, les stations amont présentant de fortes concentrations en nitrates, elles s’opposent aux stations avals. Les résultats de simulation sur la période 2003 à 2011 ont montré qu’une grande partie de l'eau de surface de la Tafna provient du ruissellement de surface (59%) et du flux latéral (40%), alors que la contribution des eaux souterraines est insignifiante (1%). La Tafna transfère une quantité annuelle moyenne de 2942 t.an-1 de sédiment vers la mer entre 2003 et 2011. SWAT a montré aussi que les cinq barrages de la Tafna stockent une grande quantité de sédiments, plus de 27 000 t.an-1 (90% des sédiments transportés par la Tafna). Le modèle reproduit aussi le flux de nitrates dans les stations de mesure entre 2003 et 2011. La Tafna transporte une quantité moyenne annuelle de 37 à 86 tN.an-1 jusqu'à la Méditerranée. Sur la base de ces résultats, l’approche de modélisation a pemis d’effectuer un changement d’échelle vers les grands bassins du nord-africain. La simulation a montré que les barrages de la Tafna stockent une quantité importante de nitrates [80 à 486 tN.an- 1], qui représente en moyenne 62% de la quantité totale de nitrates transportée annuellement par le fleuve. L’application du modèle sur les plus grands fleuves d'Afrique du Nord, Moulouya (55 860 km2), Tafna (7 200 km2), Chélif (44 694 km2) Oued el Kbir-Rhumel (8824 km2) et Medjerda (23 213 km2) a permis de représenter les flux d'eau, de sédiments et de nitrate. Les fleuves de cette région transportent une quantité de sédiments de 706 .103tan-1 vers la Mer ou le fleuve Oued Kbir-Rhumel qui contribue à la plus grande quantité (55%).Le flux de nitrates transportés vers la mer est de 5330 tN.an-1, le fleuve Moulouya qui contribue à la plus grande quantité (32%). Les barrages de cette région stockent une grande quantité de sédiments (98%) et de nitrates (39%) transportés par les fleuves vers la Méditerranée. / In semi-arid and highly anthropized area characterization and quantification of pollutant transfers in surface waters and to the sea is important to understand and to help better management of water resources, especially in North Africa, where the of obtaining data in quantity and sufficient quality is difficult. This study investigates mechanisms transfer of nitrate and sediment in the Tafna watershed (7200 km2) and at the biggest coastal rivers in North Africa with an approach, coupling in situ measurements and SWAT model (Soil and Water Assessment Tool). The in situ part made it possible to quantify the quantity of nitrates exported by the different Tafna sub-basins. It also highlighted the contribution of the watershed to eutrophication of the Mediterranean Sea by measuring the quantities of nitrates transported to marine waters. The quantities transferred are important in the high water season while they decrease in the rivers during the dry season. The results highlighted the factors that influence the nitrates flux and nitrate concentrations; the land use has a great influence on the nitrate flux in the Tafna. The study stations were classified according to the eutrophication gradient. The upstream stations with high nitrates concentration, oppose downstream stations. Modeling results showed that Tafna's surface water comes from surface runoff (59%) and lateral flow (40%), while the contribution from groundwater was insignificant (1%). Tafna has transferred an average annual amount of 2942 t.yr-1 of sediment to the sea. SWAT has also shown that dams store a large amount of sediment, over 27 000 t.yr-1 (90% of the sediments transported by Tafna). The model also reproduced the nitrates flux at the gauging stations between 2003 and 2011. The Tafna carries 37-86 tN.yr-1 to the Mediterranean Sea and Tafna dams store a large quantity of nitrates [80 and 486 tN.yr-1], which represents an average 62% of the total amount of nitrates transported annually by the river. The application of the model at largest scale in North Africa, Moulouya (55,860 km2), Tafna (7,200 km2), Chelif (44,694 km2) Oued el Kbir-Rhumel (8,824 km2) and Medjerda (23,213 km2) showed that the rivers of this region carry 706.103t.yr-1 of sediments to the Sea. The Oued Kbir-Rhumel River contributes with the greatest quantity (55%), whereas the nitrate flows transported towards the sea is 5330 tN.yr-1 and the Medjerda River contributes with the largest amount (32%). Dams in this region store a large amount of sediment (98%) and nitrate (39%) transported by rivers to the Mediterranean.

O objetivo deste trabalho foi avaliar a desmineralização do esmalte após períodos de 4, 7 e 10 dias de acúmulo de placa bacteriana in situ, além da influência de fatores salivares e microbiológicos no processo. Após a avaliação do fluxo salivar estimulado, capacidade tampão da saliva e níveis salivares de estreptococos do grupo mutans, 13 voluntários utilizaram dispositivos intrabucais palatinos contendo 4 blocos de esmalte bovino (4 X 4 X 2 mm) polidos, cobertos por uma tela plástica, durante 3 períodos cruzados de 4, 7 e 10 dias. Os voluntários utilizaram um dentifrício não fluoretado e gotejaram 10 vezes ao dia uma solução de sacarose a 20% sobre os blocos de esmalte. A microdureza superficial dos blocos foi determinada antes e após a desmineralização in situ, utilizando um penetrador Knoop e carga de 50 g por 5 s. Ao final de cada fase experimental, a placa bacteriana formada sobre os blocos foi coletada e a porcentagem de estreptococos mutans em relação ao número de bactérias totais foi determinada. Houve diferença estatisticamente significante, segundo o teste “t" pareado, entre a microdureza inicial e final nos três períodos estudados (p<0,05). A porcentagem de perda de dureza superficial (%PDS) foi em média de 13,8%, 19,3% e 48,3%, nos períodos de 4, 7 e 10 dias. Não houve diferença estatisticamente significante entre a %PDS com 4 e 7 dias, de acordo com a análise de variância e teste de Tukey (p<0,05). A correlação entre a %PDS e os fatores salivares e microbiológicos avaliados foi determinada por meio dos coeficientes de correlação de Pearson e Spearman, com p<0,05. Não foi encontrada correlação estatisticamente significante entre a %PDS e os fatores salivares avaliados inicialmente. Em aproximadamente metade das análises de placa bacteriana não foram detectados estreptococos mutans, embora tenha ocorrido desmineralização do esmalte. A porcentagem de estreptococos mutans na placa bacteriana não apresentou correlação com os níveis salivares dessas bactérias. Não foi encontrada correlação entre a %PDS e a porcentagem de estreptococos mutans na placa bacteriana formada sobre os blocos de esmalte. Before the beginning of the in situ phase, a stimulated whole saliva sample was collected from 13 volunteers for 5 min and the salivary secretion rate, buffering capacity and numbers of mutans streptococci were estimated. A palatal appliance containing 4 polished bovine enamel blocks (4 X 4 X 2 mm) covered by a plastic mesh was worn on 3 separate periods of 4, 7 and 10 days, throughout which time a non-fluoridated dentifrice was used. A 20% sucrose solution was dripped extra-orally onto the enamel blocks 10 times a day. All enamel blocks were evaluated by the analysis of enamel surface microhardness before and after in situ demineralization, using a 50 g, Knoop load for 5 s. At the end of each period, plaque covering the enamel blocks was collected and analyzed for the % of mutans streptococci of the total viable counts. Paired-t test showed that initial and final microhardness were statistically different at the three periods evaluated (p<0.05). Change in surface microhardness (%CSMH) was 13.8%, 19.3% and 48.3% during the 4, 7 and 10-day periods, respectively. There was no statistically significant difference on the %CSMH between 4 and 7 days, according to analysis of variance and Tukey’s test (p<0.05). Correlation between %CSMH and the salivary and bacteriological factors evaluated was determined using Pearson’s and Spearman’s coefficient of correlation (p<0.05). The %CSMH was not significantly correlated to the salivary factors determined initially. In about half of the plaque samples analyzed mutans streptococci were not detected, although the enamel showed some demineralization. The % of mutans streptococci in plaque samples was not correlated to its numbers in saliva. There was no statistically significant correlation between the %CSMH and the % of mutans streptococci in plaque over the enamel blocks. The presence of these bacteria in plaque significantly increased enamel demineralization when compared to the blocks where they were not detected only at 4 days, according to Mann-Whitney test (p<0.05). The results showed in situ enamel demineralization after short periods of plaque accumulation, which was not related to the salivary factors evaluated nor to the numbers of mutans streptococci in saliva or plaque.

Les constructions traditionnelles en bois en France et à Madagascar témoignent de l’utilisation ancienne d’abondantes ressources forestières locales. Le bois peut être considéré comme un matériau naturel et renouvelable dans la mesure où les ressources forestières sont gérées durablement. L’utilisation du bois dans la construction contribue largement à la maîtrise du cycle du carbone atmosphérique et à la limitation des émissions de gaz à effet de serre. Ce travail présente les principales caractéristiques physiques du matériau bois et des systèmes constructifs associés, montrant l’importance et l’influence du revêtement en bois sur l’environnement intérieur des habitations. La caractérisation des phénomènes hygrothermiques à l’échelle de la pièce d’habitation a été détaillée : présence d’eau dans le bois, comportement de l’équilibre hygroscopique, transfert thermique et transfert de masse, les paramètres physiques du bois et d’une ambiance intérieure. Enfin, ce travail apporte des connaissances sur l’évaluation des interactions entre le confort thermique, l’ambiance intérieure et le revêtement en bois, grâce à des campagnes de mesure in situ, à des mesures en laboratoire et à des modélisations de scénarios numériques. / Traditional wood buildings in France and in Madagascar use abundant local forest resources in architecture. Woodcan be considered as renewable and natural material when coming from sustainably managed forest. Using wood in construction contributes significantly in controlling the atmospheric carbon cycle and the emission of greenhouse gas. This study presents the main physical characteristics of wood materials and building systems showing the influence of wood coating on indoor environment. Hygrothermal phenomena across the living space are characterized and presented in this work : the presence of water in the wood, the behavior of equilibrium moisture, heat transfer and mass transfer, the physical parameters of wood material and physical parameters of indoor environment. Thermal comfort is similarly characterised. This work contributes knowledge on the evaluation of interactions between thermal comfort, indoor air and wood coating by means of in situ and laboratory measurements and numerical model simulations.

Application of Plant Extracts for the Prevention of Dental Erosion: An in situ/in vitro Study

Objectives: Antiadherent and antibacterial effects of certain plant extracts have been proven to be beneficial in preventive dentistry. In the present in situ/in vitro crossover study, the impact of plant extracts rich in polyphenols on the erosion-protective properties of the in situ pellicle was evaluated. Methods: Individual splints were prepared for 12 subjects for intraoral exposure of bovine enamel specimens. Following formation of a 1-min pellicle, watery plant extracts(leaves of the wild form of Ribes nigrum , the wild form of Origanum as well as a combination of both) were administered for 10 min in situ. Alternatively, a mouth rinse with fluorides (Elmex Kariesschutz) was performed for 1 min. After further oral exposure for 19/28 min, respectively, slabs were removed and incubated with HCl in vitro over 120 s (pH 2, 2.3, 3). The resulting calcium and phosphate release was quantified photometrically. Slabs with and without a 30-min in situ pellicle served as controls. The modification of pellicle ultrastructure was evaluated by transmission electron microscopy (TEM). Results: Plant extracts modulated the erosion-protective properties of the native in situ pellicle in all test groups in a pH-dependent manner. The combination of R. nigrum leaves and Origanum enhanced the protective properties of the pellicle at all pH values; the administration of this preparation was comparable, yet superior, to the effect of the fluoridated mouth rinse. TEM images indicated that rinsing with R. nigrum leaves /Origanum yielded a distinctly thicker and more electron-dense pellicle. Conclusion: The combination of certain plant extracts offers a novel approach to the complementary prevention of dental erosion.
