A depressão em idosos é uma desordem persistente e recorrente, resultado do stress psicossocial ou efeito de doenças fisiológicas, que podem acarretar a desabilidade do indivíduo, aumento dos sintomas das doenças clínicas, na maior utilização dos serviços de saúde e altas taxas de suicídios.A monitorização terapêutica permite a ndividualização do regime de dosagem, assegurando a eficácia clínica e minimizando os efeitos adversos dos fármacos, prescritos na clínica. Os antidepressivos têm sido monitorados, pois, apresentam intervalos terapêuticos bem estabelecidos, ou seja, a maioria dos pacientes, que apresentam concentrações plasmáticas dentro deste intervalo fixo, tem as desordens psiquiátricas mantidas sob controle e efeitos adversos aceitáveis. Os antidepressivos tricíclicos (ADTs): imipramina, amitriptilina, nortriptilina e desipramina, embora eficazes e ainda muito utilizados, apresentam efeitos adversos, não desejáveis. Os antidepressivos, inibidores seletivos da recaptação de serotonina (SSRIs): citalopram, fluoxetina, paroxetina e sertralina, apresentam eficácia clínica comparável aos clássicos ADTs, mas destituídos dos efeitos adversos associados aos mesmos. Os métodos convencionais, empregados no tratamento de amostras biológicas, para análises de antidepressivos por técnicas cromatográficas, têm sido a extração líquido-líquido e extração em fase sólida. A microextração em fase sólida tem sido empregada em diferentes análises em fluidos biológicos, porém essa técnica apresenta certas limitações como, o número limitado de fases extratoras disponíveis no comércio que sejam adequadas para a análise de compostos não iônicos. A avaliação de novas fases extratoras, mais seletivas, estáveis e de baixo custo tem sido requerida para a análise de fármacos. O interesse no uso de poli(pirrol) (PPY), como fase extratora para SPME, está relacionado às diferentes interações dos fármacos (hidrofóbicas, -, com o grupo funcional polar, troca iônica, ácido-base) aos grupos multifuncionais deste polímero. Sua polimerização pode ser alcançada tanto por oxidação química, quanto por eletrodeposição em meio aquoso ou orgânico. O método eletroquímico apresenta algumas vantagens, tais como: o polímero ou mistura de polimeros podem ser revestidos diretamente em um metal, incorporação de diferentes grupos funcionais, entre outras, a polimerização pode ser eletroquimicamente controlada através da voltametria cíclica. Neste trabalho o poli(pirrol) (PPY) foi eletrodepositado em eletrodo de aço inox e empregado para a microextração em fase sólida dos antidepressivos: paroxetina, fluoxetina, mirtazapina, duloxetina, sertralina e citalopram. As variáveis da eletrodeposição, número de ciclos e contra-íon empregado no processo de eletrodeposição foram otimizadas. Assim como as variáveis SPME: tempo, temperatura, pH e volume de amostra, e tempo e solvente de dessorção; almejando maior sensibilidade para o método SPME-PPY/LC UV proposto. A extração sortiva em barra de agitação (SBSE), técnica recente de preparo de amostras, para a pré-concentração de compostos orgânicos presentes em amostras biológicas, baseia-se na extração estática, através do polímero polidimetilsiloxano (PDMS), no qual ocorre a dissolução (sorção) do analito. Neste trabalho, as técnicas SBSE e cromatografia líquida de alta eficiência com detecção UV (SBSE/LC UV) foram avaliadas para a análise simultânea de antidepressivos em amostras de plasma para fins de monitorização terapêutica. As condições cromatográficas de análise, assim como as variáveis SBSE de extração (tempo, temperatura, força iônica, pH da matriz) e tempo de dessorção, foram otimizadas, visando adequada sensibilidade analítica. A validação analítica foi realizada segundo normas da ANVISA, para ambos os métodos propostos, em diferentes concentrações plasmáticas, as quais contemplam o intervalo terapêutico. Segundo os parâmetros de validação avaliados, os métodos SBSE/LCUV e SPMEPPY/LCUV padronizados poderão ser empregados nas análises dos antidepressivos, para fins de monitorização terapêutica. / Depression in the elderly is a persistent and recurrent disorder resulting from psychosocial stress or physiological effect or disease. This condition can lead to disability, cognitive impairment, enhanced symptoms of medical illnesses, increased use of health care services and, increased of suicide rates. Therapeutic monitoring allows individualization of the dose regimen, ensuring clinical effectiveness and minimizing the adverse effects of drugs, prescribed at the clinic. Antidepressants have been monitored because they present well - established therapeutic intervals; in other words, most of the patients present plasmatic concentrations within this fixed range, so that their psychiatric disorders are kept under control and the adverse effects are acceptable. The tricyclic antidepressants (ADTs) imipramine, amitriptyline, nortryptiline, and desipramine, have adverse effects. The selective serotonin reuptake inhibitors (SSRIs) antidepressants citalopram, fluoxetine, paroxetine, and sertraline, are clinically effectiveness as the classic ADTs, but they do not lead to the adverse effects associated to the latter. The conventional methods employed in the treatment of biological samples for analysis of antidepressants by chromatographic techniques have been the liquid-liquid extraction (LLE) and solid phase extraction (SPE) techniques. Solid-phase microextraction (SPME) has been used in various analyses of biological fluids. However, this technique has limitations such as the small number of comemercially available extracting phases that are appropriate for the analysis of non-ionic compounds. Investigation of new extraction phases that are more selective and stable, as well as inexpensive, has been requested for drug analysis. The interest in the use of poly(pirrole) (PPY) as an extraction phase for SPME is related to the different interactions of the drugs (hydrophobic, - , with the polar functional group, ionic exchange and acid-base) with the multifunctional groups on this polymer. The PPY polymerization can be achieved by chemical oxidation or electropolymerization in aqueous solution or organic solvents. The electrochemical method has advantages such as, the polymer or a mixes of polymers can be directly deposited on a metal wire, different functional groups can be incorporated, polymerization can be electrochemically controlled by cyclic voltammetry. In this work poly(pyrrole) was electropolymerized on stainless steel electrodes and employed for the solid phase microextraction of the antidepressants paroxetine, fluoxetine, mirtazapine, duloxetine, sertraline, and citalopram. The electropolymerization variables, the number of cycles and counterion employed in the process were optimized, as well as the SPME variables, time, temperature, pH, sample volume, and desorption solvent; aiming at a better sensibility for the proposed method SPME-PPY/LC-UV. The stir bar sorptive extraction (SBSE), recently established technique for samples preparation, that targets the pre concentration of organic compounds present in biological samples. It is based on static extraction, through the polymeric polydimethylsiloxane (PDMS), where there is analyte dissolution (sorption). In this work, the SBSE technique and liquid chromatography with UV detector (SBSE/LC-UV) were evaluated for the simultaneous determination of antidepressants in plasma samples for therapeutic monitoring purposes. The cromatographic conditions, the SBSE extraction variables (time, temperature, ionic strength and matrix pH), and the dessorption time were optimized for appropriate analytical sensibility. The analytical validation was accomplished according to the norms of ANVISA for both of the proposed methods, in different plasmatic concentrations, which contemplate the therapeutic interval. According to the evaluated validation parameters, the standardized methods SBSE/LC-UV and SPME-PPY/LC-UV can be used in the analyses of antidepressants for therapeutic drug monitoring purposes.

Desenvolvimento de métodos para coleta e determinação de poluentes em fase gasosa da atmosfera utilizando amostragem por difusão com membranas capilares microporosas / Development of methods for sampling and determination of gaseous pollutants in the atmosphere using diffusional microporous capillary membrane scrubber

A tese enfatiza o desenvolvimento e aplicação de amostradores difusionais para pré-concentração de traços de poluentes gasosos e sua conjugação com técnicas de separação e determinação, especialmente, a eletroforese capilar. Foram implementadas novas aplicações para um amostrador com múltiplos filamentos microporosos de polipropileno, previamente desenvolvido pelo grupo de pesquisa, e para o novo dispositivo de coleta de dimensões reduzidas, composto por monofilamento microporoso de polipropileno (CMDS) com volume interno de 30 µL. Para o estabelecimento da vazão estável em 1,0 µL min-1 de fase aceptora pelo capilar poroso, recorreu-se à pressurização do reservatório com uma bomba pneumática de aquário e regulagem de vazão por um capilar de sílica fundida e uma válvula de agulha. Outra bomba pneumática foi utilizada para aspirar o fluxo de ar amostrado. As amostras foram coletadas seqüencialmente em frascos de 200 µL, mantidos sob temperatura reduzida em unidade de refrigeração do tipo Peltier acoplado ao amostrador. O CMDS monofilamentar, mais compacto e robusto, apresentou alta eficiência de coleta para a pré-concentração de formaldeído (CH2O), ácidos fórmico e acético e amônia da fase gasosa da atmosfera. Para as espécies com elevada constante de Henry, água deionizada serviu como fase aceptora, enquanto que para amônia, recorreu-se ao deslocamento do equilíbrio por um aceptor ácido, de modo a reter o analito na forma de NH4 +. A concentração das espécies em fase líquida foi determinada posteriormente por eletroforese capilar com detecção condutométrica sem contato (CE-C4D). Os limites de detecção, em fase líquida, para formiato, acetato, formaldeído (determinado na forma do aduto hidroximetanosulfonato HMS) e amônio foram estimados em 1,0, 1,5, 1,2 e 1,2 µmol L-1, respectivamente (equivalentes à 0,9, 3,0, 1,0 e 0,7 µg m-3 dessas espécies na atmosfera, respectivamente). Para a determinação de CH2O por injeção em fluxo (FIA) acoplada à detecção amperométrica em eletrodo de ouro platinizado, fez-se uso do coletor multifilamentar, por atender melhor à necessidade de volume de amostra. A interferência de espécies comumente presentes na atmosfera, como H2O2 e SO2, pôde ser contornada realizando coletas em presença de peróxido de hidrogênio para promover a oxidação do S(IV) à S(VI) e, posteriormente, destruindo o oxidante em reator contendo enzima catalase imobilizada. A determinação de CH2O foi implementada com sucesso por FIA com detecção por amperometria em eletrodo de ouro platinizado. Um CMDS portátil que funciona à pilha, próprio para amostragens em campo, foi desenvolvido utilizando, para o deslocamento do ar amostrado, uma bomba de pistão retirada de aparelho automático para medida de pressão arterial. O amostrador foi empregado na coleta de H2S, SO2 e alquil-mercaptanas em fase gasosa da atmosfera, utilizando fase aceptora alcalina para promover a desprotonação e conseqüente fixação das espécies em meio líquido. Os analitos foram determinados por CE-C4D ou por microextração em fase sólida (SPME) seguida de determinação por GC. O uso conjunto do antioxidante ascorbato para conservação das amostras e de etanol para fixação dos compostos voláteis em meio aquoso permitiu o estabelecimento de um protocolo completo para coleta e detecção dessas espécies reduzidas de enxofre, com sensibilidade suficiente para monitorar emissões de origem biogênica, o que foi exemplificado na prática coletando amostras próximo a um córrego contaminado com esgoto. A perfeita combinação do diminuto volume de fase aceptora do CMDS com a demanda de nanolitros de amostra da CE-C4D culminou com o desenvolvimento de um sistema de análise total (TAS) com gerenciamento de fluidos baseado em propulsão por bombas de aquário (de baixo custo e alta durabilidade) e válvulas solenóide de estrangulamento, controladas por computador. Como exemplo de aplicação inovador e bem sucedido do TAS desenvolveu-se a análise concomitante e em tempo quase real de ácidos fórmico e acético no ar, com freqüência de 10 pares de dados por hora. As vantagens do sistema CMDS-CE-C4D incluem simplicidade, versatilidade, consumo de reagentes e amostra e geração de resíduos minimizada, robustez e rapidez enquanto uma amostra é coletada, o eletroferograma da anterior é adquirido sem necessidade de bombas de alta pressão ou colunas dispendiosas como as requeridas para HPLC / The development and application of porous membrane diffusion samplers for fast and efficient pre-concentration of an array of trace air pollutants was emphasized in this thesis, in conjunction with compatible separation and determination techniques, especially capillary electrophoresis. New applications were found for a formerly developed diffusion scrubber based on a bundle of microporous hollow polypropylene capillary membranes and for a scaled down version with a single core capillary membrane diffusion scrubber (CMDS) comprising an internal volume of 30 µL, were used for sampling of the trace level pollutants to an adequate liquid acceptor. The low-flow of acceptor solution, 1.0 µL min-1, required by the CMDS was satisfied by pressurization of the reservoir with an aquarium pump combined with flow regulation by a silica capillary as hydrodynamic resistor and a needle valve. Another aquarium pump was used for the aspiration of the sampled air through the sampler. The low volumes collected in the CMDS were stored in 200 µL vials inserted in a cooling plate of a Peltier device. The robust and compact system was used for sampling of formaldehyde (CH2O), formic acid, acetic acid and ammonia in the gaseous phase of the atmosphere. For chemical species with high Henry´s constant, deionized water suffices as acceptor phase. Otherwise, equilibria displacement to a non-volatile ion, like NH4 + for NH3 sampling, promoted quantitative retention in the acceptor phase. The concentrations of the analytes in the liquid phase were determined by capillary electrophoresis with capacitively coupled contactless conductometric detection (CEC4D). The detection limits obtained in the liquid phase for formate, acetate, formaldehyde (in the form of the adduct hydroxymethanesulfonate HMS) and ammonium were 1,0, 1,5, 1,2 e 1,2 µmol L-1 respectively (what corresponds to 0,9, 3,0, 1,0 e 0,7 µg m-3 of the respective gaseous species in the air). The higher volume of acceptor phase provided by the sampler with a bundle of microporous membrane capillaries (~600 µL) are in tune with the needs of flow injection analysis (FIA), as demonstrated for amperometric detection of CH2O on a platinized gold electrode. The interferences from SO2 and H2O2 were overcome by adding H2O2 to the acceptor solution to promote the oxidation of S(IV) to S(VI) and destruction of the oxidant afterwards in a column with immobilized catalase enzyme. The aquarium pump used for gas aspiration was substituted to a piston pump, taken from arterial blood pressure meter fed with 8 V D.C., to turn possible field collections with the CMDS device. The system was used for the sampling of H2S, SO2 e alkyl-mercaptans in the gaseous phase of the atmosphere. To succeed on that, the samplings were performed in alkaline media to promote deprotonation of the species (and stabilization in non-volatile forms). The collected analytes were determined by CE-C4D or by solid phase microextraction (SPME) followed by GC analysis. The joint use of the antioxidant ascorbate for sample preservation and ethanol for fixation of the volatile compounds allowed the establishment of a complete protocol for sampling, storage and detection of the sulfur reduced species with enough sensitivity for monitoring biogenic emissions from waste discharge. Perfect matching of the low-volume characteristics of the CMDS device and the CE-C4D equipment led to the conception of a low-cost automatic CMDS-CE-C4D total analysis system (TAS) and its innovative and successful application to near-realtime simultaneous analysis of formic acid and acetic acid in air. During the evaluation of one sample, the TAS collects a new one, with valves, pumps and high voltage delivery under software control. Advantages include rapidity (10 data points per hour for each analyte), high preconcentration efficiency, simplicity and versatility, minimum sample and reagent consumption and residue generation (green analytical method), no need of costly high pressure pumps and separation columns like those used in HPLC

Redistribuição postmortem de antidepressivos e seus produtos de biotransformação em tecidos biológicos humanos / Postmortem redistribution of antidepressants and their metabolites in human biological tissues.

Os antidepressivos pertencem a uma importante classe de medicamentos investigados na toxicologia forense. Em casos de amostras provenientes de cadáveres, o intervalo entre o óbito e a obtenção da espécie biológica pode proporcionar a redistribuição postmortem destes fármacos. Com o objetivo de elucidar esse fenômeno, métodos analíticos foram desenvolvidos e aplicados utilizando sangue total (ST), humor vítreo (HV) e fígado. Para as amostras de ST e HV, o método de extração escolhido e validado foi a microextração em fase líquida (LPME) trifásica. Fibras ocas constituídas de polipropileno, com a extensão de 8 cm cada, foram tratadas com o solvente orgânico dodecano (fase orgânica), resultando em um membrana com permeabilidade seletiva. No lúmen destas fibras, adicionou-se ácido fórmico 0,1 mol/L (fase aceptora). Em frasco de fundo chato com 5 mL de capacidade, pipetou-se 3,5 mL de NaOH 0,1 mol/L (fase doadora) e 0,5 mL de ST ou HV. Ao término da extração, as amostras foram introduzidas no GC-MS, sem a necessidade de reações de derivatização. O estudo com ST contemplou os antidepressivos amitriptilina (AMI), nortriptilina (NTR), imipramina (IMI), desipramine (DES), clomipramina (CLO), desmetilclomipramina (DMC), fluoxetina (FLU) e norfluoxetina (NFL). Os limites de quantificação para estas substâncias ficaram inferiores aos níveis terapêuticos (20 ng/mL). As médias dos coeficientes de variação intradia e interdia foram, respectivamente, de 9,7 e 9,8%. As curvas de calibração apresentaram linearidade entre as concentrações de 20 até 1200 ng/mL. A validação do parâmetro integridade da diluição assegurou a mensuração de quantidades superiores ao limite apresentado na curva de calibração. O método foi aplicado em sete amostras reais postmortem e em apenas um caso foi observada uma diferença significativa (300%) entre os valores quantificados no ST periférico e central. Os antidepressivos tricíclicos AMI, NTR, IMI e DES foram avaliados no HV e o efeito matriz foi detectado para os dois últimos analitos. O método foi otimizado e validado utilizando solução salina adicionada de AMI e NTR. O limite de detecção igual a 5 ng/mL, foi obtido com a redução da voltagem da fonte de íons do espectrômetro de massa para 50 eV. Coeficientes de variação foram inferiores a 15%. Os procedimentos validados foram aplicados em seis amostras reais de HV. A relação encontrada entre os valores obtidos no ST periférico e HV foi de aproximadamente 0,1. A extração acelerada por solvente (ASE) e, posteriormente, a extração em fase sólida (SPE) foram as técnicas de separação dos analitos da matriz fígado. Ao término das citadas extrações, os antidepressivos foram analisados no GC-MS. Para esta matriz sólida, são necessários mais estudos, pois os valores encontrados nos ensaios analíticos estão em desacordo com as diretrizes utilizadas na validação dos métodos. / Antidepressants belong to an important class of drugs investigated in forensic toxicology. In cases of samples from corpses, the interval between death and obtaining the biological specimens can provide the postmortem redistribution of these drugs. Aiming to elucidate this phenomenon, analytical methods were developed and applied using whole blood (WB), vitreous humor (VH) and liver. For samples of WB and HV, the extraction method chosen and validated was the three-phase liquid phase microextraction (LPME). Hollow fibers consist of polypropylene, with a length of 8 cm each were treated with dodecane organic solvent (organic phase) resulting in a membrane with selective permeability. Into the lumen of these fibers was added formic acid 0.1 mol/ L (acceptor phase). In the vial containing 3.5 mL of NaOH 0.1 mol / L (donor phase) was spiked 0.5 ml of biological fluids (WB or VH). Subsequently, the samples were injected in GC-MS without derivatization reactions. The study of the ST included antidepressants amitriptyline (AMI), nortriptyline (NTR), imipramine (IMI), desipramine (DES), clomipramine (CLO), desmethylclomipramine (DMC), fluoxetine (FLU) and norfluoxetine (NFL). The quantification limits for these substances were below the therapeutic levels (20 ng / ml). The mean coefficients of variation and separate intradays were respectively 9.7 and 9.8%. The calibration curves showed linearity between concentrations of 20 to 1200 ng / mL. The validation of the integrity of the dilution parameter assured measurement higher than the limit shown in the calibration curve quantities. The method was applied to seven real postmortem samples and in one case a significant difference (300%) between the measured values in the peripheral and central ST was observed. The tricyclic antidepressants AMI, NTR, IMI and DES were evaluated in VH and the matrix effect was detected in the last two analytes. The method was optimized and validated using saline spiked AMI and NTR. The limit of detection (5 ng/ml) was obtained by reducing the voltage of the ion source of the mass spectrometer 50 eV. Coefficients of variation were below 15%. The procedures were validated in six real samples of HV. The relationship found between the values obtained in the peripheral ST and HV was approximately 0.1. Accelerated solvent extraction (ASE) and subsequently the solid phase extraction (SPE) were the techniques of separation of analytes liver matrix. At the end of the cited extractions, antidepressants were analyzed in GC-MS. To this solid tissue, further studies are needed, because the values found in the analytical tests were not in accordance with the guidelines used in the validation of the methods.

Eletrofiação de nanofibras de fibroína da seda como dispositivos adsorventes para microextração em fase sólida / Silk fibroin nanofibers electrospun as adsorbents device for solid phase microextraction

The parameters of electrospinning process were evaluated through factorial experimental design 22. This study showed the variables flow of solution and capillary-collector distance were statistically significant in the medium diameter response. The model obtained was validated through variance analysis (ANOVA) and response surface methodology. The material was used at solid phase micro extraction (SPME) in the extraction of samples containing a small chain alcohol, applied at gas chromatography (GC). The recovered device was characterized through Fourier transform infrared spectroscopy with attenuated total reflectance (FTIR-ATR) and thermal analysis (DSC and TGA). The RSF nanofibers covered fibers were morphologic evaluated through scanning electronic spectroscopy (MEV). The results of MEV showed that was possible to obtain fibers in nanoscale, medium diameter around 304 ± 46 nm. The fibers were also submitted to simulated thermal treatment (100°C to 250°C). There was no fusion between the fibers until 250°C (4 hours of treatment), and a little structural damage. The thermal analysis of FRS and covered FRS nanofibers device showed that the electrospun material maintained the thermal stability, with loss due thermal degradation from 250°C. The GC assay demonstrated the standard SPME covered device didn´t interfere on the suggested method to the alcohol isopropyl analysis (AIS). The times of extraction and desorption were optimized in 20 minutes and 10 minutes, respectively. The methodology was evaluated about linearity and specificity. In the concentration between 10 to 500 ppm of AIS, the method was linear, with R2= 0,9927 and selective, with high resolution between peaks of methanol, AIS and ethyl acetate. Electrospinning process of RSF as covering device showed great potential to use in SPME due its high surface area, thermal stability and easiness process, as well as potential to application in extraction of alcohols and GC analysis. / A técnica de eletrofiação foi aplicada no revestimento de fibras de sílica fundida por nanofibras de fibroína de seda regenerada (FSR). Os parâmetros do processo de eletrofiação foram avaliados por meio de delineamento experimental fatorial 2². Este estudo mostrou que as variáveis vazão da solução e distância entre capilar-coletor foram estatisticamente significativas na resposta diâmetro médio. O modelo obtido foi validado através da análise de variância (ANOVA) e metodologia de superfície de resposta. O material foi empregado em Microextração em Fase Sólida (Solid Phase Microextraction SPME ) na extração de amostras contendo um álcool de cadeia pequena, aplicado em cromatografia gasosa (CG). O dispositivo recoberto foi caracterizado por meio de espectroscopia na região do infravermelho (FTIR-ATR) e análises térmicas (DSC e TGA). As fibras recobertas com nanofibras de FSR foram avaliadas morfologicamente através de microscopia eletrônica de varredura (MEV). Por meio dos resultados das análises de MEV, observou-se que foi possível obter fibras em escala nanométrica, com diâmetro médio em torno de 304 ± 46 nm. As fibras também foram submetidas a tratamento térmico em forno (100°C a 250°C). Não houve fusão das fibras até 250°C (até 4 h de tratamento), e poucos danos estruturais. As análises térmicas da FSR e do dispositivo recoberto com as nanofibras de FSR mostraram que o material eletrofiado manteve a estabilidade térmica, com perda por degradação térmica a partir de 250 °C. Os ensaios em CG demonstraram que o branco com o dispositivo de SPME recoberto não interferiu no método proposto para a análise de álcool isopropílico (AIS). Os tempos de extração e dessorção do analito foram otimizados em 20 min e 10 min, respectivamente. A metodologia foi avaliada quanto à linearidade e especificidade. Na faixa de concentração de 10 a 500 ppm de AIS, o método foi linear, com R=0,9927 e seletivo, apresentando alta resolução entre os picos de metanol, AIS e acetato de etila. O processo de eletrofiação da FSR como recobrimento do dispositivo mostrou grande potencial para emprego em SPME devido a sua grande área superficial, estabilidade térmica e facilidade no processo, bem como potencial para aplicação em extração de alcoóis e análise em CG.

Etude des interactions matière organique dissoute : contaminants organiques dans l'environnement aquatique

Suite à une anthropisation généralisée des ressources aquatiques et des écosystèmes associés, on retrouve de nombreux contaminants chimiques organiques à l’état de traces dans l’environnement aquatique. En parallèle de ces contaminants, dans les milieux aquatiques on retrouve également des molécules rassemblées sous la dénomination « matière organique dissoute (MOD) », qui sont des mélanges complexes de molécules et macromolécules, qui peuvent s’associer à ces contaminants et influencer leur comportement et leur distribution. Les interactions entre MOD et contaminants peuvent s’avérer difficiles à étudier étant donné qu’elles sont dépendantes de nombreux facteurs tels que la nature des contaminants et de la MOD, ainsi que des paramètres physico-chimiques du milieu. L’objectif de ces travaux était donc d’étudier ces interactions. Les contaminants organiques choisis ont été les Hydrocarbures Aromatiques Polycycliques (HAP) et les substances pharmaceutiques. D’autre part, plusieurs types de MOD d’origine naturelle ou commerciale ont été utilisés. Pour étudier les interactions, plusieurs techniques analytiques ont été développées dont la micro-extraction sur phase solide couplée à la chromatographie en phase gazeuse et à la spectrométrie de masse et l’extinction de fluorescence. / In relation with a widespread human impact on aquatic resources and ecosystems, many organic chemical contaminants are present at trace levels in aquatic environments. In parallel to these contaminants, dissolved organic matter (DOM), which is composed of complex mixtures of molecules and macromolecules, is also found and could be associated with these contaminants and influence their behavior and distribution. Interactions between DOM and contaminants can be very difficult to understand because they depend on many factors such as the nature of contaminants and DOM, as well as on environmental physico-chemical parameters. The aim of this work was thus to study these interactions. Organic contaminants studied were Polycyclic Aromatic Hydrocarbons (PAHs) and pharmaceuticals. On the other hand, several types of natural and commercial DOM have been used. To study the interactions, several analytical techniques have been developed like solid phase microextraction coupled to gas chromatography and mass spectrometry and fluorescence quenching.

Eletroquímica de interface óleo/água : aplicações e estudos fundamentais de parâmetros cinéticos e termodinâmicos

Primeiramente, pretendeu-se compreender as diferencas na oxidacao de uma molecula modelo (ferroceno) por meio de estudos eletroquimicos relacionados a cinetica e termodinamica em diferentes situacoes. A primeira situacao enfoca a oxidacao direta do ferroceno em um eletrolito de suporte convencional aquoso e a segunda e relacionada ao ferroceno sendo adicionado diretamente em oleo mineral que quando em contato com o eletrolito de suporte aquoso, forma uma interface do tipo oleo/agua. Experimentos adicionais tambem foram conduzidos em uma emulsao, que foi constituida por ferroceno em oleo misturados com eletrolitos convencionais e cloreto de tetrabutilamonio (CTBA). Uma placa de diamante dopado com boro (DDB) foi usada como eletrodo de trabalho. Observou-se um deslocamento de aproximadamente 0,07 V quando a oxidacao do ferroceno foi conduzida na interface oleo/agua em comparacao com a oxidacao direta na fase aquosa convencional. Esta mudanca de potencial tambem foi calculada usando a Teoria do Funcional da Densidade (DFT) como ferramenta teorica. Uma comparacao adicional foi realizada em relacao a interface e as emulsoes, constatando que a forca ionica e um fator determinante nesses tipos de estudos. Finalmente, as curvas de Tafel indicaram que as coordenadas da reacao do ferroceno sao diferentes quando o eletrodo e colocado na interface versus diretamente na fase aquosa, sendo que a energia de ativacao obtida para o ferroceno na agua foi de 129 kJ mol L-1 e na interface igual a 53,0 kJ mol L-1. Posteriormente, mostrou-se o uso da microextracao liquido-liquido dispersiva (DLLME) combinada com a eletroquimica de interface oleo/agua, utilizando ferroceno como molecula- modelo. Uma mistura de 150 ¿ÊL de etanol (solvente dispersante) e 50 ¿ÊL de 1-undecanol (solvente de extracao) foi injetada em 9 mL de uma solucao aquosa de ferroceno preparada em H2SO 4 1,0 x10 -3 mol L -1. Uma gota da fase organica contendo ferroceno foi inserida diretamente na superficie de um eletrodo de carbono impresso, e uma curva analitica foi obtida, ponto a ponto, com boa linearidade. O eletrodo de DDB foi utilizado para promover o calculo do coeficiente de particao, em que foi possivel calcular as concentracoes finais de ferroceno nas fases aquosa e organica como sendo iguais a 4,2 ~ 10 -4 mol L-1 e 0,15 mol L -1, respectivamente. Ao usar estes valores, o coeficiente de particao (log P) de 2,5 foi calculado. Por fim, a eletroquimica de interface oleo/agua foi aplicada para estudar a influencia do acido naftenico na corrosao do aco API-5L-X70 a temperatura ambiente a partir de um sistema bifasico constituido por petroleo/solucao aquosa. Um estudo in-situ baseou-se na espectroscopia de impedancia eletroquimica (EIS), enquanto medidas localizadas de impedancia eletroquimica (LEIS) e microscopia eletroquimica de varredura (SECM) foram utilizadas para estudar o efeito da pre-exposicao do aco ao sistema bifasico. Os resultados de LEIS e SECM mostraram que a pre-exposicao ao petroleo com acido naftenico produz uma superficie mais ativa do que apenas a solucao de sulfato. A adicao de acido naftenico ao oleo provoca corrosao. A morfologia da corrosao e caracterizada por tres regioes: i) na fase de oleo; ii) na fase aquosa e iii) em torno da interface. A corrosao foi mais evidente em torno da interface. Propoe-se um mecanismo no qual as moleculas de acidos naftenicos se auto-organizam na interface oleo/agua, gerando acidificacao local no contato trifasico entre oleo / fase aquosa / metal. / This work describes the realization of fundamental electrochemical studies at a liquid / liquid interface. Firstly, it was intended to understand the differences in the oxidation of a model molecule (ferrocene) through electrochemical studies related to kinetics and thermodynamics in different situations. The first situation focuses the direct oxidation of ferrocene in an aqueous conventional support electrolyte and the second one is when ferrocene is directly placed in mineral oil, subsequently placed in contact with an aqueous support electrolyte thereby forming an oil/water interface. Additional experiments were also conducted in an emulsion, which was constituted by ferrocene in oil and mixed with conventional electrolytes and tetrabutylammonium chloride (TBAC). A Boron-Doped Diamond Electrode (BDD) was used as the working electrode. We observed an approximately 0.07 V shift when the ferrocene oxidation was conducted at the oil/water interface compared with the direct oxidation in the conventional aqueous phase. This potential shift was also calculated using the Density Functional Theory (DFT) as a theoretical tool. An additional comparison was conducted related to the interface and emulsions, finding that the ionic strength is determining factor in these types of studies. Finally, Tafel plots indicated that reaction coordinates of the ferrocene are different when the electrode is placed at the interface versus directly in the aqueous phase, in which the activation energy obtained for ferrocene in water was 129 kJ mol L-1 and at the interface equal to 53.0 kJ mol L-1. Posteriorly, we showed the use of dispersive liquid-liquid microextraction (DLLME) combined with organic/water interface electrochemistry by using ferrocene as a model-molecule. A mixture of 150 ¿ÊL of ethanol (disperser solvent) and 50 ¿ÊL of 1-undecanol (extractant solvent) was injected into 9 mL of a ferrocene aqueous solution prepared in 1.0~10.3 mol L.1 H2SO4. Afterward, a drop of the organic phase containing ferrocene was directly inserted on a screen-printed carbon electrode and the analytical curve was obtained, point-to-point, with a good linearity. Besides that, a BDD electrode was used to promote the partition coefficient calculation, and it was possible to calculate the final concentrations of ferrocene in the aqueous and organic phases as being equal to 4.2~10.4 mol L.1 and 0.15 mol L.1, respectively. By using these values, the partition coefficient (log P) was calculated as 2.5. Finally, the oil/water interface was applied to study the role of naphthenic acid in the API.5L.X70 steel corrosion at room temperature from a biphasic system consisting of petroleum/aqueous solution. An in-situ study was based on electrochemical impedance spectroscopy, while localized measures of electrochemical impedance (LEIS) and scanning electrochemical microscopy (SECM) were used for the effect of pre-exposure from steel to biphasic system. The results LEIS and SECM showed that the pre-exposure to oil with acid produces a more active surface than just sulfate medium. Corrosion morphology is characterized by three regions: i) in oil phase; ii) in the aqueous phase, and iii) around the interface. Naphthenic acid corrosion was restricted to the area around the interface. A mechanism is proposed in which naphthenic acids self-assemble at the oil/water interface, generating local acidification at the oil/aqueous phase/metal three-phase contact.

Synthetic musk compounds are artificial organic substances commonly used as fragrant constituents of personal care products as parfums, cosmetics, detergents, in-house cleaning and washing agents. Given their large area of applications and their ability to be persistent, they leaked into all parts of ecosystem, especially aquatic one. There have been a big focus on studiyng these compounds, their properties and fate in the different parts of ecosystems in the last years. The aim of master's thesis was carry out the determination of five representatives (habanolide, exaltolide, ambrettolide, musk MC4 and ethylene brassylate) of macrocyclic musk compounds in waste water samples from influent and effluent of three water treatment plants (WWTP Brno-Modřice, WWTP Lednice and WWTP Mikulov). One of the main tasks was to perform a method optimisation for the determination of selected musk compounds in waste water and evaluation and interpretation of the results. Solid phase microextraction (SPME) technique was used for the extraction of analytes, consequently gas chromatography-mass spectrometry was used for analysis of selected analytes. Following evaluation the removal efficiency of musk compounds in the waste water treatment plant was assessed.

This work was focused on the determination of musk compounds in various components of the environment. Synthetic musk compounds are organic substances commonly used as fragrant constituents of detergents, perfumes, cosmetics and other personal care products (PCPs). These compounds contaminate the aquatic environment through municipal wastewater from sewage treatment plants. The result is accumulation of these analytes in the environment and their occurrence in food chain. Synthetic fragrances are commonly detected also in human body; these substances generally belong among relatively new persistent organic pollutants (POPs) because of their persistence, bioaccumulation, and potential toxicity. The aim of this work was to optimize the methods for the determination of selected synthetic fragrances in wastewater (municipal, industrial) and in fish muscle. These analytes were determined in wastewater [the wastewater treatment plant (WWTP) Brno – Modřice, cosmetic manufacturing plant] and in fish Squalius cephalus that had been caught in the river Svratka. The selected analytes (14 substances) originate from the group of nitromusk, polycyclic musk and linear musk compounds. Solid phase microextraction (SPME) [for wastewater samples] and pressurized solvent extraction (PSE) [for fish samples] were used for isolation of analytes from real samples. The identification and quantification of analytes were carried out by high resolution gas chromatography coupled with mass spectrometry detection (GC-MS). The results show that galaxolide (g.L-1) and tonalide (tenths of g.L-1) were polycyclic musk compounds which were found at the highest concentration level in influent and also in effluent samples from the WWTP Brno – Modřice. The most extended nitromusk compound was musk ketone (hundredths of g.L-1). Linear musk compounds were detected in all influent samples (g.L-1). The overall removal efficiency of synthetic musk compounds from wastewater in the WWTP varies within the following ranges: nitromusk compounds 34.4 % – 85.4 %, polycyclic musk compounds 36.7 % – 53.3 % and linear musk compounds 90.5 % – 99.9 %. On the basis of the results it can be stated that wastewater (effluent from the WWTP) is the primary source of musk compounds, especially of the polycyclic ones in the environment. It has been also shown that ultrafiltration used in a cosmetic plant is a highly efficient technology for removing nitromusk and polycyclic musk compounds from industry wastewater. The results from evaluation of the fish contamination by musk compounds show that galaxolide was the substance which reached the highest concentration levels (tens of g.kg-1 l.w.). The results corroborate ubiquitous occurrence of musk compounds in the environment and reduction of using of nitromusk compounds due to their potential toxic properties. It has been shown that the insufficiency of the purifying procedures used in the WWTP Brno – Modřice has a significant influence on the concentration level of polycyclic musk compounds (tonalide, galaxolide) in fish caught from the river Svratka.

Identification of Tobacco-Related Compounds in Tobacco Products and Human Hair

Indiana University-Purdue University Indianapolis (IUPUI) / Analyses of tobacco products and their usage are well-researched and have implications in analytical chemistry, forensic science, toxicology, and medicine. As such, analytical methods must be developed to extract compounds of interest from tobacco products and biological specimens in order to determine tobacco exposure. In 2009, R.J. Reynolds Tobacco Co. released a line of dissolvable tobacco products that are marketed as a smoking alternative. The dissolvables were extracted and prepared by ultrasonic extractions, derivatization, and headspace solid phase microextraction (SPME) with analysis by gas chromatography-mass spectrometry (GC-MS). The results show that the compounds present are nicotine, flavoring compounds, humectants and binders. Humectant concentrations vary among different tobacco types depending on the intended use. Humectants were quantified in various tobacco types by GC and “splitting” the column flow between a flame ionization detector (FID) and an MS using a microfluidic splitter in order to gain advantage from the MS’s selectivity. The results demonstrated excellent correlation between FID and MS and show that MS provides a higher level of selectivity and ensures peak purity. Chemometrics was also used to distinguish products by tobacco type. Hair is a common type of evidence in forensic investigations, and it is often subjected to mitochondrial DNA (mtDNA) analysis. Preliminary data was gathered on potential “lifestyle” markers for smoking status as well as any indications of subject age, gender, or race by investigating the organic “waste” produced during a mtDNA extraction procedure. The normally discarded organic fractions were analyzed by GC-MS and various lipids and fatty acids were detected. At this point, a total vaporization-SPME (TV-SPME) method was theorized, developed, and optimized for the specific determination of nicotine and its metabolite, cotinine. The theory of TV-SPME is to completely vaporize an organic extract which will eliminate the partitioning between the sample and the headspace, thereby simplifying the thermodynamic equilibrium. Parameters such as sample volume, incubation temperature, and extraction time were optimized to achieve the maximum analyte signal. Response surface methodology (RSM) is a statistical model that is very useful in predicting and determining optimum values for variables to ensure the ideal response. RSM was used to optimize the technique of TV-SPME for the analysis of nicotine and cotinine. Lastly, quantitation of nicotine and cotinine in human hair typically requires large sample sizes and extensive extraction procedures. Hence, a method using small sample sizes and a simple alkaline digestion followed by TV-SPME-GC-MS has been developed. Hair samples were collected from anonymous volunteers and nicotine and cotinine were identified and quantitated in the hair of tobacco users.
