Desenvolvimento e validação de métodos para análise de Ractopamina na matéria-prima, no produto Ractosuin®, no complexo vitamínico-mineral e na ração para suínos em terminação / Development and validation of methods for analysis of ractopamine in raw material, feed additive Ractosuin®, vitamin-mineral complex and ration for swine in termination.

Ellen Figueiredo Freire 16 August 2010 (has links)
Several drugs, especially B-adrenergic agonists, have been used in animal production and due to new forms of intensive livestock their use has grown exponentially. Among the B-adrenergic agonists, ractopamine (RAC) is the most used as a nutrient repartitioning agent, it promotes weight gain, feed efficiency, deposition of muscle and carcass yield of pigs, cattle, sheep and turkeys. In this context, the development of analytical methods able to detect and quantify RAC in the raw material, feed additive, in pre-formulation (feed additive) enriched with nutrients and minerals and in the ration is very important in quality control of pharmaceutical formulation marketed by veterinary industries for greater security for animals and to consumers of meat. Thus, this project aims to develop and validate analytical methods able to quantify the drug in these 4 stages (raw material, feed additive Ractosuin®, pre-mixed with vitamin mineral complex and the mixture of these components with the ration). In the first chapter of this work is described the method for quantification of RAC in the raw material and feed additive Ractosuin®. This method showed linearity over the concentration range from 160.00 to 240.00 ug mL-1 (r > 0.99). The coefficients of variation in studies of intra-and interassay precision were less than 2,0% and intra and interassay accuracy ranged from 97 to 100%. In the second chapter, we used liquid-liquid extraction and ethyl acetate as solvent extractor for the analysis of RAC in complex vitamin-mineral. The calibration curve was linear over the concentration range from 90.00 to 210.00 ug mL-1 (r > 0.99). The coefficients of variation in studies of intra and interassay precision were less than 1.5%, intra and interassay precision varied from 98 to 102% and the recovery was around 94%. The third chapter shows the method developed and validated for analysis of RAC in the ration. The RAC was extracted from the matrix by liquid-liquid extraction with ethyl acetate. The analytical curve was linear over the concentration range 6.50 to 14.40 ug mL-1 (r > 0.99), the limit of detection (LD) was 2.00 ug mL-1 and limit of quantification (LQ) was 4.00 ug mL-1.The coefficients of variation in studies of intra and interassay precision were less than 5.0%, intra and interassay accuracy ranged from 90 to 98% and the recovery was 84%. Thus, the methods developed and validated can be applied in routine analysis for quality control of veterinary industries for the quantification of RAC in the raw material, feed additive Ractosuin®, vitamin mineral complex and in the ration.

Substituição de milho por ingredientes alternativos na dieta de tourinhos confinados na fase de terminação / Substitution of byproducts for ground corn in feedlot finishing diets

Eduardo Menegueli Pereira 26 August 2005 (has links)
In order to evaluate the effects of the substitution of different byproducts for ground corn, two trials were performed with finishing yearling bulls. Rations were formulated according to NRC (1996) to be isonitrogenous and have positive rumen degradable and metabolizable protein balances. In trial 1 seventy two Canchim yearling bulls (308 initial body weight and 18 months of age) were used in a randomized complete block design, with 4 treatments and 6 replications. Rations contained 70% concentrate and 30% sugarcane silage, treated with microbial additive Lactobacillus buchneri 50788 (Lalsil®). Treatments were 100% ground corn (M), 50% ground corn + 50% dried citrus pulp (P50), 25% ground corn + 75% dried citrus pulp (P75), and 100% dried citrus pulp (P), as energy sources. Feed efficiency (P<0,05) and net energy for maintenance and gain (P<0,10) were greater for P50 compared to M. There was a linear reduction (P<0,05) in daily gain and dry matter intake when citrus pulp replaced 75% and 100% of ground corn, compared with P50. According to the present data, the NRC(1996) and NRC(2001) underestimated the energy value of the citrus pulp, compared to ground corn. Carcass fat depth and dressing percentage were not different (P<0,05) among treatments. In trial 2 thirty six yearling bulls (18 Nelore and 18 Canchim), averaging 382 kg initial live weight and 14 months of age, were used in a randomized complete block design. Animals were grouped by breed and initial live weight. Experimental period lasted 87 days, divided in 3 sub-periods. Rations contained 24% Tanzania grass silage and 76% concentrate, composed by citrus pulp, soybean meal, urea, vitamin-mineral mix, sodium bicarbonate and sodium monensin, plus one of the energy sources tested. Ingredients tested were: fine ground corn, wheat middling and dried corn gluten feed. All rations contained a 50:50 citrus pulp:energy source ratio. Substitution of corn gluten feed for ground corn reduced dry matter intake, neutral detergent fiber intake (P<0,05) and daily gain (P<0,10), but did not affect feed efficiency (P>0,05). Neutral detergent fiber intakes were different (P<0,05) between the ground corn and wheat middling treatments, but dry matter intake, daily gain and feed efficiency were not altered (P>0,05). Observed values for diets net energy of maintenance and gain did not differ (P>0,05) among treatments, what indicates that NRC (2001) and NRC (1996) underestimated the byproducts energy value, compared to fine ground corn.

The effects of cellulosic fiber charges on polyelectrolyte adsorption and fiber-fiber interactions

Horvath, A. Elisabet January 2006 (has links)
The surface charges of cellulosic fibers contribute to several papermaking operations that influence the manufacture and final properties of paper. This thesis investigates the effect of the surface charges on wet-end chemistry, e.g. through the interaction of cationic polyelectrolytes with the fiber surface charges, and on the network strength of pulp suspensions. The polyelectrolyte titration method was used to investigate the interaction of the fiber charges with cationic polyelectrolytes. Techniques were developed to fluorescent label the adsorbing cationic polyelectrolyte in order to visualize the adsorption behavior. Fluorescent confocal laser scanning microscopy (CLSM) was used to determine the extent to which the cationic polyelectrolyte adsorbs into the porous fiber wall. It was shown that the polyelectrolyte charge density limits the adsorption to the surface under electrolyte-free conditions. Adsorption into the fiber wall only occurs for two conditions: 1) if the molecular mass is sufficiently low or 2) the electrolyte concentration is high enough to screen the charges along the polyelectrolyte backbone but not the interactions between the polyelectrolyte and the fiber charges. Aside from the polyelectrolyte properties, the fiber charge density contributes to the adsorption behavior of cationic polyelectrolytes. The fiber charge profile was altered by bulk and surface carboxymethylation. The electrolyte concentration at which a deviation from 1:1 stoichiometry occurs was shown to be dependent on the amount of surface charges, such that the deviation in stoichiometry occurs at a higher electrolyte concentration for pulps having a higher surface charge. A hypothesis was developed to test the conditions at which the deviation in adsorption stoichiometry occurs, which was defined as the critical electrolyte concentration (CEC). It was found that the CEC corresponded to the electrolyte concentration at which the distance between the fiber charges was on the order of the Debye length. Electron spectroscopy for chemical analysis (ESCA) was used as an independent calibration procedure to validate for which a 1:1 stoichiometry occurs. The analysis with ESCA agreed well with the polyelectrolyte titration method for measurement of fiber surface charges. When measured under appropriate conditions, i.e. electrolyte concentration and molecular properties, the fiber surface charge can accurately be measured by the polyelectrolyte titration method. The charge profiles of various pulp types and treatments were also examined. Having been established as a valid technique, the polyelectrolyte titration method was again used to measure the surface charge while conductometric titration was used to measure the total charge content. The amount of bulk and surface charges vary depending on the pulping method and type of wood, although the ratio between the bulk and surface charge (i.e. the charge ratio) is similar for chemical pulps. The mechanical pulp has a higher charge ratio because it contains more fines material than chemical pulp. Bleaching of the chemical pulp decreases the amount of bulk and surface charges, although the charge ratio remains essentially constant. However, methods such as beating or carboxymethyl cellulose (CMC) grafting are available to increase the charge ratio. The effect of the charge profile on fiber-fiber interactions was studied on both a microscopic and macroscopic level. Colloidal probe microscopy (CPM) was used to investigate the microscopic interactions between two cellulose surfaces. Cellulose surfaces, prepared by spin-coating a dissolving pulp onto silica, were used to model the fiber surface, which is too rough for surface force measurements. The charge density of the model surface was increased by CMC grafting. Results showed that increasing the surface charge density created large electrosteric repulsions, due to CMC the chains protruding out from the surface. These interactions on the microscopic scale affect the fiber network strength, which was measured with a parallel plate rheometer. When the repulsion is increased between the fibers, caused by the increase in the surface charge, fiber flocs break apart more easily due to a reduced friction between the fiber surfaces. The forces acting on the fiber network can also be mechanical in origin. The fiber length and flexibility were altered in order to study the influence of mechanical surface linking and elastic fiber bending on the fiber network strength. Using the storage modulus (G’0) as a measure of fiber network strength, longer fibers were found to create a stronger network due to an increased amount of fiber contacts. Flexible fibers have a lower network strength than stiff fibers because the fibers come to rest in a less strained position such that the the influence of elastic fiber bending on the fiber network strength is predominant. / QC 20100831

Optimalizace krmné dávky vybraných druhů lemurů / Diet optimalisation of selected species of lemurs

ŽAHOUROVÁ, Petra January 2012 (has links)
Ring-tailed lemur Lemur catta, red ruffed lemur Varecia rubra and black and white ruffed lemur Varecia variegata belongs to the family Lemuridae, living at Madagaskar as endemits. In wildlife is folivorous and frugivorous food strategy at the both genus ? in the process g. Varecia is more frugivorous. Study was divided into two parts ? the first about ration of feeding chosen species of lemurs in several czech zoos and the second about ethology feeding behaviour lemurs in zoo Jihlava. For nutritional assessment were further chosen zoos Lešná, Jihlava, Ústí nad Labem, Ohrada and Chleby. From ration of feeding for chosen species of lemurs was counted average personal ration per day. This average was used to calculate nutritional values of dietary (fats, protein, carbohydrates, fiber, calcium, phosporus and iron) for each kind of menu or particularly day a week. For ethologic observation was made method of straight observation with used behaviour sampling ? scanning of a behaviour. Observation was done from July to December 2011 in whole extent 56 hours. Study solved out that ration of feeding are very different in each zoo like nutritional values of dietary and amount. In each zoo has larger ration of feeding Varecia rubra than Lemur catta. In average this diference beetwen that makes 144,25 g. Observation (zoo Jihlava) showed that Lemur catta spends foraging behaviour 19,75 minutes per day and handling 42,75 minutes per day in average. All in all they spending their feeding behavior only 62,5 minutes (1,04 hours) per day. Further more was discovered diference in length feeding behaviour when in their habitation more tree branches were. Adding branches to there increased length of feeding behaviour about of 22,67 minutes per day.

Produção, composição do leite e desempenho reprodutivo de ovelhas Santa Inês alimentadas com rações contendo óleo de canola ou linhaça / Milk yield, milk composition and reproductive performance of Santa Inês ewes fed diets containing canola or linseed oil

Cristine Paduan Nolli 23 April 2012 (has links)
The objective in this study was to evaluate the effect of the addition of canola oil (CO) or linseed oil (LO) on milk production and composition and reproductive performance of Santa Inês ewes. In the experiment I, 33 Santa Inês ewes were fed diets containing 50% of roughage (Cynodon dactylon hay). Experimental diets included a control (no oil) and two remaining diets with the addition of 3% of CO or LO (dry matter basis). Ewes were mechanically milked once a week and milk production, in a 3 hours interval, was recorded and sampled for composition and fatty acid (FA) profile determination. The addition of oils decreased DM intake and incresead serum non-esterified FA concentration. Lamb performance, milk yield and chemical composition were not altered by the diets. Feeding oils decreased short and medium chain FA, saturated FA (SFA), 9-dessaturase enzyme activity and atherogenicity index; and increased C18:0, C18:1 n-9, C18:2 cis-9, trans-11 C18:2 (CLA), C18:1 trans-11, C18:3 n-3, long chain, mono and polyunsaturated FA (PUFA) in milk fat. In addition, both oils improved FA profile of milk fat, and LO increased the concentration of healthier FA (trans-11 C18:1, cis-9 trans-11 CLA, total CLA and n-3 FA). In the experiment II, 222 Santa Inês ewes were assigned, according to body weight (BW) and body condition score, to one of three experimental diets to determine the effects of fat sources on pregnancy and estrus index, BW, time to estrus and number of lambs per ewe. Ewes were fed diets containing 40% of sugarcane bagasse as the roughage source and the experimental diets included a control (no oil) and two remaining diets with the addition of 3% of CO or LO (dry matter basis). All ewes were fed the experimental diets 45d before the estrus synchronization. The synchronization protocol consisted on vaginal insertion of a progesterone (P4) device for 5d (D0), rams introduction, i.m. injection of eCG (1.5 mL) and PGF2 (2.0 mL) on the 5th day (D5). At D0 the ewes BW was obtained. The time of P4 device withdrawal (D5) was recorded and estrus observation lasted 72 hours. The 2nd service began at D16 and lasted 13 days. The BW and number of lambs per ewe were not affected by diets. The rates of onset of estrus, pregnancy at 1st and 2nd services and time for the onset of estrus were not affected by diets. The total pregnancy rate was higher in animals from the control diet compared to those fed LO. Supplementation of oil rich in oleic acid or linolenic acid did not improve reproductive rates of Santa Inês ewes.
