This diploma thesis is dealing with design of explorative robot named Gloin, which is designed especially for the exploration and mapping of the underwater part of Hranická propast, which is the second deepest fresh-water abbys in the world. This thesis is concentrating on design of algorithms needed for control and navigation of robot in the abyss. The designed algorithms are verified by simulation.

Lokalizace metylačních míst transposonů / Localization of Methylation Sites in Transposons

This master's thesis deals with the creation of a tool for the extraction of methylation level from transposon sequences. Transposons are DNA elements with ability to move or copy themselves and their activity is regulated by DNA methylation. Sequence methylation information is stored in the bisulfite data and their processing is done with parts of two existing tools in a combination with implemented modules. Created tool takes into consideration unique challenges brought in the methylation calling process by transposable elements and it's functionality is presented on a set of experiments with simulated and real data.

Mapování rozšíření kříženců jelenů evropských a jelenů sika v ČR s využitím občanské vědy a audionahrávek / Mapping of distribution of red deer and sika deer hybrids in Czechia using audio recordings - citizen science project

In the Czech Republic, as in other parts of Europe (e.g., British Isles), the native European red deer (Cervus elaphus) has been hybridising with sika deer (C. nippon), which was introduced from East Asia. The hybrids are fertile, and, therefore, can backcross with their parental species and threaten their genetic integrity. Offsprings of such multiple crosses are very difficult to identify according to their morphological traits, while more reliable genetic methods are financially and logistically demanding. This work follows the study of Long et al. from 1998, which showed clear differences in vocalization between red deer, sika deer and the first generation of their hybrids, and thus suggested the third way how to identify hybrids - bioacoustic analysis. We decided to test this method in the Czech Republic in a citizen science project, the aim of which was to map the distribution of parent species and their hybrids using audiorecordings of rut vocalisation. To allow more detailed analysis, we also recorded our own recordings on a professional recording device with a directional microphone. This work documents the first case of idiopathic AA amyloidosis in humans caused by mutation in the promoter region of SAA1 gene. Knowledge of the mechanism of the disease may be an indication for targeted treatment in the future. Mutations in the SAA1 promoter should be considered in all cases of idiopathic forms of AA amyloidosis in which neither the immune nor the inflammatory component of the disease are clearly present.

The diploma thesis focuses on subject description unification methods in records aggregated from different sources in digital repositories, using the example of the National Repository of Grey Literature (NRGL). After presenting experiences with systems BASE and LASSO abroad, I describe the current situation in NRGL, where the automatic indexing is used to assign each record a unified subject heading from the Polythematic Structured Subject Heading System (PSSHS). The thesis then presents how the MeSH thesaurus and Conspectus categorization scheme were mapped to PSSHS. These mappings were then applied to records from the National Medical Library. The aim of the experiment was to compare the subject description consisting of PSSHS subject headings created by automatic indexing, and the subject description created by mapping. (in English): The diploma thesis deals with the activities of Czech Caritas organizations in the field of humanitarian assistance and development cooperation abroad. The aim of the work is to explore the possibilities and limits of transferring and sharing "good practice". The work consists of a theoretical and practical part. The theoretical part is structured in three main chapters, which deal with humanitarian aid and development cooperation topics, describe the selected Czech Caritas organizations and define "good practice". The practical part consists of a descriptive research of the selected Czech Caritas organizations activities in the field of humanitarian assistance and development cooperation abroad executed between 2000 and 2016 and qualitative research of employee's approaches to mutual sharing of "good practice". Research questions are answered by combining questionnaire survey and interviews which are conducted with the help of the manual. The main results of this thesis is a summary table containing descriptions of activities in the field of humanitarian assistance and development cooperation abroad and presentation of employee's attitudes to "good practice" and its sharing.

5 Abstract: Aims: The main objective of this work was to analyze in detail the effects of acute temperature changes on the function of isolated chick embryonic heart in vitro in comparison with natural conditions in ovo. Methods: The effects of temperature change (34 řC, 37 řC and 40 řC - hypo-, normo- and hyperthermia, respectively) on calcium dynamics in four days old isolated chick hearts in vitro were investigated by high-speed calcium optical imaging. For comparison and validation of in vitro measurements, experiments were also performed in ovo using videomicroscopy. Artificial electrical stimulation experiments were performed in vitro and in ovo to uncover conduction limits of different heart segments. Results: We observed almost linear dependence of sinus frequency on temperature in our temperature range. Sinus frequency during hypothermia and hyperthermia in vitro and in ovo changed about 20% in comparison with normothermia. We observed no significant changes in amplitude of calcium transients during temperature change to hypothermia but hyperthermia caused a significant decrease in amplitude of calcium transients (atria 35%, ventricles 38%). Protein misfolding is considered to be the major pathogenic mechanism in homocystinuria due to cystathionine beta-synthase (CBS) deficiency. The aim of this work was to study molecular mechanisms underlying protein misfolding of CBS mutants. Firstly, we studied spatial arrangement of normal human CBS protein. Using data from differential covalent labeling of solvent-exposed aminoacid residues, we identified interdomain contact area between the catalytic core and the regulatory domain in human CBS, and we subsequently generated the structural model of the full-length CBS. In the next step, we studied evolutionary divergence of CBS protein structures. We performed phylogenetic analysis that revealed unique spatial arrangement of CBS enzyme in nematodes; the domain architecture of CBS in Caenorhabditis elegans was studied experimentally in more detail. Finally, we determined conformational properties of a representative set of human CBS mutants that exhibited in various extent affected formation of tetramers and decreased catalytic activity. Using thermolysin-based proteolytic techniques for analysis of nine mutants expressed in E.coli, we found that an unfolded structure is a common intermediate occurring in CBS misfolding. Group of specialized cells that form cardiac conduction system is responsible for generation and coordinated propagation of the electrical impulse in the heart. Changes in its development can be connected with arrhythmias; therefore, a good level of knowledge is necessary and relevant for basic science and clinical practice. For correct development of the conduction system are important genes coding gap junctions proteins, ion channels, transcription factors and other molecules involved in signaling cascades (endothelin, neuregulin). Development of conduction system is determined in addition to genetic factors also by epigenetics and environmental factors. This thesis with its individual papers on which it is based is addressing different aspects of conduction system development, which appears to be a complex process. Another feature which is linking all papers together, is the methodological approach enabling us to study function of the conduction system - optical mapping. In the first publication we studied by the means of in vitro organ culture the impact of work load without interfering hemodynamics on the conduction system maturation in the chick embryonic heart.
