Integrin subunits: expression and function in early development of Strongylocentrotus purpuratus

Integrins are heterodimeric transmembrane receptors composed of an α and a β subunit, that are expressed on the surface of all metazoan cells. These bidirectional signaling molecules are involved in many well-known aspects of cell function, although the role of integrins in early embryonic development remains a mystery. The purpose of this study was to characterize S. purpuratus integrins and determine if they are necessary for early embryonic development. Full length cDNA sequences for four incomplete gene predictions, αC, αD, αF, and βD, were determined by amplifying overlapping fragments and sequencing EST clones. Each cDNA has a single open reading frame predicting a protein with canonical integrin features. QPCR results show αC, αD, and βD are expressed in the embryo at relatively constant levels during the first 96 hours of development. αF is expressed in blastulae, during morphogenesis and tissue differentiation, at up to 35 times the levels of mRNA in the egg. Using a morpholino antisense oligonucleotide to block translation of αC results in a higher than normal mortality rate (57.1%) by 24 hours of development and 36.7% of embryos during this period have defects in aspects of cell division. These results indicate that αC is an essential gene for early development and that it may function in coordination of mitosis and cytokinesis. The expression of multiple subunits and the demonstration that αC has an essential role suggests that there are several non-overlapping functions for integrins in early embryonic development.

Baixos níveis séricos de 25(OH)D3 e 1,25(OH)2D3 (calcitriol) podem estar associados à incidência e prognóstico do câncer de mama. Além disso, vários estudos indicam que a vitamina D tenha um efeito antiproliferativo em linhagens celulares de câncer de mama expostas a concentrações supra-fisiológicas de calcitriol (1,25(OH)2D3, 100nM). A suplementação com vitamina D é indicada a mulheres pós-menopausadas para prevenção de osteoporose e observamos previamente que a suplementação de calcitirol a pacientes pós-menopausadas com câncer de mama causa redução do índice proliferativo tumoral. Entretanto, não há estudos até o momento que avaliam o efeito da vitamina D na expressão gênica global in vivo. Incluímos 31 pacientes pós-menopausadas com câncer de mama. Estas pacientes realizaram suplementação com calcitriol (0,5g/dia, dose indicada para prevenção de osteoporose) por um curto período de tempo (mediana de 32 dias). A amostra tumoral foi coletada por ocasião da biópsia (présuplementação) e da ressecção tumoral (pós-suplementação). Os perfis de expressão gênica de 16 pacientes foram analisados a partir de 100ng de RNA total no gene chip U133 Plus 2.0 Affymetrix. Observamos redução na expressão de Ki-67 após a suplementação. Dentre os genes diferencialmente expressos encontram-se EGR1, FOS, DUSP1, MMP12 e RGS1, os quais foram mais expressos em amostras pós-suplementadas. Genes modulados pela vitamina D estão associados à resposta inflamatória e à membrana. Nossos resultados indicam que a suplementação com vitamina D reduz o índice de proliferação tumoral, sendo a mesma envolvida em vias importantes na regulação da resposta inflamatória / Low 25(OH)2D3 or 1,25(OH)2D3 serum levels may be associated with breast cancer incidence and prognosis. Additionally, the antiproliferative effects of vitamin D are observed in breast cancer cell lines exposed to phamacological doses of calcitriol (1,25(OH)2D3, 100nM). Vitamin D supplementation is indicated for post-menopausal women to prevent osteoporosis and a previous study from our group observed a reduced tumor proliferative index after calcitriol supplementation on post menopausal breast cancer patients. However, there is no study that verifies the effect of vitamin D on gene expression profile in vivo so far. Thirty one post menopausal breast cancer patients were included on our analysis. They were supplemented with calcitriol after tumor biopsy (0.50g/day, indicated dose for osteoporosis prevention) for a short period of time (median 32 days). Tumor samples were collected during biopsy (before supplementation) and breast surgery (after supplementation). Gene expression profile of 16 patients was analyzed using the U133 Plus 2.0 Affymetrix Gene Chips from 100ng of total RNA. After supplementation, a reduced expression of Ki-67 was observed. Among the differentially expressed genes, EGR1, FOS, DUSP1, MMP12 and RGS1 were upregulated after calcitriol supplementation. Differentially expressed genes were involved in inflammatory response or were associated with the membrane. Our results indicate that calcitriol supplementation diminish tumor proliferation index regulating inflammatory pathways .

Au cours de la dernière décennie, les anticorps conjugués à des médicaments cytotoxiques ou des oligonucléotides ont acquis une grande attention dans la communauté scientifique en raison des propriétés uniques des anticorps, tels que leur long temps de circulation dans le sérum et leur sélectivité élevée par rapport à leur cible. Par exemple, les conjugués d'anticorps (ACs) sont de plus en plus appliqués en thérapie ciblée contre le cancer ou en bioimagerie. Par conséquent, le développement de méthodologies fiables pour la préparation des AC est actuellement en pleine expansion. Cependant, la conjugaison et la préparation contrôlables des ACs avec une structure définie rencontrent encore de nombreux obstacles en raison de l'excès élevé et de la variété des groupes réactifs dans la structure des anticorps, qui sont accessibles pour la conjugaison. En outre, les technologies de liaison actuelles sont basées sur la réaction de maléimide-thiol, produisant des adduits, qui sont instables dans le sang. Ce travail se concentre sur les approches chimiques pour la fonctionnalisation fiable des anticorps, qui permettent la préparation d'ACs stables présentant un ratio anticorps/principe actif bien défini. La première partie est consacrée à la conception et au développement du réactif maléimide-dioxane, solution auto-hydrolysable et stable dans le sérum, comme alternative à la chimie classique du maléimide. La deuxième partie est consacrée à l'évaluation de la réactivité sélective des différents acides aminés portés par les anticorps par spectrométrie de masse native à haute résolution. Finalement, une nouvelle technologie permettant d’obtenir des ACs stables avec un ratio anticorps/principe actif contrôlé est présentée au lecteur dans une 3ème partie. / Within the last decade, antibodies conjugated to cytotoxic drugs or oligonucleotides have gained a great attention in scientific community owing to the unique properties of the antibodies, such as their long circulation time in serum and high selectivity against their target. For instance, antibody conjugates (ACs) are increasingly applied for targeted cancer therapy or bioimaging. Consequently, the development of reliable methodologies for ACs preparation is currently of high demand. However, the controllable conjugation and preparation of ACs with defined structure are still challenging due to high excess and variety of reactive groups in antibody structure, which are accessible for conjugation. Moreover, current linker technologies are based on the maleimide-thiol reaction, yielding adducts, which are unstable during circulation in blood.This work is focused on chemical approaches for the reliable antibody functionalisation, which enable the preparation of stable ACs with well-defined payload to antibody ratios. The first part is devoted to design and development of maleimide-dioxane reagents as self-hydrolysable and serum-stable alternative to classical maleimide chemistry. The second part is dedicated to a screening approach for evaluation of residue-selective functionalities in reactions with an antibody using high resolution native mass spectrometry. Finally, in the third part the reader is introduced with a novel technology, which enables efficient preparation of stable ACs with a defined degree of conjugation and particularly mono-functionalisation of antibodies.

Neurochemical markers in CSF of adolescent and adult SMA patients undergoing nusinersen treatment

Background: There is limited information on neurochemical markers being used to support and monitor the affection of motoneurons in patients with spinal muscular atrophy (SMA). The objective of this study was to examine neurochemical markers in cerebrospinal fluid (CSF) under treatment with the antisense-oligonucleotide (ASO), nusinersen. Methods: We measured markers of axonal degeneration [neurofilament light chain (NfL) and phosphorylated neurofilament heavy chain (pNfH)] along with basic CSF parameters in 25 adolescent and adult SMA type 2 and 3 patients at baseline and after four intrathecal injections of nusinersen. Neurochemical markers were compared with controls. In addition, neurochemical markers in SMA patients were related to the Hammersmith Functional Rating Scale Expanded (HFMSE). Results: No significant difference in neurofilament (Nf) values was observed between SMA and control group, neither at baseline nor after four injections of nusinersen. NfL, protein and quotients of albumin (Qalb) increased slightly in SMA patients after the fourth injection. The slight increase of NfL could be related to the development of mild CSF flow change. No relations were observed between changes in Nf and HFMSE. Conclusion: We assume that Nf levels in CSF in these patients may result from slow disease progression in this stage of disease, pre-existing loss of motoneurons due to long disease duration besides affection of the LMN only. Therefore, we conclude that Nf levels in CSF do not seem useful as diagnostic and monitoring markers in adolescent and adult SMA type 2 and 3 patients.
