Estes materiais podem ser processados na forma de filmes finos e dependendo da técnica utilizada pode-se obter filmes organizados e altamente reprodutíveis. Uma das técnicas de deposição que pode proporcionar estas propriedades é a de Langmuir Blodgett (LB) e Langmuir Schaefer (LS). Este trabalho de mestrado teve como objetivo fabricar e caracterizar materiais/filmes com capacidade para aplicação em dispositivos fotovoltaicos Os materiais caracterizados, com este objetivo, foram polímeros de baixo valor de band gap:poli[(4,4-bis(2-etilhexil)-ciclopenta-[2,1-b:3,4-b’]ditiofeno)2,6-diil-alt-(2,1,3-benzotiadiazol)-4,7-diil], PCPDTBT e poli[(4,4’-dioctilditieno[3,2-b:2’,3’d]silol-2,6-diil)-alt-(2,1,3-benzotiadiazol)-4,7-diil)], PDTSBT. Para a fabricação dos filmes ultrafinos foi realizado um estudo de isotermas de pressão superficial por área do monómero (π-A) em uma cuba de Langmuir. As técnicas de LB e LS permitiram o processamento molecular dos materiais através das informações obtidas das isotermas (π-A). As caracterizações ópticas (UV-Vis) foram realizadas para análise de crescimento e da organização das camadas dos filmes finos. Para as medidas elétricas foi utilizado uma fonte de corrente continua para se obter a condutividade e fotocondutividade elétrica dos filmes depositados por LB e LS sobre eletrodos interdigitados. Os filmes fabricados foram utilizados como camadas ativas dos dispositivos fotovoltaicos em uma heterojunção planar, com estrutura basicamente formada por ITO/Polímero/PCBM/Alumínio. Foram realizadas medidas de densidade de corrente versus tensão (J vs V) sob iluminação de um Simulador Solar para o cálculo de eficiência da célula. / The conjugated polymers applied as active layer in optoelectronic devices, have high technological potential and have been widely studied in recent decades. These materials can be processed in the form of thin films, and depending on the technique used one can obtain organized and highly reproducible films. One of the deposition techniques that can provide these properties is Langmuir Blodgett (LB) and Langmuir Schaefer (LS). This master 's work was designed to manufacture and characterize materials / films with potential for application in photovoltaic devices. The materials characterized for this purpose, were low bandgap value polymers: poly [(4,4-bis (2-ethylhexyl) cyclopenta- [2,1-b: 3,4-b '] dithiophene) 2,6-diyl-al- (2,1,3-benzothiadiazole) - 4,7-diyl] PCPDTBT and poly [(4,4'-dioctyldithieno [3,2-b: 2 ', 3'd] silol-2,6-diyl) -alt- (2,1,3-benzothiadiazol) -4,7- ] PDTSBT. For the production of ultrafine films, a study of surface pressure isotherms by area of the monomer (π-A) in a Langmuir trough was performed. The LB and LS techniques allowed the molecular processing of the materials through the information obtained from the isotherms (π-A). The optical characterizations (UV-Vis) were performed for analysis of the growth and organization of thin film layers. For the electrical measurements, a DC current source was used to obtain the electrical conductivity and photoconductivity of the films deposited by LB and LS on interdigitated electrodes. The films manufactured were used as active layers of the photovoltaic devices in a planar heterojunction, with structure basically formed by ITO / Polymer / PCBM / Aluminum. A study of the adsorption of some polymers and Langmuir Blodgett film systems by inelastic electron tunneling spectroscopy (IETS)

The work in this thesis can be split into three areas. In the first, inelastic electron tunnelling spectroscopy (IETS) is used to investigate resistive and bias polarity dependent effects on vibrational mode energy in metal - aluminium oxide - metal tunnel junctions. It is shown that even when four point-probe techniques are used the ratio of width to thickness of the electrodes used in IET junctions has an effect on both the breadth and position of spectral lines. A simple treatment based upon work done by Giaever allows for such effects to be corrected. Work done on top metal and polarity effects in undoped IET junctions investigated the effect on the position of the 450 meV mode on reversing the applied bias. The investigation revealed that any dependence on the nature of the top-metal electrode was outside the accuracy of the work. Calculation of q/e for the hydroxyl group on alumina within the tunnel junction has been extended to include other electrode materials. A new and exciting facility within the university provided the impetus for the second area of work. A class 100 clean room housing a Langmuir Blodgett (LB) trough offered the opportunity to produce metal - insulator - Langmuir Blodgett film - metal tunnel junctions. It was realised at the outset that IETS using LB films would be difficult, previous workers had tried using a home-made tank with only limited success. However, the added sophistication of the new tank did not improve matters as was hoped. Although the results were disappointing, only a few junctions had resistances low enough to be usable in the spectrometer, the investigation produced some of the very few IET specra using junctions doped with LB films. The results also revealed the important role that imperfections and pin-holes play in the tunnelling process. The last area used IETS to investigate two commercially important and interesting polymers, hydrogels and polymeric electrolytes. Hydrogel have many applications in the field of implants, prosthetic, and cosmetics and have been studied and developed for many years. Polymeric electrolytes have many commercial applications especially in the field of solid-state batteries and conducting polymers. The way in which hydrogels swell as they absorb water is important, as is the way they adsorb onto a surface and much work has been done to investigate these characteristics using bulk samples. The study done by this group is the first to investigate the swelling and adsorption behaviour of a monolayer of the hydrogel poly 2-hydroxylethyl methacrylate. The results from both investigations indicate that ester cleavage occurs in p-HEMA and that water incorporated within a hydrogel has a limited structure with the first layer being thinner than the second and subsequent layers.

NASAâ s VAriable Specific Impulse Magnetoplasma Rocket Engine (VASIMR) will provide a highly efficient propulsion source that can dramatically reduce Martian transit times, provide for more abort contingencies, and protect astronauts from space radiation with its highly radiation-absorbent hydrogen fuel. The VASIMR is still in its developmental infancy and requires many years of research before its initial operational capability. Much is still unknown about the complex plasma interactions in the exhaust. A Langmuir probe was designed, constructed, and operated to determine current density radial profiles and total particle flux at various stages in the exhaust of VASIMR. The Langmuir probe results proved that the exhaustâ s radial profile is Gaussian, experimentally validated predictions of magnetic field line dragging, and verified the ionization efficiency of VASIMR.

Investigating effects of hypertonic saline solutions on lipid monolayers at the air-water interface

More than 70,000 people worldwide suffer from cystic fibrosis, a genetic disease characterized by chronic accumulation of mucus in patients’ lungs provoking bacterial infections, and leading to respiratory failure. An employed age-old treatment to prevent the symptoms of the disease is inhalation of hypertonic saline solution, NaCl at concentrations higher than in the human body (~150 mM). This procedure clears the mucus in the lungs, bringing relief to the patient. However, the biophysical mechanisms underlying this process are not entirely clear. We undertook a new experimental approach to understand the effects of sprayed saline solutions on model lung surfactants towards understanding the mechanisms of the treatment. The surface of lungs contains mainly 1,2-Dipalmitol-sn-glycero-3-phosphocoline (DPPC). As previously assumed by others, we considered that monolayer of DPPC at the air-water interface serves as model system for the lungs surface; we employed a Langmuir-Blodgett (LB) trough and PM-IRRAS to measure surface-specific infrared spectra of the surfactant monolayers and effects on the interfacial tensions. We investigated spraying hyper-saline solutions onto surfactant monolayers at the airwater interface in two parts: (i) validation of our methodology and techniques with stearic acid and (ii) experiments with DPPC monolayers at the air-water interface. Remarkably, when micro-droplets of NaCl were sprayed to the monolayer of stearic acid, we observed enhanced organization of the surfactant, interpreted from the intensities of the CH2 peaks in the surface-specific IR spectra. However, our results with DPPC monolayers didn’t show an effect with the salt added as aerosol, possibly indicating that the experimental methodology proposed is not adequate for the phenomena studied. In parallel, we mimicked respiratory mucous by preparing salt solutions containing 1% (wt%) agar and measured effects on their viscosities. Interestingly, we found that NaCl was much more effective than NaI and NaClO4. This thesis reports structural dynamics of monolayers of stearic acid and DPPC at the airwater interfaces and those of aqueous solutions towards understanding mechanisms underlying the most commonly employed treatment for cystic fibrosis. Our methodology has never been reported before; but requires further modifications to gain deeper insights into the effects of salt sprays on model lung systems.

Interação de capsaicinóides com sistema modelo de membrana celular / Capsaicinoids interaction of with cell model membrane system

Este trabalho visa ao entendimento da interação de capsaicinóides com membranas celulares utilizando sistemas modelo. Dentre os alcalóides derivados de plantas do gênero Capsicum, a capsaicina e a dihidrocapsaicina respondem por 90% dos capsaicinóides, que são usados como analgésicos e antiinflamatórios, devido a sua interação específica com receptores. O mecanismo neurofarmacológico já foi bastante estudado, mas o modo de ação não neural ainda não foi elucidado. Usamos extratos brutos (EBs) de pimenta malagueta e de dedo-de-moça, que têm atividade superficial, e afetaram monocamadas de Langmuir de fosfatidil colina de dipalmitoíla (DPPC) e fosfatidil glicerol de dipalmitoíla (DPPG). Tais efeitos não tiveram dependência expressiva com a carga, pois o EB de dedo-de-moça interagiu mais fortemente com o DPPC do que com o DPPG, ao passo que o contrário se verificou para o EB de malagueta. Também não houve diferença significativa entre os EBs das duas pimentas. Nas monocamadas de Langmuir representativas para a bactéria S. aureus, ambos os EBs tiveram efeito, tanto nas isotermas de pressão quanto nos resultados de espectroscopia de absorção e reflexão no infravermelho com modulação de polarização (PM-IRRAS), sem distinção significativa entre malagueta e dedo-de-moça. No entanto, as medidas de vazamento com lipossomos mostraram maior interação com o EB de dedo-de-moça, o que é consistente com a atividade bactericida para S. aureus. De fato, a concentração inibitória mínima (MIC) foi 0,13 mg mL-1 para o EB da pimenta dedo-de moça e 4,0 mg mL-1 para o EB de malagueta. Para a E. coli, os EBs interagiram com as monocamadas de Langmuir sem diferenças dignas de nota para as duas pimentas, e nas medidas de vazamento o efeito maior foi para a dedo-de-moça. Não houve efeito bactericida para nenhum dos extratos. Isso se explica porque bactérias gram-negativas, como a E. coli, têm uma camada externa protetora de lipossacarídeos (LPS). Das medidas de monocamadas de Langmuir representativas da camada de LPS, observou-se pouca incorporação dos EBs. Conclui-se, assim, que os EBs não conseguem causar rompimento da camada de LPS. Do conjunto dos resultados, infere-se que o mecanismo de ação para a S. aureus envolve solubilização parcial da membrana, e não há relação entre pungência e atividade bactericida, pois a pimenta dedo-de-moça, que é menos pungente, teve maior efeito do que a malagueta. Depreende-se, também, que a ação de extratos de pimenta deve depender da interação com receptores na membrana, o que explica porque o uso de tais extratos tem sido principalmente em aplicações tópicas. / This study is aimed at understanding the interaction of capsaicinoids with cell membranes using model systems. Among the alkaloids derived from plants of the genus Capsicum, capsaicin and dihydrocapsaicin account for 90% of capsaicinoids, which are used as analgesic and anti-inflammatory due to their interaction with specific receptors. The neuropharmacological mechanism has been well studied, but the non-neural mode of action has not been elucidated. Here, we use crude extracts (EBs) of malagueta and dedo-de-moça chilli peppers, which are surface active, and affected Langmuir monolayers of dipalmitoyl phosphatidylcholine (DPPC) and dipalmitoyl phosphatidyl glycerol (DPPG). Such effects did not depend on the charge, since EB from dedo-de-moça interacted more strongly with DPPC than with DPPG, while the opposite applied for malagueta. In addition, there was no significant difference between the two EBs. For Langmuir monolayers representing the bacteria S. aureus, both EBs affected the surface pressure isotherms and the polarizationmodulated infrared reflection-absorption spectroscopy (PM-IRRAS) data, without significant distinction between dedo-de-moça and malagueta. However, in the leakage measurements with liposomes the EB from dedo-de-moça was more efficient in rupturing the liposome, which is consistent with the bactericidal activity for S. aureus. In fact, the minimum inhibitory concentration (MIC) was 0.13 mg mL-1 for dedo-de-moça and 4.0 mg mL-1 for malagueta. For the Langmuir monolayers mimicking the E. coli membrane, the EBs interacted much in the same way, while the EB from dedo-de-moça caused larger leakage in liposomes. There was no bactericidal effect of the EBs. This is explained by the fact that gram-negative bacteria, such as E. coli, have a protective outer layer of liposaccharides (LPS). In monolayers representing LPS, there was little incorporation of EBs, from which one infers that the EBs cannot cause disruption of the LPS layer. Taking all these results together, it appears that the mechanism of action for S. aureus involves partial solubilization of the membrane. Furthermore, there is no relationship between pungency and bactericidal activity because dedo-de-moça, which is less pungent, had greater effect than malagueta. It seems also that the action of pepper extracts must depend on the interaction with membrane receptors, which explains why the use of such extracts has been essentially in topical applications.

Estudo de filmes de Langmuir e Langmuir-Blodgett de aminas anfifílicas utilizando a espectroscopia SFG / The interaction of water with Langmuir films of amphiphilic amines as a function of pH probed by Pi-A isotherms and SFG spectroscopy

O estudo da interação de moléculas orgânicas com a água é de grande importância para diversas áreas das ciências, como por exemplo, na compreensão de sistemas biológicos. Uma elevada diversidade de estudos nesta área já foram realizados, no entanto, alguns aspectos do estudo de filmes finos de aminas anfifílicas não foram satisfatoriamente esclarecidos. Este trabalho aborda filmes de Langmuir de octadecilamina em função do pH e da força iônica de subfases aquosas, investigando a estrutura da monocamada orgânica e sua interação com a água, em diversos graus de ionização. Foi utilizada a Espectroscopia Vibracional por Geração de Soma de Frequências (Sum Frequency Generation - SFG), uma técnica que utiliza o princípio da óptica não-linear para estudar superfícies e interfaces, com o objetivo de investigar a conformação molecular dos filmes de Langmuir de aminas anfifílicas de cadeias longas, seu grau de ionização e a estrutura da água interagindo com a monocamada orgânica. Pôde-se acompanhar a ionização da monocamada em função do pH da subfase pela intensidade do modo de estiramento do grupo NH3+, e verificou-se que ela depende do pH local da interface, que por sua vez é alterado pela ionização da monocamada e é sensível à força iônica da subfase. A água interage com a monocamada via ligações de H com os grupos NH2/NH3+ e forma uma camada com alto grau de orientação perpendicular à superfície, mas relativamente desordenada do ponto de vista da rede de ligações de H (como na água líquida). A conformação das cadeias alquila depende da ionização do filme de Langmuir, sendo bastante compacta e ordenada a altos pHs (filme neutro) e mais expandida e desordenada a baixos pHs. Além disso, foram fabricados filmes de Langmuir-Blodgett (LB) de octadecilamina para comparar a estrutura dos filmes na superfície da água e em substratos sólidos. / The study of the interaction of organic molecules with water is of great importance for many areas of science, such as the understanding of biological systems. A large diversity of studies in this area have been performed, however some aspects of the study of thin films of amphiphilic amines have not been satisfactorily clarified. This dissertation focuses on Langmuir films of Octadecylamine as a function of pH and ionic strength of the aqueous subphase, investigating the structure of the organic monolayer and its interaction with water at various degrees of ionization. Sum-Frequency Vibrational Spectroscopy (SFG spectroscopy), a non-linear optical technique to study surfaces and interfaces, was used in order to investigate the molecular conformation of the Langmuir films, its degree of ionization and the structure of water interacting with the organic monolayer. The monolayer ionization could be monitored by the intensity of the NH3+ group stretch mode, and it was found that it depends on the local pH at the interface, which in turn is changed by the monolayer ionization and is sensitive to the subphase ionic strength. Water interacts with the monolayer by H bonds with NH2/NH3+ groups, forming a layer with a high degree of orientational order along the surface normal, but relatively disordered regarding the H-bonding network (like in the bulk liquid). The conformation of the alkyl chains depends on the monolayer ionization, being compact and all-trans at high pHs, but more expanded and disordered at low pHs.
