The Effects of Early Postnatal PCP Administration on Performance in Locomotor Activity, Reference Memory, and Working Memory Tasks in C57BL/6 Mice

There is a growing consensus, based on several converging lines of evidence, which suggests schizophrenia is the product of a developmental insult occurring in the late 2 nd or early 3 rd trimester. Additionally, it has been observed that adults who abuse the noncompetitive NMDA antagonist PCP present with symptoms that mimic schizophrenia, such as hallucinations, formal thought disorder, delusions, unstable or flattened affect, social withdrawal, and impaired cognition. Thus, several labs have attempted to use early postnatal PCP administration in rodents as a drug model of schizophrenia. The current study investigated the cognitive effects of early postnatal PCP administration in C57BL/6 mice. Mouse pups received daily administrations of either 10.0 mg/kg PCP or saline on postnatal (PN) days 5-15. After weaning, pups were assessed in locomotor activity, a reference memory task in the Morris water maze, and a spatial delayed alternation task in the T-maze. Additionally, pups were subjected to a pharmacological challenge with PCP in the delayed alternation task. In males, No significant differences were detected between PCP- and saline-treated animals in locomotor activity. However, in the reference memory task, PCP-treated males had significantly longer path lengths, and displayed a non-significant trend towards increased thigmotaxia. Furthermore, males treated with PCP displayed significantly reduced accuracy in the working memory task without differences in choice latency, and were more sensitive to the acute effects of PCP than saline controls. Finally, these deficits were associated with a 29% increase in NR1 subunit expression in the hippocampus. Interestingly, PCP-treated female mice were not significantly different from saline-treated controls in locomotor activity, reference memory task performance, or delayed alternation performance, did not have a significantly different reaction to pharmacological challenge with PCP in the delayed alternation task, and did not demonstrate any changes in NR1 subunit expression. The present study provided the first evidence that early postnatal PCP administration in C57BL/6 mice can produce selective memory pairments. However, this effect was limited to the male mice, suggesting that the female mice were protected somewhat from these effects.

Effet des macrophytes aquatiques sur le traitement d’eau contaminée à l’ACC et au PCP

L’objectif principal de ce projet est d’évaluer le potentiel d’un système de marais filtrants combinés pour le traitement d’un lixiviat contaminé aux agents de préservations du bois - l’arséniate de cuivre chromaté (ACC) et le pentachlorophénol (PCP) - en portant une attention particulière au rôle des plantes. Pour ce faire, une expérience en pot faisant varier la concentration d’exposition de quatre espèces de macrophyte (Typha angustifolia, Phalaris arundinacea, Phragmites australis americanus et Phragmites australis australis) a été effectuée. Le suivi de quatre marais filtrants à écoulement horizontal sous-surfacique, plantés d’une des quatre espèces de macrophyte et d’un bassin planté de saules (Salix Miyabeana SX67) a également été réalisé. La résistance des plantes au milieu contaminé, la capacité de bioaccumulation des macrophytes et les efficacités de traitement des différents systèmes ont été analysés. Nos résultats montrent que la concentration d’exposition n’influence pas la capacité des plantes à croître en milieu contaminé. Par contre, il existe une relation dose-réponse entre la concentration d’exposition et la capacité de bioaccumulation des macrophytes. Les quatre marais pilotes ont tous des efficacités de traitements supérieures à 55% en 2013 et 82% en 2014 pour les contaminants à l’étude. Le bassin de saule a la capacité théorique d’évapotranspirer jusqu’à 1200 L par jour. De plus, ses efficacités de traitements sont supérieures à 59% pour tous les composés à l’étude. L’utilisation de marais filtrants pour le traitement d’un lixiviat contaminé aux agents de préservations du bois est donc une alternative intéressante aux méthodes de traitement conventionnel. / The main objective of this project is to evaluate the capacity of a combined constructed wetland system to treat a leachate polluted with two wood preservatives-chromium copper arsenate (CCA) and pentachlorophenol (PCP)- according a particular importance to the role of plants. The effect of the exposure concentration on four macrophytes species (Typha angustifolia, Phalaris arundinacea, Phragmites australis americanus and Phragmites australis australis) was evaluated using a pot experiment. Four HSSF pilot constructed wetlands, planted with one of these plants species, were monitored. The role of an HSSF constructed wetland, planted with willow (Salix Miyabeana SX67), placed at the end of the treatment chain was also assessed. The plants growth and survival and their bioaccumulation capacity were measured, and the system treatment efficiencies were monitored. There was no effect of the concentration on plants growth but the higher exposure concentration resulted in a higher bioaccumulation in the roots of the four macrophytes. All four pilots CW showed treatment efficiencies superior to 55% in 2013 and to 82% in 2014 for the pollutants tested. The willows CW have the theorical capacity to evapotranspirate up to 1200 L of water. Its treatments efficiencies were over 59% for all the pollutants tested. A combined constructed wetland system is a good alternative to conventional methods to treat a leachate polluted with wood preservatives.

In this final thesis, an existing method for analysis of chlorophenols (CP) in bottom sediments has been updated and adjusted for analysis of chlorophenols in soil. The covalent bonds between the chlorophenols and the soil matrix were broken through basic hydrolysis and the chlorophenols were then separated from the water phase through addition of sulphuric acid followed by ether extraction. The chromatography was improved through extractive acetylation of the chlorophenols. The updated method was then applied on soil samples from a contaminated area (a former sawmill in Hyttsjö, Östergötland, Sweden). The analyse was preformed by GC/MS with respect to 2-MonoCP, 4-MonoCP, 2,4-DiCP, 2,6-DiCP, 2,4,6-TriCP, 2,3,4,6-TetraCP and pentachlorophenol (PCP). Contamination of chlorophenols in nature can be explained by the former use of wood preservative chemicals based on chlorophenols. In the 1960s and the 1970s these chemicals were used in Sweden, but due to their toxicity they were banned by the Swedish government in 1978. In Hyttsjö a pentachlorophenol-based product named Santobrite was used for several years. The concentration of PCP in the soil samples from Hyttsjö varied from 0.2->1.8 ng/mg dry substance. 2,3,4,6- Tetrachlorophenol was also detected in some of the soil samples.

Functionalization and processing of porous powders into hierarchically porous monoliths

Inorganic porous materials are widely used in a number of applications, where is a need to functionalize and produce materials with a multiscale porosity. The first part of the thesis describes how a novel and facile powder processing approach, using pulsed current processing (PCP) or, as it is commonly called, spark plasma sintering (SPS), has been employed to produce mechanically stable, hierarchically porous bodies from different porous powders. Surfactant-templated mesoporous spheres were PCP-treated to yield meso/macro porous monoliths. The bimodal pore size can be tailored by choice of templating molecules in the aerosol-assisted synthesis process and by the particle size of the spheres. Diatomite powders were used to produce macro/macroporous monoliths. The densification behaviour of this inexpensive and renewable macroporous raw material was evaluated in detail, and an optimum temperature range was identified where the PCP process yields mechanically strong monoliths. Binder-less, hierarchically porous zeolite monoliths were produced from various zeolite powders, e.g. silicalite-1, ZSM-5 and zeolite Y. Line-broadening analysis of X-ray powder diffraction data by the Rietveld method and electron microscopy showed that the formation of strong interparticle bonds during the PCP process is associated with a local amorphization reaction that is induced by the high contact stress and temperature. Xylene isomerisation studies showed that binder-less ZSM-5 monoliths display a high catalytic selectivity. Direct (in-situ) nanoparticle functionalization of surfactant templated mesoporous silica particles has also been demonstrated. Pre-synthesized TiO2 nanoparticles were dispersed in a precursor solution, containing surfactant and silica source, and processed in an aerosol-generator to produce spherical nanoparticle-functionalized mesoporous particles.

Controlling cracking in precast prestressed concrete panels

Precast, prestressed concrete panels (PCPs) have been widely used in Texas as stay-in-place formwork in bridge deck construction. Although PCPs are widely popular and extensively used, Texas is experiencing problems with collinear cracks (cracks along the strands) in panels. One reason for the formation of collinear cracks is thought to be the required level of initial prestress. Currently, PCPs are designed assuming a 45-ksi, lump-sum prestress loss. If the prestress losses are demonstrated to be lower than this value, this could justify the use of a lower initial prestress, probably resulting in fewer collinear cracks. For this purpose, 20 precast, prestressed panels were cast at two different plants. Half of those 20 panels were fabricated with the current TxDOT-required prestress of 16.1 kips per strand, and the other half were fabricated with a lower prestress of 14.4 kips per strand based on initially observed prestress losses of 25 ksi or less. Thirteen of those panels were instrumented with strain gages and monitored over their life time. Observed losses stabilized after five months, and are found to be about 24.4 ksi. Even with the reduced initial prestress, the remaining prestress in all panels exceeds the value now assumed by TxDOT for design. / text

Planar cell polarity (PCP) is the organization of cellular characteristics within the plane of a tissue. PCP manifests both structurally, as in the directionality of insect bristles or mammalian skin hair, or dynamically, as in vertebrate neurulation, gastrulation, and oriented cell division in the kidney. Two well-conserved pathways are known to regulate PCP in invertebrates and in vertebrates: the Frizzled/PCP pathway and the Fat-Dachsous (Ft-Ds) pathway. The latter consists of the cadherins Ft and Ds, along with the Golgi kinase Four-jointed (Fj) and the transcriptional co-repressor Atrophin (Atro). Ft and Ds can bind each other, suggesting a mechanism for signal transduction. Fj phosphorylates Ft and Ds, modulating their binding affinities for each other. Atro is proposed to link Ft-Ds signaling with downstream events in the nucleus during eye development. The details of Ft-Ds binding, and the consequences of their interactions with other members of the pathway are poorly understood. In this work, I quantitatively analyzed Ft-Ds pathway mutant clones for their effects on ommatidial polarity in the Drosophila eye. My findings suggest that the Ft-Ds pathway regulates PCP independently of asymmetric cellular accumulation of Ft or Ds. I found that Atro has a position-specific role in regulating polarity in the eye, that Fj dampens clonal polarity signals, and that asymmetric accumulation of the atypical myosin Dachs is not essential for production and propagation of a long-range PCP signal. My observations suggest that Ft and Ds interact to modulate a secondary signal that regulates long-range polarity, that signaling by the Ds intracellular domain is dependent on Ft, and that ommatidial fate specification is genetically separable from long-range signaling.

Tumour-stroma Signalling in Cancer Cell Motility and Metastasis

The tumour-associated stroma, consisting of fibroblasts, inflammatory cells, vasculature and extracellular matrix proteins, plays a critical role in tumour growth, but how it regulates cancer cell migration and metastasis is poorly understood. The Wnt-planar cell polarity (PCP) pathway regulates convergent extension movements in vertebrate development. However, it is unclear whether this pathway also functions in cancer cell migration. In addition, the factors that mobilize long-range signalling of Wnt morphogens, which are tightly associated with the plasma membrane, have yet to be completely characterized. Here, I show that fibroblasts secrete membrane microvesicles of endocytic origin, termed exosomes, which promote tumour cell protrusive activity, motility and metastasis via the exosome component Cd81. In addition, I demonstrate that fibroblast exosomes activate autocrine Wnt-PCP signalling in breast cancer cells as detected by the association of Wnt with Fzd receptors and the asymmetric distribution of Fzd-Dvl and Vangl-Pk complexes in exosome-stimulated cancer cell protrusive structures. Moreover, I show that Pk expression in breast cancer cells is essential for fibroblast-stimulated cancer cell metastasis. Lastly, I reveal that trafficking in cancer cells promotes tethering of autocrine Wnt11 to fibroblast exosomes. These studies further our understanding of the role of the tumour-associated stroma in cancer metastasis and bring us closer to a more targeted approach for the treatment of cancer spread.
