Development of foster mother-child attachmentKorhonen, J. (Jasmi) 26 May 2014 (has links)
The impact of early attachment relationships and child-adult attachment relationships to children’s social and emotional development has been recognised for a long time. Since the pioneering attachment theories of John Bowlby and Mary Ainsworth highlighting the importance of secure child-adult attachments, many other theories from various perspectives have risen to fill in the gaps. These theories are examined with the framework of foster care in mind.
In Finland, the primary solution for children in insufficient care is family-based foster care. In practice, a child is placed in a new family, while maintaining contact with their biological family. Previous studies have shown that young children tend to form new attachment relationships in the foster family (Cole, 2005). The formation of new attachment relationships in foster care settings can be affected by many external factors, such as foster parent behaviour (Schofield & Beek, 2008; Dozier et al., 2006), foster parents’ attitude towards biological parents (Andersson, 2008) and age of the child at the time of placement (Smyke, Zeanah, Fox, Nelson & Guthrie, 2010). Some studies also suggest that keeping contact with biological parents may in fact increase the foster child’s chances of forming secure attachments in the foster family and identifying with the foster family (Haight et al., 2003).
In addition to existing theories and foster care studies and interventions, this thesis utilises the narratives of three Finnish foster mothers in answering the research question: “How do foster mother-child attachment relationships develop in foster families?” The foster mothers were instructed to write about their relationship with their foster child(ren), with reference to the development of their relationship, the nature of the current relationship, and what they think their relationship will be in the future. The three narratives are analysed from a phenomenological perspective, with the help of Giorgi’s Psychological Phenomenological Method.
All of the three narratives give accounts of attachment formation between the foster mother and foster children, and include reference to themes such as age at time of placement, the process of forming an attachment, and keeping contact with the biological family. They also bring out the paradoxical nature of fostering a child, of not being the real parent but having to behave as one, and how this affects the foster mother emotionally. All three foster mothers highlight the impact of the foster child’s previous attachment relationships to the formation of their relationship with the child. Furthermore, the foster mothers’ experiences give reason to believe that the younger the child is at the time of placement, the easier it is for them to form an attachment to the foster mother.
Uusien kiintymyssuhteiden muodostumiseen voivat vaikuttaa monet ulkoiset tekijät, kuten sijasvanhemman käytös (Schofield & Beek, 2008; Dozier et al., 2006), sijaisperheen asenne lapsen biologisia vanhempia kohtaan (Andersson, 2008) ja lapsen ikä sijoituksen alkaessa (Smyke, Zeanah, Fox, Nelson & Guthrie, 2010).

Application software development via model based designHaapala, O. (Olli) 09 April 2015 (has links)
This thesis was set to study the utilization of the MathWorks’ Simulink® program in model based application software development and its compatibility with the Vacon 100 inverter. The target was to identify all the problems related to everyday usage of this method and create a white paper of how to execute a model based design to create a Vacon 100 compatible system software.
Before this thesis was started, there was very little knowledge of the compatibility of this method. However during the practical experiments, it became quite quickly clear that this method is very compatible with the Vacon 100. Majority of the problems expected before this thesis was started proved to be wrong. The only permanent problem that came up during this thesis was that Vacon 100 supports only the 32-bit floating-point precision while Simulink uses the 64-bit floating-point precision by default.
Even though this data type incompatibility prevents usage of some of the Simulink’s blocks, it is by no means a limiting factor that restricts usage of this method. During the practical research no problems were found that completely prevent the usage of this method with the Vacon 100 inverter.
The Simulink PLC Coder was in the spotlight during the practical research work. Even though the code generator worked better than was expected, the biggest problems limiting the usage of model based design relates to the PLC Coder.
All in all based on this study, it is easy to say that model based design is a very usable tool for everyday design work. The method does not necessarily show its advantages in a small scale design work and the implementation phase can take some time before the company has collected the support model library needed. However in the future, the software requirements will increase, which grows the importance of model based design. A well-executed model based design can have a significant improving effect on financial, time and quality aspects.
Tulevaisuudessa kuitenkin suunnittelutyön vaatimusten kasvaessa, mallipohjaisen ohjelmistotuotannon merkitys tulee kasvamaan.

Stakeholder analysis in software-intensive systems developmentKelanti, M. (Markus) 18 October 2016 (has links)
Abstract
A stakeholder analysis is commonly a part of the requirements engineering process in the development of software systems. It contributes to identifying, analysing, negotiating and validating requirements from multiple stakeholder viewpoints that do not necessary share the same views on a system under development and do not necessary express themselves using a similar language. Stakeholder analysis is often integrated into a used development method or practice and doesn’t necessarily appear as a separate process.
The increase in software size, availability and use in different appliances, however, requires more from the stakeholder analysis than has been recognized in Software Engineering literature. The increasing scale of software systems and connections to other systems increase the number of involved stakeholders complicating the stakeholder analysis. In addition, how the actual stakeholder analysis should be implemented in large scale software development and how it supports the development effort is problematic in practice.
The purpose of this thesis is to study the role and purpose of a stakeholder analysis in a large-scale software-intensive systems development. In this thesis, an empirical approach is taken to study the large-scale software-intensive systems development as phenomena in order to observe it as a whole. This approach allows this thesis to analyse the phenomena from different perspectives in order to identify and describe the nature and purpose of a stakeholder analysis in large-scale software-intensive systems development.
The contribution of this thesis is the following. First, the thesis contributes to both the practical and scientific community by describing the role of stakeholder analysis in the software-intensive systems development process. Secondly, it demonstrates how a stakeholder analysis can be implemented in a large-scale software-intensive systems development process. / Tiivistelmä
Sidosryhmäanalyysi on yleensä osa vaatimusmäärittelyprosessia ohjelmistojärjestelmien kehityksessä.
Tutkimus on toteutettu empiirisellä lähestymistavalla tarkkailemalla suuren mittakaavan ohjelmistointensiivisten järjestelmien tuotantoa kokonaisuutena. Tämä lähestymistapa mahdollistaa kokonaisuuden analysoinnin eri näkökulmista, jotta sidosryhmäanalyysin luonne ja tarkoitus voidaan tunnistaa ja kuvata suuren mittakaavan ohjelmistointensiivisten järjestelmien tuotannossa.
Väitöskirjan tulosten kontribuutio jakautuu kahteen osaan. Ensimmäiseksi väitöskirjan tulokset auttavat sekä tiedeyhteisöä ja käytännön työtä tekeviä kuvaamalla sidosryhmäanalyysin suuren mittakaavan ohjelmistointensiivisten järjestelmien tuotannossa. Toiseksi tulokset havainnollistavat miten sidosryhmäanalyysi voidaan toteuttaa suuren mittakaavan ohjelmistointensiivisten järjestelmien tuotekehitysprosessissa.

Développement d'un système "générique" de production d'anticorps murins et recombinants par bioingénierie / Development of a generic system for the production of murine and recombinant antibodies by bioengineeringYakoub, Walid 25 October 2017 (has links)
Monoclonal Antibodies (mAbs) are proteins with specific antigen recognition used for development of both therapeutic and diagnostic reagents. Commercial production is achieved by growing host cells in specific bioreactors. Cell density and cell metabolism are key parameters for high yield of mAbs. Hollow fiber bioreactor (I-IFB), a cartridge containing packed porous fibres, is one of the system for production commercially available. If the cell density achieved can lead to high yield, the cost of the whole device, including pumps and very expensive cartridges prevents its use of small units. As an economical alternative, we proposed here to investigate the potential of commercial polysulfone dialysis modules, classically employed in the treatment of end stage renal failure. However, the native polysulfone membrane demonstrated a significant non-specific protein adsorption detrimental to mAbs production. Moreover normative issues are added to these scientific and techno-economic issues, with the case of standards (ISO 13485 / AC S99-104 / GMP FDA / BPF ...) which impose standard working methods. This thesis consists of the design of a disposable bioreactor, based on a hollow-fiber medical dialysis cartridge. This system must offer all the guarantees in terms of production, ease of use, sterility, and allow to concentrate the cytoculture products. Scientific methodology has been coupled with a quality approach. The management of this project was coupled with the risk analysis. Indeed this project was divided into its 5 elementary components describe by Ishikawa by the 5M method. The risk analysis consisted in the calculation of the criticality index by the AMDEC method of each of its families of risks. This approach allowed us to formalize two research axes: i) the implementation of an effective oxygenation of the culture medium (chapter 4) and ii) the means of limiting the clogging in the hollow fiber module to obtain a culture consistent with the objectives (Chapter 5). The rate of oxygenation is a factor to be taken into account in a cell culture process. Oxygen can be supplemented in two modes, passive mode or active mode [Ozturk and Palsson 1990; Zhang S. et al 19921. There are two types of oxygenation system: the so-called passive system or the exchanges are made through a silicone wall and an active system by direct aeration in the culture medium. This system is by far the simplest operation for providing oxygen. However, when it is used to supply oxygen to mammalian cell cultures, this can cause cellular damage. Chemical protective agents can be used to reduce cell damage and DKamase and Moo-yung 1990 foam formation; van Der pol L.A. et al 1993 Cl. Our studies have shown that the addition of antifoaming agents can lead to a decrease in the liquid phase (K2) mass transfer coefficient of D Kamase and Moo-yung 1990C]. We have established the effectiveness of using a silica / silicone polymer to remove foam on bacterial and mammalian cell cultures. In order to limit these adsorption phenomena, polysulfone fibers were treated with several surfactants (pluronic acid F 127, D-limonen, and different silicone oils) which led to a significant decrease in protein adsorption. The effect of such surfactants on the filtration performances and on cytotoxicity were investigated. Some of the them did not influence these parameters while some presented negative effects. Finally, different cell culture parameters (cells densities, production yield, flow properties, fouling) were studied, as well as the performance of the bioreactor in perfusion continuous mode. The bioreactor was maintained in continuous mode for fifteen days and the production yield per batch was 250 mg of AcMs. The results obtained in this work allowed us to define the next steps to be taken, and are the subject of the Perspectives section.

Nutrient Signaling, Mammalian Target of Rapamycin and Ovine Conceptus DevelopmentGao, Haijun 2009 May 1900 (has links)
This research was conducted to test the hypothesis that select nutrients including
glucose, leucine, arginine and glutamine stimulate conceptus development by activating
mTOR (mammalian target of rapamycin; HGNC approved gene name: FRAP1, FK506
binding protein 12-rapamycin associated protein 1) signaling pathway.
First, temporal changes in quantities of select nutrients (glucose, amino acids,
glutathione, calcium, sodium and potassium) in uterine lumenal fluid from cyclic (Days
3 to 16) and pregnant (Days 10 to 16) ewes were determined. Total recoverable glucose,
Arg, Gln, Leu, Asp, Glu, Asn, His, beta-Ala, Tyr, Trp, Met, Val, Phe, Ile, Lys, Cys, Pro,
glutathione, calcium and sodium was greater in uterine fluid of pregnant compared to
cyclic ewes between Days 10 and 16 after onset of estrus. Of note were remarkable
increases in glucose, Arg, Leu and Gln in uterine flushings of pregnant ewes between
Days 10 and 16 of pregnancy.
Second, effects of the estrous cycle, pregnancy, progesterone (P4) and interferon
tau (IFNT) on expression of both facilitative (SLC2A1, SLC2A3 and SLC2A4) and
sodium-dependent (SLC5A1 and SLC5A11) glucose transporters, cationic amino acid
transporters (SLC7A1, SLC7A2 and SLC7A3), neutral amino acid transporters (SLC1A4,
SLC1A5, SLC3A1, SLC6A14, SLC6A19, SLC7A5, SLC7A6, SLC7A8, SLC38A3,
SLC38A6 and SLC43A2) and acidic amino acid transporters (SLC1A1, SLC1A2 and
SLC1A3) in ovine uterine endometria from Days 10 to 16 of the estrous cycle and Days 10 to 20 of pregnancy as well as in conceptuses from Days 13 to 18 of pregnancy were
determined. Among these genes, SLC2A3 and SLC7A6 were detectable only in
trophectoderm and endoderm of conceptuses. The abundance of mRNAs for SLC2A1,
SLC2A4, SLC5A1, SLC5A11, SLC7A1, SLC7A2, SLC1A4, SLC1A5, SLC43A2 and
SLC1A3 changed dynamically in ovine uterine endometria according to day of the
estrous cycle and early pregnancy. Expression of mRNAs for SLC2A1, SLC5A11 and
SLC7A1 in endometria was induced by P4 and further stimulated by IFNT with shortterm
treatment (12 days), while expression of SLC7A1 and SLC1A5 in endometria
required long-term treatment (20 days) with P4 and IFNT.
Third, effects of the estrous cycle, pregnancy, P4 and IFNT on expression of
nitric oxide synthase (NOS1, NOS2 and NOS3), GTP cyclohydrolase (GCH1), ornithine
decarboxylase 1(ODC1), insulin-like growth factor II (IGF2), FRAP1 complexes
(FRAP1, LST8, MAPKAP1, RAPTOR, RICTOR), regulators (TSC1, TSC2, RHEB) and an
effector (EIF4EBP1) of FRAP1 signaling in ovine uterine endometria from Days 10 to
16 of the estrous cycle and Days 10 to 20 of pregnancy as well as in conceptuses from
Days 13 to 18 of pregnancy were determined. All of these genes were expressed in
ovine uterine endometrium and conceptuses. Among these genes, expression of NOS1,
IGF2, RHEB and EIF4EBP1 changed dynamically due to day of the estrous cycle and
early pregnancy. Progesterone stimulated NOS1 and GCH1 expression while IFNT
inhibited NOS1 expression in uterine endometria, and P4 and IFNT stimulated
expression of RHEB and EIF4EBP1 in uterine endometria.
Collectively, these results indicate that: 1) the availability of select nutrients in
the ovine uterine lumen increases to support the rapid growth and elongation of the
conceptus during the peri-implantation stage of pregnancy; 2) P4 and/or IFNT
stimulate(s) glucose and amino acid transporters to facilitate their transport from
maternal tissues and/or blood into the uterine lumen during early pregnancy; 3) the
FRAP1 cell signaling pathway mediates interactions between the maternal uterus and
peri-implantation conceptus and both P4 and IFNT affect this pathway by regulating expression of RHEB and EIF4EBP1. Expression of NOS, ODC1 and IGF2 appear to be
linked to FRAP1 signaling in both uteri and peri-implantation conceptuses.
