Etudes d'interactions moléculaires par RMN dans les systèmes complexes : utilisation de la technologie HR-MAS pour l'étude de l'interaction protéine ligand / Molecular interactions study in complex systems by NMR

La RMN est un outil analytique extrêmement puissant pour l’analyse quantitative et structurale, et est très utilisée en biologie structurale. C’est dans ce contexte que nous avons choisi d’étudier les interactions par RMN. Le premier système choisi est un système de polymères, les PEO. Ils sont caractérisés par plusieurs grandeurs physiques dont l’indice de polydispersité. Cet indice représente la distribution de taille d’une population de polymères. Nous avons développé une nouvelle méthode de diffusion par RMN, la DOSY afin d’accéder à cet indice : J. Viéville et al. / Journal of Magnetic Resonance 212 (2011)169–173 Le deuxième système complexe étudié est un type de molécules chimiques mimétiques de l’ADN, les PNA, acides nucléiques peptidiques. Les PNA possèdent des bases nucléiques leur permettant de former des liaisons avec l’ADN ou l’ARN. Nous avons montré par des analyses de RMN que les PNA en solution peuvent former des complexes très stables avec des températures de fusion élevées. Ces analyses RMN sont complétées par des analyses de dichroïsme circulaire.Afin de compléter notre panel d’études sur les interactions moléculaires par RMN, nous avons étudié les interactions protéine-ligand avec greffage de la protéine sur phase solide : J.M.P. Viéville et al. / Journal of Pharmaceutical and Biomedical Analysis 89 (2014) 18–23 / NMR is a powerfull technique we decided to extend to follow interactions in complex mixtures. First part is about polydispersity index of polymer which is an important physical parameter when working with polymers. We developed here a new method, based on PEO analysis, using diffusion experiments (DOSY) by NMR to assess the polydispersity index: J. Viéville et al. / Journal of Magnetic Resonance 212 (2011) 169–173. In a second time, we worked on peptidic nucleic acids, PNA. These chemicals molecules are designed to bind DNA or RNA for clinical studies. We studied PNA in solution, by NMR to showwhich kind of interactions they form on themselves. We found very stable complexes with high fusion temperatures. Circular dichroism measurements were helpful for fusion temperature determination and structural studies. To complete this panel, we were interested in the study of protein-ligand interaction. Social reasoning in multi-agent systems with the expectation-strategy-behaviour framework

Multi-agent Systems (MAS) provide an increasingly relevant field of research due to their many applications to modelling real world situations where the behaviour of many individual, self-motivated, agents must be reasoned about and controlled. The problem of agent social reasoning is central to MAS, where an agent reasons about its actions and interactions with other agents. This is the most important component of MAS, as it is the interactions, cooperation and competition between agents that make MAS a powerful approach suited for tackling many complex problems. Existing work focuses either on specific types of social reasoning or general purpose agent practical reasoning - reasoning directed toward actions. This thesis argues that social reasoning should be considered separately from practical reasoning. There are many possible benefits to this separation compared to existing approaches. Principally, it can allow general algorithms for agent implementation, analysis and bounded reasoning. This viewpoint is motivated by the desire to implement social reasoning agents and allow for a more general theory of social reasoning in agents. This thesis presents the novel Expectation- Strategy-Behaviour (ESB) framework for social reasoning, which provides a generic way to specify and execute agent reasoning approaches. ESB is a powerful tool, allowing an agent designer to write expressive social reasoning specifications and have a computational model generated automatically. Through a formalism and description of an implemented reasoner based on this theory it is shown that it is possible and beneficial to implement a social reasoning engine as a complementary component to practical reasoning. By using ESB to specify, and then implement, existing social reasoning schemes for joint commitment and normative reasoning, the framework is shown to be a suitable general reasoner. Examples are provided of how reasoning can be bounded in an ESB agent and the mechanism to allow analysis of agent designs is discussed. Finally, there is discussion on the merits of the ESB solution and possible future work.

Evaluation of Agreement among Respondents to Anecdotal Assessments and Correspondence between Anecdotal and Experimental Analysis Outcomes

Study 1 evaluated agreement among five respondents using the Functional Analysis Screening Tool (FAST), the Motivation Assessment Scale (MAS) and Questions About Behavioral Function (QABF). Respondents provided ratings for 20 target behaviors exhibited by 10 individuals. At least 4/5 raters agreed on the primary maintaining variable in 80% of cases with the FAST, 70% of cases with the MAS, and 55% of cases with the QABF. Study 2 evaluated correspondence between results of anecdotal assessments and experimental functional analysis for 10 target behaviors selected from Study 1. Correspondence between the experimental functional analyses was 60% with the FAST and the MAS, 50% with the QABF.

Progressing From Multiple-respondent Anecdotal Assessments to Test-control Analyses of Problem Behavior

The current study was designed to evaluate the utility of progressing sequentially from multiple-respondent anecdotal assessments through test-control treatment analysis as an effective and efficient method of identifying the environmental determinants of problem behavior. the goal of the study was to evaluate overall agreement among multiple respondents on the primary function of aberrant behavior using the Motivation Assessment Scale (MAS) and Questions About Behavioral Function (QABF) and, if agreement was obtained, conduct a test-control evaluation to confirm anecdotal assessment findings while simultaneously evaluating the effects of function-based treatment. for 4 individuals, at least 4 of 5 respondents to the anecdotal assessments agreed (both within and across assessments) on the probable maintaining consequence for their problem behaviors. Test-control multielement evaluations were then conducted in which baseline sessions, corresponding to the suspected operant function of each individual’s problem behavior, were alternated with sessions in which the identified contingency was arranged for alternative behavior. Each evaluation showed substantial decreases in problem behavior and maintenance of alternative responses.

Within and beyond boundaries in Henri Bosco's Le mas Théotime

Spiritual, mystic and natural boundaries haunt the writings of Henri Bosco. Critics such as Bachelard have studied Bosco's interpretation of the natural elements and noted his sensitive portrayal of the protagonists with a focus on their unconscious desires by the use of this device. In Le Mas Theotime Bosco has unleashed his artistic and poetic creativity in addition to his passion for nature. Indeed, of all of Bosco's novels, perhaps this one exemplifies the quintessential harmony of man with nature. Le Mas Theotime incorporates Bosco's true genius as a writer who brings to his reader the possibility of a personal rendition and view of the story. His is not a straightforward and evident recollection of events, but rather a voyage through and involvement in the lives and minds of the characters.

Orientador: Antonio Carlos Cicogna / Resumo: Introdução: Nos últimos anos, investigadores têm demonstrado que há a possibilidade de estimular a proliferação de cardiomiócitos adultos. Atualmente, a comunidade científica está voltada para três principais estratégias para a regeneração cardíaca: uso de células tronco, inibição de genes que regulam o sistema de controle do ciclo celular e o aperfeiçoamento de mecanismos endógenos. Um exemplo de mecanismo endógeno é a estimulação de vias de sinalização que permite a reentrada dos miócitos no ciclo celular e sua posterior divisão. O treinamento físico (TF) é um fator estimulatório para a proliferação das células cardíacas por meio da angiotensina-(1-7) (Ang-(1-7)) via receptor Mas. Além disso, o TF promove cardioproteção e aumenta os níveis deste peptídeo em animais com sobrecarga pressórica. Existem evidências, recentes, que o TF promove proliferação de miócitos em diferentes modelos experimentais e atua sobre os níveis de Ang-(1-7) em corações disfuncionantes. Entretanto, não foram encontrados estudos que avaliaram, concomitantemente, os efeitos do TF sobre a função, níveis de Ang-(1-7) e receptor Mas miocárdicos e proliferação de cardiomiócitos em modelo de sobrecarga pressórica e deterioração da função cardíaca. Objetivo: Testar a hipótese que o TF atenua a queda do desempenho cardíaco, acarreta proliferação de cardiomiócitos e eleva os níveis de Ang-(1-7) e do receptor Mas miocárdicos em ratos com estenose aórtica supravalvar (EAo) e disfunção ventricular esquerda.
