The extent of Aflatoxin and Aspergillus section Flavi, Penicillium spp. and Rhizopus spp. contamination of peanuts from households in western Kenya and the causative factors of contamination.Mutegi, Charity Kawira. January 2010 (has links)
Peanuts contribute significantly to food security in western Kenya due to their high nutritional value and cash crop potential. However, the crop is highly susceptible to aflatoxin contamination. Yet little information is available on the extent of contamination in the region. This study explores the level and extent of contamination of peanuts by aflatoxins, Aspergillus section Flavi, Rhizopus and Penicillium spp. in western Kenya. A survey of 769 households was carried out in the Busia and Homa bay districts of Kenya. Information on peanut pre- and post-harvest practices was collected through person-to-person interviews. Aflatoxin levels of samples collected from each household were determined by indirect competitive ELISA method. Isolation of Aspergillus section Flavi, Penicillium and Rhizopus spp. was done on Modified Dichloran Rose Bengal (MDRB) agar, while identification of specific fungal species was done on Czapek yeast extract agar (CYA). Screening isolates of A. flavus and A. parasiticus for aflatoxin production was done in high sucrose yeast extract (YES) liquid medium, and the aflatoxin types identified on TLC plates, using analytical grades of aflatoxin B1, B2, G1 and G2 as reference standards. Common household preparation techniques (roasting, making peanut paste and boiling peanuts) were evaluated for effectiveness in reducing aflatoxin levels in peanuts. The boiling procedure was modified to test the effect of magadi (locally available salt used mainly to soften legumes, vegetables or maize while cooking), ammonium persulphate and sodium hypochlorite during soaking. Magadi, sodium bicarbonate and locally prepared ash was subsequently used to boil the nuts after soaking. Aflatoxin levels ranged from zero to 7525 ìg/kg. Most samples were safe to consume, based on the European Union and Kenya Bureau of Standards tolerance levels, with 63.7 per cent of all samples having undetectable levels, and only 7.54 per cent being contaminated based on KEBS standards. Peanuts from the Busia district, which has more of Lower Midland 1 (mean annual rainfall of 1600-1800 mm) and Lower Midland 2 (mean annual rainfall of 1300-1700 mm) agro-ecological zones had significantly (÷2=14.172; P=0.0002) higher levels of aflatoxin compared to the Homa bay district, that has more of the drier Lower Midland 3 agroecological zone (mean annual rainfall of 900-1500mm). Improved cultivars had significantly (÷2=9.748; P=0.0018) lower levels of aflatoxin compared to local cultivars. Over 60 per cent of all samples had A. flavus S-strain, A. flavus L-strain and A. niger. A. flavus S-strain was positively correlated with aflatoxin levels. As expected, grading of peanuts post-harvest significantly reduced the incidence of A. flavus S- and L-strains, while peanuts collected from farmers who belonged to producer marketing groups had a significantly lower incidence of A. flavus S- and L-strains, A. niger and Rhizopus spp. The incidence of A. flavus L-strain, A. niger and Rhizopus spp. was significantly higher in local landraces compared to the improved cultivars. Over 60 per cent of isolates produced Aflatoxin B1. Intermediate processes such as sorting and dehusking led to a significant decline in levels of aflatoxin. Soaking peanuts in water, magadi, NaOCl and ammonium persulphate significantly reduced aflatoxin levels by 27.7, 18.4, 18.3 and 1.6 per cent respectively; while boiling the peanuts in magadi, local ash, baking powder and water reduced aflatoxin levels by 43.8, 41.8, 28.9 and 11.7 per cent respectively. Using magadi during boiling increased the acceptability of the boiled peanuts while reducing the aflatoxin levels. The impact of aflatoxin levels in peanuts studied in this research is within safe limits except a few samples, and therefore aflatoxin contamination of peanuts at household level is not a serious threat. Contamination by aflatoxin and post-harvest fungi can be reduced by focusing on improved control strategies for wetter and more humid zones such as planting improved peanut cultivars and controlling pre-harvest pest damage. Conventional household peanut preparation techniques should be explored as possible aflatoxin management strategies in Kenya. A study of the quality and feasibility of Sibusiso, a ready-to-use food.Mahlangu, Zodwa Nita. January 2012 (has links)
A ready-to-use food (RUF), Sibusiso, has the potential to alleviate protein- energy malnutrition. However, its nutritional composition, physical properties, consumer acceptability, and economic feasibility for use are unknown.
This study aimed to determine the nutritional composition and physical properties of a RUF, Sibusiso. As well as to assess the consumer acceptability of Sibusiso to healthy and HIV infected children on antiretroviral (ARV) medication, and the caregiver‟s attitudes towards Sibusiso. The feasibility of using Sibusiso for nutrition rehabilitation was also determined.
Methodology: Four samples of Sibusiso and a peanut butter (control) were analysed for their nutritional composition and physical properties. The consumer acceptability of Sibusiso to healthy children and HIV infected children on ARVs (ART group) was determined using a five-point facial hedonic rating scale. Focus group discussions were conducted to assess the attitudes and perceptions of caregivers surrounding Sibusiso. These caregivers had children who were either malnourished or at risk of malnutrition. The financial feasibility of using Sibusiso for nutrition rehabilitation was determined using published data.
The results revealed that Sibusiso was a good source of energy (2624 kJ/100 g) and quality protein (15.7 g/100 g). The nutritional composition of Sibusiso met the WHO/WFP/SCN/UNICEF recommendations for RUF. Instrumental colour analysis indicated that both Sibusiso and the peanut butter had a brown colour, although Sibusiso was slightly lighter. Sibusiso had the same spreadability or hardness as the peanut butter, but it was stickier than the peanut butter. Based on the sensory evaluation, Sibusiso was found acceptable to both healthy (n=121) and HIV infected children (n=51). Over 65% of the children in both the healthy and ART group liked the taste, smell and mouthfeel of Sibusiso. The caregivers also found Sibusiso acceptable and were willing to buy it, but at half its current price (60 ZAR/ 500 g). The price of Sibusiso and perception of the caregivers that Sibusiso was a peanut butter were the main factors that affected their willingness to buy the product. Financial feasibility analysis showed that the estimated cost (5.99 ZAR/day) of rehabilitating a child using Sibusiso was higher than the KwaZulu-Natal Department of Health nutrition budget of 0.02 ZAR/day.
Sibusiso is a good source of energy and quality protein, and it is fairly acceptable to children with HIV and caregivers. This indicates that Sibusiso has a potential to alleviate protein-energy malnutrition in the targeted groups. Yet, Sibusiso is expensive. Knowledge of the dissociation constants of the ionizable protons of weak acids in aqueous media is of fundamental importance in many areas of chemistry and biochemistry. The pKa value, or equilibrium dissociation constant, of a molecule determines the relative concentration of its protonated and deprotonated forms at a specified pH and is therefore an important descriptor of its chemical reactivity. Considerable efforts have been devoted to the determination of pKa values by different experimental techniques. Although in most cases the determination of pKa values from experimental is straightforward, there are situations where interpretation is difficult and the results ambiguous. It is, therefore, not surprising that the capability to provide accurate estimates of the pKa value has been a central goal in theoretical chemistry and there has been a large effort in developing methodologies for predicting pKa values for a variety of chemical systems by differing quantum chemical techniques. A prediction accuracy within 0.5 pKa units of experiment is the desirable level of accuracy. This is a non-trivial exercise, for an error of 1 kcal/mol in estimates of the free energy value would result in an error of 0.74 pKa units.
In this thesis ab initio Car-Parrinello molecular dynamics (CPMD) has been used for investigating the Brϕnsted acid-base chemistry of weak acids in aqueous solution. A key issue in any dissociation event is how the solvating water molecules arrange themselves spatially and dynamically around the neutral and dissociated acid molecule. Ab initio methods have the advantage that all solvent water molecules can, in principle, be con- sidered explicitly. One of the factors that has inhibited the widespread use of ab initio MD methods to study the dissociation reaction is that dissociation of weak acids are rare events that require extremely long simulation times before one is observed. The metady- namics formalism provides a solution to this conundrum by preventing the system from revisiting regions of configuration space where it has been in the past. The formalism allows the system to escape the free-energy minima by biasing the dynamics with a history dependent potential (or force) that acts on select degrees of freedom, referred to as collective variables. The bias potentials, modeled by repulsive inverted Gaussians that are dropped during propagation, drive the system out of any free-energy minima and allow it to explore the configurational space by a relatively quick and efficient sampling. The the- sis deals with a detailed investigation of the Brϕnsted acid-base chemistry of weak acids in aqueous solutions by the CPMD-metadynamics procedure. In Chapter 1, current approaches for the theoretical estimation of pKa values are summarized while in Chapter 2 the simulation methodology and the metadynamics sampling techniques used in this
study are described.
The potential of the CPMD-metadynamics procedure to provide estimates of the acid dissociation constant (pKa) is explored in Chapter 3, using acetic acid as a test sys- tem. Using the bond-distance dependent coordination number of protons bound to the dissociating carboxylic groups as the collective variable, the free-energy profile for the dissociation reaction of acetic acid in water was computed. Convergence of the free-energy profiles and barriers for the simulations parameters is demonstrated. The free-energy profiles exhibit two distinct minima corresponding to the dissociated and neutral states of the acid and the deterrence in their values provides the estimate for pKa. The estimated value of pKa for acetic acid from the simulations, 4.80, is in good agreement with the experiment at value of 4.76. It is shown that the good agreement with experiment is a consequence of the cancellation of errors, as the pKa values are computed as the divergence in the free energy values at the minima corresponding to the neutral and dissociated state. The chapter further explores the critical factors required for obtaining accurate estimates of the pKa values by the CPMD-metadynamics procedure. It is shown that having water molecules sufficient to complete three hydration shells as well as maintaining water density in the simulation cell as close to unity is important.
In Chapter 4, the CPMD-metadynamics procedure described in Chapter-3 has been used to investigate the dissociation of a series of weak organic acids in aqueous solutions. The acids studied were chosen to highlight some of the major factors that influence the dissociation constant. These include the influence of the inductive effect, the stabilization of the dissociated anion by H-bonding as well as the presence of multiple ionizable groups. The acids investigated were aliphatic carboxylic acids, chlorine-substituted carboxylic acids, cis- and trans-butenedioic, the isomers of hydroxybenzoic acid and ophthalmic acids and its isomers. It was found that in each of these examples the CPMD-metadynamics procedure correctly estimates the pKa values, indicating that the formulism is capable of capturing these influences and equally importantly indicating that the cancellation of errors is indeed universal. Further, it is shown that the procedure can provide accurate estimates of the successive pKa values of polypro tic acids as well as the subtle difference in their values for different isomers of the acid molecule.
Changes in protonation-deprotonation of amino acid residues in proteins play a key role in many biological processes and pathways. It is shown that CPMD simulations in conjunction with metadynamics calculations of the free energy profile of the protonation- deprotonation reaction can provide estimates of the multiple pKa values of the 20 canonical α-amino acids in aqueous solutions in good agreement with experiment (Chapter 5). The distance-dependent coordination number of the protons bound to the hydroxyl oxygen of the carboxylic and the amine groups is used as the collective variable to explore the free energy profiles of the Brϕnsted acid-base chemistry of amino acids in aqueous solutions. Water molecules, sufficient to complete three hydration shells surrounding the acid molecule were included explicitly in the computation procedure. The method works equally well for amino acids with neutral, acidic and basic side chains and provides estimates of the multiple pKa values with a mean relative error with respect to experimental results, of 0.2 pKa units.
The tripeptide Glutathione (GSH) is one of the most abundant peptides and the major repository for non-protein sulfur in both animal and plant cells. It plays a critical role in intracellular oxidative stress management by the reversible formation of glutathione disulfide with the thioldisulfide pair acting as a redox buffer. The state of charge of the ionizable groups of GSH can influences the redox couple and hence the pKa value of the cysteine residue of GSH is critical to its functioning. In Chapter 6, it has been reported that ab initio Car-Parrinello Molecular Dynamics simulations of glutathione solvated by 200 water molecules, all of which are considered in the simulation. It is shown that the free-energy landscape for the protonation - deprotonation reaction of the cysteine residue of GSH computed using metadynamics sampling provides accurate estimates of the pKa and correctly predicts the shift in the dissociation constant values as compared to the isolated cysteine amino acid.
The dissociation constants of weak acids are commonly determined from pH-titration
curves. For simple acids the determination of the pKa from the titration curves using the Henderson-Hasselbalch equation is relatively straightforward. There are situations, however, especially in polyprotic acids with closely spaced dissociation constants, where titration curves do not exhibit clear inflexion and equivalence stages and consequently the estimation of multiple pKa values from a single titration curve is no longer straightfor-
ward resulting in uncertainties in the determined pKa values. In Chapter 7, the multiple dissociation constant of the hexapeptide glutathione disulfide (GSSG) with six ionizable groups and six associated dissociation constants has been investigated. The six pKa values of GSSG were estimated using the CPMD-metadynamics procedure from the free-energy profiles for each dissociation reaction computed using the appropriate collective variable. The six pKa values of GSSG were estimated and the theoretical pH-titration curve was then compared with the experimentally measured pH-titration curve and found to be in excellent agreement. The object of the exercise was to establish whether interpretation of pH-titration curves of complex molecules with multiple ionizable groups could be facilitated using results of ab initio molecular dynamics simulations.

Previous issue date: 2010-11-03 / The present work aims to study the technical and economic feasibility of the interaction between sugarcane and soybean / peanut in the areas of crop rotation in the sugar and ethanol mill. For this purpose, it presents a study, conducted in a mill located in the country side of São Paulo, which has an area of approximately 4,500 ha for the annual crop rotation (50% of this area being used for the plantation of soybeans and 50% for the cultivation of peanuts). The analysis of the technical feasibility of the project is based on data supplied by the Sistema de Monitoramento Agrometeorológico (Agritempo), Embrapa, used for calculating the climatic risk for the crop to be planted in the area. With the use of meteorological and agrometeorological data provided by Agrotempo, it is possible to obtain 80% success in the crops, if such planting is done for the indicated periods. To demonstrate the economic feasibility, this research uses the classical model of Economic Engineering, with the rates and fees observation, such as hurdle rate (TMA), Net Present Value (NPV), Internal Rate of Return (IRR), Period Recovery of Invested Capital (Payback) and Sensitivity Analysis. This study demonstrates that the practice of crop rotation in sugarcane areas provides not only agronomic, but economic and social gains as well. Among the social benefits identified it includes: the sustainability of the rural sector through the production of more food, the generation of more income in the productive chain of agribusiness, improvement to the image of the sugarcane sector in urban and rural society, the greater employment generation, reduction of competitiveness for land, the increase in tax collection, etc. Under the economic gains, some benefrits are important, such as: the diversification of income and reduction in the costs of sugar cane production, made it possible by the rotation of crops, the promotion of economies of scale in production as a result of the synergy with the existing structure in the are of cane cultivation, to name but a few. / O presente trabalho tem por objetivo estudar e apresentar as viabilidades técnica e econômica da interação entre cana e soja/amendoim, nas áreas de rotação de cultura, em usina de açúcar e etanol. Para isso, apresenta um estudo de caso, realizado em uma usina localizada no interior do Estado de São Paulo, que possui área de aproximadamente 4.500 ha destinada anualmente à rotação de cultura (sendo 50% desta área utilizada para o plantio de soja e 50%, para o cultivo de amendoim). A análise da viabilidade técnica do projeto baseia-se em dados fornecidos pelo Sistema de Monitoramento Agrometeorológico (Agritempo), da Embrapa, empregado para o cálculo do risco climático para a cultura a ser plantada na região considerada. Com o emprego desses dados meteorológicos e agrometeorológicos fornecidos pelo Agritempo, é possível que se obtenha 80% de sucesso com as culturas, se realizado o referido plantio nos períodos indicados. Para demonstrar a viabilidade econômica, esta pesquisa recorre ao modelo clássico de Engenharia Econômica, com a observação de índices e taxas, como: Taxa Mínima de Atratividade (TMA), Valor Presente Líquido (VPL), Taxa Interna de Retorno (TIR), Período de Recuperação do Capital Investido (Payback) e Análise de Sensibilidade. Este estudo de caso demonstra que, com a prática da rotação de culturas nas áreas canavieiras, além de ganhos agronômicos, há ganhos econômicos e sociais. Dentre os benefícios sociais identificados, destacam-se: a sustentabilidade do setor rural, por meio da produção de mais alimentos; a geração de mais renda na cadeia produtiva do agronegócio; a melhora para a imagem do setor sucroalcooleiro na sociedade urbana e rural; a maior geração de empregos; a redução da competitividade por terras; o aumento na arrecadação de impostos; dentre outros. No âmbito dos ganhos econômicos, são importantes a diversificação da renda e a redução dos custos de produção da cana-de-açúcar, possibilitados pela rotação de culturas; a promoção de ganhos de escala de produção, em consequência da sinergia com a estrutura já existente na área de cultivo de cana; dentre outros.

Determination of aflatoxins in peanut (Arachis hypogaea L.) collected from Kinshasa, Democratic Republic of Congo and Pretoria, South Africa : a comparative studyKamika, Ilunga 16 April 2013 (has links)
This study assessed the mycological and aflatoxin contamination of peanuts collected from Kinshasa, DRC and Pretoria, South Africa. Forty peanut samples were collected randomly at informal markets in the two cities and analysed for mycoflora and aflatoxins (B1, B2, G1 and G2) using standard methods. The results indicated that 95% and 100% of peanut samples collected from Kinshasa and Pretoria, respectively were contaminated with aflatoxigenic fungi with Kinshasa’s samples being the most contaminated (up to 49, 000 CFU/g). Seventy percent (70 %) of Kinshasa-samples and 35% of Pretoria-samples exceeded the maximum allowable limit of aflatoxin B1 set by JECFA (5 ppb). Statistical evidence showed a significant positive correlation between mycoflora and aflatoxin level for Kinshasa-samples (r = 0.4743, p < 0.005) while Pretoria-samples showed no correlation. The study reveals that high level of contamination in Kinshasa-samples could be due to the tropical nature of the climate and poor storage conditions as compared to Pretoria which is sub-tropical and sanitary regulations are enforced. / Life and Consumer Sciences / M. A comparative study of natural contamination with aflatoxins and fumonisins in selected food commodities from Botswana and ZimbabweMupunga, Innocent 06 1900 (has links)
Mycotoxins are toxic secondary metabolites produced by filamentous fungi. Aflatoxins and fumonisins are among the most toxic mycotoxins. They are a significant risk factor for a cocktail of chronic health conditions including cancer of the liver, oesophagus and kidney, teratogenicity, neural tube defects, interference with lipid metabolism, a weakened immune system and a negative impact on micronutrient absorption in both man and animals. This study compared natural contamination of peanuts, peanut butter and sorghum from Gaborone, Botswana and Bulawayo, Zimbabwe with aflatoxins and fumonisins. In total 34 peanut samples, 34 sorghum samples and 11 peanut butter samples were collected randomly from retail shops and informal markets in the two cities. Fungal contamination was determined using standard mycology methods. Aflatoxin and fumonisin contamination was determined using HPLC-FLD. A. flavus/parasiticus species were detected in 66% and 100% of randomly analysed peanut samples from Bulawayo and Gaborone respectively and 27% (3/11) of peanut butter samples from Bulawayo. 67% of randomly analysed sorghum samples from Bulawayo showed A. flavus/parasiticus and Fusarium species contamination while none of the randomly analysed sorghum samples from Gaborone showed any fungal contamination. Furthermore aflatoxins were not detected in any of the sorghum samples; however 61% (11/18) of the Bulawayo sorghum samples showed fumonisin contamination (Range: 8 – 187 ng/g). Three of the peanut samples from Bulawayo were contaminated with aflatoxins (range: 6.6 – 622 ng/g) and no aflatoxins were detected in Gaborone peanuts. All 11 peanut butter samples from Bulawayo were contaminated with aflatoxins (Mean: 73.5 ng/g, Range: 6.8-250 ng/g) and AFB1 was the most prevalent. These preliminary results indicate that peanut butter and peanuts from Bulawayo are contaminated with high levels of aflatoxins. Stricter policing of regulations should be implemented to ensure compliance by manufacturers and public health interventions implemented in vulnerable communities. / Life and Consumer Sciences / M. Molecular Characterization of Groundnut Bud Necrosis Virus Encoded Non Structural Protein m (NSm)Singh, Pratibha January 2014 (has links) (PDF)
Chapter 3
Groundnut Bud Necrosis Virus (GBNV) is a tripartite ambisense RNA plant virus that belongs to serogroup IV of Tospovirus genus. Non-Structural protein-m (NSm), which functions as movement protein in tospoviruses, is encoded by the M RNA. In this chapter, we demonstrate that despite the absence of any putative transmembrane domain, GBNV NSm associates with membranes when expressed in E. coli as well as in N. benthamiana. Incubation of refolded NSm with liposomes ranging in size from 200-250 nm resulted in changes in the secondary and tertiary structure of NSm. A similar behaviour was observed in the presence of anionic and zwitterionic detergents. Furthermore, the morphology of the liposomes was found to be modified in the presence of NSm. Deletion of coiled coil domain resulted in the inability of in planta expressed NSm to interact with membranes. Further, when the C-terminal coiled coil domain alone was expressed, it was found to be associated with membrane. These results demonstrate that NSm associates with membranes via the C-terminal coiled coil domain and such an association may be important for movement of viral RNA from cell to cell. Further NSm was shown to be phosphorylated by N. benthamiana and tomato crude sap as observed in other movement proteins.
Chapter 4
This chapter deals with localization of NSm to PD and identification of domain involved in localization. For this purpose NSm and its mutants were cloned in pEAQ:GFP vector and transiently expressed in N. benthamiana by infiltration of transformed Agrobacteria. The GFP tagged NSm was visualized by confocal microscopy. The results demonstrated that NSm forms punctate structures and localizes to PD as confirmed by colocalization of mCherry: PDLP1a, a PD marker which resides in PD, with GFP:NSm. To find out the domain involved in PD localization, sequential deletion mutants were made. It was found that C-terminal domain is involved in PD localization. On the other hand, N-terminal unfolded region was dispensable for PD localization. This is the first report of a coiled coil domain shown to be involved in PD localization. It has also been demonstrated that GBNV NSm interacts with NP. Further, membrane floatation assay carried in presence of NP suggested that interaction of NSm and NP affected membrane association of NSm. These results were further confirmed by localization studies of NSm in presence of NP. It was found that there was considerable relocalization of both NSm and NP. NSm was observed to be present in cytoplasm as well as on the membrane. At the same time, NP was observed on membrane apart from being present in the cytoplasm. When N-terminal 50 amino acids (unfolded) region of NSm was deleted and colocalization studies were carried out, it was found that NSm and NP do not colocalize, suggesting that NSm interacts with NP via the unfolded region and helps in the relocalization of NP to the membrane.

This chapter deals with the pathway of targeting NSm to PD. To decipher the pathway, followed by NSm, an inhibitor of endomembrane or vesicle mediated transport, Brefeldin A (BFA) was used. When GFP-NSm was expressed it was observed to form punctate structure at PD as before. Upon treatment with BFA, green islands were observed in the cytoplasm suggesting that ER was involved in targeting NSm to PD. Similarly, LatB, inhibitor of actin mediated targeting of protein to membrane, also abrogated the localization of NSm to PD. In order to further understand the role of ER in targeting NSm to PD, an ER marker, ER-GFP (GFP fused to HDEL peptide that directs it to ER) was coexpressed with GBNV NSm fused to mCherry. It was observed that NSm colocalizes with ER-GFP as yellow puncta on PD. The puncta appeared as patches and the whole ER-network was converted to vesicles. This was further confirmed by coexpressing ER-GFP with NSm without any tag. The green fluorescent vesicles were observed preferentially near cell membrane. To delineate the region of NSm involved in vesicle formation, point mutants and deletion mutants of NSm were generated without the tag and coexpressed with ER-GFP. When N-terminal 203 amino acids were deleted, NSm was able to transform ER membranes to vesicles suggesting that these residues are dispensable for vesicle formation. Interestingly, the deletion of coiled coil domain leads to cytosolic location of NSm. Furthermore, the C-terminal coiled coil domain when expressed alone was capable of inducing vesicle formation. This is the first report of involvement of such a domain in ER membrane association and vesicle formation.

Glycerol‑bound oxidized fatty acids: formation and occurrence in peanutsStörmer, Lars, Globisch, Martin, Henle, Thomas 08 April 2024 (has links)
For peanuts, roasted at 170 °C, the formation of selected glycerol-bound oxidized fatty acids (GOFAs), namely 9-oxononanoic acid (9-ONA), azelaic acid (AZA) and octanoic acid, was observed by GC-MS (EI). The content of octanoic acid as well as AZA increased with continuous roasting time (from 59 mg/kg peanut oil to 101 mg/kg peanut oil and from not detectable to 8 mg/kg peanut oil, respectively), whereas the content of 9-ONA initially decreased from 25 mg/kg peanut oil to 8 mg/kg peanut oil (20 min) and increased again up to 37 mg/kg peanut oil following roasting for 40 min. Due to its aldehyde function, 9-ONA could contribute to amino acid side chain modifications as a result of lipation, which could directly influence the functional properties of peanut proteins. Both 9-ONA and octanoic acid are potential markers of thermal processes. Furthermore, in model experiments using methyl linoleate and methyl oleate, up to 18 oxidized fatty acids could be identified as methyl esters, 9-ONA as well as octanoic acid as major components and a faster formation of GOFAs under roasting conditions (170 °C, 20 min). In addition, 9-ONA contributes to the formation of AZA and octanoic acid in both free and bound form as a result of oxidative subsequent reactions in presence of iron (III).

Investigation of marketing of small-scale peanut agribusinesses in the Vulamehlo District, Southern KwaZulu-Natal.Sonjica, Ivy. January 2008 (has links)
The main objective of the study was to investigate the marketing strategies needed for the development of a peanut agribusiness for small-scale farmers producing peanuts in the Vulamehlo District, southern KwaZulu-Natal (KZN). The study was part of the extension programme of the KwaZulu-Natal Department of Agriculture and Environmental Affairs (KZNDAEA). Another aim of the study was to examine the market requirements for peanuts purchased from Vulamehlo farmers in relation to the processing and sale of peanuts and whether or not market outlets for peanut products in KZN were available. In addition, current links between farmers, peanut processors, agents and retailers were to determine, if the marketing of peanut butter produced by small-scale farmers could be improved. Small-scale farmers were targeted because they play an important role in rural areas for community development and for poverty alleviation and also to generate income. There is little research on small-scale agribusinesses. Small-scale farmers have unreliable supply chains and markets. The study was conducted between May and November 2005. The data for the development of agribusiness for small-scale farmers in the Vulamehlo district were gathered from four wards of Vulamehlo: Mkhunya, Qiko, Mzimlilo and Mazabangwane. Questionnaires, a focus group discussion and observation were used to collect data from farmers and informal processors of peanut butter. Interviews were conducted with informal traders, formal traders and a formal peanut butter processor, to collect information about their marketing of peanut products. Six peanut producing farmers’ associations out of ten were randomly selected from the list of farmers’ associations in the Vulamehlo District. Details were obtained from the Department of Agriculture office. The study found that the marketing requirements needing to be met by small-scale farmers include availability of adaptable certified peanut seeds, appropriate tools and equipments for cultivation, reliable supply of raw material, grading and testing of level of aflatoxin in peanuts, knowledge of processing regulation to food safety and hygiene, proper business and management skills and relevant technology and marketing information. The study revealed that there were no seed companies around Vulamehlo sold peanut seeds and there were no commercial peanut-producing farmers in KZN to assist small-scale farmers. Farmers were using manual tools for the peanut production process. This limited production and expansion and lowered the yield. Constant supply may be a problem for the business processing enterprises. The markets for Vulamehlo peanut producing farmers were informal and insecure. The peanut butter co-operative lacked processing equipment, processing know-how, energy-saving technology and marketing information. It is recommended that developing agribusiness linkages with suppliers, traders, farmers and processors would improve the small-scale farmers’ negotiating skills. The KZNDAEA should improve links with large-scale farmers, traders, suppliers and processors, in order to obtain more information on peanut production. alimentares como para suprir parte da demanda de energia renovável, porém nos
últimos cinco anos tem sido o maior importador de azeite de oliva do mundo, devido
aos seus benefícios para a saúde. Neste contexto objetivou-se, com este trabalho, a
elaboração de misturas especiais de óleos vegetais para fins alimentícios, com base nos
óleos de amendoim, gergelim, maracujá e soja. Os óleos de amendoim e gergelim como
alternativa por serem culturas produzidas no Brasil, constituem uma fonte de ácidos
graxos e antioxidantes tão pouco explorados na alimentação dos brasileiros, tal como o
óleo de maracujá como co-produto, descartado pela indústria de suco e ainda o óleo de
soja cuja matéria-prima abundante, tornam o produto acessível aos consumidores de
todas as classes sociais. Os óleos de amendoim e gergelim foram extraídos de sementes
das cultivares BRS-Havana e BRS-Seda, respectivamente, cedidas pela Embrapa
Algodão enquanto o óleo de maracujá foi adquirido no comércio de São Paulo e o de
soja no de Campina Grande. As misturas foram elaboradas com base em uma matriz de
planejamento experimental e, em seguida, submetidas às análises de composição de
ácidos graxos e ensaios físico-químicas (umidade, índice de acidez, índice de iodo,
índice de refração, densidade e viscosidade) segundo a metodologia do IAL (2008). As
medidas obtidas foram utilizadas para estudar e otimizar a elaboração das misturas de
óleos vegetais e como variáveis para predição por espectrometria no infravermelho
próximo (NIR) foram utilizados os mínimos quadrados parciais (PLS) como
metodologia de calibração multivariada. Os resultados foram submetidos à análise de
variância (ANOVA), à análise de variância multivariada (MANOVA), à análise de
correlação canónica e à análise de correlação de Pearson, pelo software SAS 9.1.3; na
análise de componentes principais (PCA) e regressão por mínimos quadrados parciais
(PLS) utilizou-se o software Uncrambler 9.8®. Concluiu-se que a mistura equivalente a
40% de óleo de amendoim, 20% de óleo de gergelim, 20% de óleo de maracujá e 20%
de óleo de soja apresentou-se como a melhor mistura para fins alimentares, com base na
composição de ácidos graxos. Os métodos de predição por NIR foram eficazes para a
estimativa não destrutiva, rápida, de baixo custo e direta, dos ácidos graxos palmítico,
esteárico, oleico, linoleico, araquídico e linolênico e para as medidas físico-químicas de
densidade, índice de refração, viscosidade, índice de acidez e índice de iodo. / Brazil has great potential to produce vegetable oils as a source of raw materiais for food
and energy. But in the last five years has been the largest importer of olive oil in the
worid due to its health benefits and wide divulgation. In this context, the aim of this
work was the development of special blends of vegetable oils as an alternative to olive
oils based on peanut, sesame, soy and passion fruit. The peanut and sesame oils because
they are a source of fatty acids and antioxidants in the diet of little explored in Brazil,
passion fruit oil as co-product of the juice industry, and soybean oil abundant raw
material which makes the product available to consumers of ali social classes. The
peanut and sesame oils were extracted from seeds of BRS-Seda and BRS-Havana, while
passion fruit oil was purchased commercially of city São Paulo-SP and trade soybean in
the city of Campina Grande-PB. The mixtures were prepared based on an array of
experimental design and then submitted to analysis of fatty acid composition and
physico-chemical (moisture, acid value, iodine value, refractive index, density and
viscosity). Moreover, were used as reference variables for prediction via infrared
spectroscopy (NIR) using the partial least squares (PLS) and multivariate calibration
methodology. The results were subjected to analysis of variance (ANOVA),
multivariate analysis of variance (MANOVA), canonical correlation analysis and
Pearson correlation analysis by SAS 9.1.3 software, the principal component analysis
(PCA) and Partial least squares regression (PLS) was used Uncrambler 9.8 ® softwarlt
was concluded that the mixture equivalent to 40% peanut oil, 20% sesame oil, 20%
passion fruit oil, and 20% soybean oil was presented as the best mix for food
composition based on fatty acids. The prediction by NIR methods were effective for
estimating non-destructive, rapid, inexpensive and direct fatty acids palmitic, stearic,
oleie, linoleic, linolenic and arachidic and physico-chemical measurements of density,
refractive index, viscosity, acid value and iodine value.
