Microcavidades ópticas à base de silício: projeto, confecção e propriedades / Silicon based optical microcavities: project, construction and properties

Estruturas fotônicas à base de silício têm despertado grande interesse por permitirem uma perfeita integração entre processos ópticos e eletrônicos em um único chip. Além de compatíveis com a atual indústria microeletrônica, acredita-se, que tais estruturas possam aumentar consideravelmente a velocidade de processamento de informações. Uma microcavidade óptica é um exemplo de estrutura fotônica simples. Feita à base de silício, e dopada com íons terra-rara, tal microcavidade pode intensificar a emissão gerada pelo íon e resultar em um dispositivo de importância tecnológica. O projeto-construção deste tipo de microcavidade deve considerar a sua região de funcionamento e os materiais a serem utilizados. Deve contemplar, ainda, algumas condições externas que, eventualmente, possam alterar o seu funcionamento. Uma dessas condições é descrita pelo chamado coeficiente termo-óptico que indica a dependência do índice de refração com a temperatura. Dentro desse contexto a presente Tese de Doutorado foi dedicada ao estudo de microcavidades ópticas com janelas de transmissão em 650 nm e em 1550 nm – correspondendo às regiões de menores perdas de fibras ópticas de plástico e de sílica. As microcavidades compreenderam espelhos de Bragg [camadas alternadas de silício amorfo (a-Si) e nitreto de silício amorfo (a-SiN)], um filme de a-SiN como espaçador, e foram depositadas sobre substratos de sílica pela técnica de sputtering. As cavidades MC-Er e MC-ErYb tiveram como espaçadores filmes de a-SiN dopados com Er e Er + Yb, respectivamente. Medidas de fotoluminescência da MC-ErYb na região do infravermelho próximo indicaram um aumento de 48 vezes na emissão dos íons Er3+ (em ~1535 nm) comparado a um filme de a-SiN dopado com érbio devido: (1) a presença do itérbio e, (2) às múltiplas reflexões sofridas pela luz nos espelhos de Bragg. As microcavidades cujos espaçadores eram a-SiN puro foram submetidas a medidas de transmissão óptica em função da temperatura de medida. O deslocamento da janela de transmissão devido às variações de temperatura permitiu determinar o valor do TOC do a-SiN como: (6.2±0.1)×10-5 ºC-1 (em ~ 620 nm) e, (4.7±0.1)×10-5 ºC -1 (em ~ 1510 nm). Até onde sabemos, o TOC do a-SiN no VIS foi determinado pela primeira vez neste trabalho. / Silicon photonic based structures have attracted great interest for allowing a perfect integration between optical and electronic process in a single chip. Besides being compatible with the actual microelectronic industry, it is believed that such structures can considerably increase the information processing speed. An optical microcavity is an example of a simple photonic structure. Made based on silicon, and doped with rare-earth ions, such microcavity may enhance the emission generated by the ion and become a device of technological importance. The project-construction of this kind of microcavity has to consider the operation region and the materials used. It still has to take into account external conditions that may, eventually, change its operation. One of these conditions is described by the thermo-optic coefficient (TOC) that shows the dependence of the refractive index with the temperature. Within this context the present PhD thesis was dedicated to the study of optical microcavities with transmission windows at 650 nm and at 1550 nm – corresponding to the low losses regions of the plastic and silica optical fibers. The microcavities comprised Bragg mirrors [alternated layers of amorphous silicon (a-Si) and amorphous silicon nitride (a-SiN)], one film of a-SiN as spacer, and were deposited on silica substrates by the sputtering technique. The MC-Er and MC-ErYb cavities had Er and Er+Yb-doped a-SiN films as spacers, respectively. Photoluminescence measurements of the MC-ErYb in the near infrared showed an enhancement of 48 times in the emission of the Er3+ ions (at ~ 1535 nm) compared with an Er-doped a-SiN film owing to: (1) the presence of ytterbium and, (2) the multiple reflections experienced by the light at the Bragg mirrors. The microcavities whose spacers were pure a-SiN were submitted to optical transmission measurements as a function of the measurement temperature. The shift of the transmission window due to variations in the temperature allowed determining the a-SiN TOC: (6.2±0.1)×10-5 ºC-1 (at ~ 620 nm) and, (4.7±0.1)×10-5 ºC -1 (at ~ 1510 nm). To the best of our knowledge, the a-SiN TOC in the visible was determined for the first time in this work.

Efeito da irradiação com laser de diodo sobre adesivos dentinários / Effect of diode laser irradiation on dentin-bonding agents

A irradiação de sistemas adesivos com laser tem obtido resultados promissores, no entanto, seu mecanismo de ação, bem como a longevidade da camada híbrida obtida após a irradiação com laser ainda não estão bem estabelecidos na literatura. A presente tese teve como objetivo: avaliar a absorvância do laser de Diodo por diferentes sistemas adesivos por meio de espectroscopia no infravermelho próximo; avaliar a evaporação de água e solventes proporcionada pelo laser por meio da variação da massa analisada em balança de precisão; avaliar a resistência de união por meio de teste de microtração após envelhecimento de 12 meses. Os dados obtidos foram analisados por modelos de ANOVA, seguidos de teste Tukey (p<0,05) e os resultados demonstraram que: a irradiação no infravermelho próximo, área do espectro eletromagnético que compreende o laser de Diodo, não apresenta interação com os sistemas adesivos testados; o laser de Diodo é capaz de proporcionar maior alteração de massa, mesmo após contínua evaporação dos solventes com jatos de ar por 60s; sistemas adesivos simplificados apresentaram valores de resistência de união imediato semelhantes aos valores obtidos após 12 meses de envelhecimento. Os dados obtidos neste estudo sugerem que a irradiação com laser de Diodo pode contribuir favoravelmente para a adesão em dentina com sistemas adesivos simplificados após 1 ano. / The laser irradiation of adhesive systems has obtained promising results, however, its mechanism of action, as well as the longevity of the hybrid layer obtained after laser irradiation are still not yet well established in the literature. This thesis aimed to: assess the diode laser interaction with different adhesive systems by means of near infrared spectroscopy; evaluate the evaporation of solvents and water provided by the laser through the mass variation analyzed at a precision balance; evaluate the bond strength through microtensile bond strength test after 12 months of aging. The data was analyzed through ANOVA models, followed by Tukey test (p<0.05), and the results demonstrated that: in the near-infrared irradiation area, the electromagnetic spectrum comprising the diode laser, shows no interaction with the adhesive systems tested; the diode laser was able to provide greater mass variation, even after continual evaporation of solvents with air jets for 60s; simplified adhesive systems presented immediate bond strength values similar to the ones verified after 12 months of aging. The data obtained in this study suggest that the Diode laser irradiation can contribute favorably for the adhesion between dentin and simplified adhesive systems after 1 year.

O aumento no consumo de energia induz um acréscimo na demanda por glicose e oxigênio no tecido extravascular. Fatores bioquímicos e neuronais induzem variações nos vasos sanguíneos que resultam em alterações de uxo sanguíneo, volume e oxigenação. Embora extensivamente investigada, esta cascata de eventos ainda é pouco compreendida. Neste projeto, procuramos descrever o acoplamento entre os níveis celular, metabólico e vascular associado à ativação funcional do cérebro. Usando medidas eletro fisiológicas, modelamos os sistemas neuro-vascular e neuro-metabólico para descrever a hemodinâmica cerebral medida através de técnicas ópticas. Resultados obtidos em ratos durante a estimulação de um fio de bigode mostraram que a determinação de uma função resposta para cada sistema, assumido como linear, descreve bem o comportamento hemodinâmico e possibilita o estudo dos estados vascular e metabólico caracterizados pelos parâmetros medidos. A partir de experimentos multimodais de NIRS e fMRI, desenvolvemos metodologias inovadoras para a determinação de imagens metabólicas, capazes de prever variações do consumo de oxigênio com boa resolução espacial e temporal. Por fim, analisamos a in u^encia de parâmetros fisiológicos no sinal óptico, mostrando a contribuição importante da pressão sanguínea na composição deste. Medidas de correlação temporal foram projetadas para gerar mapas de correlação vascular que podem ser aplicados ao estudo da conectividade vascular cerebral, tanto em indivíduos normais como em pacientes com patologias cerebrais. / Abstract: Task-associated neuronal activity leads to a complex chain of biological events within the brain. The increased energetics gives rise to elevated glucose and oxygen consumption in the tissue. Biochemical and neuronal factors induce changes in blood vessels and variations in blood ow, volume and oxygenation. Although it has been extensively investigated, this cascade of events is still poorly understood and highly debated. In this project, the aim was to describe the coupling among the cellular, metabolic and vascular levels associated to functional brain activation. Using electrophysiological measurements, we modeled the neuro-vascular and neuro-metabolic systems in order to describe cerebral hemodynamics as seen through optical techniques. Results obtained in rats during whisker-barrel stimulation showed that the determination of a response function for each system, assumed as linear, can describe the hemodynamic behavior and allow the study of the vascular and metabolic states characterized by the measurements. From multimodal experiments of NIRS and fMRI, we developed unique methods to the determination of metabolic images, which can predict changes in oxygen consumption with good temporal and spatial resolution. Finally, we analyzed the in uence of the physiology in the optical signal, and showed the importance of taking into account blood pressure oscillations into this signal. Variações na circulação sanguínea cerebral decorrentes de ativação neuronal podem ser detectadas e registradas através da técnica óptica denominada NIRS (near infrared spectroscopy). Neste trabalho, desenvolvemos estudos sobre a aplicabilidade da técnica NIRS sob três diferentes abordagens, envolvendo indivíduos saudáveis e portadores de uma condição patológica específica. Os estudos com indivíduos saudáveis foram focados na aplicabilidade da técnica NIRS para investigar alterações hemodinâmicas associadas à estimulação do córtex visual e à realização de exercício físico. No caso envolvendo condição patológica, investigamos a aplicabilidade clínica desta mesma técnica para avaliar a perfusão cerebral de pacientes com estenose carotídea. Nos estudos sobre estimulação do córtex visual, buscou-se estabelecer uma relação entre a frequência do estímulo apresentado ao voluntário e parâmetros obtidos das curvas hemodinâmicas. Nos experimentos associados a exercícios físicos, ciclistas semiprofissionais e indivíduos fisicamente ativos realizaram testes em bicicletas simulando tipos específicos de provas, envolvendo tarefas abertas (teste progressivo) e fechadas (teste contra relógio em uma prova de 4Km), em diferentes condições (controle, ingestão de placebo ou cafeína), buscando caracterizar diferenças em aspectos da hemodinâmica cerebral a elas associadas. Por fim, nos experimentos envolvendo indivíduos com estenose carotídea, foram realizadas medidas de NIRS, durante a realização de testes de apneia com duração de 30 segundos, buscando estabelecer a aplicabilidade da técnica na avaliação clínica deste tipo de patologia. Para cada uma dessas abordagens, foram estabelecidos parâmetros associados às respostas hemodinâmicas obtidas através de tarefas e estímulos específicos, que permitiram caracterizar e quantificar os processos fisiológicos envolvidos em cada tipo de experimento, demonstrando assim a aplicabilidade da técnica NIRS para o estudo da atividade cerebral sob as condições experimentais em questão / Abstract: Brain activity involves a complex network of neurophysiological processes whose energy demand requires constant supply of glucose and oxygen which is provided by the cerebral microcirculation. Changes in cerebral blood flow due to neuronal activation can be detected and recorded by the optical technique called NIRS (near infrared spectroscopy). In this work, we developed studies on the applicability of the NIRS technique under three different approaches, involving healthy subjects and patients with a specific pathological condition. Studies in healthy subjects were focused on the applicability of the NIRS technique to investigate hemodynamic changes associated with stimulation of the visual cortex and the performance of physical exercise. In the case involving pathological condition, we investigated the clinical applicability of this same technique to evaluate cerebral perfusion in patients with carotid stenosis. In studies of stimulation of the visual cortex, we sought to establish a relationship between the frequency of the stimulus presented to volunteer and parameters obtained from the hemodynamic curves. In the experiments associated with exercise, semi-professional cyclists and physically active subjects performed tasks on bicycles simulating specific types of tests, involving open tasks (progressive test) and closed (a 4km test against the clock), under different conditions (control, placebo or caffeine intake), seeking to characterize differences in aspects of cerebral hemodynamics associated with them. Finally, in experiments involving individuals with carotid stenosis, NIRS measurements were carried out during apnea tests lasting 30 seconds, from which we sought to establish the applicability of the technique in the clinical evaluation of this type of pathology. Novel perspectives in near-infrared optical imaging with lanthanide based molecules, macromolecules and nanomaterials / Nouvelles perspectives en imagerie optique proche-infrarouge avec des molécules, macromolécules et nanomatériaux incorporant des cations lanthanides

Le domaine du proche infrarouge a une importance majeure pour l'imagerie optique en raison de la faible autofluorescence des systèmes biologiques et de la diffusion limitée de la lumière permettant l’amélioration du rapport signal-sur-bruit et une pénétration plus profonde dans les tissus. Les sondes fluorescentes proche-infrarouges les plus couramment utilisées à ce jour sont les fluorophores organiques ou les nanocristaux semi-conducteurs qui sont affectés par un certain nombre d’inconvénients. Plusieurs cations lanthanide possèdent des propriétés optiques uniques comme des bandes d'émissions étroites dont les longueurs d'ondes sont très peu sensibles à l’environnement, de grandes différences d’énergie entre longueurs d’onde d’excitation et d'émission et une forte résistance au photoblanchiment. Afin de bénéficier de la luminescence des lanthanides, les faibles absorbances des cations lanthanides libres doivent être compensées par l'utilisation de groupes chromophores appropriés fonctionnant comme sensibilisateurs. On parle d’effet d’"antenne". Nous présentons dans ce travail plusieurs familles de sondes à base de lanthanides (i) de petites molécules, plus spécifiquement des complexes monométalliques constitués par la coordination de Ln³⁺ par un ligand de type TTHA-anthraquinone et des métallacrowns polymétalliques Ln³⁺/Zn²⁺ auto-assemblés; (ii) de nouveaux nanomatériaux basés sur des billes de polystyrène possédant deux types de chargements différents, à savoir (i) un complexe bimétallique d-f Cr³⁺-Ln³⁺ ainsi que (ii) des antennes dérivées d’hydroxyanthraquinone et des cations lanthanide libres; (iii) des macromolécules de type dendrimère polyamidoamine de génération 3 fonctionnalisés avec sensibilisateurs aza-BODIPY en leurs périphéries et qui encapsulent des lanthanide luminescents. Les propriétés photophysiques, la cytotoxicité et l'incorporation cellulaire ont été décrites et discutées pour chacun des types de sondes. En outre, ces sondes ont été testées en imagerie in vitro visible et/ou proche-infrarouge par des expériences de microscopie confocale et d’épifluorescence. L’ensemble des sondes décrites dans ce travail, en raison de leurs propriétés avantageuses et nouvelles, constituent des avancées majeures pour le développement de nouvelles générations d'agents d'imagerie optique. / The near-infrared (NIR) region has a significant importance for optical imaging due to the minimal autofluorescence and reduced light scattering thus allowing for improved signal-to-noise ratio and deeper penetration through tissues. Nowadays, the most commonly used NIR-emitting fluorescent probes rely mainly on organic fluorophores or quantum dots and exhibit drawbacks. Several lanthanide(III) ions (Ln³⁺) possess unique optical properties e.g. sharp emission bands the wavelengths of which have minimal sensitivity to the microenvironment, large differences between excitation and emission wavelengths and strong resistance toward photobleaching. In order to obtain the luminescence of lanthanides, the low absorbances of the free Ln³⁺ have to be overcome by the use of appropriate chromophoric groups functioning as a sensitizing “antenna”. We present here several families of Ln³⁺-based probes: (i) small molecules, in particular monometallic complexes constituted by the TTHA-anthraquinone moiety and Ln³⁺, and polymetallic self-assembled Ln³⁺/Zn²⁺ metallacrowns; (ii) novel nanomaterials based on polystyrene beads with two different loadings, i.e. d-f bimetallic Cr³⁺- Ln³⁺+ complex and hydroxyanthraquinone antennae and Ln³⁺ ; (iii) macromolecular generation-3 polyamidoamine dendrimers functionalized with aza-BODIPY sensitizers at their periphery and encapsulating the luminescent Ln³⁺. The photophysical properties, cytotoxicity and cellular uptake were reported and discussed for each of the presented types of Ln³⁺-based probes. Moreover, these probes were successfully tested for visible and/or NIR in vitro imaging by confocal or epifluorescence microscopy experiments. Finally, the reported Ln³⁺-based probes, due to the number of advantageous properties, represent significant breakthroughs toward the developments of new generations of optical imaging agents.

Synthèses et propriétés de dibenzoBODIPYs : orthogonalité et chiralité / Syntheses and properties of dibenzoBODIPYs : orthogonality and chirality

Lors de ces trois années de thèse, le développement de nouvelles molécules luminescentes, reposant sur la stabilisation d’un squelette dibenzoBODIPY a été réalisé. Dans un premier temps, la modulation des propriétés photophysiques a été effectuée (par introduction de groupement aromatique et/ou donneur soit lors de la formation du dibenzoBODIPY, soit par couplage croisé). Dans un second temps, nous nous sommes intéressés à la synthèse de dibenzoBODIPYs dissymétriques. La dissymétrie des composés a été réalisée soit par condensation mixte de dicétone soit par mono-couplage des dibenzoBODIPYs. L’utilisation de ces derniers a permis d’obtenir des dibenzoBODIPYs chiraux (soit par formation de liaison C-B, soit par formation de liaison B-O). La mono-fonctionnalisation intramoléculaire par le groupement phénol formé in situ est la première à ce jour. Enfin la formation de nouveau BODIPYs β-fusionnés par un hétérocycle a été entrepris. Nous avons synthétisé le premier BODIPY β-fusionné par un benzofurane. / During this PhD, the development of new luminescent molecules based on the stabilization of a dibenzoBODIPY scaffold was performed. Firstly, the modulation of the photophysical properties was carried out by the introduction of an aromatic and/or a donor group, either during the formation of dibenzoBODIPY or by cross-coupling. Secondly, we were interested in the synthesis of asymmetrical dibenzoBODIPYs. The dissymmetry of compounds was done either by mixed condensation of a diketone or by dibenzoBODIPYs mono-coupling. The use of the latters yielded chiral dibenzoBODIPYs either by C-B bond formation, or by B-O bond formation. Intramolecular mono-functionalization phenol formed in situ has never been reported so far. Finally, the synthesis of β‑fused BODIPYs with a heterocycle was undertaken. Déshydratation de la théophylline : analyse gravimétrique statique et suivi par spectroscopie proche infrarouge du séchage par contact sous vide en lit agité / Dehydration of theophylline : static gravimetric analysis and monitoring of a contact vacuum stirred bed drying by near infrared spectroscopy

Le but de ce travail était d'étudier le comportement des cristaux de théophylline lors de la déshydratation statique en monocouche dans différentes conditions hygrothermiques et le comportement du même produit lors de la déshydratation séchage en lit agité dans un séchoir par contact sous vide dans des conditions hygrothermiques semblables. Tout d'abord, la forme hydratée et les formes déshydratées de la théophylline ont été identifiées par DSC, ATG et DRXP. Ensuite, des essais gravimétriques statiques à température et humidité relative constantes ont été effectués et les évolutions de teneur en eau au cours du temps ont été enregistrées à des températures de 20 à 80 °C et des humidités relatives de 4 à 50%. Les données à l'équilibre thermodynamique ont été utilisées pour tracer les isothermes de désorption et établir le diagramme de phase solide-solide de la théophylline. Enfin, les cinétiques de déshydratation ont été étudiées. Il a été constaté que la constante de vitesse de déshydratation augmente exponentiellement avec la température et diminue de façon exponentielle avec l'humidité relative. Un modèle semi-empirique permettant la prédiction de l'évolution dans le temps de la teneur en eau du produit pour différentes conditions opératoires a été développé. Dans la seconde partie du travail, il a été d'abord démontré que la SPIR couplée avec la régression PLS permet de suivre en ligne et in situ la composition exacte du mélange des différentes formes de la théophylline (et d'eau) pendant le séchage. Ensuite, l'influence des paramètres de fonctionnement (température et l'activité de l'eau) sur les cinétiques de transformation de l'état solide a été étudiée. Il a été observé que la déshydratation commence d'abord par la formation de la forme anhydre métastable puis, après un temps de latence, continue par la formation de la forme anhydre stable. Il s'est avéré aussi que la température a été le principal facteur contrôlant la vitesse globale du procédé, mais aussi la teneur finale de la forme anhydre stable et de la forme métastable, ceci étant valable pour le séchoir étudié et pour les conditions opératoires considérées. Finalement, un modèle semi-empirique permettant la simulation de l'état du produit (température, teneur en eau, teneur en forme hydratée, anhydre métastable et stable) au cours du temps pour différentes conditions opératoires du séchage (pression dans la cuve et température du fluide caloporteur) a été proposé / The aim of this work was to investigate the dehydration process of a single static layer of theophylline's crystals under different hygrothermal conditions and the dehydration-drying process of a stirred bed of theophylline in a vacuum contact dryer in nearly the same hygrothermal conditions. First of all, the hydrated and dehydrated forms of theophylline were identified using DSC, TGA and PXRD devices. Next, the static gravimetric experiments at constant temperature and relative humidity were carried out and the water content evolutions with time were recorded at temperatures from 20 to 80°C and humidities from 4 to 50%. Then, the thermodynamic equilibrium data were reported as desorption isotherms and solid-solid phase diagrams. Finally, the dehydration kinetics were analyzed. The dehydration rate constant was found to increase exponentially with temperature and to decrease exponentially with relative humidity. A simple semi-empirical model allowing for prediction of the product water content time evolution time for different processing parameters was developed. In the second part of the work, it was first proven that NIRS coupled with PLS regression enabled to monitor inline and in-situ the exact composition of the mixture of different forms of theophylline (and of water) during drying. Second, the influence of operating parameters (temperature and water activity) on the kinetics of solid state transformations was investigated. It was observed that the dehydration begun by the formation of the metastable anhydrate and, after a time delay, continued by the formation of the stable one. It was also pointed out that the temperature was the main process factor controlling not only the overall process rate but also the final contents of the stable and metastable anhydrates, this observation being valid for the considered dryer and for the considered operating conditions. Finally, a semi-empirical model allowing for simulation of the product state (temperature, hydrate content, metastable and stable anhydrate contents and water content) time evolution for different processing parameters (pressure in the vessel and temperature of the heating fluid) was proposed

L’évaluation de la performance des ouvrages hydrauliques en remblai est un enjeu majeur pour les gestionnaires en charge de leur sécurité. Qu'il s'agisse de digues ou de barrages, la présence de végétation arborescente représente un facteur de fragilité important. Deux types de mécanismes de détérioration peuvent ainsi être initiés ou aggravés : l’érosion interne et l’érosion externe. Or, ce facteur n’a été pris en compte que partiellement dans les modèles d’évaluation existants. De plus, les petits barrages en remblai peuvent avoir des dispositifs d’étanchéité différents et ne sont pas toujours équipés de drains et d’instruments d’auscultation: il est alors nécessaire d’adapter les modèles à chaque cas. L’objectif de la thèse est de répondre à ces verrous scientifiques. Les résultats de nos recherches concernent l’établissement de modèles d’évaluation de la performance basés sur des approches à base de connaissances.La démarche a été appliquée sur des ouvrages réels : 3 tronçons de digues de protection contre les inondations ainsi que 7 barrages en remblai ce qui nous a permis de montrer sa pertinence et donc son potentiel d’utilisation opérationnelle. / The performance assessment of embankment hydraulic works is a major challenge for managers in charge of their security. For both dikes and dams, the presence of trees represents an important fragility factor. Two types of deterioration mechanism can thus be initiated or aggravated: internal erosion or scouring. However, this factor has been taken only partially into account in existing assessment models. In addition, small embankment dams can have different sealing devices and are not always equipped with drains and monitoring instruments: it is thus necessary to customize the models in each case. The aim of the thesis was to remove these scientific obstacles. The results of our research concern the design of performance assessment models relying on knowledge-based approaches.The approach was implemented for real works: three sections of river dikes and seven small embankment dams which allowed us to show its relevance and therefore its operational potential.

Integrating near-infrared spectroscopy to synchronous multimodal neuroimaging:applications and novel findings

Abstract Brain disorders such as epilepsy, dementia and other mental illnesses induce increasing costs on health care systems with aging populations. The most effective treatment of these disorders would be either prevention or intervention of the disorder before irreversible damage develops. However, despite the increased interest in different brain diseases, many of them are still detected too late. One reason for this is the lack of appropriate functional imaging modality that can critically sample the targeted physiological phenomenon. Furthermore, it has been shown that one imaging modality is not enough to cover brain functionality properly; a multimodal approach is required. The main goal of this thesis was to validate near-infrared spectroscopy (NIRS) for brain measurement and to integrate it into a multimodal neuroimaging setup that can critically sample basic human physiological phenomena. A novel key element was the combined use of NIRS with ultra-fast magnetic resonance encephalography (MREG), electroencephalography (EEG), continuous non-invasive blood pressure and anesthesia monitoring as a synchronous system. This unique multimodal neuroimaging set-up with a new functional magnetic resonance imaging sequence, MREG, can sample human brain physiology at 10 Hz sampling rate without cardiorespiratory aliasing. The implemented setup was successfully used in scanning multiple patient and control populations. With the help of critical sampling rate, non-stationarity between the measured signals reflecting brain pulsations could be detected. Combined NIRS and EEG showed the capability to monitor therapeutic opening of the blood-brain barrier during treatment of central nervous system lymphoma for the first time in humans. Furthermore, our multimodal neuroimaging setup enabled the mapping of the recently described brain avalanches and glymphatic pulsation mechanisms of the brain. In conclusion, the ultra-fast multimodal laboratory with integrated NIRS offers novel and more comprehensive views on basic brain physiology. The measures from this thesis also have the potential to offer new, quantitative biomarkers for the detection of different brain disorders prior to irreversible damage. / Tiivistelmä Aivosairaudet kuten epilepsia, dementia ja muut mielenterveyden häiriöt aiheuttavat kasvavissa määrin kuluja ikääntyvien ihmisten terveydenhuollossa. Näiden tautien tehokkain hoitokeino olisi joko ennaltaehkäisy tai varhainen havaitseminen ennen peruuttamattomien kudosvaurioiden kehittymistä. Lisääntyneestä kiinnostuksesta huolimatta monet aivosairaudet havaitaan edelleen liian myöhään. Osasyy tähän on sopivan toiminnallisen kuvausmenetelmän puuttuminen, jolla voitaisiin kuvata haluttu fysiologinen ilmiö riittävän nopeasti. Onkin osoitettu, ettei yksittäinen kuvausmenetelmä riitä aivojen toiminnan riittävän tarkkaan ymmärtämiseen, vaan siihen tarvitaan eri menetelmien yhdistämistä. Tämän väitöskirjatutkimuksen päätarkoituksena oli arvioida lähi-infrapunaspektroskopian (NIRS) soveltuvuutta aivojen toiminnan mittaamisessa sekä integroida se osaksi multimodaalista neurokuvantamisjärjestelmää. Uutena elementtinä NIRS:iä käytettiin yhdessä ultranopean magneettiresonanssienkefalogrammin (MREG), aivosähkökäyrän (EEG), jatkuva-aikaisen kajoamattoman verenpaineen mittauksen ja anestesiamonitoroinnin kanssa samanaikaisesti, ajallisesti synkronoituna. Yhdessä uuden toiminnallisen magneettikuvaussekvenssin, MREG:n, kanssa tällä ainutlaatuisella multimodaalisella neurokuvantamisjärjestelmällä voidaan kuvata ihmisen aivojen perusfysiologiaa 10 Hz näytteistysnopeudella ilman sydämen sykkeen ja hengityksen laskostumista. Toteutetulla multimodaalisella mittausjärjestelmällä tehtiin useita onnistuneita kuvauksia eri potilasryhmillä ja terveillä koehenkilöillä. Kriittisen näytteistämisen ansiosta voitiin havaita epästationaarisuutta aivojen pulsaatioita heijastelevien signaalien välillä. NIRS:n ja EEG:n samanaikainen mittaaminen mahdollisti ensimmäistä kertaa ihmisen veriaivoesteen aukeamisen monitoroinnin keskushermostolymfoomapotilaiden hoidossa. Lisäksi multimodaalinen neurokuvantamisjärjestelmä mahdollisti hiljattain havaittujen aivojen vyöryjen (engl. avalanches) ja glymfaattisten pulsaatioiden kartoittamisen. Yhteenvetona voidaan todeta, että väitöskirjatyön aikana toteutettu multimodaalinen laboratorio yhdessä NIRS:n kanssa mahdollistaa aivojen perusfysiologian edistyksellisen ja tarkan tutkimisen. Nyt kehitetyt mittarit saattavat myös tarjota uusia, kvantitatiivisia biomarkkereita eri aivosairauksiin ennen vakavien vaurioiden syntymistä.

Stacked Dual Narrowband Organic Near-Infrared Photodetectors

Compared with the detector devices made of inorganic semiconductors, organic photodetectors are granted with additional strengths, such as flexibility, high scalability and bio-compatibility. However, in the family of organic optoelectronic devices, the detectors that are capable of detecting photons at two or multiple specific wavelengths are still missing. Such photodetectors are highly interesting because they could identify the target objects or materials much more precisely by detecting the reflected, transmitted or emitted photons at two or multiple characteristic wavelengths. In this thesis project, the optical simulations using Transfer Matrix Method (TMM) were performed on the organic devices to achieve the dual wavelength narrowband detection in the near-infrared (NIR) spectral range of 700 ~ 1100 nm. The devices use the fact that, at the interface of the blended organic electron donating and accepting materials, the charge-transfer (CT) states with the transition energies that are lower than the optical gap of the neat materials are formed. Combined with a Fabry-Perot resonant cavity, the CT absorption can be dramatically enhanced at certain wavelengths. The simulation results show that the two detection wavelengths can be tuned independently from 650 to 1100 nm. The spectral resolution (full with at half maximum - FWHM) of the detection bands varies between 10 and 30 nm. The simulated external quantum efficiency (EQE) is ~35% at 700 nm and ~10% at 1000 nm, respectively. A possible application of such photodetectors is for example moisture detection, where two of the characteristic absorption peaks of water are located at around 750 and 960 nm. By optimizing the thickness of the two photo-absorbing layers in a tandem device structure, the detection bands can be tuned to match with those two wavelengths for simultaneous and precise detection.
