The effect of the 5-HT1A receptor agonist, 8-OH-DPAT, on motion-induced emesis in Suncus murinus.

No / In the present study we evaluated the role of 5-HT1A receptors in mediating the inhibitory action of 8-OH-DPAT, a 5-HT1A receptor agonist, in motion sickness in Suncus murinus. 8-OH-DPAT (0.1 mg/kg, i. p) attenuated motion-induced emesis which was associated with an increase in the latency of the onset to the first emetic episode. Pre-treatment with methysergide (a 5-HT1/2/7 receptor antagonist, 1.0 mg/kg, i. p.), WAY-100635 (a 5-HT1A receptor antagonist, 1.0 mg/kg, i. p.), SB269970A (a 5-HT7 receptor antagonist, 1.0 and 5.0 mg/kg, i. p.), ondansetron (a 5-HT3 receptor antagonist, 1.0 mg/kg, i. p) or GR13808 (a 5-HT4 receptor antagonist, 0.5 mg/kg, i. p) failed to modify the inhibitory action of 8-OH-DPAT on motion sickness. Furthermore, the application of either methysergide, WAY-100635, SB269970A, ondansetron or GR13808 alone had no effect on motion sickness in its own right. These data indicate that neither 5-HT1A nor any 5-HT2 receptor subtypes, 5-HT3, 5-HT4 and 5-HT7 receptors are likely to be involved in the inhibition of motion-induced emesis mediated by 8-OH-DPAT.

Le mécanisme de la réaction d’oxydation de l’hydrogène (HOR) dans Ni (111) est bien connu et se fait par étapes de Volmer-Heyrovsky, en milieu alcalin. Il a été proposé que la formation d'eau puisse jouer un rôle important. Dans cette thèse, j'ai étudié les surfaces de nickel et de nickel bimétallique en utilisant la théorie de la densité fonctionnelle (DFT). J'ai calculé des magnitudes thermodynamiques (comme les énergies libres d'adsorption de Gibbs) et des propriétés cinétiques (comme des barrières d'activation pour la formation d'eau). Plusieurs surfaces Ni / Cu ont été analysées. Celle qui contient 25% de Cu (sur la couche supérieure) a les meilleures performances: 1) l’énergie d’activation est de 0,2 eV, et 2) OH et H ne doivent pas être fortement adsorbés dans la plage de potentiel HOR. / The mechanism of hydrogen oxidation reaction (HOR) in Ni(111) is well-known and it happens through Volmer-Heyrovsky steps, in alkaline media. However it was proposed that water formation could play an important role. In this thesis, I have studied nickel and bimetallic nickel surfaces using density functional theory (DFT). I calculated thermodynamical magnitudes (like Gibbs energies of adsorption) and kinetic properties (like activation barriers for water formation). Several Ni/Cu surfaces were analyzed. The one with 25% of Cu (on top layer) has the best performance because: 1) the activation energy is 0.2 eV, and 2) OH and H are not to strongly adsorbed on the HOR potential range.

Este trabalho apresenta a caracterização espectroscópica de dois conjuntos de vidros sódio aluminofosfatos co-dopados com Yb3+ e Tm3+, na região do visível e infravermelho, preparados em atmosfera de nitrogênio e ambiente. Nestas amostras, os íons de Yb3+ são empregados como doadores eficientes, da excitação em 980nm, com transferência de energia subseqüente para os íons Tm3+. Os resultados são discutidos em termos do conteúdo de OH- presentes em cada conjunto das amostras, e em função da concentração Tm2O3. As propriedades radiativas foram analisadas usando a teoria de Judd Ofelt, e empregando os resultados experimentais e teóricos, foram avaliadas as eficiências quântica das luminescências (h) dos diversos níveis. Os resultados indicam que as amostras preparadas em atmosfera do N2, com um conteúdo de OH- menor, apresentam propriedades superiores do que aquelas preparadas em atmosfera ambiente. Entre estas propriedades destacamos a transmissão no infravermelho, a qual é 30% mais extensa, as intensidades de emissão são mais elevadas, e a diminuição dos efeitos de supressão da luminescência, caracterizando-os como materiais potenciais para a geração do laser alta potência, bem com conversão ascendente de energia eficiente. Também foram estudados os processos de transferência de energia, usando os modelos teóricos de Förster, Dexter e Miyakawa, calculando-se os parâmetros de transferência de energia. Os processos principais de transferência de energia analisados foram: migração da energia entre íons de Yb3+, transferência de energia entre íons Yb3+ e Tm3+, e interações com radicais OH-. Os resultados indicam que transferência de energia Yb-Tm favorece a emissão em 1800 nm, e não há nenhuma evidência da supressão da luminescência até concentração de 2% de Tm2O3. / This work presents a throughout near-infrared and visible spectroscopic characterization of two sets of Yb3+ and Tm3+ codoped sodium aluminophosphate glasses, prepared in nitrogen and ambient atmospheres. Desenvolvimento de um espectrofotômetro a laser de centro de cor aplicado ao estudo de cristais de NaF:Cu+:OH- / Development of a Color Center Laser spectrophotometer to study NaF:Cu+:OH- crystals

Este trabalho se encontra dividido em duas partes. Em uma parte inicial, relatamos a construção e caracterização de um laser de centro de cor (LCC) com meio ativo de KCl:Li+, operando de forma sintonizável no infravermelho na região em torno de 2,7 um. São descritas três configurações distintas para a cavidade do laser, e fazemos um estudo comparativo do desempenho do LCC para os três casos. Na segunda parte, o laser de centro de cor foi transformado em um espectrofotômetro de duplo feixe de alta resolução (largura de linha OMEGA-V<O,2 cm-1), com o qual passamos a estudar amostras cristalinas de NaF multiplamente dopadas com impurezas moleculares OH- e monoatômicas Cu+. Relatamos novas características desse material, que surgem em virtude da interação entre as referidas impurezas. Dentre essas características, temos o surgimento de um espectro complexo de linhas deslocadas para menores energias com relação a banda de absorção do modo de estiramento do OH-. Parâmetros de anarmonicidade são analisados, os quais sugerem a existência de anarmonicidade elétrica em adição a anarmonicidade mecânica. Estudos da dependência das larguras das bandas de absorção com a temperatura juntamente com medidas de TSDC ( corrente de despolarização termo-estimulada), revelaram a existência de um comportamento diferente daquele de rotor quase livre encontrado para o OH- em vários halogenetos alcalinos. Os resultados mostraram que no cristal de NaF tanto o OH- isolado quanto o acoplado estão permanentemente orientados / This work has two parts. The starting one is related with the construction and characterization of a tunable color center laser (CCL) with an active medium of KCl:Li+ which works in the mid infrared region around 2,7 um. It is discussed three different configurations set up for the laser cavity, and the profile of each case is compared between each other. For the second part, the color center laser is used as the source of a high-resolution double beam spectrophotometer (line width OMEGA-V<O,2 cm-1). The spectrophotometer is used to study multiple doped NaF crystal samples, with OH- molecules and monoatomic Cu+ impurities. New optical properties are found due to assumed coupling interaction between both impurities, which gives rise to a more complex spectrum where several lines are displaced to lower energies relative to the main OH- stretching mode absorption. Some anharmonic effects are observed, which suggests the existence of electric anharmonicity added to the mechanical one. Further study of the bandwidth temperature dependence, together with thermally stimulated depolarization current measurements, reveal a different behavior of the usual OH- quasi-free rotor found in several alkali halides. The results show that for the NaF crystal the isolated and coupled OH- ions are permanently oriented

Analys av 25-hydroxyvitamin D i primärvården

Background: The interest of vitamin D has increased in the last years. That is because there is so many possible positive effects of vitamin D and also because many individuals has vitamin D deficiency. Modern man spends much time indoors which leads to lower levels of vitamin D. People who have emigrated from a sunny climate to a Nordic climate often gets a deficiency due to a more pigmented skin which requires a larger amount of UVB to get an adequate synthesis of vitamin D. Aim: The aim with this study is to compare and evaluate how similar the instrument mini VIDAS measures 25(OH)D total against the current existing method cobas e 602. A discussion about if 25(OH)D total has a place in primary health care is included in the study. Method: The comparison was based on 39 samples. The samples was analyzed on cobas e 602 and mini VIDAS. A precision test was performed. External controls from DEQAS was also included in the study. The results have been presented with simple linear regression analysis, mean value, SD and CV. Results: The comparison between cobas e 602 and mini VIDAS gave a coefficient of determination of 81,34 %. mini VIDAS was closest to the external controls target values. Conclusion: There is no obvious conclusions about if mini VIDAS fulfills the requirement to be introduced to primary health care. The coefficient of determination of 81,34 % should be at least 95 %. However is mini VIDAS closer to the external controls target values then cobas e 602. There is factors that implies that 25(OH)D total has a place in primary health care with regards to demand, use and because many individuals has vitamin D deficiency. The instrument is also user-friendly to a primary health care laboratory. / Bakgrund: Intresset för vitamin D har ökat de senare åren. Det beror dels på att det finns många eventuella positiva effekter av vitamin D och dels på att många individer har brist på vitamin D. Nutidens människa spenderar mycket tid inomhus vilket leder till lägre nivåer av vitamin D. Personer som har utvandrat från ett soligt klimat till nordiskt klimat får ofta brist på grund av en mer pigmenterad hud som behöver större mängd UVB för att få en adekvat syntes av vitamin D. Syfte: Syftet med den här studien är att jämföra och utvärdera hur lika det patientnära instrumentet mini VIDAS mäter 25(OH)D total mot befintlig metod cobas e 602. Diskussion om analysen 25(OH)D total har en plats i primärvården ingår även i studien. Metod: Jämförelsen baserades på 39 st prover. Proven analyserades på cobas e 602 och mini VIDAS. Ett precisionsförsök gjordes. Externkontroller från DEQAS inkluderades även i studien. Resultaten har presenterats genom enkel linjär regressionsanalys, medelvärde, SD och CV. Resultat: Jämförelsen mellan cobas e 602 och mini VIDAS gav en förklaringsgrad på 81,43 %. mini VIDAS var närmst externkontrollernas målvärden. Slutsats: Det går inte att dra självklara slutsatser ifall mini VIDAS uppfyller kraven att införas i primärvården. Förklaringsgraden som är på 81,43 % bör vara minst 95 %. Däremot överensstämmer mini VIDAS med externkontrollerna bättre än cobas e 602. Det finns faktorer som tyder på att analysen 25(OH)D har en plats i primärvården med avseende på efterfrågan, användningsområde och antal individer med brist. Instrumentet är dessutom användarvänligt för ett primärvårdslaboratorium.

Change particularities of the blood serum levels of vitamin D, calcitonin and parathyroid hormone were analysed in healthy cows of different feeding, age and productivity and in sick cows with metabolic diseases. It was investigated how biochemical indicators vary in cows with metabolic diseases (parturient paresis, osteomalacia and mastitis) and the most informative indicators for diagnostics of these diseases (for levels of calcium, phosphorus and magnesium) were determined. Obtained findings were processed statistically. The method of electrochemiluminescence analysis used for analysis of the blood serum levels of PTH in humans was applied. Compared with radioimmunic method, this method is not expensive and quite fast; in addition, this method is sensitive and reliable, because it permits to detect low levels of PTH in the blood serum (up to 0.127 pmol/l). The method of chemiluminescence immunometric analysis used to find the level of CT in humans was tested. Also the method of immunoferment analysis (ELISA) was tested to measure the blood serum level of 25-OH vitamin D in cattle using human antibodies.

Neue photokatalytisch aktive Verbundmaterialien zur Eliminierung von pharmazeutischen Wirkstoffen aus Wässern: Verfahrenschemische Untersuchungen und Analytik

Schwerpunkt der vorliegenden Arbeit war die Erstellung, Anpassung und Anwendung einer Methode, die es über die Quantifizierung gebildeter OH-Radikale ermöglicht, sowohl den Einfluss verschiedener Materialmodifikationen (physikalisch dotierte Nanokatalysatoren bzw. Biokompositmaterialien) als auch die Auswirkungen von Matrixbestandteilen oder hydrochemischen Randbedingungen auf die photokatalytische Effizienz der Materialien zu untersuchen. Zudem wurde angestrebt, dass sich die Anwendbarkeit der Methode nicht nur auf die Photokatalyse beschränkt, sondern auch auf andere AOPs ausgeweitet werden kann. Des Weiteren wurde über die Umsetzung der Modellspurenstoffe Carbamazepin und Diclofenac die Wirksamkeit der Katalysatormaterialien untersucht. Anhand von Versuchen im Labormaßstab werden unter Anwendung der entsprechenden Methode zur Quantifizierung der OH-Radikale bisherige zugrundeliegende Hypothesen zur photokatalytischen Erzeugung von OH-Radikalen (Einfluss von pH und Oberfläche) überprüft und modifiziert. Dabei werden neue Ansätze zur Oberflächenabhängigkeit der OH-Radikalbildung in AOPs (EAOP Diamantelektrode, UV/VUV), die Effizienz von UV/VUV im Vergleich zu UVA-Photokatalyse, die Anwendung von S-Layer-Proteinen in photokatalytischen Biokompositmaterialien sowie eine photokatalytische Umsetzung von Carbamazepin unter Nutzung neuartiger Katalysatoren und Sonnenlicht untersucht. Mit Hilfe der gewonnenen Erkenntnisse ist es möglich, photokatalytisch aktive Materialien über die OH-Radikalbildungskapazität, als Basisprozess einer photokatalytischen Eliminierung von pharmazeutischen Wirkstoffen aus Wässern, mit Hinblick auf ihre Effizienz und Haltbarkeit zu untersuchen. Daneben bietet das erstellte Konzept zur analytischen Anwendung der OH-Radikalbestimmung neben der Gelegenheit für interessante Vergleiche diverser AOP-Systeme auch die Möglichkeit einer Charakterisierung und Optimierung der einzelnen AOPs. Zudem konnte gezeigt werden, dass die Anwendung von nano-Biokompositmaterialien unter Verwendung von S-Layer-Protein zur Herstellung multifunktionaler photokatalytischer Beschichtungen vielversprechend ist. Die Ergebnisse der Arbeit unterstreichen, dass die analytische Erfassung von Transformationsprodukten aus photokatalytischen bzw. oxidativen Umsetzungen im Allgemeinen von großer Bedeutung ist, jedoch allein nicht ausreicht, um hinreichend sichere Aussagen über eine mögliche Gefährdung für Mensch bzw. Ökosystem zu erhalten. / The current work was focused on the preparation, adaption and application of an analytical method for the determination of OH radicals for the comparison of the activity of different photocatalytic materials in relation to the material modification (i.e. physically doped nanomaterials or biocomposite materials) and the composition of the water matrix. Furthermore, the application of the OH radical assay should be extended on other AOPs. The degradation of the model compounds carbamazepine and diclofenac was examined to determine the efficiency of the novel photocatalysts. By using appropriate OH radical assays in laboratory scale experiments, present hypotheses in relation to the photocatalytic formation of OH radicals (i.e. influence of pH or surface) were examined and modified. New approaches on the formation of OH radicals with respect to the surface within AOPs (EAOP diamond electrodes or UV/VUV), the efficiency of UV/VUV in relation to photocatalysis using UVA irradiation, the application of S-layer proteins in biocomposite materials and the photocatalytic degradation of carbamazepine applying novel photocatalysts and natural sunlight were examined. Based upon the findings, it was possible to compare photocatalytic materials regarding efficiency and stability by means of the capacity to form OH radicals as the base process for the oxidative degradation of pharmaceutical trace compounds. The analytical concept offers the possibility to compare different AOPs and to characterize or optimize a single AOP. Furthermore, it was shown that the implementation of nanoscale biocomposite materials using S-layer proteins for the preparation of multi-functional coatings for photocatalytic applications is promising.
