This thesis focuses on the various adaptations of Arthur Conan Doyles novel The Hound of the Baskerville and accentuate the episode The Hounds of Baskervilles from the BBC TV series Sherlock. Thus, this thesis is decribing the theme, which was not fully covered in Czech literature. The author used different sources, books, internet pages and cinematographic pieces. The thesis consists of two parts. In the first part the author is focusing on the theory of adaptation, Doyle's life and art, The Hound of the Baskervilles book and the TV series Sherlock. In the practical part the author narrates about the episode The Hounds of the Baskervilles and searching for definition of the adaptation process, which the makers of this episode used. This part also answers the question, why is this episode so popular and key for the whole series. The crucial final part of the thesis summarizes the main principles, which authors and film makers can use during the process of the adaptation. Proteomické rozlišení srsti psích plemen / Proteomic differentiation of hairs of different dog breeds

The goal of my thesis was to find out whether it is possible to distinguish among individual dog breeds from the protein analysis of their fur. This knowledge could be used, for example, in forensic science. In this work, there was also a comparison of the similarity of dog breeds with a wolf, which was domesticated and is considered as an ancestor of the dog. For this study, the hair of three representatives of sixteen dog breeds was collected. To analyse these samples enzyme cleavage was used a trypsin, and mass spectra were obtained by MALDI- TOF MS (Matrix-Assisted Laser Desorption/Ionization - Time of Flight Mass Spectrometry) and LC/MS-MS (Liquid Chromatography with Tandem Mass Spectrometry) methods. The obtained data were evaluated by the PCA (Principal Component Analysis) and LDA (Linear Discriminant Analysis) method. Názory žáků ZŠ na nebezpečnost vybraných savců / Opinions of Primary School Students on the Dangers Posed by Selected Mammal Species

This thesis builds on my bachelor thesis "Selected mammals in elementary and secondary education". It deals with the opinions of 8th and 9th grade pupils of elementary schools on the dangers posed by selected mammal species. Among the selected species in this work are brown bear (Ursus arktos), wolf (Canis lupus), lynx (Lynx lynx), fox (Vulpes vulpes), dog (Canis lupus familiaris), and wild boar (Sus strofa). All these animals are found in the territory of the Czech Republic. In addition to the perception of dangers posed by these mammals, the thesis also ascertains knowledge of and attitude towards these selected species. The pupils also expressed their opinions about dog breeds and stated whether they currently had a dog, or had had one in the past, and if so, what kind it was. The study was conducted on the basis of a questionnaire survey. Questionnaires were given out at primary schools in areas, where some of the beasts abide (brown bear, wolf, lynx). The work also contains information about individual mammal species that were studied as well as the introduction of the domestic dog, its domestication and an overview of dog breeds. Potential Energy Surface around the Tropylium Ion.

The formation of the tropylium ion, C7H7+, in the mass spectrum of toluene is a chemical process that has been studied extensively in the past. The advances in computational power of personal computers have made the investigation of the pathway to form this ion and its subsequent decomposition feasible at a fairly high level of theory. The calculations that we performed were at the HF/6-31G (d, p) and the B3LYP/6-311++G (2d) levels. This work will show areas of the potential energy surface around the highly symmetric tropylium ion to give a glance of possible mechanisms for its formation and decomposition. Our results have confirmed some of the mechanisms reported in the literature, and in addition new areas are explored in the report.

Oberflächenphotospannung an dünnen organischen Schichten auf Metallsubstraten / Surface photovoltage of thin organic films on metal substrates

Gegenstand dieser Arbeit ist die Untersuchung der Ober?ächen- bzw. Grenz?ächenphotospannung (SPV) an dünnen organischen Schichten auf Metallsubstraten. Besonderes Augenmerk gilt dabei dem System dünner Schichten von 3,4,9,10-Perylen- Tetracarbonsäure-Di-Anhydrid (PTCDA) auf einem Au(110)-Kristall. Für diese Untersuchungen wurde eine phasensensitive Methode auf der Basis der Photoelektronenspektroskopie (PES) mit zusätzlicher modulierter Lichteinstrahlung entwickelt, die es erlaubt, lichtinduzierte Verschiebungen des Ober?ächenpotentials mit einer Au?ösung ? 1mV zu detektieren. Diese modulierte Photoelektronenspektroskopie wird ausführlich vorgestellt. Eine makroskopische Kelvin-Sonde und die Kelvin-Sonden-Rasterkraftmikroskopie (KPFM) werden als weitere Möglichkeiten zur Bestimmung des Ober?ächenpotentials vorgestellt. Die Photospannung wird mit diesen Methoden in Bezug auf drei Parameter untersucht: Da die Barriere an der Grenz?äche, welche für die Ausbildung der Photospannung ursächlich ist, durch die Photospannung reduziert wird, geben Messungen der SPV in Abhängigkeit von der eingestrahlten Lichtleistung Informationen über die energetische Struktur der Grenz?äche, speziell über die Höhe der Barriere. Mit den ebenfalls aus diesen Methoden gewonnen Informationen über die Austrittsarbeit und Ionisationsenergie von PTCDA lässt sich ein Bandschema des Systems Au/PTCDA entwickeln. Die wellenlängenabhängigen Messungen zeigen, dass Exzitonen in verschiedenen Zuständen erzeugt werden. Diese werden mit unterschiedlichen Photonenenergien angeregt und besitzen unterschiedliche Di?usionslängen. Da die Exzitonen zum Dissoziieren an die Grenz?äche di?undieren müssen, tragen die unterschiedlichen Exzitonenzust ände mit unterschiedlichem Anteil zur Photospannung bei. Die Untersuchungen der Entstehungs- und Zerfallszeit zeigen, dass sich die Photospannung sehr schnell nach Lichteinschaltung aufbaut. Der Abbau der Photospannung nach Lichtabschaltung erfolgt vergleichsweise langsam. Dies bedeutet, dass die Ladungsträger, die nach der Ladungsträgertrennung in der organischen Schicht zur ückbleiben, in Fallenzuständen gebunden sind, aus denen sie thermisch aktiviert werden müssen. In einem separaten Abschnitt werden die Entwicklung sowie Messergebnisse eines Stimmgabel-Rasterkraftmikroskops (AFM) vorgestellt. Dieses ermöglicht die hochau ?ösende topographische Abbildung der Ober?äche mit einer vertikalen Sensitivität im Ångström-Bereich. Das Ziel, mit diesem Gerät KPFM zu betreiben, konnte beim Einsatz an Luft nicht verwirklicht werden. / Subject of this work is the investigation of surface and interface photovoltage (SPV) of thin organic films on metal substrates. Special attention is focused on the system of thin layers of 3,4,9,10-perylenetetracarboxylic-dianhydride (PTCDA) on an Au(110) crystal. For this investigation a novel phase sensitive method was developed based on photoelectron spectroscopy (PES) under additional modulated illumination. It provides the possibility to detect light induced changes in the surface potential with a resolution of about 1mV. This modulated photoelectron spectroscopy is described in detail. Macroscopic Kelvin probe and Kelvin probe force microscopy (KPFM) are presented as further possibilities to measure the surface potential. The photovoltage is investigated by these techniques regarding three parameters: Due to the fact that the barrier at the interface is responsible for the formation of the photovoltage and that the height of this barrier is reduced by the photovoltage, the measurement of the dependence of the SPV on the intensity of the incident light provides information about the energetic structure of the interface. Together with the values of the work function and the ionisation energy of PTCDA, also gained with this methods, a band diagram of the interface can be developed. The wavelength dependent measurements show that excitons can be generated in multiple states. They are excited at different photon energies and have different diffusion lengths. The excitons have to diffuse to the interface to dissociate. Therefore the different excitation states contribute to SPV with different amounts. The investigations upon the generation and decay of the photovoltage shows that the SPV signal appears immediately after switching on the illumination. The decay of the photovoltage after switching off the light is much slower. This implies that the charge carriers are trapped as they remain in the organic film after charge separation at the interface. They have to be thermally activated from this traps. In an extra chapter the development and measurement results of a tuning fork scanning force microscope (AFM) are described. This AFM features high resolution topography images with a vertical sensitivity in the range of single angstroms.

Among recent studies, the leaf is the most utilized organ for the understanding of morphoanatomical relationships between individuals and the environment, while works focusing stem and root are yet incipient. Thus, the aim of this work was to characterize the morpho-anatomy of adventitious roots and stems of Blutaparon portulacoides (A. St.-Hil.) Mears (Amaranthaceae), Canavalia rosea (Sw.) Dc. (Fabaceae), Ipomoea pes-caprae (L.) Stweet (Convolvulaceae), Remirea maritima Aubl. (Cyperaceae) e Scaevola plumieri (L.) Vahl. (Goodeniaceae) occurring in a Halophile-Psammophila Reptant formation of the Paulo César Vinha State Park, relating to environment parameters; to describe the secretory structures and to identify the classes of secondary metabolites present in the secretion of leaves of Ipomoea pes-caprae and Ipomoea imperati. Soil was sampled and micro environmental measurements were performed. For anatomical evaluations, roots, stems and leaves of different regions were collected and processed following usual techniques in plant anatomy. The soil had few nutrients and organic matter. The micro environmental data showed that herbaceous plants are susceptible to wind speed conditions, relative humidity and evaporation similar to that of sub shrubby species. Morpho-anatomically, the species presented distinct adaptive strategies,specially stolons or rhizomes, aquiferous tissue, aerenchyma in the roots and starch reserve in both stem and root. In Ipomoea pes-caprae and Ipomoea imperati,mucilaginous trichomes and laticifers were described on leaf lamina, and in I. pescaprae, nectaries on petiole. The morpho-anatomical structures found in stems, roots and leaves of the studied species were outstanding features which allowed the survival and competitiveness of these species in the Non-flooded Herbaceous Formation, once they provide adaptive capacity to high temperature, strong wind, high salinity, lack of nutrients, among others. / Restinga é um ecossistema associado à Mata Atlântica, com as espécies próximas ao mar adaptadas a condições climáticas e edáficas adversas. Dentre os estudos atuais, a folha é o órgão mais utilizado para o entendimento das relações morfoanatômicas dos indivíduos com o ambiente, sendo incipientes os trabalhos sobre caules e raízes. Desta forma, o objetivo foi caracterizar a morfoanatomia de raízes adventícias e caules de Blutaparon portulacoides (A. St.-Hil.) Mears (Amaranthaceae), Canavalia rosea (Sw.) Dc. (Fabaceae), Ipomoea pes-caprae (L.) Stweet (Convolvulaceae), Remirea maritima Aubl. (Cyperaceae) e Scaevola plumieri (L.) Vahl. (Goodeniaceae) ocorrentes na formação Halófila-Psamófila Reptante do Parque Estadual Paulo César Vinha, relacionando com parâmetros ambientais; descrever as estruturas secretoras e identificar as classes de metabólitos secundários presentes no secretado das folhas de Ipomoea pes-caprae e Ipomoea imperati. Foram realizadas coletas de solo e medições microambientais. Para avaliações anatômicas, raízes, caules e folhas de diferentes regiões foram coletados, e processados segundo técnicas usuais em anatomia vegetal. O solo apresentou-se pobre em nutrientes e em matéria orgânica. Os dados microambientais demonstraram que as plantas herbáceas estão sujeitas a velocidade do vento, umidade relativa do ar e evaporação similares às da espécie subarbustiva. Em relação à morfoanatomia, as espécies apresentaram estratégias adaptativas distintas, com destaque para presença de estolões ou rizomas, tecido aquífero e aerênquima nas raízes e reserva de amido tanto no caule quanto na raiz. Em Ipomoea pes-caprae e Ipomoea imperati, foram descritas, na lâmina foliar, tricomas mucilaginosos e laticíferos; e em I. pes-caprae, nectários no pecíolo. As estruturas morfoanatômicas encontradas nos caules, raízes e folhas das espécies estudadas foram características marcantes que possibilitaram a sobrevivência e competitividade dessas espécies na formação Halófila-psamófila Reptante, uma vez que fornecem capacidade adaptativa à alta temperatura, vento forte, alta salinidade, falta de nutrientes, dentre outros.

The diploma thesis focuses on the issue of guide dogs and their owners from in view of the general public across various generations through qualitative research. Theoretical part of the thesis introduces guide dog in world and Czech history, relevant legislative measures, fully describes course of dog training, and behavior of a guide dog in public. Practical part describes qualitative research, a questionnaire aimed at public in the Czech Republic. The survey examines public knowledge of legal measures about guide dogs, appropriate behavior towards a guide dog and its owner, and about guide dog's tasks. Research provides out look of the public 's knowledge and suggests targeted education for generations studied. The diploma thesis also brings method of educating respondents during data acquisition. Fonctionnalisation de polymères et applications dans les domaines de l’énergie, de la catalyse, de la cosmétique et de la santé / Functionalization of polymers and applications in the fields of energy, catalysis and health

Les polymères sont à l’heure actuelle étudiés dans de nombreux domaines comme la chimie, la biochimie, les nanotechnologies, l'électronique, la médecine ou encore les sciences des matériaux et trouvent des applications dans des domaines comme l’industrie automobile, la chimie fine. L’objectif de cette thèse est de réaliser la fonctionnalisation de polymères et de modifier les propriétés de ces matériaux afin d’envisager des nouvelles applications. Nous nous sommes intéressés à des polymères de la famille des poly(aryle éther) et plus particulièrement au poly(éther éther cétone) (PEEK). Ce polymère est connu pour ses propriétés mécaniques, thermiques, électriques ou encore pour sa résistance aux produits chimiques. Dans le premier chapitre, il est question de la fonctionnalisation des différents polymères de départ par des fonctions chlorures de sulfonyle, acides sulfoniques et sulfonamides. Le second chapitre est consacré à la synthèse et à l’étude électrochimique de nouveaux électrolytes polymériques et à de nouvelles membranes pour d’éventuelles applications dans le domaine des batteries au lithium et au sodium, ainsi que dans le domaine des piles à combustible. Dans un troisième chapitre, la préparation de nouveaux catalyseurs métalliques dérivés d’acides sulfoniques polymériques est discutée. Une étude de l’activité catalytique de ces différents catalyseurs a été réalisée sur la réaction d’acylation de Friedel-Crafts. Le quatrième chapitre est consacré à la préparation de nouveaux matériaux ayant des propriétés optiques intéressantes. Enfin dans un cinquième chapitre, la préparation et l’étude de nouveaux matériaux ayant des propriétés antibactériennes sont exposées. / Polymers are now being studied in many fields such as chemistry, biochemistry, nanotechnology, electronics, medicine or material science and have applications in areas such as automotive industry, food industry, fine chemistry. The objective of this thesis is to achieve the functionalization of polymers and modify the properties of these materials in order to consider new applications. We were interested in polymers with the poly(aryl ether) motif, more particularly poly(ether ether ketone) (PEEK). This polymer is known for its mechanical, thermal, electrical properties and for its resistance to chemicals. In the first chapter, we present the functionalization of different polymers by sulfonyl chloride, sulfonic acid and sulfonamide functions. The second chapter is devoted to the synthesis and electrochemical study of novel polymeric electrolytes and new membranes for potential applications in the field of lithium and sodium batteries, as well as in the field of fuel cells. In the third chapter, the preparation of new metal catalysts derived from polymeric sulfonic acids is discussed. A study of the catalytic activity of these different polymeric catalysts was carried out on the Friedel-Crafts acylation reaction. The fourth chapter is devoted to the preparation of new materials with interesting optical properties. Finally, in the fifth chapter, the preparation and the study of new materials with antibacterial properties are reported.
