Místní akční skupina (MAS) Říčansko jako nástroj rozvoje venkova v daném regionu / The Local Action Group (LAG) Říčansko as a tool for rural development in the region

The thesis contains an evaluation of LAG as a tool for development of the rural region. The default hypothesis is that the LAG Říčansko has of its region and the influence of this effect can be evaluated positively. It followed the trends and developments in the field of rural development with a focus on the LEADER approach. Furthermore, for the final evaluation analyzes the structure of the LAG, the selection of supported projects and the implementation of cooperation projects. Part of the work is devoted to the characteristics and the current situation in the region LAG Říčansko. As a basis for the design of projects in problematic areas include a description of activities of some foreign LAG in Germany and Austria. The aim is achieved through interviews with workers LAG Říčansko and compares its structure and focus of the recommended and mandatory requirement for LAG. It was also the focus of supported projects compared with regard to problem areas in the region. The main part of the job evaluation LAG was a questionnaire survey among inhabitants of the region, partners and holders LAG regional brand Zápraží original product, which was focused on knowledge of the activities of the LAG and satisfaction with the treatment. Economic efficiency projects LAG was assessed using a cost-benefit analysis (CBA) of the selected project. Due to the nature of the project, its methodology was amended and as a result of its work is proposed for evaluating the effectiveness of cooperative projects using space CBA cost utility analysis (CUA). Knowledge about the work of LAG Říčansko are summarized in the conclusion that the MAS Říčansko has on the region a positive effect. Conclusion also includes the realization that this fact is only sustainable if the LAG will respond to changes in the region and its activities will involve local stakeholders in all sectors in sufficient representation.

As caracterizações físico-químicas dos vanadosilicatos foram realizadas utilizando-se técnicas como difração de raios-X (DRX), microscopia eletrônica de varredura (MEV), espectroscopia no infravermelho (FT-IR), espectroscopia Raman, espectroscopia no UV – Visível e ressonância magnética nuclear com rotação em torno do ângulo mágico (RMN-MAS) no estado sólido para os núcleos de 13C, 29Si e 51V. Os resultados experimentais indicam que as moléculas orgânicas podem atuar como agentes direcionadores de estrutura (SDAs). Os SDAs 1, 3, 4 e 5 direcionaram a formação dos vanadosilicatos VN1, VN3, VN4 e VN5 isomorfos ao titaniosilicato ETS-10 e ao vanadosilicato do tipo AM-6, enquanto os SDAs 2, 6, 7 e 8 direcionam a estrutura de novos vanadosilicatos (VN2, VN6, VN7 e VN8). Evidências experimentais da incorporação de vanádio na estrutura cristalina dos materiais foram obtidas por FT-IR, UV – Visível, 29Si RMN-MAS. Na espectroscopia no infravermelho (FT-IR) o sinal localizado na frequência 870 cm-1 é proveniente da vibração da ligação V-O e o sinal centrado na frequência 1030 cm-1 é proveniente da vibração dos grupos vanadila. Na espectroscopia no UV – Visível o comprimento de onda 600 nm representa os grupos vanadila e os comprimentos onda 450 e 545 nm indicam a presença de vanádios em coordenação octaédrica distorcida (VN2) e ordenada (VN1, VN3 e VN4), respectivamente. E no caso do RMN de 29Si o sinal centralizado em -93 ppm é proveniente de núcleos de silício da forma Si(3Si, 1V), estes dados comprovam a incorporação do vanádio na rede cristalográfica do material microporoso. A espectroscopia no UV – Visível juntamente com a ressonância magnética nuclear de 29Si sugerem que os vanadosilicatos isomorfos ao titaniosilicato VN1, VN3, VN4 e VN5 apresentam uma rede cristalográfica mais ordenada que os vanadosilicatos VN2, VN6, VN7 e VN8. Visto que a espectroscopia no UV – Visível indica que os octaedros VO6 dos vanadosilicatos isomorfos ao ETS-10 são mais regulares que os octaedros de vanádio dos novos vanadosilicatos, e os resultados experimentais de RMN-MAS de 29Si revelam que os vanadosilicatos isomorfos ao ETS-10 apresentam somente o sinal centralizado em -93 ppm, proveniente de núcleos de silício da forma Si(3Si, 1V), enquanto os novos vanadosilicatos apresentam, além do sinal centralizado em -93 ppm, um novo sinal centralizado em -110 ppm, proveniente de núcleos de silício da forma Si(4Si, 0V). / New vanadosilicates were synthesized in this work. They have been synthesized using organic molecules derivatives of 3,5-dimethylpiperidine (SDAs 1 – 4) and 2,6-dimethylpiperidine (SDAs 5 – 8) in their reactional mixture. The vanadosilicates were named using the same numbers of the SDA used in their synthesis (VN1 to VN8). Physicochemical characterization of the new vanadosilicates was carried out by X-ray diffraction (XRD), scanning electron microscopy (SEM), infrared spectroscopy (FT-IR), Raman spectroscopy, UV – Visible spectroscopy (UV – Vis), solid state 13C, 29Si and 51V magic angle spinning nuclear magnetic resonance (MAS NMR) spectroscopy. The experimental results suggest that the organic molecules can act as structure directing agents (SDAs). The SDAs 1, 3, 4 and 5 directed the synthesis to the formation of vanadosilicates VN1, VN3, VN4 and VN5 isomorphs to titanosilicate ETS-10 and vanadosilicate AM-6; on the other hand the SDAs 2, 6, 7 and 8 show selectivity to the formation of new vanadosilicates (VN2, VN6, VN7 and VN8). Experimental evidences of the vanadium incorporation into the vanadosilicate framework were obtained by FT-IR, UV – Vis and 29Si MAS NMR. The infrared spectroscopy has a signal at the frequency of 870 cm-1 designed to the vibration of V-O bond and the signal at the frequency of 1030 cm-1 designed to the vibration of vanadyl groups. The UV – Vis spectroscopy reveals the presence of vanadyl groups at 600 nm and the wavelengths 450 and 545 nm indicate the presence of distorted (VN2) and ordely (VN1, VN3 and VN4) octahedrally coordinated vanadium, respectively. And the 29Si MAS NMR shows a chemical shift centred at -93 ppm assigned to the Si(3Si, 1V) neighbourhood. UV – Vis spectroscopy and 29Si MAS NMR evidence that the vanadosilicates VN1, VN3, VN4 and VN5, isomorphs to ETS-10, show their framework more crystalline than the new vanadosilicates framework (VN2, VN6, VN7 and VN8). This affirmative is supported by both UV – Vis and 29Si MAS NMR data. UV – Vis data indicates that VO6 octahedra in vanadosilicates isomorphs to ETS- 10 are more regular than the VO6 octahedra in the new vanadosilicates. 29Si MAS NMR experimental data shows that vanadosilicates isomorphs to ETS-10 have only one chemical shift centred at -93 ppm, assigned to the Si(3Si, 1V) neighbourhood, while the new vanadosilicates have two chemical shifts that one centred at -93 ppm and other centred at -110 ppm assigned to Si(4Si, 0V) neighbourhood.

Atualmente, a comunidade científica está voltada para três principais estratégias para a regeneração cardíaca: uso de células tronco, inibição de genes que regulam o sistema de controle do ciclo celular e o aperfeiçoamento de mecanismos endógenos. Um exemplo de mecanismo endógeno é a estimulação de vias de sinalização que permite a reentrada dos miócitos no ciclo celular e sua posterior divisão. O treinamento físico (TF) é um fator estimulatório para a proliferação das células cardíacas por meio da angiotensina-(1-7) (Ang-(1-7)) via receptor Mas. Além disso, o TF promove cardioproteção e aumenta os níveis deste peptídeo em animais com sobrecarga pressórica. Existem evidências, recentes, que o TF promove proliferação de miócitos em diferentes modelos experimentais e atua sobre os níveis de Ang-(1-7) em corações disfuncionantes. Entretanto, não foram encontrados estudos que avaliaram, concomitantemente, os efeitos do TF sobre a função, níveis de Ang-(1-7) e receptor Mas miocárdicos e proliferação de cardiomiócitos em modelo de sobrecarga pressórica e deterioração da função cardíaca. Objetivo: Testar a hipótese que o TF atenua a queda do desempenho cardíaco, acarreta proliferação de cardiomiócitos e eleva os níveis de Ang-(1-7) e do receptor Mas miocárdicos em ratos com estenose aórtica supravalvar (EAo) e disfunção ventricular esquerda. Métodos: Ratos Wistar machos foram separados em dois grupos: controle operado (Sham) e EAo. Após 18 semanas do procedimento cirúrgico, os ratos EAo e Sham foram redistribuídos em 4 grupos e randomizados quanto a presença ou ausência do TF por 10 semanas; (Sham, n = 20), (EAo, n = 19), (ShamTF, n = 20) e (EAoTF, n = 15). A remodelação cardíaca foi caracterizada pelas análises estrutural e funcional por ecocardiograma na 18ª e 28ª semana e estudo macroscópico post mortem. A análise da capacidade funcional dos animais foi realizada pelo teste de esforço progressivo. A avaliação da proliferação dos cardiomiócitos, pelo conteúdo de DNA, foi feita por citometria de fluxo. O sistema renina angiotensina cardíaco foi analisado pela expressão proteica dos receptores Mas e AT1 por Western blot. Resultados: O TF promoveu melhoria na capacidade funcional dos animais Sham e EAo quanto a velocidade de exaustão, tempo total e distância percorrida. Ao iniciar o TF na 18º semana os animais EAo apresentavam disfunção diastólica, sistólica e hipertrofia ventricular esquerda concêntrica. Na 28ª semana, este grupo manteve o mesmo padrão de disfunção cardíaca. O TF não acarretou alteração na função diastólica e melhorou a estrutura e função sistólica dos animais EAo. Os animais ShamTF mostraram aumento da função sistólica comparados com o Sham. Não foram observadas diferenças entre os quatro grupos estudados em relação ao ciclo celular dos cardiomiócitos e expressão do receptor Mas no miocárdio. Houve diminuição da expressão do receptor AT1 no grupo EAo, e o TF não alterou este padrão no EAoTF. Conclusão: O treinamento físico melhorou a capacidade funcional e a função sistólica dos animais, sem repercussões benéficas sobre a função diastólica, proliferação de cardiomiócitos e expressão proteica do receptor Mas. / Introduction: In the last years, researchers have showed that it is possible to induce adult cardiomyocytes proliferation. Currently, the scientific community is focused on three main strategies for cardiac regeneration: stem-cell use, inhibition of genes that regulate cell-cycle control system and improvement of endogenous mechanisms. An example of endogenous mechanism is the stimulation of signaling pathways that allow the reentry of myocytes into the cell cycle and their posterior division. The physical training (PT) is a stimulatory factor for the proliferation of cardiac cells through angiotensin-(1-7) (Ang-(1-7)) via Mas-receptor. In addition, PT promotes heart protection and increases this peptide levels in animals with pressure overload. Recent evidences showed that PT promotes myocyte proliferation in distinct experimental models and acts on Ang-(1-7) levels in dysfunctional hearts. However, no studies were found that evaluated, concurrently, the effects of PT on cardiac function, myocardial levels of Ang-(1-7) and Mas receptor and cardiomyocyte proliferation in a model of pressure overload with deteriorated cardiac function. Objective: To test the hypothesis that PT attenuates the impairment in cardiac performance, leads to cardiomyocyte proliferation and increases myocardial Ang-(1- 7) and Mas-receptor levels in rats with supravalvar aortic stenosis (AS) and left ventricular dysfunction. Methods: Male Wistar rats were separated into two groups: operated control (Sham) and AS. After 18 weeks of the surgical procedure, AS and Sham rats were redistributed into four groups and randomized for the presence or absence of PT for 10 weeks; (Sham, n=20), (AS, n=19), (ShamPT, n=20) and (ASPT, n=15). Cardiac remodeling was characterized by structural and functional analysis by echocardiogram at the 18th and 28th week and by post-mortem macroscopic evaluation. The functional capacity of the groups was analyzed by the progressive treadmill running test. The cardiomyocyte proliferation was evaluated by DNA content using flow cytometry. The cardiac renin-angiotensin system was analyzed by protein expression of Mas and AT1 receptors by Western blot. Results: The PT improved the functional capacity of Sham and AS animals regarding the exhaust speed, total time and treadmill running distance. At the beginning of PT, at the18th week, the AS group presented diastolic and systolic dysfunction and concentric left ventricular hypertrophy. At the 28th week, AS group maintained the same pattern of cardiac dysfunction. The PT was not alter diastolic function and improved structure and systolic function in AS group. The ShamPT rats showed increased systolic function in relation to animals of Sham group. No differences were detected among the four groups regarding the cardiomyocyte cell cycle and the myocardial Mas-receptor expression. The expression of AT1 receptor was reduced in the AS group, and PT did not modify this pattern in ASPT group. Conclusion: The PT improved the functional capacity and systolic function of the rats, without beneficial repercussions on diastolic function, cardiomyocyte proliferation and protein expression of Mas receptor.

This thesis deals with rural development actors' social networks and individual social capital of mayors in municipalities belonging to local action groups Rozvoj Krnovska and Rýmařovsko. In my work I examine if there is any correlation between evaluation of funcionality of local action groups and degree of individual social capital of mayors in municipalities belonging to local action groups and between certain characteristics of actors' social networks. In the first part of the thesis I deal with rural areas' problems, development factors, social capital and social network in general. In the second part of the thesis I analyze results of survey, which took part in March 2017 between 34 mayors of municipalities belonging to local action group Rozvoj Krnovska and Rýmařovsko respectively. On the basis of survey results I continue with analyzing of mayors' individual social capital and types of social capital - bonding, bridging, linking. The study confirmed the key role of individual social capital (and bonding and bridging social capital) of rural municipality mayors for funcionality of local action groups. 17-O NMR on Crystalline Hydrades Hydrates: Impact of Hydrogen Bonding

The water molecules in inorganic hydrate salts adopt different geometries and are involved in different hydrogen bond interactions. In this work, magic-angle spinning (MAS) and static 17O solid-state NMR experiments are performed to characterize the 17O electric field gradient (EFG) and chemical shift (CS) tensors of the water molecules in a series of inorganic salt hydrates which include: oxalic acid hydrate, barium chlorate hydrate, sodium perchlorate hydrate, lithium sulphate hydrate, and potassium oxalate hydrate, which were all enriched with 17O water. Data were acquired at magnetic field strengths of 9.4, 11.75, and 21.1 T. Gauge-including projector-augmented-wave density functional theory (GIPAW DFT) calculations are performed on barium chlorate hydrate and oxalic acid hydrate where structural changes including the Ow-H•••O distance, H-O-H angle, and O-H distance are employed to understand their impact on the NMR parameters. Furthermore, simplified molecular models consisting of a metal cation and a water molecule were built to establish the effect the M-Ow distance has on the parameters. The computational studies are then used to understand the experimental results. The 17O quadrupolar coupling constant ranged from 6.75 MHz in K2C2O4•H2O to 7.39 MHz in NaClO4•H2O while the asymmetry parameter ranged from 0.75 in NaClO4•H2O to 1.0 in K2C2O4•H2O and the isotropic chemical shift ranged from -15.0 ppm in NaClO4•H2O to 19.6 ppm in BaClO3•H2O. The computational results revealed the trends for each parameter, where there is an increasing trend for quadrupolar coupling constant and span as a function of increasing hydrogen bond distance, decreasing trend for the three chemical shift tensors as a function of increasing M-Ow distance and unclear trends for asymmetry parameter and skew due to competing electronic factors. Overall, this study provides benchmark 17O NMR data for water molecules in crystalline hydrates, including the first measurement of 17O chemical shift anisotropy for such materials.

UAV units are by many researchers and aviation specialists considered the future and cutting edge of modern flight technology. This thesis discusses methods for efficient autonomous environmental mapping in a multi-agent domain. An algorithm that emphasizes on team work by sharing the agents local map information and exploration intentions is presented as a solution to the mapping problem. General theories on how to model and implement rational autonomous behaviour for UAV agents are presented. Three different human and tactical behaviour modeling techniques are evaluated. The author found the CxBR paradigm to be the most interesting approach. Also, in order to test and quantify the theories presented in this thesis a simulation environment was developed. This simulation software allows for UAV agents to operate in a visual 3-D environment with mountains, other various terrain types, danger points and enemies to model unexpected events.

Multi-Agent Based Control and Reconfiguration for Restoration of Distribution Systems with Distributed Generators

Restoration entails the development of a plan consisting of opening or closing of switches, which is called reconfiguration. This dissertation proposes the design of a fast and efficient service restoration with a load shedding method for land-based and ship systems, considering priority of customers and several other system operating constraints. Existing methods, based on centralized restoration schemes that require a powerful central computer, may lead to a single point of failure. This research uses a decentralized scheme based on agents. A group of agents created to realize a specific goal by their interactions is called a Multi-Agent System (MAS). Agents and their behaviors are developed in Java Agent DEvelopment Framework (JADE) and the power system is simulated in the Virtual Test Bed (VTB). The large-scale introduction of Distributed Generators (DGs) in distribution systems has made it increasingly necessary to develop restoration schemes considering DG. The separation of utility causes the system to decompose into electrically isolated islands with generation and load imbalance that can have severe consequences. Automated load shedding schemes are essential for systems with DGs, since the disconnection of the utility can lead to instability much faster than an operator intervention can repair. Load shedding may be the only option to maintain the island when conditions are so severe as to require correction by restoration schemes. Few algorithms have been reported for the problem of maintaining the island, even though load shedding has been reported for power systems using underrequency and under-voltage criteria. This research proposes a new operational strategy for sudden generator-load imbalance due to loss of utility that dynamically calculates the quantity of load to be shed for each island and the quantity of load that can be restored. Results presented in this dissertation are among the first to demonstrate a state-of-the-art MAS for load shedding under islanded conditions and restoration of the shed loads. The load shedding and restoration schemes developed here have behaviors that can incorporate most of the distribution topologies. Achieving service restoration with DG is complicated but new automated switch technologies and communications make MAS a better scheme than existing schemes.

An ex ante economic impact analysis of developing low cost technologies for pyramiding useful genes from wild relatives into elite progenitors of cassava

This study conducts an ex-ante economic impact evaluation of developing low cost technologies for pyramiding useful genes from wild relatives into elite progenitors of cassava in Nigeria, Ghana and Uganda. More specifically, it estimates the change in economic surplus generated by introducing cassava varieties with tolerance to cassava mosaic disease, green mites, whiteflies, and delayed post-harvest deterioration. It compares the economic benefits of marker-assisted selection (MAS) to conventional breeding for these traits. Results indicate that varieties developed with marker-assisted breeding that incorporate all three traits are worth US$2.89 billion in Nigeria, $854 million in Ghana, and $280 million in Uganda over 20 years. If these varieties were to be developed with tolerance to CMD and Green mites alone they would be worth US$1.49 billion in Nigeria, $675 million in Ghana, and $52 million in Uganda if developed through MAS. If developed solely by conventional breeding they would be worth about US$676 million in Nigeria, $304 million in Ghana, and $18 million in Uganda. The difference is mostly due to the faster timing of release for the varieties developed with MAS and the higher probability of success. Several sensitivity analyses were conducted and benefits for MAS range from US$1.7 billion to US$4.3 billion for all three traits depending on assumptions. MAS förklaras ingående i uppsatsens inledande kapitel, men är en del i Hofstedes mer övergripande kulturdimensionsteori, som ämnar att förklara kulturella skillnader mellan olika länder (De Moij & Hofstede, 2010). Kulturdimensionsteorin och MAS har prövats i flertalet tidigare interkulturella jämförelsestudier, där man jämfört medieinnehåll i två eller fler länder. Många av dessa studier har varit av kvantitativt slag, och har genom statistiskt säkerställda resultat kunnat legitimera teorin i flera avseenden (Wiles, Wiles & Tjernlund, 1995; An & Kim, 2007; Stoica, Miller & Ardelea, 2011). I vår studie har vi med hjälp av en kvalitativ innehållsanalys jämfört reklambilder publicerade i livsstilsmagasin i Sverige och USA. Vi har genom att titta på hur genus konstrueras i reklambilder i de båda länderna funnit att reklambilder i svenska livsstilsmagasin innehåller mer icke-stereotypa framställningar av män och kvinnor jämfört med reklambilder funna i amerikanska livsstilsmagasin. Studiens teoretiska grund bygger på ett socialkonstruktionistiskt ramverk, där genusteori och stereotyper utgör en fundamental utgångspunkt för studien. Analysen av materialunderlaget har gjorts med hjälp av bland annat Goffmans (1979) klassifikationer för könsframställning, där även feministisk blickanalys har inkluderats i analysverktyget. Studiens resultat är inte entydiga då det finns många likheter mellan reklambilder publicerade i svenska och amerikanska livsstilsmagasin, men det finns en märkbar skillnad i hur genus konstrueras i reklambilder i de båda länderna, något som går att härleda till den teoretiska grunden i Hofstedes MAS-dimension. / This study aims to – through a qualitative method – examine the reliability of Geert Hofstede’s theory of cultural dimensions. More specifically, we are interested in Hofstede’s dimension masculinity/femininity (MAS), which for one tries to explain differences regarding the view on sex and gender between different cultures – a more thorough explanation of MAS follows in the initial chapter of this thesis. There have been several prior studies testing the reliability of MAS; using a quantitative method they have compared media content and advertising in two or more countries, and the results have in most cases legitimized MAS and the theory of cultural dimensions (Wiles, Wiles & Tjernlund, 1995; An & Kim, 2007; Stoica, Miller & Ardelea, 2011). In this study we have compared advertising images published in lifestyle magazines in Sweden and the US using a qualitative text/content analysis. Through examining how gender is constructed in advertising images found in magazines from the two countries, we have found that images published in Swedish magazines contain more non-stereotypical portrayals of men and women than those published in American magazines. When we say more, we don’t mean as in a higher frequency, that would take a quantitative method to determine, but there are more unique themes regarding the construction of gender in Swedish advertising images compared to American. Gender theory and stereotypes have been theoretical foundations of this study, and the actual analysis of images has been conducted using feminist gaze analysis together with Goffman’s (1979) classifications of gender portrayal. The results are not unequivocal since we found similarities regarding gender construction in advertising images in the two countries; however, despite the similarities, there is also a noticeable difference in the construction of gender that can be linked to the theoretical foundation in Hofstedes MAS-dimension.
