This thesis was prepared as a set of current knowledge and solution of the issues of biological safety in relation to occurrence of dangerous biological agents mainly in individuals. The possibility of occurrence of ``highly dangerous infection{\crqq} disease is real in the CR and the health care system together with the integrated emergency system services have to be prepared, actually at the moment when a suspicion of its occurrence is pronounced by a competent person. The questions processed here relate to an assessment of the level of cooperation between the integrated emergency system services and further involved entities of South Bohemian region on dealing with this type of emergency situation that might occur in an air transport passenger. The thesis also deals with the issue of current provision of protective means needed for biological protection of the intervening services staff. Topicality of this issue in South Bohemian Region is among others given by the reconstruction of the South Bohemian Airport in České Budějovice, which is being prepared for international operation. And the development of travelling of people who use air transport to distant and exotic parts of the world where they might get infected and return back to the CR still in the incubation period using the quick air transport, is one of the reasons why this danger must not be underestimated. Personal interviews with representatives of the integrated emergency system services of the South Bohemia Region and related literary and information sources were used for obtaining data and information for the thesis. A tabling method was also used for organization of the data and the data were consequently subject to systemic and causal analysis. An outline of specific protective means available to the individual entities was formed within the analysis of current protective means specified for biological protection of intervening staff, and the outline was assessed in line with applicable European standards. Thanks to efficient cooperation between the integrated emergency system services and the involved entities and thanks to performed exercises readiness for dealing with situations that might occur in consequence of suspicion of dangerous infection occurrence is very good in the South Bohemia Region. The system of solving such situations as well as availability of protective means is at a good level in the South Bohemia Region, however some disproportions were found and the thesis provides proposals for possible measures of solving them.

The aim of the thesis was to characterize chosen expression vectors used in biotechnologically important bacterial species, Corynebacterium glutamicum, and to test their use in studies of promoter activity control by sigma factors of RNA polymerase. Different properties of these vectors (level of expression of the cloned gene, leaky expression without inducer, dependence of expression level on inducer concentration and cell population homogeneity) were found by determination of expression level of the model gfpuv gene by fluorescence intensity assay of the produced protein and by gfpuv-expressing C. glutamicum cell population analysis using flow cytometry. The vector pEC-XT99A was chosen for testing the bi-plasmid system for assignment of a sigma factor to the chosen promoter. Although the level of expression provided by pEC-XT99A was not high, the vector showed no leaky expression, expression from the vector was comparable for a wide range of IPTG concentrations and the cell population was homogenous concerning the gene expression. Using pEC-XT99A from which individual stress sig genes were expressed, the σD factor was clearly assigned to the up-to-now unknown Pcg0420 promoter. On many structures there ocurred a problem of expanded polystyrene-based thermal insulation material degradation caused by high temperatures close to glazed areas. In a theoretical part of this Master’s Thesis there are the most common thermal insulation materials described, followed by a brief desciption of flat roof waterproofing materials and a thermal stress of structures. An experimental part describes a design of a methodology for evaluation of thermal resistance properties of foamed insulants in flat roofs and several experiments carried out according to the methodology.

Využití magnetických částic pro izolaci a purifikaci DNA z výrobků pro dětskou výživu / The use of magnetic particles for isolation and purification of DNA from products for children nutrition

Isolation of plant DNA is complicated due to the presence of polyphenols, polysaccharides and other metabolites, that are isolated together with DNA. These compounds can affect the yield and quality of DNA and can inhibit a polymerase chain reaction (PCR). A modern, simple, and fast method of DNA isolation in molecular biology laboratories is magnetic separation based on reversible DNA immobilization on magnetic particles. These methods allow to obtain DNA of high quality and purity. In the experimental part, magnetic microparticles PGMA 2 mg/ml and magnetic nanoparticles functionalized by polylysine (PLL) 0,2 mg/ml were used for isolation of plant DNA from vegetables (carrots), fruits (pear, apple, lemon, mango) and heat treated food products for children (Hami first carrot, Nestlé fruit pocket, dmBIO pear carrot apple, Hello fruit snack with apples and Hello fruity snack with mango). The efficiency of separation of magnetic particles with bound DNA using a magnetic needle and a magnetic separator were compared. The quality and quantity of isolated DNA were verified by spectrophotometric analysis. The amplificability of isolated DNA was tested in a conventional PCR using primers specific for plant ribosomal DNA (rDNA). PCR products were analyzed by agarose gel electrophoresis. Major fluorescent bands were of 700, 350 and 220 bp corresponding to three different rDNA amplicons. DNA was isolated from heat treated food products for children in a PCR-ready quality. Only 220 bp long PCR products were detected, that indicated DNA degradation. The identity of PCR products was determined by restriction fragment lenght analysis (RFLP) using NlaIII enzyme cutting the rDNA subregion (ITS1) of Daucus carota (carrot). The digestion profiles were in a good agreement with those predicted from bioinformatic analysis confirming, thus, the specificity of the developed PCR method.

The isolation of high quality DNA is nessecary for many molecular biology applications. However, plant DNA contains high amonts of polysaccharides, polyphenols and various secondary metabolites, which decrease yield and quality of isolated DNA. The aim of this study was preparation of samples and different food matrices for DNA isolation DNA by magnetic particles. It was about 5 species of vegetable and 10 species of processed plant food. Homogenization of samples was performed in CTAB buffer. Isolation of plant DNA was performed by magnetic particles covered with carboxyl groups. All DNAs were isolated in conventional PCR qualities using primers for 700 bp amplicons, in the case of heat processed products for 220 bp ampilicons and for real time PCR. The efficiancy of separation of magnetic particles with DNA by magnetic separator and magnetic needle was compared. It was find out that DNA of higher purity was isolated using magnetic needle. The micromethod of isolation of plant DNA from homogenates with CTAB with magnetic particles is suitable for different processed food.

Due to a European energy concept for reducing energy consumption and also the concept of sustainable development, there is a growing demand for reduced energy consumption during the operation of the building and hence increasing demands on the thermal and mechanical properties of the building envelope. For this reason, it is necessary to look for materials that could meet both thermal and mechanical properties, as well as mechanical resistance and loadability, especially for the application for the foundations of the house, the base of the house and for application to structural details for elimination of the thermal bridges between interior and exterior boundaries. The main motivation for choosing the topic of thesis was to find materials derived from recycled or secondary raw materials that would be suitable for manufacturing composite applicable for structural details in the envelope of the building and for insulating in humid environment. An economic and environmental aspect plays an important role in the choice of material. The main theme of the thesis is the laboratory manufacturing of a composite with a thermoplastic matrix derived from recycled plastic materials and waste foam as a phase. Has been developed a unique Waste-based Particle Polymer Composite (WPPC) made from recycled foam and polypropylene. However, before WPPC can be reliably used by construction designers, physical properties of WPPC must be accurately identified. Therefore, it was designed laboratory manufacturing system and sample testing system, it was studied thermal, mechanical, thermomechanical and moisture absorptivity of WPPC. Application of finished composite material with satisfying thermal insulation properties to structural details to eliminate the thermal bridge, was the next step. The objective of the presented diploma thesis is to evaluate the influence of modification on surface layer of products with their base structure from organical fibers and on the ability to resist increased humidity. Emphasis is puted on the description of humidity propagation in fiber materials ant he approximation of secondary waste from the textile industry as a potencional material for thermal insulation. The end of the theoretical part focuses on the types of possible surface treatment to prevent the ingress water into the structure of the material. The practical part verifies the selected type of modification of the surface layer of three materials choosen as test ensemble. After applying surface treatment, the effect of treatment on humidity sensitivity and water absorption was assessed. The determination of the thermal conductivity coefficient, the short-term absorption at partial immersion and the determination of the sorption humidity were made.

Development, production and durability of vacuum insulating panels are described in the theoretical part as well as principles of heat transfer. Method for production of core of VIP, created using waste fibers from textile industry and agriculture, is described in the practical part. Verification of behavior during normal and reduced pressure (even to vacuum) was carried out on experimentally made core insulants.
