The trend in today's civil engineering are growing demands for quality and comfort constructed buildings and spaces in them. This trend causes a change in the approach to designing buildings and building new buildings. One of the main directions great importance is attached dedicated reconstruction of buildings and their technical adjustments to meet the ever increasing demands being norms and laws. One of the most monitored parameters are thermal-technical characteristics of objects - i.e. better quality means saving on heating costs. In this connection there is a big trend of the modern construction industry and building insulation, mainly ETICS systems. Dissertation focuses on the basic (reinforced) layer of the ETICS and examines the effect of low temperatures, acting on the matter in the implementation of the system, the properties of this layer. This coating is one of the key elements of the system, which more or less determines the quality and durability of the system. The first part deals with the current state of the problem and generally insulation systems, basic types and the advantages and disadvantages of these systems. In the main part of the work is solution focused work, its objectives and methodology for testing materials using laboratory tests and are mentioned outputs of experiments in terms of results. The final section addresses the evaluation of the data and results and output for construction practice. The theme of work was also addressed in the specific research. The works are also numerical and graphical attachments to records of experiments and calculations.

This Diploma thesis deals with the preparation of the realization of the railway viaduct in Ostrov nad Oslavou. Realization of major technological stages is addressed in building technological study. Technological processes of each sub-job phases are supplemented by technical regulations, the design of the main building machines and mechanisms, inspection and test plan, schedule of occupational health and safety, time and financial plan construction, construction budget, a plan for securing material resources. I also worked out the technical report of the building sites facilities including drawings needed. I also worked out design rehabilitation of the substructure of the railway viaduct, which is consist of technical regulations for remediation work and drawings.

Stavba horského hotelu na bázi dřeva / Wooden Based Hotel in Mountains

The subject of my thesis is the design of new mountain hotel based on wood, which is located at the top of Little Maple, cadastral area Trinec-Tyra, the city of Trinec, Frydek-Mistek level documentation for construction. The building has a partial basement, with three floors. The supporting system consists of three structural systems. The structural system consists of basement formwork, the first floor is made of ceramic blocks heluz, second and third floor is designed as a timber frame panel. Roof structure is made up of lattice wood trusses and gabled aisle. Proposal mountain hotel respects the territorial plan of Trinec. Furthermore, emphasis is placed on the layout, including ensuring the design to the static, architectural, energy savings and safety in the use of the building.

The thesis discusses the operating costs and the possibilities for their optimization. Specifically, it focuses on one of the greatest possible operating costs and it's heating costs. In the first part of the thesis are defined in general expenses, the price and the production, operating costs, optimization of these costs, life cycle of buildings and technological methods to improve the thermal and technical properties of the object. Finally, the theoretical part describes the terms for calculating the energy performance of the building and the payback period, which evaluates the effectiveness of investments. The practical part is assessed according to specific building energy performance, and especially in terms of its demand for heating. The building is designed for six variants of insulation, which is again judged on their energy performance. Subsequently these measures for better thermal insulating properties of the object assessed according to their cost efficiency and return on investment of individual variants.

Analýza DNA Lactobacillus s využitím PCR v reálném čase a HRM analýzy / Lactobacillus DNA analysis using real-time PCR and HRM analysis

The rapid and accurate identification of the bacterium of the genus Lactobacillus, which are an important part of the normal gastrointestinal microflora and fermented dairy products are currently mainly used amplification methods. The aim of the study was to analyze the possibility of resolution of selected bacterial strains of the genus Lactobacillus, using the metod of polymerase chain reaction in the real time combined with high resolution melting curve analysis (qPCR HRM). It was tested five primers designed for qPCR-HRM analysis of lactic acid bacteria. The specificity of the primers was also verified simultaneously using bioinformatic analysis. On the basis of analysis of the DNA were selected as the most appropriate primers P1V1/P2V1, V3F/V3R and V6F/V6R. The suitability of the primers V3F/V3R and V6F/V6R was verified on a complex sample of food supplement from which the DNA was isolated using magnetic particles. The presence of bacteria of the genus Lactobacillus was performed using high resoluting melting analysis curves. The obtained results were in agreement with the information given by the manufacturer.

Probiotic food products - food additives contain different species of probiotic bacteria. Accurate species identification with their characteristics is very important from the view of products quality. Methods of DNA diagnostics are used for these purposes. In this thesis DNA was isolated from 4 probiotic products. The presence of bacterial of genus Lactobacillus and species L. acidophilus, L. casei, L. plantarum, L. rhamnosus were detected in three products by PCR. This information was in accordance with the data provided by the manufacturer. Two sets of primers were used for identification of species. Using other primers sequences of genes such as bsh, lai and odc were detected in DNA isolated from the products. Differences were estimated among products concerning the detection of lai gene Lactobacillus acidophilus.

Antimicrobial substances, or bacteriocins are substances produced by probiotic lactic acid bacteria. They have a positive effect on the gastrointestinal tract of humans and are especially suitable for the food, but also the pharmaceutical industry. The aim of the thesis was the molecular identification of lactic acid bacteria of the Lactobacillus genus, species and their subsequent inclusion PCR method. Using the PCR method were tested also genes responsible for the production of bacteriocins. To confirm the production of bacteriocins has been selected the microbiological method, agarose droplet spot-test. In the present study also included the bioinformatics part to assess the specificity and non-specificity of the primers using in Primer-BLAST program.

The thesis deals with the layout, building and construction, buildings, so that it effectively uses renewable energy and efficiently work with them.

Dermatofyty izolované ze srsti volně žijících hlodavců / Dermatophytes isolated from the hair of free-living rodents

Dermatophytes (order Onygenales, Ascomycota) are microscopic filamentous keratinophilic fungi that can cause skin infections known as dermatophytosis. The most diverse but not very studied genus Arthroderma has been revised recently (Míková 2018) which was essential for further research. This genus comprises mostly species with a supposed reservoir in soil. Lack of information about their ecology and frequent isolation of some species from the hair of free- living mammals (mainly rodents) may testify a strong host association. Rodents could thus represent the hidden reservoir of this species. For this thesis, I have chosen three ecologically distinct rodent species: Mus musculus, Apodemus flavicollis, and Clethrionomys glareolus. I obtained the material by brushing the hair of asymptomatic individuals and used this material for cultivation on selective medium. I identified the isolates of dermatophytes (n = 30) using molecular methods. I used sequences of three highly variable loci (ITS, tubb a tef1α) to incorporate these isolates in the phylogenetic analysis based on the monography of the genus Arthroderma (Míková 2018). I characterized the phenotype of selected strains based on morphological and physiological data including the ability to utilize keratin and the production of siderophores. The...
