Problémy se stravováním v MŠ / Eating problems in kindergarten

This thesis deals with nutrition of preschool children. It concentrates on the issue of eating in nursery schools. The theoretical part contains characteristics of a preschool child development and information on the basic components of nutrition and its importance to the human body. I mentioned the influence of dietary habits on the healthy development of the child population and analysed the negative factors that could adversely affect this development. The practical part includes directed interviews with staff of kindergartens in district Prachatice and comparison between current diets and diets before 1989 in these institutions. The intention was to find differences between these diets. With the change in the political system new possibilities opened up. School canteens can experiment with new recipes, diets include new healthy foods.

ZÁSOBOVÁNÍ SCHWARZENBERSKÉ DOMÁCNOSTI VE DRUHÉ POLOVINĚ 17. STOLETÍ / The supplying of Schwarzenberg households in the second half of the 17th century

The thesis consists of several parts discussing the issue of the supplying of aristocratic homes in the Baroque period. Everything was done on the example of the Schwarzenberg households between the years 1656 and 1683, during the reign of Jan Adolf I. of Schwarzenberg. The first part involves the organization of the servants in his household. The second part deals with looking at the residence of his wife Marie Justine from Schwarzenberg, born in Starhemberg, Prague, around 1658. On the basis of their personal accounts, the authors presented this reconstruction supplying her household in Prague and a wider analysis of potential internal view of the expenditures in the category of alms. The following chapter deals with food supplies for Jan Adolf I's wife's household at Trebon. The last part relates to the analysis of transport of food to Schwarzenberg households in Vienna in the third quarter of the 17th century, part of which is closer to the probe focused on local supply and consumption of alcohol.

This diploma thesis deals with fiber intake and possibilities of its increase. The main aim of the thesis is to evaluate the actual intake of fiber in the population and to find out whether people actually know what fiber is and what should be its daily intake. The theoretical part is focused on fiber itself. It describes its characteristics, distribution, properties and sources. The effect of fiber on human health and the consequences of insufficient or excessive fiber intake are discussed as well. The practical part is focused on the real intake of fiber in the population. It is done in the form of a questionnaire survey, in which I try to find out what knowledge of fiber people have, but also how often they consume foods containing fiber. Most of the questionnaires were handed out to customers of one unnamed pharmacy and healthy nutrition center in Prague and other passers-by. Altogether, I managed to collect 84 questionnaires. The questionnaire results show that vast majority of respondents have an idea of what fiber is and which foods contain it most; however, they do not know what the recommended daily intake is. The aim of this thesis is to describe current trends in global retail, the current state of retail in Czech Republic and possible need of bringing innovation to the outburst of Millennials generation economic activity. Introduction is devoted to theoretical description of retail and its meaning. As part of the thesis, there is a significant part containing relations within the worldwide retail and also its current trends and development. Furthermore, part of the thesis is devoted to the development and ongoing evolution of retail in the Czech Republic. Theoretical definition in the third chapter deals with buying behavior as well as the factors which have an impact on it. Millennials are the bearers of the innovation need in the world retail sector. The thesis put emphasis on this generation essential features characteristic, as well as their purchasing behavior. Quantitative research includes evaluation of their behavior and attitudes on the czech market in relation to information about groceries and consumer goods, then to their use of technology (smartphones) in the buying process of groceries and consumer goods, and also evaluation and description of their specific attitudes to purchase behavior and different characteristic aspects resulting from global characteristics of Millennials.

The goal of this masters thesis is focused on segmentation of the organic food market in the Czech Republic based on analysis of similarities or differences among consumers in Czech organic food market and also find out who is a typical organic food consumer. The thesis is divided into three parts, the theoretical, the methodological and the practical part. Process of the market segmentation, methods and approaches of the segmentation, the basic concepts and legislation on the matter are described in the theoretical part. Furthermore, mapping the situation of the contemporary state of the Czech organic food market. Marketing research is introduced in methodological part of the thesis. The practical part includes market segmentation by using secondary data MML - TGI (with Data Analyzer software) and primary data from quantitative research (with SPSS software). Variables were reduced to the four factors by using factor analysis. Then consumers were put into five clusters based on cluster analysis. Segments were characterized in detail using a general analysis, contingency tables, MCART analysis and multivariate statistical methods. Finally, there were elaborated appropriate marketing recommendations for the individual segments to effective marketing communication with them.

Optimalizace nové mikrometody izolace DNA z potravin / Optimalisation of a new micromethod of DNA isolation from foods

The thesis were focused on the optimalization of micromethod for isolation of DNA in quality for polymerase chain reaction (PCR) using magnetic microparticles from plant food products. There were chosen a red beetroot (fresh, frozen, dried and sterilized) for the analysis and food products containing red beetroot. Different approaches of processing of homogenates were compared and optimized. The homogenates were prepared in lysis buffer with cetyltrimethylammonium bromide (CTAB) with different amounts of NaCl with or without addition of organic extraction agents chloroform-octanol and isopropanol. Microisolation of DNA was performed using magnetic particles P(GMA). The concentration of NaCl and polyethylene glycol (PEG) 6000 in separation mixtures was tested. The influence on quantity and purity of isolated DNA was compared and the optimum amounts of NaCl in CTAB buffer and optimal concentration of PEG 6000 in separation mixtures were compared. The optimized separation mixture for the DNA isolation from red beetroot was applied to food products containing red beetroot. Amplifiability of DNA was tested in conventional PCR using specific primers for plant DNA. PCR products of length 700 bp were detected by agarose gel electrophoresis.

Počet obyvatel trpících různými infekčními, zánětlivými a alergickými nemocemi stejně jako výskyt laktózové nesnášenlivosti a vysoké hodnoty krevního cholesterolu, má narůstající tendenci. Některé z těchto zdravotních problémů jsou způsobeny nevyváženou střevní mikroflorou. Probiotika jsou pak chápána (nejen) jako potravní komponenty, které přispívají k ustanovení mikrobiální rovnováhy (Parker, 1974) mezi zdraví prospěšnými a škodlivými bakteriemi. Z tohoto důvodu, terapie založená na podávání probiotik pacientům přitáhla zájem ze strany vědců. Vhodný probiotický kmen se pak volí v závislosti na požadovaném zdravotním účinku (příp. zdravotním problému, který má být probiotickou terapií léčen). Lactobacillus plantarum 299v již prokázal své blahodárné účinky na lidech a zároveň byla i potvrzena jeho zdravotní bezpečnost, díky čemuž může tato bakterie být kategorizována jako probiotický kmen (Probi AB, Sweden). I díky tomu je Lactobacillus plantarum 299v ve značné oblibě přidáván do mnoha fukčních potravin a prodáván na trhu pod různými jmény, probiotický nápoj ProViva je jedním takovým příkladem. Cílem této práce bylo studovat fermentační proces na žitném, ječmenném a sojovém substrátu pomocí kmene Lactobacillus plantarum 299v, přičemž zvýšená pozornost byla věnována právě soji a ječmeni jako potenciálně novým substrátům pro výše uvedenou bakterii. Hlavními záměry bylo zkoumání růstu a metabolické aktivity bakterie Lactobacillus plantarum 299v v asociaci s různými cereálními substráty, a později bylo studováno totéž také ve směsi fermentované cereální komponenty s běžně dostupným ovocným džusem. K tomu, aby se dosáhlo optimálních podmínek fermentace, je třeba vzít v úvahu několik aspektů. Hlavní role při konceptování nového fermentovaného produktu patří především zpracování a taktéž kompozici surového materiálu, růstové kapacitě a produktivitě bakteriální kultury a stabilitě finálního produktu během skladování (De Vuyst, 2000). Tyto parametry jsou důležité hlavně ze strany výrobců. Krom toho jsou tu ale i zákazníci, pro něž je přijatelnost produktu založena z velké části na organoleptických vlastnostech finálního probiotického produktu, tj. aromatu a chuti. Přítomnost a dostupnost různých jednotlivých nutrientů, která byla obsažena ve fermentačním médiu výsledkem rozdílných použitých cereálních substrátů, pravděpodobně vyústila v odlišnosti metabolických drah, což pak později mohlo způsobit rozdíly v organoleptických vlastnostech finálního produktu.
