Avaliação do teste intradérmico em equinos com extratos alergênicos de pólens, insetos e três concentrações de histamina / Evaluation of intradermal test in horses with allergen extracts of polens, insects and three histamine concentrations

O teste alérgico intradérmico (TID) é uma importante ferramenta no diagnóstico dos alérgenos implicados nas reações de hipersensibilidade mediada por IgE. O objetivo deste estudo foi avaliar o TID em eqüinos não alérgicos com extratos alergênicos de Cynodon dactylon, Lolium multiflorum, Paspalum notatum, Culex sp. e Aedes aegipty, e três concentrações de histamina, em dois volumes diferentes, e baseado nos resultados, verificar a acurácia do TID em eqüinos alérgicos. Foram realizados TID em 17 eqüinos não alérgicos com três concentrações de cada extrato (1:2000v/w, 1:4000v/w e 1:8000v/w) e três concentrações de histamina (0,1mg/ml, 0,05mg/ml e 0,025mg/ml), em dois volumes (0,1 e 0,05ml). Baseado nos resultados obtidos, foram realizados TID em seis eqüinos alérgicos utilizando os extratos na concentração de 1:2000v/w, a histamina na concentração de 0,025mg/ml, em um volume de 0,05ml. Nos eqüinos não alérgicos, nenhuma das três concentrações dos extratos testados nos dois volumes mostrou-se irritativa. Concentrações maiores de histamina provocaram halos maiores e levemente mais túrgidos, e quanto maior o volume injetado, maiores as reações Os TID realizados em eqüinos alérgicos mostraram confiabilidade dos resultados por evidenciar hipersensibilidade individual e não provocar reações irritativas. Em conclusão, as diferentes concentrações dos estratos testados em eqüinos não alérgicos não provocaram reações falso positivas (irritantes). O volume de 0,05ml é mais recomendado para realização dos TID pois as aplicações com 0,1ml produziram reações maiores, prejudicando a sensibilidade do teste. A concentração de 0,025mg/ml da solução de histamina provocou a formação de halos menores, permitindo maior acurácia do cut off no TID. Os extratos alergênicos testados em equinos alérgicos não provoraram reações em todos os animais, o que poderia ser considerado uma reação irritativa. Os equinos alérgicos apresentaram reações positivas diferentes, de acordo com a hipersensibilidade individual, demonstrando sensibilização alérgica verdadeira. / The intradermal test (IDT) is an important tool in the diagnosis of allergens involved in IgE-mediated hypersensitivity reactions. The aim of this study was to evaluate the IDT in nonallergic horses with allergenic extracts of Cynodon dactylon, Lolium multiflorum, Paspalum notatum, Culex sp. and Aedes aegypti, and three histamine concentrations, in two different volumes, and based on the results, verify the accuracy of IDT in allergic horses. IDT was performed on 17 nonallergic horses with three concentrations of each extract (1:2000v/w, 1:4000v/h and 18000v/w) and three histamine concentrations (0,1mg/ml, 0,05mg/ml and 0,025mg/ml) in two volumes (0,1 and 0,05ml). Based on the results obtained, IDT was performed in six allergic horses using extracts at the concentration of 1:2000v/w, histamine at a concentration of 0.025mg/ml, in a volume of 0,05ml. In nonallergic horses, none of the three concentrations of the extracts tested in the two volumes were irritant. Larger concentrations of histamine provoked larger and slightly more turgid wheals, and the larger the volume injected, the greater the reactions The IDT performed in allergic horses showed reliability of the results because they demonstrated individual hypersensitivity and did not provoke irritative reactions. In conclusion, the different concentrations of extracts tested in nonallergic horses did not provoke false positive (irritant) reactions. The volume of 0,05ml is more recommended for execution of IDT because the applications with 0,1ml produced larger reactions, impairing the sensitivity of the test. The concentration of 0,025mg/ml of the histamine solution caused the formation of smaller wheals, allowing a better accuracy of the cut off in the IDT. The allergenic extracts tested in allergic horses did not provoke reactions in all the animals, what could be considered an irritative reaction. Allergic horses presented different positive reactions, according to individual hypersensitivity, demonstrating true allergic sensitization.

No entanto, a maioria das cultivares oficiais presentes no mercado precisam ser dessecadas antes do final do ciclo para permitir uma adequada época de semeadura para culturas de estação quente. Alguns materiais não certificados apresentam ciclos mais precoces, porém também apresentam resistência a dessecantes e rendimentos de forragem insatisfatórios. Neste sentido a Embrapa lançou a cultivar BRS Integração com a proposta de um ciclo mais precoce, todavia, sem resistência a herbicidas, bem como com elevado rendimento forrageiro. Contudo, se desconhece o efeito do manejo de desfolha nesta cultivar para a colheita de forragem e seus impactos na produção e qualidade de sementes e no comprimento do ciclo produtivo. Deste modo o objetivo do presente estudo foi avaliar o efeito da desfolha na produção de forragem e de sementes do azevém anual BRS Integração. O experimento foi realizado na Estação Experimental Terras Baixas da Embrapa Clima Temperado com delineamento de blocos ao acaso com quatro repetições. Foram testados cinco níveis de desfolha (sem, uma, duas, três e quatro desfolhas) para a verificação da produção de forragem (fração de folhas, colmos e material morto); das características morfogênicas e estruturais; dos componentes de rendimento, do rendimento e da qualidade fisiológica das sementes. Verificou-se que o número de desfolhas afetou as características morfogênicas e estruturais da cultivar BRS Integração e, por conseguinte, o rendimento e a qualidade fisiológica de sementes. A execução de três desfolhas permitiu a colheita de aproximadamente 5t de MS.ha-1 e afetou positivamente o rendimento e a qualidade fisiológica das sementes, as quais foram colhidas ainda nos primeiros dias de novembro, o que possibilita o uso da área para a adequada semeadura de cultivos de estação quente. A realização de quatro desfolhas afetou negativamente as características morfogênicas, a estrutura pré-colheita, os componentes de rendimento de sementes, bem como o rendimento real de sementes. No entanto, a ação de quatro desfolhas (colheita total de aproximadamente 7t de MS.ha-1), não afetou negativamente a qualidade fisiológica da semente e ainda possibilitou um rendimento de sementes 15 vezes superior a densidade de semeadura recomendada. / The crop-livestock integration with the use of annual ryegrass is a common practice in southern Brazil. However, most official cultivars on the market need to be desiccated before the end of the cycle to allow adequate time for sowing warm season crops. Some not certified materials present earlier cycles, but also show resistance to desiccants and poor forage yields. In this sense, Embrapa launched “BRS Integração” with the proposal of an earlier cycle, however, without herbicide resistance as well as high forage yield. However, it is unknown the effect of defoliation management in this cultivar for forage crop and its impact on production and quality of seeds and the length of the production cycle. Thus the purpose of this study was to evaluate the effect of defoliation in the production of fodder and seeds of annual ryegrass “BRS Integração”. The experiment was conducted at the Estação Experimental Terras Baixas in Embrapa Clima Temperado with randomized block design with four replications. Five levels of defoliation were tested (no, one, two, three and four defoliation) for verification of forage production (leaves fraction, stems and dead material); morphogenic and structural characteristics; the yield components, yield and seed quality. It was found that the number of defoliation affect the morphogenic and structural characteristics of the “BRS Integração”, and therefore, the yield and physiological quality of seeds. The execution of three defoliation allowed the harvest of approximately 5 tons of dry matter per hectare and positively affected the yield and seed quality, which were still harvested in early November, what allow the use of the area for proper seeding of warm season crops. The performance of four defoliation adversely affected the morphogenic characteristics, pre-harvest structure, seed yield components and the real yield seeds. However, the action of four defoliation (total harvest about 7 tons of dry matter per hectare) didn’t affect negatively the physiological quality of the seed and still allowed a yield of seeds 15 times the recommended seeding rate.

Altura de manejo do pasto e suas conseqüências sobre a produção animal e a dinâmica de pastagens anuais de inverno / Managemen sward height and his consequence in animal production and dynamics of anual winter pasture

O objetivo do experimento foi compreender e quantificar a mudança estrutural de pastagens anuais típicas do período hibernal do RS e avaliar o potencial produtivo e as características de carcaças de novilhos superprecoces. O experimento foi conduzido em área pertencente à Fazenda Espinilho, localizada no município de São Miguel das Missões – RS. Foram impostos quatro tratamentos por meio de diferentes alturas de manejo da pastagem: 10, 20, 30 e 40 cm, obtidas a partir da aplicação de diferentes cargas animais. O delineamento foi o de blocos completos casualizados com três repetições. Foram utilizados animais, com cerca de 10 meses de idade, machos inteiros, uniformes, sem padrão racial definido, com peso médio inicial de 190 kg. As variáveis estudadas foram: massa de forragem (MF), taxa de acúmulo de forragem (TAC), produção total de forragem (PTMS), relação lâmina foliar/colmo + bainha, ganho médio diário (GMD), ganho por área (GPA). Observou-se um aumento linear da MF com o aumento da altura do pasto, onde para cada cm de aumento na altura acima de 10 cm, correspondem um incremento de cerca de 108 kg/há na MF do pasto. Não houve efeito dos tratamentos para a TAC, bem como para a PTMS, cujos valores médios foram de 55,8 kg/há.dia e 8210 kg/ha, respectivamente. O aumento no GMD foi condicionado pelo incremento na qualidade e/ou na quantidade de forragem disponível, já que as OF diárias para os tratamentos de 10, 20, 30 e 40 cm de altura foram de 6, 7, 13 e 19 kg de MS/100 kg de PV, respectivamente. Portanto, o modelo de resposta do GMD em relação às alturas resultou em 0,96 e 1,24 kg/animal nos tratamentos de menor e maior GMD, respectivamente, que foram de 10 e 20 cm de altura. O maior GPA observado no tratamento 10 cm (515 kg de PV/ha) deveu-se à uma maior carga animal utilizada, e ambas apresentaram respostas lineares decrescendo com o aumento da altura de pastejo. O peso vivo dos animais antes do abate não foram incrementados com o aumento da altura do pasto (P>0,05). Houve um forte e abrupto decréscimo da relação lâmina/colmo+bainha colmo nos tratamentos 30 e 40 cm a partir de setembro. Investigou-se o ajuste de quatro tipos teóricos de distribuição das freqüências de altura em cada tratamento e em seis diferentes datas de observação, bem como o potencial de predição da MF por intermédio da altura do pasto. A distribuição das freqüências de altura se ajustou ao modelo Normal em apenas uma das noventa e seis séries analisadas. A distribuição tipo Gamma foi a que mais freqüentemente se ajustou aos dados de altura, porém, uma vez iniciado o pastejo, o incremento da heterogeneidade no pasto foi tal que a distribuição de freqüências não se ajustou a nenhum dos modelos estudados. Sugerese que as metas de altura de manejo devam ser variáveis ao longo do ciclo de pastejo, com o intuito de se administrar a heterogeneidade causada pelo animal. / Aiming to comprehend and quantify the structural changes in typical temperate annual pastures from RS and evaluate the potential production and characteristics of young beef steers carcass, this experiment was conducted at Fazenda Espinilho, located in São Miguel das Missões. Treatments were four sward height management targets (10, 20, 30 and 40 cm), using continuous variable stocking. A completely randomized block design with three replicates was applied. Beef steers weighting 190 Kg were used and they were ten months old, non-castrated males with no defined breed. The investigated variables were herbage mass (HM), herbage growth rate (GR), total dry matter production (TDMP) and laminae/stem+sheath relation, average daily gain (ADG), gain per area (GPA). Results indicated a linear increase relating herbage mass and sward height, where each cm on sward height above 10 cm increases herbage mass in 108 kg/ha. Treatments did not differ in GR and the TDMP, average values being 55,8 kg DM/ha and 8210 kg DM/ha, respectively. The increase on the average daily gain (ADG) was due to the increase of quality/amount of available forage and the herbage allowance for the treatments of 10, 20, 30, 40 cm, was 6, 7, 13, 19 DM kg/ 100 kg LW, respectively. So, the ADG was 0.96 and 1.24 kg/animal for the treatments of lowest and highest ADG, which corresponds to 10 cm and 20 cm. Once ADG was low, the highest GPA observed on the 10 cm treatment (515 Kg PV/ha) was due to a higher stocking rate and both presented a negative linear response, decreasing with the increase in sward height. Concerning final live weight there was no difference with increasing sward height (P>0,05). A pronounced decrease in laminae/stem+sheath relation for 30 and 40 cm treatments occurred by September. In addition, four theoretical distributions were fitted to sward height data for each treatment in six different sample dates. Sward frequency height distributions fitted Normal distribution only in one of ninety six data series analyzed. Gamma distribution was more frequently adjusted to sward height data, however, once grazing started, pasture heterogeneity was so increased that sward height did not fitted neither of the investigated models. It is suggested that sward height targets should be variable along the grazing season aiming to manage the heterogeneity caused by the animal.

Contamination atmosphérique par les hydrocarbures aromatiques polycycliques : toxicité et devenir du phénanthrène dans des systèmes sol-plante-microorganismes

Les hydrocarbures aromatiques polycycliques (HAP) sont des polluants organiques persistants potentiellement mutagènes et cancérigènes. Leur transfert de l'atmosphère vers les écosystèmes, notamment vers les plantes, conditionne leur entrée dans les chaines alimentaires mais les modalités de ce transfert restent encore mal connues. L'objectif de ce travail était donc de caractériser le transfert et d'identifier les effets biologiques des HAP atmosphériques sur un système sol-plante-microorganismes symbiotiques.Un dispositif expérimental a été conçu afin de recréer en laboratoire une pollution atmosphérique par les HAP avec comme HAP modèle le phénanthrène (PHE). Le dispositif a fait l'objet d'une validation et d'une calibration élaborée de manière originale par une double approche mêlant l'expérimental à la simulation mathématique. Les niveaux d'exposition en polluant (150 µg m-3), contrôlés par des échantillonneurs passifs, se sont avérés pertinents par rapport aux conditions in situ. Ce dispositif a donc été utilisé pour exposer durant un mois des microsystèmes sol-plante-microorganismes au PHE par voie atmosphérique.Les différentes études ont mis en évidence un transfert du PHE depuis l'atmosphère vers tous les compartiments du microsystème, avec une accumulation majeure vers les feuilles de trèfle ou de ray-grass (respectivement 170 et 70 µg g־ ¹MS) et un transfert phloémien vers les racines est suggéré. Chez le trèfle, la mycorhization n'a pas été affectée, tandis que le nombre de nodules actifs a diminué de manière significative. Contrairement aux racines, la biomasse aérienne du trèfle a été significativement affectée (environ - 25%) par l'exposition au PHE atmosphérique, suggérant un impact sur le métabolisme carboné de la plante. Une expérience de marquage des trèfles au ¹³C-CO2 a effectivement montré un impact négatif du PHE atmosphérique sur la croissance, l'allocation de biomasse et l'allocation carbonée. Pour conclure, ces études ont permis non seulement de caractériser les effets biologiques et physiologiques des HAP atmosphériques sur les végétaux mais également de proposer l'utilisation du potentiel mycorhizien comme indicateur de pollution atmosphérique par les HAP.

Contribution à l'étude de l'allocation des photoassimilats récents dans la plante et la rhizosphère chez une graminée pérenne <br />(Lolium perenne L.)

L'objectif de notre travail a été étudié le transfert de C dans la plante et vers la rhizosphère de Lolium perenne afin d'expliciter le couplage entre le fonctionnement de la plante et la rhizodéposition.<br />Nous avons d'abord déterminé la contribution du carbone récemment assimilé par la plante à la rhizodéposition. Nous avons ainsi réalisé un double marquage long des parties aériennes des plantes (14CO2 et 13CO2) afin d'estimer la répartition dans la plante et les compartiments rhizosphériques du C récent et le C plus anciennement assimilé. Contrairement à ce qui est admis généralement, le C ancien, probablement issu de la remobilisation de composés de réserve, contribue significativement à la rhizodéposition.<br />Nous avons ensuite évalué sur des dispositifs expérimentaux de terrain l'influence de contraintes subies par les parties aériennes de la plante sur les flux de C récent vers la rhizosphère (augmentation de la concentration en CO2 atmosphérique et défoliation). La disponibilité en azote dans le sol est un facteur déterminant de la réponse des plantes à l'élévation des concentrations en CO2 ou à la défoliation. A l'automne, une diminution de la rhizodéposition de C est observée pour les plantes soumises au doublement de CO2. Les composés organiques libérés par les racines sont minéralisés plus rapidement sous CO2 élevé que sous CO2 ambiant. La défoliation n'induit pas de variations notables de la disponibilité en C dans la rhizosphère, suggérant aucune influence significative de ce facteur sur la rhizodéposition. <br />Les interactions entre les facteurs de l'environnement et de gestion de la parcelle cultivée déterminent le fonctionnement de la plante et modulent les efflux de C à partir des racines, ce qui rend la rhizodéposition difficile à prédire et à modéliser. Il s'avère que les effets nets du doublement de CO2 ou de la défoliation sur la rhizodéposition sont beaucoup moins marqués au champ qu'en conditions contrôlées. Au delà de la contribution quantitative de la rhizodéposition au bilan de C dans le sol, les interactions entre la nature biochimique des rhizodépôts et l'activité biologique des sols nécessitent d'être approfondies.

Genetic material of Lithuanian origin varieties, breeding lines and wild ecotypes representing P. pratense, P. bertolonii and P. phleoides species were studied for the most important agro-morphological and feeding value indicators as well as at the genetic level employing different biochemical–molecular markers. The water stress experiment clearly demonstrated the existence of different levels of water stress response in Phleum species, suggesting that P. phleoides might have evolved under conditions of limited water availability. “Blind” mapping by HRM was used successfully to map water stress response genes of timothy in the perennial ryegrass mapping population. Correlation of fecal ergovaline, lolitrem B, and their metabolites in steers fed endophyte infected perennial ryegrass straw

Perennial ryegrass (PRG, Lolium perenne) is a hardy cool-season grass that is infected with the endophytic fungus Neotyphodium lolii, which enables the plant to be insect repellant and drought resistant, lowering the use of insecticides and fertilizers. However, this fungus produces the compound lolitrem B (LB, m/z 686.4) which causes the tremorgenic neurotoxicity syndrome 'ryegrass staggers' in livestock consuming forage which contains <2000 ppb LB. Ergovaline (EV, m/z 534) is a vasoconstrictor normally associated with tall fescue (Festuca arudinacea), but has also been found in endophyte-infected PRG. Past research has shown a strong linear correlation between levels of LB and EV in PRG. The purpose of this study was to examine the linear relationship between EV and LB in feces and determine common metabolites. To accomplish this, four groups of steers (n=6/group) consumed endophyte- infected PRG over 70 days consumed the following averages of LB and EV: group I 2254ppb LB/633 ppb EV; group II 1554ppb LB/ 373ppb EV, group III 1011ppb LB/259ppb EV, and group IV 246ppb LB/<100ppb EV. Group I in week 4 was inadvertently given a washout period at which time the steers consumed the amount of LB and EV given to group IV (control). Both feed and feces samples were extracted using difference solid phase extraction methods and quantified by HPLC-fluorescence for LB and EV. Concentrations of EV and LB obtained through HPLC-fluorescence in both PRG and feces showed a linear relationship. Additional screening for metabolites was conducted LC-MS/MS and showed possible oxidation and reduction metabolites for both toxins. / Graduation date: 2013

This thesis defines digestion kinetics for perennial ryegrass (Lolium perenne L.), which is the main component of diets fed to dairy cows in New Zealand. Chemical composition and digestion kinetics were measured in fresh minced ryegrass as it matured and leaf, stem and inflorescence of several grass species. In sacco and in vitro incubations were used to define rates of degradation and nutrient release. Two short-term grazing trials were used to evaluate contrasting silages as supplements for cows fed restricted amounts of summer pasture. The minced preparation of ryegrass resulted in a similar distribution of dry matter (DM) between particle size fraction and rumen digesta from cows fed pasture. Mincing released 0.46 – 0.80 of crude protein into the soluble fraction, with highest proportions for mature grasses which had low CP concentrations (about 8 g CP/100 g DM). In contrast, the majority of fibre remained in the insoluble fraction but rates of degradation (k) approximately halved as grass matured. In vitro yield of VFA was similar for immature and mature minced ryegrass (after 12 hours VFA was equivalent to about 30% of DM), even though ammonia concentration declined to very low values for stem and mature grass. This suggests the rapid initial microbial growth was able to sustain a high level of DM degradation to VFA with mature grass. The summer pasture used for silage supplementation was of uncharacteristically good quality so the expected contrasts between maize, pasture, sulla (Hedysarum coronarium), lotus (Lotus corniculatus) and sulla/maize silage mixtures were less than expected. Milk responses to lotus silage supplements were greater than other silages (e.g.: 290 g milksolids from 54 MJ ME by lotus versus 110 g milksolids from about 50 MJ ME supplied by other silages). Pasture substitution was low (0.06 – 0.33). The Cornell Net Carbohydrate and Protein System (CNCPS) was chosen for evaluation of cow trial data because it uses feed degradation parameters as input variables to estimate nutrient supply. Model prediction of milk yield matched observed values when cows maintained liveweight. Milk yield was underestimated at low intakes and overestimated at high intakes because no allowance is made for nutrient partitioning between milk production and liveweight change.
